| 2 | Review on supermolecules as chemical drugs显示文摘Supramolecular medicinal chemistry field has been a quite rapidly developing,increasingly active and newly rising interdiscipline which is the new expansion of supramolecular chemistry in pharmaceu-tical sciences,and is gradually becoming a relatively independent scientific area. Supramolecular drugs could be defined as medicinal supermolecules formed by two or more molecules through non-covalent bonds. So far a lot of supermolecules as chemical drugs have been widely used in clinics. Supermolecules as chemical drugs,i.e. supramolecular chemical drugs or supramolecular drugs,which might have the excellences of lower cost,shorter period,higher potential as clinical drugs for their successful research and development,may possess higher bioavailability,better biocompatibility and drug-targeting,fewer multidrug-resistances,lower toxicity,less adverse effect,and better curative effects as well as safety,and therefore exhibit wide potential application. These overwhelming advan-tages have drawn enormous special attention. This paper gives the definition of supramolecular drugs,proposes the concept of supramolecular chemical drugs,and systematically reviews the recent ad-vances in the research and development of supermolecules,including organic and inorganic complex ones as chemical drugs in the area of antitumor,anti-inflammatory,analgesic,antimalarial,antibacterial,antifungal,antivirus,anti-epileptic,cardiovascular agents and magnetic resonance imaging agents and so on. | 3 | Adaptive subsequence adjustment with evolutionary asymmetric path-relinking for TDRSS scheduling显示文摘Due to the limited transmission resources for data relay in the tracking and data relay satellite system(TDRSS), there are many job requirements in busy days which will be discarded in the conventional job scheduling model. Therefore, the improvement of scheduling efficiency in the TDRSS can not only help to increase the resource utilities, but also to reduce the scheduling failure ratio.A model of nonhomogeneous parallel machines scheduling problems with time window(NPM-TW) is firstly built up for the TDRSS,considering the distinct features of the variable preparation time and the nonhomogeneous transmission rates for different types of antennas on each tracking and data relay satellite(TDRS). Then,an adaptive subsequence adjustment(ASA) framework with evolutionary asymmetric path-relinking(Ev APR) is proposed to solve this problem, in which an asymmetric progressive crossover operation is involved to overcome the local optima by the conventional job inserting methods. | 4 | Implications from protein uptake kinetics onto dextran-grafted Sepharose FF coupled with ion exchange and affinity ligands显示文摘Our previous studies have reported the presence of 'chain delivery' effects of protein adsorption onto ion exchangers with polymer-grafted ion-exchange groups, such as dextran-grafted and poly(ethylenimine)-modified Sepharose gels. However, it is unclear if the 'chain delivery' occurs on affinity adsorption with specific interactions. This work is designed to address this issue. A dextran-grafted Sepharose gel was prepared, and then the matrix was modified using diethylaminoethyl, a typical ion-exchange group, or octapeptide(FYCHWQDE), an affinity ligand for human immunoglobulin G(h Ig G) to prepare ion-exchange or affinity adsorbents, respectively.Results of h Ig G adsorption showed that the uptake rate represented by the effective diffusivity of h Ig G onto the dextran-grafted ion exchangers was obviously enhanced by the dextran grafting, indicating the presence of'chain delivery' of the bound proteins on the charged groups on the dextran chains. By contrast, the effective diffusivity of h Ig G changed little as ligand density increased on the dextran-grafted FYCHWQDE adsorbents.Their adsorption capacities decreased and effective diffusivities were not accelerated by the dextran grafting.Thus, this work clarified that grafted dextran could not accelerate h Ig G uptake rate on the affinity resins, or in other words, chain delivery did not occur on the specific interaction-based affinity adsorption. | Aiying Xue Linling Yu Yan Sun | 2017 | Chinese Journal of Chemical Engineering2017,25,7: | 10 |
| 5 | Protein adsorption onto diethylaminoethyl dextran modified anion exchanger:Effect of ionic strength and column behavior显示文摘Our previous studies on bovine serum albumin(BSA) adsorption to diethylaminoethyl dextran(DEAE dextran,DexD, grafting-ligand) and DEAE(D, surface-ligand) modified Sepharose FF resins found that all the grafted resins(FF-DexD and FF-D-DexD) exhibited extremely fast uptake rate(effective diffusivity, D_e, D_e/D_O> 1.4),which was six times greater than the ungrafted resins(D_e/D_O < 0.3). In this work, the influence of ionic strength(IS) on 6 typical DEAE dextran-grafted resins was investigated. Bath adsorption equilibria and kinetics, breakthrough, and linear gradient elution experiments were conducted. Commercial DEAE Sepharose FF was used for comparison. It is found that protein adsorption capacities on DEAE dextran-FF resins and the commercial resin decreased with increasing IS, but DEAE dextran-FF resins exhibited much higher capacity sensitivity to salt concentration. Besides, steeper decrease of adsorption capacities could be obtained at higher graftingligand or surface-ligand density. It is worth noting that the facilitating role of surface-ligand to the 'chain