| 3 | Developing a bacteriophage cocktail for biocontrol of potato bacterial wilt显示文摘Bacterial wilt is a devastating disease of potato and can cause an 80% production loss. To control wilt using bacteriophage therapy, we isolated and characterized twelve lytic bacteriophages from different water sources in Kenya and China. Based on the lytic curves of the phages with the pathogen Ralstonia solanacearum, one optimal bacteriophage cocktail, P1, containing six phage isolations was formulated and used for studying wilt prevention and treatment efficiency in potato plants growing in pots. The preliminary tests showed that the phage cocktail was very effective in preventing potato bacterial wilt by injection of the phages into the plants or decontamination of sterilized soil spiked with R. solanacearum. Eighty percent of potato plants could be protected from the bacterial wilt(caused by R. solanacearum reference strain GIM1.74 and field isolates), and the P1 cocktail could kill 98% of live bacteria spiked in the sterilized soil at one week after spraying.However, the treatment efficiencies of P1 depended on the timing of application of the phages, the susceptibility of the plants to the bacterial wilt, as well as the virulence of the bacteria infected,suggesting that it is important to apply the phage therapy as soon as possible once there are early signs of the bacterial wilt. | 4 | Analysis on Traditional Chinese Medicine syndrome elements and relevant factors for senile diabetes显示文摘OBJECTIVE: To explore the laws governing the distribution ofTraditional Chinese Medicine (TCM) syndrome elements (SEs) of senile diabetes (SD) and their relationship to relevant factors. METHODS: An investigation of patients who met the inclusion criteria was conducted by trained doctors, using case report forms. All related data were collected, including body mass index, glycated hemoglobin, illness course, complications, symptoms, and tongue and pulse manifestation. The SEs of each patient were judged by three qualified associate chief physicians independently. RESULTS: The main SEs of SD are Yin deficiency, Qi deficiency, blood stasis, and phlegm turbidity. Yindeficiency, Qi deficiency, blood stasis, and phlegm turbidity are most commonly seen among 4-SE combinations. Yang deficiency is typically related to illness course and BMI, phlegm turbidity to hypertension and hyperlipidemia, excessive heat to diabetic microangiopathy, and blood stasis to illness course and diabetic macroangiopathy. CONCLUSION: SD pathogenesis has a deficiency in origin and excess in superficiality. Deficiency syndrome mainly manifests as deficiency of both Qi and Yin, and concurrently in Yang deficiency. Excess syndrome is characterized by blood stasis and phlegm turbidity. | 6 | Bacteriophages Isolated in China for the Control of Pectobacterium carotovorum Causing Potato Soft Rot in Kenya显示文摘Soft rot is an economically significant disease in potato and one of the major threats to sustainable potato production.This study aimed at isolating lytic bacteriophages and evaluating methods for and the efficacy of applying phages to control potato soft rot caused by Pectobacterium carotovorum.Eleven bacteriophages isolated from soil and water samples collected in Wuhan,China,were used to infect P.carotovorum host strains isolated from potato tubers showing soft rot symptoms in Nakuru county,Kenya.The efficacy of the phages in controlling soft rot disease was evaluated by applying individual phage strains or a phage cocktail on potato slices and tubers at different time points before or after inoculation with a P.carotovorum strain.The phages could lyse 20 strains of P.carotovorum,but not Pseudomonas fluorescens control strains.Among the 11 phages,Pectobacterium phage Wc5r,interestingly showed cross-activity against Pectobacterium atrosepticum and two phage-resistant P.carotovorum strains.Potato slice assays showed that the phage concentration and timing of application are crucial factors for effective soft rot control.Phage cocktail applied at a concentration of 1×10^9 plaque-forming units per milliliter before or within an hour after bacterial inoculation on potato slices,resulted in>90%reduction of soft rot symptoms.This study provides a basis for the development and application of phages to reduce the impact of potato soft rot disease. | Peter Muturi Junping Yu Alice Nyambura Maina Samuel Kariuki Frantis B.Mwaura Hongping Wei | 2019 | Virologica Sinica2019,34,3: | 4 |
