1Protein adsorption onto diethylaminoethyl dextran modified anion exchanger:Effect of ionic strength and column behavior显示文摘Our previous studies on bovine serum albumin(BSA) adsorption to diethylaminoethyl dextran(DEAE dextran,DexD, grafting-ligand) and DEAE(D, surface-ligand) modified Sepharose FF resins found that all the grafted resins(FF-DexD and FF-D-DexD) exhibited extremely fast uptake rate(effective diffusivity, D_e, D_e/D_O> 1.4),which was six times greater than the ungrafted resins(D_e/D_O < 0.3). In this work, the influence of ionic strength(IS) on 6 typical DEAE dextran-grafted resins was investigated. Bath adsorption equilibria and kinetics, breakthrough, and linear gradient elution experiments were conducted. Commercial DEAE Sepharose FF was used for comparison. It is found that protein adsorption capacities on DEAE dextran-FF resins and the commercial resin decreased with increasing IS, but DEAE dextran-FF resins exhibited much higher capacity sensitivity to salt concentration. Besides, steeper decrease of adsorption capacities could be obtained at higher graftingligand or surface-ligand density. It is worth noting that the facilitating role of surface-ligand to the 'chain delivery' effect was weakened after adding salt, leading to the less improvement in uptake rate by increasing surface-ligand density at higher IS. Although the uptake rates of the DEAE dextran-FF resins increased first and then decreased with increasing IS, they kept the extremely high level of De values(D_e/D_O > 1.1) at the their working/binding IS range. Moreover, the DEAE dextran-FF resin displayed much higher adsorption capacities and De values than commercial ungrafted resin in their working condition. Furthermore, the column results of DEAE dextran-FF resins presented higher dynamic binding capacities than and similar elution ISs with DEAE Sepharose FF to achieve similar(or even higher) recoveries suggest the excellent chromatographic column performance of the DEAE dextran-FF resins. Finally, both high recovery and purity of BSA and γ-globulin could be easily achieved using the typical DEAE dextran-FF column, FF-D60-DexD160, to separate their binary mixtures,by step gradient elution. 2Fractional pyrolysis of Cyanobacteria from water blooms over HZSM-5 for high quality bio-oil production显示文摘Fractional pyrolysis and one-step pyrolysis of natural algae Cyanobacteria from Taihu Lake were comparatively studied from 200 to 500°C.One-step pyrolysis produced bio-oil with complex composition and low high heating value(HHV 30.9 MJ/kg). Fractional pyrolysis separated the degradation of different components in Cyanobacteria and improved the selectivity to products in bio-oil. That is, acids at 200°C, amides and acids at 300°C, phenols and nitriles at 400°C, and phenols at 500°C, were got as main products, respectively. HZSM-5 could promote the dehydration, cracking and aromatization of pyrolytic intermediates in fractional pyrolysis. At optimal HZSM-5 catalyst dosage of 1.0 g, the selectivity to products and the quality of bio-oil were improved obviously. The main products in bio-oil changed to nitriles(47.2%) at 300°C,indoles(51.3%) and phenols(36.3%) at 400°C. The oxygen content was reduced to 7.2 wt% and 9.4 wt%, and the HHV was raised to 38.1and 37.3 MJ/kg at 300 and 400°C, respectively. 3A novel secretagogin/ATF4 pathway is involved in oxidized LDL-induced endoplasmic reticulum stress and islet β-cell apoptosis显示文摘Excessive accumulation of cholesterol inβcells initiates endoplasmic reticulum(ER)stress and associated apoptosis.We have reported that excessive uptake of cholesterol by MIN6 cells decreases the expression of secretagogin(SCGN)and then attenuates insulin secretion.Here,we aimed to determine whether cholesterol-induced SCGN decrease is involved in the modulation of ER stress and apoptosis in pancreaticβcells.In this study,MIN6 cells were treated with oxidized low-density lipoprotein(ox-LDL)for 24 h,and then intracellular lipid droplets and cell apoptosis were quantified,and SCGN and ER stress markers were identified by western blot analysis.Furthermore,small interfer RNA(siRNA)-mediated SCGN knockdown and recombinant plasmid-mediated SCGN restoration experiments were performed to confirm the role of SCGN in ER stress and associated cell apoptosis.Finally,the interaction of SCGN with ATF4 was computationally predicted and then validated by a co-immunoprecipitation assay.We found that ox-LDL treatment increased the levels of ER stress markers,such as phosphorylated protein kinase-like endoplasmic reticulum kinase,phosphorylated eukaryotic initiation factor 2 alpha,activating transcription factor 4(ATF4),and transcription factor CCAAT-enhancer-binding protein homologous protein,and promoted MIN6 cell apoptosis;in addition,the expression of SCGN was downregulated.siRNA-mediated SCGN knockdown exacerbatedβ-cell ER stress by increasing ATF4 expression.Pretreatment of MIN6 cells with the recombinant SCGN partly antagonized ox-LDL-induced ER stress and apoptosis.Furthermore,a co-immunoprecipitation assay revealed an interaction between SCGN and ATF4 in MIN6 cells.Taken together,these results demonstrated that pancreaticβ-cell apoptosis induced by ox-LDL treatment can be attributed,in part,to an SCGN/ATF4-dependent ER stress response.Li Wu Yuncheng Lv Ying Lv Sunmin Xiang Zhibo Zhao Ziqing Tang Linling Ou Bin Yan Xinhua Xiao Gebo Wen Renxian Cao Jing Yang 2021Acta Biochimica et Biophysica Sinica2021,53,1:2