delivery' effect was weakened after adding salt, leading to the less improvement in uptake rate by increasing surface-ligand density at higher IS. Although the uptake rates of the DEAE dextran-FF resins increased first and then decreased with increasing IS, they kept the extremely high level of De values(D_e/D_O > 1.1) at the their working/binding IS range. Moreover, the DEAE dextran-FF resin displayed much higher adsorption capacities and De values than commercial ungrafted resin in their working condition. Furthermore, the column results of DEAE dextran-FF resins presented higher dynamic binding capacities than and similar elution ISs with DEAE Sepharose FF to achieve similar(or even higher) recoveries suggest the excellent chromatographic column performance of the DEAE dextran-FF resins. Finally, both high recovery and purity of BSA and γ-globulin could be easily achieved using the typical DEAE dextran-FF column, FF-D60-DexD160, to separate their binary mixtures,by step gradient elution. | 7 | Fractional pyrolysis of Cyanobacteria from water blooms over HZSM-5 for high quality bio-oil production显示文摘Fractional pyrolysis and one-step pyrolysis of natural algae Cyanobacteria from Taihu Lake were comparatively studied from 200 to 500°C.One-step pyrolysis produced bio-oil with complex composition and low high heating value(HHV 30.9 MJ/kg). Fractional pyrolysis separated the degradation of different components in Cyanobacteria and improved the selectivity to products in bio-oil. That is, acids at 200°C, amides and acids at 300°C, phenols and nitriles at 400°C, and phenols at 500°C, were got as main products, respectively. HZSM-5 could promote the dehydration, cracking and aromatization of pyrolytic intermediates in fractional pyrolysis. At optimal HZSM-5 catalyst dosage of 1.0 g, the selectivity to products and the quality of bio-oil were improved obviously. The main products in bio-oil changed to nitriles(47.2%) at 300°C,indoles(51.3%) and phenols(36.3%) at 400°C. The oxygen content was reduced to 7.2 wt% and 9.4 wt%, and the HHV was raised to 38.1and 37.3 MJ/kg at 300 and 400°C, respectively. | 9 | A novel secretagogin/ATF4 pathway is involved in oxidized LDL-induced endoplasmic reticulum stress and islet β-cell apoptosis显示文摘Excessive accumulation of cholesterol inβcells initiates endoplasmic reticulum(ER)stress and associated apoptosis.We have reported that excessive uptake of cholesterol by MIN6 cells decreases the expression of secretagogin(SCGN)and then attenuates insulin secretion.Here,we aimed to determine whether cholesterol-induced SCGN decrease is involved in the modulation of ER stress and apoptosis in pancreaticβcells.In this study,MIN6 cells were treated with oxidized low-density lipoprotein(ox-LDL)for 24 h,and then intracellular lipid droplets and cell apoptosis were quantified,and SCGN and ER stress markers were identified by western blot analysis.Furthermore,small interfer RNA(siRNA)-mediated SCGN knockdown and recombinant plasmid-mediated SCGN restoration experiments were performed to confirm the role of SCGN in ER stress and associated cell apoptosis.Finally,the interaction of SCGN with ATF4 was computationally predicted and then validated by a co-immunoprecipitation assay.We found that ox-LDL treatment increased the levels of ER stress markers,such as phosphorylated protein kinase-like endoplasmic reticulum kinase,phosphorylated eukaryotic initiation factor 2 alpha,activating transcription factor 4(ATF4),and transcription factor CCAAT-enhancer-binding protein homologous protein,and promoted MIN6 cell apoptosis;in addition,the expression of SCGN was downregulated.siRNA-mediated SCGN knockdown exacerbatedβ-cell ER stress by increasing ATF4 expression.Pretreatment of MIN6 cells with the recombinant SCGN partly antagonized ox-LDL-induced ER stress and apoptosis.Furthermore,a co-immunoprecipitation assay revealed an interaction between SCGN and ATF4 in MIN6 cells.Taken together,these results demonstrated that pancreaticβ-cell apoptosis induced by ox-LDL treatment can be attributed,in part,to an SCGN/ATF4-dependent ER stress response. | Li Wu Yuncheng Lv Ying Lv Sunmin Xiang Zhibo Zhao Ziqing Tang Linling Ou Bin Yan Xinhua Xiao Gebo Wen Renxian Cao Jing Yang | 2021 | Acta Biochimica et Biophysica Sinica2021,53,1: | 2 |

| 12 | QoE-Driven Social Aware Caching Placement for Terrestrial-Satellite Networks显示文摘In remote terrestrial-satellite networks, caching is a very promising technique to alleviate the burden of space cloudlet(e.g., cache-enabled satellite user terminal) and to improve subscribers' quality of experience(Qo E) in terms of buffering delay and achievable video streaming rate. In this paper, we studied a Qo E-driven caching placement optimization problem for video streaming that takes into account the required video streaming rate and the social relationship among users. Social ties between users are used to designate a set of helpers with caching capability, which can cache popular files proactively when the cloudlet is idle. We model the utility function of Qo E as a logarithmic function. Then, the caching placement problem is formulated as an optimization problem to maximize the user's average Qo E subject to the storage capacity constraints of the helpers and the cloudlets. Furthermore, we reformulate the problem into a monotone submodular optimization problem with a partition matroid constraint, and an efficient greedy algorithm with 1-1 e approximation ratio is proposed to solve it.
