2Prospective study of biofeedback retraining in patients with chronic idiopathic functional constipation显示文摘AIM: To determine the efficacy and long-term outcome of biofeedback treatment for chronic idiopathic constipation and to compare the efficacy of two modes of biofeedback (EMG-based and manometry-based biofeedback).METHODS: Fifty consecutive contactable patients included 8 cases of slow transit constipation, 36 cases of anorectic outlet obstruction and 6 cases of mixed constipation. Two modes of biofeedback were used for these 50 patients, 30 of whom had EMG-based biofeedback, and 20 had manometrybased biofeedback. Before treatment, a consultation and physical examination were done for all the patients, related information such as bowel function and gut transit time was documented, psychological test (symptom checkJist 90, SCL90)and anorectic physiological test and defecography were applied. After biofeedback management, all the patients were followed up. The Student′s t-test, chi-squared test and Logistic regression were used for statistical analysis.RESULTS: The period of following up ranged from 12 to 24months (Median 18 months). 70% of patients felt that biofeedback was helpful, and 62.5% of patients with constipation were improved. Clinical manifestations including straining, abdominal pain, bloating, were relieved, and less oral laxative was used. Spontaneous bowel frequency and psychological state were improved significantly after treatment. Patients with slow and normal transit, and those with and without paradoxical contraction of the anal sphincter on straining, benefited equally from the treatment. The psychological status rather than anorectal test could predict outcome. The efficacy of the two modes of biofeedback was similar without side effects.CONCLUSION: This study suggests that biofeedback has a long-term effect with no side effects, for the majority of patients with chronic idiopathic constipation unresponsive to traditional treatment. 3A Genome Sequence of Novel SARS-CoV Isolates: the Genotype, GD-Ins29, Leads to a Hypothesis of Viral Transmission in South China显示文摘We report a complete genomic sequence of rare isolates (minor genotype) of theSARS-CoV from SARS patients in Guangdong, China, where the first few casesemerged. The most striking discovery from the isolate is an extra 29-nucleotidesequence located at the nucleotide positions between 27,863 and 27,864 (referredto the complete sequence of B J01) within an overlapped region composed of BGI-PUP5 (BGI-postulated uncharacterized protein 5) and BGI-PUP6 upstream ofthe N (nucleocapsid) protein. The discovery of this minor genotype, GD-Ins29,suggests a significant genetic event and differentiates it from the previously re-ported genotype, the dominant form among all sequenced SARS-CoV isolates. A17-nt segment of this extra sequence is identical to a segment of the same size intwo human mRNA sequences that may interfere with viral genome replication andtranscription in the cytosol of the infected cells. 4Clinicopathologic study of mucosa-associated lymphoid tissue lymphoma in gastroscopic biopsy显示文摘AIM: To explore and discuss the clinicopathologic characteristics of mucosa-associated lymphoid tissue (MALT)lymphoma in gastroscopic biopsy specimen. METHODS: A retrospective study of 26 cases of lymphoma diagnosed by gastroscopic biopsy during 1999 to 2001 from gastroscopy files of Xijing Hospital was made. The diagnostic criteria were adopted according to the new classification of non-Hodgkin's lymphoma. RESULTS: Twenty-six cases of primary gastric lymphoma consisting of 15 men and 11 women, aged between 23 to 76 years were recruited from 6 225 cases who Received gastroscopy. All of them were diagnosed by both endoscopic findings and histological examinations. Histologically, 23cases were MALToma (low grade) and 3 cases lymphoblastic lymphoma (high grade). Immunohistochemically, all cases were CD20 positive, while CK and EMA were negative. 7An aptamer-based shear horizontal surface acoustic wave biosensor with a CVD-grown single- layered graphene film for high-sensitivity detection of a label-free endotoxin显示文摘The thickness of the sensitive layer has an important influence on the sensitivity of a shear horizontal surface acoustic wave(SH-SAW)biosensor with a delay-line structure and lower layer numbers of graphene produce better sensitivity for biological detection.Therefore,a label-free and highly sensitive SH-SAW biosensor with chemical vapor deposition(CVD-)-grown single-layered graphene(SLG)for endotoxin detection was developed in this study.With this methodology,SH-SAW biosensors were fabricated on a 36°Y-90°X quartz substrate with a base frequency of 246.2MHz,and an effective detection cell was fabricated using acrylic material.To increase the surface hydrophilicity,chitosan was applied to the surface of the SLG film.Additionally,the aptamer was immobilized on the surface of the SLG film by cross-linking with glutaraldehyde.Finally,the sensitivity was verified by endotoxin detection with a linear detection ranging from 0 to 100 ng/mL,and the detection limit(LOD)was as low as 3.53 ng/mL.In addition,the stability of this type of SH-SAW biosensor from the air phase to the liquid phase proved to be excellent and the specificity was tested and verified by detecting the endotoxin obtained from Escherichia coli(E.coli),the endotoxin obtained from Pseudomonas aeruginosa(P.aeruginosa),and aflatoxin.Therefore,this type of SH-SAW biosensor with a CVD-grown SLG film may offer a promising approach to endotoxin detection,and it may have great potential in clinical applications.Junwang Ji Yiquan Pang Dongxiao Li Zheng Huang Zuwei Zhang Ning Xue Yi Xu Xiaojing Mu 2020Microsystems & Nanoengineering2020,6,1:3
