1Developing a bacteriophage cocktail for biocontrol of potato bacterial wilt显示文摘Bacterial wilt is a devastating disease of potato and can cause an 80% production loss. To control wilt using bacteriophage therapy, we isolated and characterized twelve lytic bacteriophages from different water sources in Kenya and China. Based on the lytic curves of the phages with the pathogen Ralstonia solanacearum, one optimal bacteriophage cocktail, P1, containing six phage isolations was formulated and used for studying wilt prevention and treatment efficiency in potato plants growing in pots. The preliminary tests showed that the phage cocktail was very effective in preventing potato bacterial wilt by injection of the phages into the plants or decontamination of sterilized soil spiked with R. solanacearum. Eighty percent of potato plants could be protected from the bacterial wilt(caused by R. solanacearum reference strain GIM1.74 and field isolates), and the P1 cocktail could kill 98% of live bacteria spiked in the sterilized soil at one week after spraying.However, the treatment efficiencies of P1 depended on the timing of application of the phages, the susceptibility of the plants to the bacterial wilt, as well as the virulence of the bacteria infected,suggesting that it is important to apply the phage therapy as soon as possible once there are early signs of the bacterial wilt. 2Bacteriophages Isolated in China for the Control of Pectobacterium carotovorum Causing Potato Soft Rot in Kenya显示文摘Soft rot is an economically significant disease in potato and one of the major threats to sustainable potato production.This study aimed at isolating lytic bacteriophages and evaluating methods for and the efficacy of applying phages to control potato soft rot caused by Pectobacterium carotovorum.Eleven bacteriophages isolated from soil and water samples collected in Wuhan,China,were used to infect P.carotovorum host strains isolated from potato tubers showing soft rot symptoms in Nakuru county,Kenya.The efficacy of the phages in controlling soft rot disease was evaluated by applying individual phage strains or a phage cocktail on potato slices and tubers at different time points before or after inoculation with a P.carotovorum strain.The phages could lyse 20 strains of P.carotovorum,but not Pseudomonas fluorescens control strains.Among the 11 phages,Pectobacterium phage Wc5r,interestingly showed cross-activity against Pectobacterium atrosepticum and two phage-resistant P.carotovorum strains.Potato slice assays showed that the phage concentration and timing of application are crucial factors for effective soft rot control.Phage cocktail applied at a concentration of 1×10^9 plaque-forming units per milliliter before or within an hour after bacterial inoculation on potato slices,resulted in>90%reduction of soft rot symptoms.This study provides a basis for the development and application of phages to reduce the impact of potato soft rot disease.Peter Muturi Junping Yu Alice Nyambura Maina Samuel Kariuki Frantis B.Mwaura Hongping Wei 2019Virologica Sinica2019,34,3:4
3Clinical impact and mechanisms of hepatitis B virus infection concurrent with non-alcoholic fatty liver disease显示文摘Chronic hepatitis B(CHB)virus infection is an important threat to global health despite the administration of vaccines and the use of antiviral treatments.In recent years,as the prevalence of obesity and metabolic syndrome has increased,non-alcoholic fatty liver disease(NAFLD)in patients with CHB has become more common.Both diseases can lead to liver fibrosis and even hepatocellular carcinoma,but the risk of dual etiology,outcome,and CHB combined with NAFLD is not fully elucidated.In this review,we assess the overlapping prevalence of NAFLD and CHB,summarize recent studies of clinical and basic research related to potential interactions,and evaluate the progressive changes of treatments for CHB patients with NAFLD.This review increases the understanding of the relationship and mechanisms of interaction between steatosis and hepatitis B virus infection,and it provides new strategies for the future clinical management and treatment of CHB combined with NAFLD.Xin Tong Yu Song Shengxia Yin Jian Wang Rui Huang Chao Wu Junping Shi Jie Li 2022Chinese Medical Journal2022,,14:4

5Enriched Opportunistic Pathogens Revealed by Metagenomic Sequencing Hint Potential Linkages between Pharyngeal Microbiota and COVID-19显示文摘As a respiratory tract virus,SARS-CoV-2 infected people through contacting with the upper respiratory tract first.Previous studies indicated that microbiota could modulate immune response against pathogen infection.In the present study,we performed metagenomic sequencing of pharyngeal swabs from eleven patients with COVID-19 and eleven Non-COVID-19 patients who had similar symptoms such as fever and cough.Through metagenomic analysis of the above two groups and a healthy group from the public data,there are 6502 species identified in the samples.Specifically,the Pielou index indicated a lower evenness of the microbiota in the COVID-19 group than that in the Non-COVID-19 group.Combined with the linear discriminant analysis(LDA)and the generalized linear model,eighty-one bacterial species were found with increased abundance in the COVID-19 group,where 51 species were enriched more than 8 folds.The top three enriched genera were Streptococcus,Prevotella and Campylobacter containing some opportunistic pathogens.More interestingly,through experiments,we found that two Streptococcus strains,S.suis and S.agalactiae,could stimulate the expression of ACE2 of Vero cells in vitro,which may promote SARS-CoV-2 infection.Therefore,these enriched pathogens in the pharynxes of COVID-19 patients may involve in the virus-host interactions to affect SARS-CoV-2 infection and cause potential secondary bacterial infections through changing the expression of the viral receptor ACE2 and/or modulate the host’s immune system.Dongyan Xiong Caroline Muema Xiaoxu Zhang Xinming Pan Jin Xiong Hang Yang Junping Yu Hongping Wei 2021Virologica Sinica2021,36,5:3

