1SPECIES DIVERSITY AND COMMUNITY STRUCTURE OF SUCKING LICE IN YUNNAN, CHINA显示文摘On the basis of investigating 9 counties (towns) in Yunnan Province of China, the species diversity and community structure of sucking lice on the body surface of small mammal hosts are studied in the paper. Species richness (S) is used to stand for the species diversity. The calculation of community diversity index and evenness are based on Shannon-Wiener's method. 2745 small mammals captured from the investigated sites belong to 10 families, 25 genera and 41 species in 5 orders (Rodentia, Insectivora, Scandentia, Logomorpha and Carnivora) while 18165 individuals of sucking lice collected from the body surface of the small mammal hosts are identified into 4 families, 6 genera and 22 species. The species of sucking lice are much less than the species of their hosts. Most species of small mammals have their fixed sucking lice on their body surface. One species of small mammals usually have few species of sucking lice (1 to 4 species). The close species of the hosts in the taxonomy are found to have the same or similar dominant species of sucking lice on their body surface. The results reveal that the species diversity of sucking lice on small mammals is very low with a very simple community structure. 2Herpes simplex vires thymidine kinase and ganciclovir suicide gene therapy for human pancreatic cancer显示文摘AIM:To investigate the in vitro effects of suicide gene therapy system of herpes simplex virus thymidine kinase gene (HSV-TK) in combination with the treatment of nucleotide analog-ganciclovir (GCV) on human pancreatic cancer, and to provide a novel clinical therapeutic method for human pancreatic cancer.METHODS: We used a replication defective recombinant retrovirus vector GINaTK (bearing HSV-TK gene) to make packaging cell PA317 produce progeny virions. We then transferred the HSV-TK gene to target cells SW1990 using these progeny virions, and treated these gene-modified tumor cells with GCV to study the sensitivity of the cells to GCV and their bystander effects by routine MTT-method.RESULTS:Packaging cell PA317/TK was successfully constructed, and we acquired SW1990/TK through virus progeny infection. These gene-modified pancreatic cancer cells were sensitive to the treatment of GCV compared with unmodified tumor cells (t=4.15,n=10,P<0.0025). 3Incidence and treatment of hepatic artery complications after orthotopic liver transplantation显示文摘AIM: To investigate the incidence and treatment of hepatic artery complications after orthotopic liver transplantation.METHODS: From February 1999 to May 2002, orthotopic liver transplantations (OLT) were performed in 72 patients with end-stage liver diseases with an average age of 40.2±13.6years (ranged from 11 to 68 years), 56 were males and 16 females. The preoperative evaluation for the 72 patients was performed using duplexsonography, abdominal CT scan,and angiography of the hepatic artery. All donor grafts were perfused and preserved in University of Wisconsin solution at 4 ℃. OLT was performed with standard techniques with or without a veno-venous bypass. Reconstructions of hepatic artery were performed between the branch patches of gastroduodenal/hepatic or splenic/common hepatic artery confluence of the donors and recipients, and an end-to-end anastomosis between other arterial vessels of the donors and recipients was done. Arterial anastomosis was performed with interrupted 7-0/8-0 monofilament polypropylene suture under 3.5 x loupe magnification. Diagnosis of the complications of hepatic artery after OLT was based on the clinical presentations, ultrasound findings and arterial angiography. All patients were followed up regularly for duplex ultrasound scan after discharge.RESULTS: The overall incidence of arterial complications in 72 patients after OLTs was 1.4 % (1/72). One 3 cm pseudoaneurysm at the side of anastomotic site of hepatic artery was found by urgent arteriogram due to hemoperitoneum secondary to bile leakage after OLT.Subsequently the pseudoaneurysm was successfully embolized and the blood flow toward the donor liver in hepatic artery remained. The overall postoperative 30-day mortality rate was 8.33 %. The one-year survival rate was 83.72 % in 50 patients with benign diseases and was 71.43 % in 22 patients with malignant diseases following OLT. 4The Structural Characterization and Antigenicity of the S Protein of SARS-CoV显示文摘The corona-like spikes or peplomers on the surface of the virion under electronicmicroscope are the most striking features of coronaviruses. The S (spike) proteinis the largest structural protein, with 1,255 amino acids, in the viral genome. Itsstructure can be divided into three regions: a long N-terminal region in the exte-rior, a characteristic transmembrane (TM) region, and a short C-terminus in theinterior of a virion. We detected fifteen substitutions of nucleotides by comparisonswith the seventeen published SARS-CoV genome sequences, eight (53.3%) of whichare non-synonymous mutations leading to amino acid alternations with predictedphysiochemical changes. The possible antigenic determinants of the S protein arepredicted, and the result is confirmed by ELISA (enzyme-linked immunosorbentassay) with synthesized peptides. Another profound finding is that three disulfidebonds are defined at the C-terminus with the N-terminus of the E (envelope) pro-tein, based on the typical sequence and positions, thus establishing the structuralconnection with these two important structural proteins, if confirmed. 5Trithiocyanuric acid derived g-C3N4 for anchoring the polysulfide in Li-S batteries application显示文摘Lithium-sulfur (Li-S) batteries have great potential as an electrochemical energy storage system because of the high theoretical energy density and acceptable cost of financial and environment.However,the shuttle effect leads to severe capacity fading and low coulombic efficiency.Here,graphitic carbon nitride(g-C3N4) is designed and prepared via a feasible and simple method from trithiocyanuric acid (TTCA) to anchor the polysulfides and suppress the shuttle effect.The obtained g-C3N4 exhibits strong chemical interaction with polysulfides due to its high N-doping of 56.87 at%,which is beneficial to improve the cycling stability of Li-S batteries.Moreover,the novel porous framework and high specific surface area of g-C3N4 also provide fast ion transport and broad reaction interface of sulfur cathode,facilitating high capacity output and superior rate performance of Li-S batteries.As a result,Li-S batteries assembled with g-C3N4 can achieve high discharge capacity of 1200 mAh/g at 0.2 C and over 800 mAh/g is remained after 100 cycles with a coulombic efficiency more than 99.5%.When the C-rate rises to 5 C,the reversible capacity of Li-S batteries can still maintain at 607mAh/g.Ziyang Jia Hongzhang Zhang Ying Yu Yuqing Chen Jingwang Yan Xianfeng Li Huamin Zhang 2020Journal of Energy Chemistry2020,29,4:4