8CHARACTERISTICS OF SEDIMENT TRANSPORT ALONG A RIVER REACH WITH A RESERVOIR显示文摘Based on long-term measurements at three gauging stations,Toudaoguai,Fugu and Hequ,and one meteorological station,this article discusses the features of discharge (Q) and sediment concentration (CS) of a river reach of the Yellow River with a reservoir located in the Loess Plateau. The impacts of the local sub-watershed between Toudaoguai and Fugu gauging stations on sediment budget to the Yellow River have been analyzed. In addition,the deposition processes in the Tianqiao Reservoir have been investigated. Results show over 80% of the precipitation that falls in the local sub- watershed is unable to contribute to the Yellow River runoff process. It is found that the annual maximum sediment concentration is usually less than 30 kg/m3 during flood seasons at Toudaoguai Gauging Station,but the sediment concentration varies dramatically at Fugu Gauging Station. About 35% of the sediment eroded in the sub-watersheds between Toudaoguai and Fugu gauging stations was produced from the Huangfuchuan sub-watershed which has a drainage area accounting only for 10% of the drainage area between Toudaoguai and Fugu gauging stations. The Tianqiao Reservoir generally has deposition during the summer flood season,and scouring during the non-flood season. On average,over 85% of deposited sediment in the reservoir occurs in the 12 km long lower reservoir reach. 10The E Protein Is a Multifunctional Membrane Protein of SARS-CoV显示文摘The E (envelope) protein is the smallest structural protein in all coronaviruses andis the only viral structural protein in which no variation has been detected. Weconducted genome sequencing and phylogenetic analyses of SARS-CoV. Based ongenome sequencing, we predicted the E protein is a transmembrane (TM) pro-tein characterized by a TM region with strong hydrophobicity and α-helix con-formation. We identified a segment (NH2-_L-Cys-A-Y-Cys-Cys-N_-COOH) in thecarboxyl-terminal region of the E protein that appears to form three disulfide bondswith another segment of corresponding cysteines in the carboxyl-terminus of the S(spike) protein. These bonds point to a possible structural association between theE and S proteins. 13A novel three primers PCR (TP-PCR) method to obtain recombinant DNA molecule independent of restriction enzyme显示文摘In this note, we report a novel and efficient three primers PCR (TP-PCR) method to rapidly generate recombinant DNA molecule at precise junction between two arbitrary DNA fragments. TP-PCR method is characterized by its reaction system with two templates and three primers, which can produce a recombinant DNA molecule in one PCR reaction. The main advantages of this method are the independence of sequences at the recombination site, the rapid-ness, and the easy establishment of adequate conditions. 15Ground-high altitude joint detection of ozone and nitrogen oxides in urban areas of Beijing显示文摘Based on observational data of ozone(O3) and nitrogen oxide(NOx) mixing ratios on the ground and at high altitude in urban areas of Beijing during a period of six days in November 2011,the temporal and spatial characteristics of mixing ratios were analyzed.The major findings include:urban O3 mixing ratios are low and NOx mixing ratios are always high near the road in November.Vertical variations of the gases are significantly different in and above the planetary boundary layer.The mixing ratio of O3 is negatively correlated with that of NOx and they are positively correlated with air temperature,which is the main factor directly causing vertical variation of O3 and NOx mixing ratios at 600-2100m altitude.The NOx mixing ratios elevated during the heating period,while the O3 mixing ratios decreased:these phenomena are more significant at high altitudes compared to lower altitudes.During November,air masses in the urban areas of Beijing are brought by northwesterly winds,which transport O3 and NOx at low mixing ratios.Due to Beijing's natural geographical location,northwest air currents are beneficial to the dilution and dispersion of pollutants,which can result in lower O3 and NOx background values in the Beijing urban area.Pengfei Chen Qiang Zhang Jiannong Quan Yang Gao Delong Zhao Junwang Meng 2013Journal of Environmental Sciences2013,25,4:1

17A Statistical Approach Designed for Finding Mathematically Defined Repeats in Shotgun Data and Determining the Length Distribution of Clone-Inserts显示文摘The large amount of repeats, especially high copy repeats, in the genomes of higher animals and plants makes whole genome assembly (WGA) quite difficult. In order to solve this problem, we tried to identify repeats and mask them prior to assembly even at the stage of genome survey. It is known that repeats of different copy number have different probabilities of appearance in shotgun data, so based on this principle, we constructed a statistical model and inferred criteria for mathematically defined repeats (MDRs) at different shotgun coverages. According to these criteria, we developed software MDRmasker to identify and mask MDRs in shotgun data. With repeats masked prior to assembly, the speed of assembly was increased with lower error probability. In addition, clone-insert size affects the accuracy of repeat assembly and scaffold construction. We also designed length distribution of clone-inserts using our model. In our simulated genomes of human and rice, the length distribution of repeats is different, so their optimal length distributions of clone-inserts were not the same.
