| 1 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 2 | Efficiency and Inheritance of Targeted Mutagenesis in Maize Using CRISPR-Cas9显示文摘CRISPR/Cas(clustered regularly interspaced short palindromic repeats/CRISPR-associated proteins) is an adaptive immune system in bacteria and archaea to defend against invasion from foreign DNA fragments. Recently, it has been developed as a powerful targeted genome editing tool for a wide variety of species. However, its application in maize has only been tested with transiently expressed somatic cells or with a limited number of stable transgenic T0 plants. The exact efficiency and specificity of the CRISPR/Cas system in the highly complex maize genome has not been documented yet. Here we report an extensive study of the well-studied type II CRISPR-Cas9 system for targeted genome editing in maize, with the codon-optimized Cas9 protein and the short non-coding guide RNA generated through a functional maize U6 sn RNA promoter. Targeted gene mutagenesis was detected for 90 loci by maize protoplast assay, with an average cleavage efficiency of 10.67%. Stable knockout transformants for maize phytoene synthase gene(PSY1) were obtained. Mutations occurred in germ cells can be stably inherited to the next generation. Moreover, no off-target effect was detected at the computationally predicted putative off-target loci. No significant difference between the transcriptomes of the Cas9 expressed and non-expressed lines was detected. | 3 | Bio-Based Hyperbranched Toughener From Tannic Acid and Its Enhanced Solvent-Free Epoxy Resin with High Performance显示文摘It is essential to design economic and efficient tougheners to prepare high-performance epoxy resin;however,this has remained a huge challenge.Herein,an eco-friendly,low-cost,and facile-fabricated bio-based hyperbranched toughener,carboxylic acid-functionalized tannic acid(CATA),was successfully prepared and applicated to the preparation of solvent-free epoxy resins.The mechanical performance,morphology,structural characterization,and thermal characterization of toughened epoxy resin system were studied.The toughened epoxy resin system with only 1.0wt%CATA reached the highest impact strength,111%higher than the neat epoxy resin system.Notably,the tensile strength and elongation at break of toughened epoxy resin systems increased moderately with increasing CATA loading.Nonphase-separated hybrids with significant toughening effect were obtained.Additionally,the thermal stabilities of toughened epoxy resin systems decreased with increasing CATA loading.This study provides an eco-friendly,cost-effective,and facile approach for the preparation of high-performance,solvent-free epoxy resins with potential for practical applications in sealing integrated circuits and electrical devices fields. | Jie Xu Jiayao Yang Hengxu Wang Peng Lin Xiaohuan Liu Jinjie Zhang Shenyuan Fu Yuxun Tang | 2019 | Journal of Renewable Materials2019,7,12: | 5 |
| 4 | Maize WI5 encodes an endo-1,4-β-xylanase required for secondary cell wall synthesis and water transport in xylem显示文摘Water transport from roots to leaves through xylem is important for plant growth and development.Defects in water transport can cause drought stress,even when there is adequate water in the soil.Here,we identi-fied the maize(Zea mays)wilty5(wi5)mutant,which ex-hibits marked dwarfing and leaf wilting throughout most of its life cycle under normal growth conditions.wilty5 seed-lings exhibited lower xylem conductivity and wilted more rapidly under drought,NaCl,and high temperature treat-ments than wild-type plants.Map-based cloning revealed that WI5 encodes an active endo-1,4-β-xylanase from gly-cosyl dehydration family 10,which mainly functions in degrading and reorganizing cell wall xylan.Reverse-transcription polymerase chain reaction andβ-glucuronidase assays revealed that WI5 is highly expressed in stems,especially in internodes undergoing secondary wall as-sembly.RNA sequencing suggested that WI5 plays a unique role in internode growth.Immunohistochemistry and electron microscopy confirmed that wi5 is defective in xylan deposition and secondary cell wall thickening.Lignin deposition and xylan content were markedly re-duced in wi5 compared to the wild-type plants.Our results suggest that WI5 functions in xylem cell wall thickening through its xylanase activity and thereby regulates xylem water transport,the drought stress response,and plant growth in maize. | Xiaojiao Hu Yang Cui Xiaomin Lu Weibin Song Lei Lei Jinjie Zhu Jinsheng Lai Lizhu E Haiming Zhao | 2020 | Journal of Integrative Plant Biology2020,62,10: | 3 |
| 5 | Comparing overall survival between first generation EGFR-TKIs and chemotherapy in lung cancer patients with Del19/L858R显示文摘Objective: Combined overall survival(OS) analysis of Lux-Lung 3 and Lux-Lung 6 demonstrated that patients with epidermal growth factor receptor(EGFR) exon 19 deletions(Del19) would benefit from first-line second generation EGFR tyrosine kinase inhibitors(TKIs) afatinib but not for those with L858 R. This study was to investigate the survival difference between first-line first generation EGFR-TKIs and chemotherapy in patients with either Del19 or L858 R, and to directly compare OS in these two mutation groups.Methods: Eligibles were all prospective and retrospective studies comparing EGFR-TKIs with conventional chemotherapy or receiving single agent EGFR-TKIs and demonstrating survival analysis based on mutation types. The primary outcome was OS measured as pooled hazard ratios(HRs). All measures were pooled using randomeffects models and 95% confidential interval(95% CI) was calculated.Results: A total of 14 studies incorporating 1,706 patients with either Del19 or