7Synthesis and characterization of super paramagnetic composite photocatalyst-Titania/silica/nickel ferrite显示文摘Composite photocatalysts with TiO2 porous shell and magnetic core were synthesized by using NiFe2O4 as a magnetic core, SiO2 as an intermediate layer and polyethylene glycol with molecular weight of 1000 (PEG 1000) as a template direction agent. The samples were characterized with N 2 adsorption-desorption, X-ray diffraction (XRD), transmission electron microscopy (TEM), scanning electron microscopy (SEM), vibrating sample magnetometry (VSM) and ultraviolet-visible (UV-vis) spectroscopy. The results showed that the obtained composites are spherical particles about 50 nm in diameter. The mesoporous TiO 2 with magnetic core calcined at 350 C, 450 C, and 550 C was found to have, respectively, different specific surface areas (94, 86, and 68 m2 /g) and average pore sizes (1.4, 2.2, and 2.8 nm). The photocatalytic activity was investigated for degradation of nitrobenzene under UV irradiation. Nitrobenzene was decomposed in 300 min under irradiation of an 8 W UV lamp with a peak wavelength of 253.7 nm. There is also widespread interest in the development of thrombolytic agents that can be used in cases of delayed presentation. 10An efficient CRISPR/Cas9 platform for targeted genome editing in rose(Rosa hybrida)显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-related nuclease 9(Cas9)system enables precise,simple editing of genes in many animals and plants.However,this system has not been applied to rose(Rosa hybrida)due to the genomic complexity and lack of an efficient transformation technology for this plant.Here,we established a platform for screening single-guide RNAs(sgRNAs)with high editing efficiency for CRISPR/Cas9-mediated gene editing in rose using suspension cells.We used the Arabidopsis thaliana U6-29 promoter,which showed high activity for driving sgRNA expression,to modify the CRISPR/Cas9 system.We used our highly efficient optimized CRISPR/Cas9 system to successfully edit RhEIN2,encoding an indispensable component of the ethylene signaling pathway,resulting in ethylene insensitivity in rose.Our optimized CRISPR/Cas9 system provides a powerful toolbox for functional genomics,molecular breeding,and synthetic biology in rose.Chengpeng Wang Yang Li Na Wang Qin Yu Yonghong Li Junping Gao Xiaofeng Zhou Nan Ma 2023Journal of Integrative Plant Biology2023,65,4:1

15First-principles investigation of the impurity-kink interaction in bcc iron显示文摘Using the first-principles self-consistent discrete variational method based on density functional theory, we investigated the effect of light impurities C and N on the electronic structure of kink on the [100](010) edge dislocation (ED) in bcc iron. Our energetic calculations show that the light impurities have a strong segregation tendency to enter the kink. The results of the charge distribution and the local den-sity of states indicate that the strong bonds between the impurity atoms and the neighboring Fe atoms are formed due to the hybridizations of impurity atoms 2p states and Fe 3d4s4p states. The introduction of light impurities can stabilize the kink system, and impedes the sideward motion of the kink in the [100](010) ED. 17Improving BDS broadcast ephemeris accuracy using ground-satellite-link observations显示文摘The BeiDou Navigation Satellite System(BDS)employs a hybrid constellation including GEO(Geosynchronous Earth Orbit),IGSO(Inclined Geosynchronous Orbit),and MEO(Medium Earth Orbit)satellites,where the GEO and IGSO satellites are critical to providing continuous and reliable Positioning,Navigation,and Timing(PNT)services in the Asia–Pacifc region.To handle the inconsistency between the satellite orbits and clocks in the broadcast ephemeris,which are determined by the Orbit Determination and Time Synchronization(ODTS)and the Two-way Satellite Time Frequency Transfer(TWSTFT)technique,respectively,we present the strategies using ground-satellite-link observations to improve the accuracy of broadcast ephemeris.The clock diferences between the ODTS and TWSTFT techniques are used for correcting the radial orbit component to derive the refned orbits,which are used to generate the refned broadcast ephemeris.The test results show the precision of the refned orbits is improved by 50–60%in the 3-h to 12-h predicted arcs for the GEO satellites,and by 40–50%for the IGSO satellites.Moreover,the validation using satellite laser ranging observations shows the mean precision of the refned broadcast ephemeris is improved by 27%compared to the original one.Applying the proposed strategies in the BDS Operational Control Segment(OCS),the time evolution of BDS Single Point Positioning(SPP)in the period from Jan.2016 to April 2021 is evaluated.The SPP accuracy is improved from 1.94,2.06 and 3.29 m to 1.39,1.85,and 2.39 m in the north,east,and up components,respectively.Further update with the inclusion of BDS-3 satellites improve the corresponding SPP precision to 0.68,0.70 and 1.91 m.Junping Chen Jungang Wang Chao Yu Yize Zhang Bin Wang 2022Satellite Navigation2022,3,1:1