| 7 | Thermal conductivity of natural rubber nanocomposites with hybrid fillers显示文摘Natural rubber nanocomposites filled with hybrid fillers of multi-walled carbon nanotubes(CNTs) and carbon black(CB) were prepared. CNTs were ultrasonically modified in mixture of hydrogen peroxide(H2O2) and distilled water(H2O). The functional groups on the surface of CNTs, changes in nanotube structure and morphology were characterized by Fourier transform infrared spectroscopy(FT-IR), Raman Spectroscopy, and transmission electron microscopy(TEM). It shows that hydroxyl(OH·) is successfully introduced. The surface defects of modified CNTs were obviously higher than those of original CNTs, and the degree of agglomeration was greatly reduced. Thermal conductivity of the composites was tested by protection heat flow meter method. Compared with unmodified CNTs/CB filling system, the thermal conductivity of hybrid composites is improved by an average of 5.8% with 1.5 phr(phr is parts per hundred rubber) of hydroxyl CNTs and 40 phr of CB filled. A three-dimensional heat conduction network composed of hydroxyl CNTs and CB, as observed by TEM, contributes to the good properties. Thermal conductivity of the hybrid composites increases as temperature rises. | 9 | Enriched Opportunistic Pathogens Revealed by Metagenomic Sequencing Hint Potential Linkages between Pharyngeal Microbiota and COVID-19显示文摘As a respiratory tract virus,SARS-CoV-2 infected people through contacting with the upper respiratory tract first.Previous studies indicated that microbiota could modulate immune response against pathogen infection.In the present study,we performed metagenomic sequencing of pharyngeal swabs from eleven patients with COVID-19 and eleven Non-COVID-19 patients who had similar symptoms such as fever and cough.Through metagenomic analysis of the above two groups and a healthy group from the public data,there are 6502 species identified in the samples.Specifically,the Pielou index indicated a lower evenness of the microbiota in the COVID-19 group than that in the Non-COVID-19 group.Combined with the linear discriminant analysis(LDA)and the generalized linear model,eighty-one bacterial species were found with increased abundance in the COVID-19 group,where 51 species were enriched more than 8 folds.The top three enriched genera were Streptococcus,Prevotella and Campylobacter containing some opportunistic pathogens.More interestingly,through experiments,we found that two Streptococcus strains,S.suis and S.agalactiae,could stimulate the expression of ACE2 of Vero cells in vitro,which may promote SARS-CoV-2 infection.Therefore,these enriched pathogens in the pharynxes of COVID-19 patients may involve in the virus-host interactions to affect SARS-CoV-2 infection and cause potential secondary bacterial infections through changing the expression of the viral receptor ACE2 and/or modulate the host’s immune system. | Dongyan Xiong Caroline Muema Xiaoxu Zhang Xinming Pan Jin Xiong Hang Yang Junping Yu Hongping Wei | 2021 | Virologica Sinica2021,36,5: | 3 |