L858 R were included. Enrolling patients with Del19 or L858 R in randomized controlled trials(RCTs), first-line first generation EGFR-TKIs were associated with no OS benefit, compared with chemotherapy(pooled HR_(TKI/Chemo) for Del19: 0.82, 95% CI: 0.64-1.06, P=0.14; pooled HR_(TKI/Chemo) for L858R: 1.15, 95% CI: 0.85-1.56, P=0.38). Direct comparison of Del19 with L858 R receiving with first-line first generation EGFR-TKIs demonstrated no significant survival difference(pooled HR_(19/21): 0.88, 95% CI: 0.67-1.16, P=0.37).Conclusions: Among patients with advanced non-small cell lung cancer(NSCLC) harboring Del19 and L858 R, first-line first generation EGFR-TKIs demonstrated no survival benefit comparing with chemotherapy. | 6 | Efficacy of rigosertib, a small molecular RAS signaling disrupter for the treatment of KRAS-mutant colorectal cancer显示文摘Objective:Mutant KRAS,the principal isoform of RAS,plays a pivotal role in the oncogenesis of colorectal cancer by constitutively activating the RAF/MEK/ERK and PI3K/AKT pathways.Effective targeted therapies are urgently needed.We investigated whether rigosertib,a benzyl styryl sulfone RAS signaling disruptor,could selectively kill KRAS-mutant colorectal cancer cells.Methods:CCK-8 was used to determine the cell viability.Patient-derived tumor and cancer cell xenograft models were used to detect the inhibitory efficacy of rigosertib.Flow cytometry was used to evaluate the apoptosis and cell cycle progression.Apoptosis and cell cycle arrest markers were detected by Western blot.DCFH-DA was used to determine the reactive oxygen species.Immunohistochemistry staining and Western blot were performed to characterize RAS signaling markers in colorectal cancer tissues and cells.Results:Rigosertib(RGS)exhibited a cytotoxic effect against colorectal cancer cells,which was greater in KRAS-mutant cells.Furthermore,RGS induced mitotic arrest and oxidative stress-dependent apoptosis in KRAS-mutant DLD1 and HCT116 cells.Besides,RGS disrupted RAS signaling,and the inhibition of RAS/MEK/ERK was independent of cellular oxidative stress.Using patient-derived xenograft models,the response and tumor inhibition of RGS were significantly higher in the KRAS-mutant subgroup,while p-MEK,p-ERK,and p-AKT levels of RGS-treated tumors were significantly decreased.Finally,in a KRAS-mutant,chemotherapy-resistant patient-derived xenograft model,RGS showed a stronger therapeutic effect than the combination standard therapy involving fluoropyrimidine+oxaliplatin/irinotecan+bevacizumab.Conclusions:These data showed that targeting RAS signaling using RGS could be a therapeutic treatment for KRAS-mutant colorectal cancer patients. | Xinyi Zhou Qian Xiao Dongliang Fu Haochen Zhang Yang Tang Jinjie He Yeting Hu Xiangxing Kong Fei Teng Xiangrui Liu Ying Yuan Kefeng Ding | 2022 | Cancer Biology & Medicine2022,19,2: | 2 |

| 8 | OsADR3 increases drought stress tolerance by inducing antioxidant defense mechanisms and regulating OsGPX1 in rice(Oryza sativa L.)显示文摘The C (Cys) 2H (His) 2-type transcription factor is one of the most important transcription factors in plants and plays a regulatory role in the physiological responses of rice to abiotic stresses.A novel rice C2H2-type zinc finger protein,abscisic acid (ABA)-drought-reactive oxygen species (ROS) 3 (OsADR3),was found to confer drought stress tolerance by enhancing antioxidant defense and regulating Os GPX1.Overexpression of OsADR3 in rice increased tolerance to drought stress by increasing ROS scavenging ability and ABA sensitivity.In contrast,CRISPR/Cas9-mediated knockout of osadr3 increased the sensitivity of rice to drought and oxidative stress.An exogenous ROS-scavenging reagent restored the droughtstress tolerance of osadr3-CRISPR plants.Global transcriptome analysis suggested that OsADR3 increased the expression of Os GPX1 under drought stress.Electrophoretic mobility shift,yeast one-hybrid,and dualluciferase reporter assays revealed that OsADR3 modified the expression of Os GPX1 by directly binding to its promoter.Knockdown of Os GPX1 repressed ROS scavenging ability under drought stress in OsADR3-overexpression plants.These findings suggest that OsADR3 plays a positive regulatory role in droughtstress tolerance by inducing antioxidant defense and associated with the ABA signaling pathway in rice. | Jiaming Li Minghui Zhang Luomiao Yang Xinrui Mao Jinjie Li Lu Li Jingguo Wang Hualong Liu Hongliang Zheng Zichao Li Hongwei Zhao Xianwei Li Lei Lei Jian Sun Detang Zou | 2021 | The Crop Journal2021,9,5: | 2 |

The thrust bearing abrasion caused by axial movement of the crankshaft will affect the force of timing gears and oil pump gears,which will result in the fracture of gears,abnormal ignition,connecting rod cracking and collision of cylinder. Simulation based on CREO software is done to build three-dimensional models of crankshaft and gears of a WP10 diesel engine. The models are imported into ADAMS to complete multi-body dynamics simulations. The force analysis of gears in different kinds of axial movements is finished and variations rules of gear dynamic load is obtained. The presented results show that the crankshaft axial movement can cause overload and vibration on gears. CdS nanoparticles were synthe-sized by using dendrimer PAMAM as inner templates. The CdS nanoparticles and glucose oxidase (GOD) were immobilized on Pt electrode via layer-by-layer (LbL) technique to fabricate a biological-inorganic hybrid system. Under ultraviolet light,the photo-effect of the CdS nanoparticles showed enhancement of the biosensor to detect glucose. Pt nanoparticles were mixed into the Nafion film to immobilize the CdS/enzyme composites and to improve the charge transfer of the hybrid.
