| 1 | TREM-1 multimerization is essential for its activation on monocytes and neutrophils显示文摘The triggering receptor expressed on myeloid cells-1(TREM-1)is a receptor expressed on innate immune cells.By promoting the amplification of inflammatory signals that are initially triggered by Toll-like receptors(TLRs),TREM-1 has been characterized as a major player in the pathophysiology of acute and chronic inflammatory diseases,such as septic shock,myocardial infarction,atherosclerosis,and inflammatory bowel diseases.However,the molecular events leading to the activation of TREM-1 in innate immune cells remain unknown.Here,we show that TREM-1 is activated by multimerization and that the levels of intracellular Ca 2+release,reactive oxygen species,and cytokine production correlate with the degree of TREM-1 aggregation.TREM-1 activation on primary human monocytes by LPS required a two-step process consisting of upregulation followed by clustering of TREM-1 at the cell surface,in contrast to primary human neutrophils,where LPS induced a rapid cell membrane reorganization of TREM-1,which confirmed that TREM-1 is regulated differently in primary human neutrophils and monocytes.In addition,we show that the ectodomain of TREM-1 is able to homooligomerize in a concentration-dependent manner,which suggests that the clustering of TREM-1 on the membrane promotes its oligomerization.We further show that the adapter protein DAP12 stabilizes TREM-1 surface expression and multimerization.TREM-1 multimerization at the cell surface is also mediated by its endogenous ligand,a conclusion supported by the ability of the TREM-1 inhibitor LR12 to limit TREM-1 multimerization.These results provide evidence for ligand-induced,receptor-mediated dimerization of TREM-1.Collectively,our findings uncover the mechanisms necessary for TREM-1 activation in monocytes and neutrophils. | Kevin Carrasco Amir Boufenzer Lucie Jolly Helene Le Cordier Guanbo Wang Albert JR Heck Adelheid Cerwenka Emilie Vinolo Alexis Nazabal Alexandre Kriznik Pierre Launay Sebastien Gibot Marc Derive | 2019 | Cellular & Molecular Immunology2019，16，5： | 13 |
| 2 | Usefulness of staging systems and prognostic scores for hepatocellular carcinoma treatments显示文摘Therapeutic management of hepatocellular carcinoma(HCC) is quite complex owing to the underlying cirrhosis and portal vein hypertension. Different scores or classification systems based on liver function and tumoral stages have been published in the recent years. If none of them is currently 'universally' recognized, the Barcelona Clinic Liver Cancer(BCLC) staging system has become the reference classification system in Western countries. Based on a robust treatment algorithm associated with stage stratification, it relies on a high level of evidence. However, BCLC stage B and C HCC include a broad spectrum of tumors but are only matched with a single therapeutic option. Some experts have thus suggested to extend the indications for surgery or for transarterial chemoembolization. In clinical practice, many patients are already treated beyond the scope of recommendations. Additional alternative prognostic scores that could be applied to any therapeutic modality have been recently proposed. They could represent complementary tools to the BCLC staging system and improve the stratification of HCC patients enrolled in clinical trials, as illustrated by the NIACE score. | 8 | Simulation and Visualisation of Functional Landscapes:Effects of the Water Resource Competition Between Plants显示文摘Vegetation ecosystem simulation and visualisation are challenging topics involving multidisciplinary aspects. In this paper,we present a new generic frame for the simulation of natural phenomena through manageable and interacting models.It focuses on the functional growth of large vegetal ecosystems,showing coherence for scales ranging from the individual plant to communities and with a particular attention to the effects of water resource competition between plants.The proposed approach is based on a model of plant growth in interaction with the environmental conditions. These are deduced from the climatic data (light,temperature,rainfall) and a model of soil hydrological budget.A set of layers is used to store the water resources and to build the interfaces between the environmental data and landscape components:temperature,rain,light,altitude,lakes,plant positions,biomass,cycles,etc.At the plant level,the simulation is performed for each individual by a structural-functional growth model,interacting with the plant's environment.Temperature is spatialised,changing according to altitude,and thus locally controls plant growth speed.The competition for water is based on a soil hydrological model taking into account rainfalls,water runoff, absorption,diffusion,percolation in soil.So far,the incoming light radiation is not studied in detail and is supposed constant.However,competition for light between plants is directly taken into account in the plant growth model.In our implementation,we propose a simple architecture for such a simulator and a simulation scheme to synchronise the water resource updating (on a temporal basis) and the plant growth cycles (determined by the sum of daily temperatures).