5Development of Specific SSR Molecular Marker of 'Huangzhou Radish' Based on Purity Identification显示文摘As an excellent local crop variety of Huanggang,'Huangzhou radish' has been widely promoted and its market influence is increasing day by day.Recently,the purity of 'Huangzhou radish' has declined deeply,so it is urgent to improve and strengthen the purity and viability of 'Huangzhou radish' including bad phenomena such as degeneration of varieties after self-pollination.At present,there was no SSR molecular marker can be used for genetic diversity identification of 'Huangzhou radish'.SSR molecular marker can provide a favorable tool to study 'Huangzhou radish'.In this study,'Xiakang','Hongbao'and 'Huangzhou radish' were used as experimental materials,100 SSR primers were screened,and 25 pairs of SSR primers were selected.Finally,two pairs of primers were selected for identification of the purity of 'Huangzhou radish' by non-denatured polyacrylamide gel electrophoresis (PAGE).The results showed that R-102 and R-112 primer pairs could amplify the special bands for 'Huangzhou radish' from above three kinds of radishes.These two pairs of primers could be developed as SSR markers to identify the specificity and purity of 'Huangzhou radish'.Furthermore,this study provides convenient conditions for promoting the economic and social value of 'Huangzhou radish'.Pingan WANG Jinlan YAO Qiuyu ZENGCHEN Linling WANG Jingcai LI Yuanping FANG Shisheng LI 2019Agricultural Biotechnology2019,8,5:2

17Sensitive semi-quantitative detection of respiratory syncytial virus by dark-field light scattering imaging of the infected host cells显示文摘A novel sensitive semi-quantitative virus detection technique was developed using the respiratory syncytial virus(RSV) as an example, through dark-field light scattering imaging of the surface state of the virusinvaded host cells. In this method, anti-RSV-antibody modified gold nanoparticles(Au NPs) could bind with the invading virus on the cell membrane of the infected host cells through the specific antibody-antigen binding. 18Characteristics of alveolar macrophages in bronchioalveolar lavage fluids from active tuberculosis patients identified by single-cell RNA sequencing显示文摘Tuberculosis(TB),is an infectious disease caused by Mycobacterium tuberculosis(M.tuberculosis),and presents with high morbidity and mortality.Alveolar macrophages play an important role in TB pathogenesis although there is heterogeneity and functional plasticity.This study aimed to show the characteristics of alveolar macrophages from bronchioalveolar lavage fluid(BALF)in active TB patients.Single-cell RNA sequencing(scRNA-seq)was performed on BALF cells from three patients with active TB and additional scRNA-seq data from three healthy adults were established as controls.Transcriptional profiles were analyzed and compared by differential gene expression and functional enrichment analysis.We applied pseudo-temporal trajectory analysis to investigate correlations and heterogeneity within alveolar macrophage subclusters.Alveolar macrophages from active TB patients at the single-cell resolution are described.We found that TB patients have higher cellular percentages in five macrophage subclusters.Alveolar macrophage subclusters with increased percentages were involved in inflammatory signaling pathways as well as the basic macrophage functions.The TB-increased alveolar macrophage subclusters might be derived from M1-like polarization state,before switching to an M2-like polarization state with the development of M.tuberculosis infection.Cell-cell communications of alveolar macrophages also increased and enhanced in active TB patients.Overall,our study demonstrated the characteristics of alveolar macrophages from BALF in active TB patients by using scRNA-seq.Qianqian Chen Chunmei Hu Wei Lu Tianxing Hang Yan Shao Cheng Chen Yanli Wang Nan Li Linling Jin Wei Wu Hong Wang Xiaoning Zeng Weiping Xie 2022The Journal of Biomedical Research2022,36,3:1

20Hallmark guided identification and characterization of a novel immune-relevant signature for prognostication of recurrence in stageⅠ-Ⅲlung adenocarcinoma显示文摘The high risk of postoperative mortality in lung adenocarcinoma(LUAD)patients is principally driven by cancer recurrence and low response rates to adjuvant treatment.Here,A combined cohort containing 1,026 stageⅠ-Ⅲpatients was divided into the learning(n Z 678)and validation datasets(n Z 348).The former was used to establish a 16-mRNA risk signature for recurrence prediction with multiple statistical algorithms,which was verified in the valida-tion set.Univariate and multivariate analyses confirmed it as an independent indicator for both recurrence-free survival(RFS)and overall survival(OS).Distinct molecular characteristics between the two groups including genomic alterations,and hallmark pathways were compre-hensively analyzed.Remarkably,the classifier was tightly linked to immune infiltrations,high-lighting the critical role of immune surveillance in prolonging survival for LUAD.Moreover,the classifier was a valuable predictor for therapeutic responses in patients,and the low-risk group was more likely to yield clinical benefits from immunotherapy.A transcription factor regulato-ry proteineprotein interaction network(TF-PPI-network)was constructed via weighted gene co-expression network analysis(WGCNA)concerning the hub genes of the signature.The con-structed multidimensional nomogram dramatically increased the predictive accuracy.There-fore,our signature provides a forceful basis for individualized LUAD management with promising potential implications.Yongqiang Zhang Zhao Yang Yuqin Tang Chengbin Guo Danni Lin d Linling Cheng Xun Hu Kang Zhang Gen Li 2023Genes & Diseases2023,10,4:0