| 11 | Long- versus short-interval follow-up after resection of hepatocellular carcinoma: a retrospective cohort study显示文摘Background:Average postoperative follow-up intervals vary in patients undergoing hepatocellular carcinoma(HCC)resection because of limited evidence regarding the optimal interval.We aimed to compare the associations of long-versus short-interval follow-up with survival and recurrence in risk-stratified HCC patients.Methods:We performed a retrospective cohort study between 2007 and 2014.In total,1227 patients treated by curative resection of Barcelona Clinic Liver Cancer stage A or B HCC were stratified as having a low(n=865)or high(n=362)risk of early recurrence(within the first 2 years after resection)based on prognostic factors identified by the least absolute shrinkage and selection operation algorithm.Patients were further classified into long-interval(every 4-6 months)and short-interval(every 2-4 months)follow-up subgroups based on follow-up within 2 years after resection(low risk,long vs.short:n=390 vs.n=475;high-risk,long vs.short:n=149 vs.n=213).Results:The short-interval follow-up did not prolong overall survival in either the low-risk(hazard ratio[HR]=1.152;95%confidence interval[CI]0.720-1.843)or high-risk(HR=1.213;95%CI 0.702-2.094)patients.Early recurrence occurred in 401 patients.For high-risk patients,the short-interval follow-up subgroup exhibited smaller intrahepatic recurrence than did the long-interval group(2.6 vs.3.5 cm,respectively,P=0.045).However,no significant difference in the rate of Barcelona Clinic Liver Cancer stage 0/A recurrence was found between the long-and short-interval follow-up groups in either low-or high-risk patients(63.1%vs.68.2%,respectively,P=0.580;31.3%vs.41.5%,respec-tively,P=0.280).The rate of curative intent treatment for recurrence(34.5%vs.39.7%,respectively,P=0.430;14.6%vs.20.3%,respectively,P=0.388)was also similar between the follow-up groups for low-and high-risk patients.Conclusions:Shortening the postoperative follow-up interval from every 4-6 months to every 2-4 months within the first 2 years after resection did not increase the rate of curative intent treatment or prolong the overall survival of patients with Barcelona Clinic Liver Cancer stage A or B HCC. | Wei He Yun Zheng Ruhai Zou Jingxian Shen Junping Yang Jiliang Qiu Qiang Tao Wenwu Liu Zhiwen Yang Yuanping Zhang Binkui Li Yunfei Yuan | 2018 | Cancer Communications2018,38,1: | 2 |

| 16 | Tenofovir disoproxil fumarate therapy in patients with chronic hepatitis B and advanced fibrosis or compensated cirrhosis显示文摘Background and aims:Tenofovir disoproxil fumarate(TDF)is the first-line therapy for chronic hepatitis B.This interim analysis presents the efficacy and safety data for TDF at Week 144 in patients with chronic hepatitis B and advanced fibrosis or compensated cirrhosis from China.Methods:Patients were assessed for incidence of newly diagnosed hepatocellular carcinoma(HCC)and disease progression,liver stiffness measurement(LSM),virological suppression(serum hepatitis B virus DNA<20 IU/mL),alanine aminotransferase normalization,hepatitis B e antigen(HBeAg)loss and seroconversion,histological liver fibrosis score,and safety at Week 144.Results:Overall,197 patients were enrolled.At Week 144,the incidence of newly diagnosed HCC was observed in 2.1%patients,and the incidence of disease progression was observed in 3.6%patients.The mean(standard deviation)change in LSM from baseline was5.1(5.85)kPa.Serum hepatitis B virus DNA<20 IU/mL was observed in 94.1%patients,alanine aminotransferase normalization in 33.5%patients,HBeAg loss in 35.6%patients,and HBeAg seroconversion in 14.4%patients.Among patients with stage F3 or F4 fibrosis at baseline by LSM,38.3%patients regressed to stage F0/1,and 22.0%of patients regressed to stage F2 at Week 144.Overall,67.7%patients experienced≥1 adverse events,13.8%patients experienced TDF-related adverse events,and 16.4%patients experienced serious(none were TDF-related).Conclusions:At Week 144 of TDF treatment,low incidence of HCC and disease progression were reported.Virological suppression was observed in 94.1%patients,which was associated with fibrosis regression.No new safety events were identified. | Huiying Rao Jia Shang Qing Xie Jianqi Lian Pujun Gao Junping Shi Xinyue Chen Jiefei Wang Min Xu Liaoyun Zhang Yingren Zhao QingMao Maorong Wang Wei Zhao Zong Zhang Jidong Jia Hong Tang Jiming Zhang Xin Zheng Chang Liu Lai Wei | 2022 | iLIVER2022,1,3: | 1 |