6Novel Constructs of Tuberculosis Gene Vaccine and Its Immune Effect on Mice显示文摘A novel tuberculosis (TB) gene vaccine containing mouse granulocyte macrophage-colony stimulating factor (mGM-CSF) and a TB antigen (Ag85A) was developed in this study. The genes encoding Ag85A and mGM-CSF were amplified by PCR respectively from the Ag85A-containing pBSby5 and pC-mGM-CSF. The genes were then cloned into two different polylinker sites of plasmid pIRES, forming a novel TB gene vaccine construct pI85AGM.Following transfection of pI85AGM plasmid into 7721 cell line by LipofectamineTM, the expression of Ag85A and GM-CSF proteins was identified by Western blotting or RT-PCR. Then Balb/c mice were inoculated with the recombinant pI85AGM, pI85A, pIGM or plasmid alone, respectively. The activities of CTL, NK cells and the Ag85A-stimulated proliferation of spleen cells were measured by MTT method. The serum antibody against Ag85A was detected by ELISA. The results showed that the Ag85A and GM-CSF proteins could be expressed in 7721 cell line and the activity of CTLs and the proliferation of spleen cells were significantly increased in the pI85AGM-immunized mice, indicating that the pI85AGM-immunized mice could generate specific immune responses to Ag85A. 7REAL-TIME THREE-DIMENSIONAL ECHOCARDIOGRAPHY FOR QUANTIFYING LEFT VENTRICULAR MASS显示文摘To test the accuracy of real-time three-dimensional echocardiography (RT3DE) imaging system for evaluating left ventricular mass (LVM) in phantom and excised canine heart. Methods Ten left ventricular (LV) wall phantoms made of two rubber-bursas, ten excised canine hearts underwent RT3DE and two-dimensional echocardiography (2DE). In RT3DE 'full volume' imaging, the myocardial volume was mea-sured using 2,4,8, and 16-plane method with the analysis software of RT3DE. Mass was then calculated by multiplying the resulting myocardial volume by specific density of myocardial tissue. In 2DE the masses were measured by area-length meth-od. The true LV wall phantom mass was measured by water displacement and the canine LVM was weighed by anatomy, which served as a reference standard. We compared RT3DE or 2DE with true mass. Results In LV wall phantoms, RT3DE correlated with true masses strongly (r = 0.813-0.994) and weakly correlated between 2DE and true masses (r = 0.628). In excised canine hearts, there is an excellent correlation between RT3DE and true masses (r = 0.764-0.991), while 2DE value showed a lesser correlation (r = 0.514). There are no difference between RT-3DE and true masses (P > 0.05) but different between 2DE and true masses (P < 0.05). In different planes, there was no difference between 8-plane and 16-plane (P > 0.05) but different between 8-plane and 2, 4-plane (P < 0.05). Conclusion RT3DE can accurately quantify LVM and provide a new tool to evaluate LV function. 8Validation of a simple automated movement detection system for tormalin test in rats显示文摘Aim: To investigate the validity and sensitivity of an automatic movement detection system developed by our laboratory for the formalin test in rats. Methods:The effects of systemic morphine and local anesthetic lidocaine on the nociceptive behaviors induced by formalin subcutaneously injected into the hindpaw were examined by using an automated movement detection system and manual measuring methods. Results: Formalin subcutaneously injected into the hindpaw produced typical biphasic nociceptive behaviors (agitation). The mean agitation event rate during a 60-min observation period increased linearly following increases in the formalin concentration (0.0%, 0.5%, 1.5%, 2.5%, and 5%, 50 μL). Systemic application of morphine of different doses (1, 2, and 5 mg/kg) 10-min prior to formalin injection depressed the agitation responses induced by formalin injection in a dose-dependent manner, and the antinociceptive effect induced by the largest dose (5 mg/kg) of morphine was significantly antagonized by systemic application of the opioid receptor antagonist naloxone (1.25 mg/kg). Local anesthetic lidocaine (20 mg/kg) injected into the ipsilateral ankle subskin 5-min prior to formalin completely blocked the agitation response to formalin injection. These results were comparable to those obtained from manual measure of the incidence of flinching or the duration time of licking/biting of the injected paw. 9FT-IR spectroscopic analysis of normal and cancerous tissues of esophagus显示文摘AIM: To investigate the special Fourier transform infraredspectroscopy (FT-IR) spectra in normal and cancerous tissuesof esophagus.METHODS: Twenty-seven pairs of normal and canceroustissues of esophagus were studied by using FT-IR and thespecial spectra characteristics were analyzed in differenttissues.RESULTS: Different spectra were found in normal andcancerous tissues. The peak at 1 550/cm was weak andwide in cancerous tissues but strong and high in normaltissues.The ratio of I1 647/I1 550 was 2.0 in normal tissuesand 2.36 in cancerous tissues (P<0.05). The ratio of Ⅰ1 550/I 1 080 was 4.5 in normal tissues and 3.4 in canceroustissues (P<0.01). 12Effect of the pore length and orientation upon the electrochemical capacitive performance of ordered mesoporous carbons显示文摘By utilizing hard template method to adjust the mesopore length, and alkali activation to generate micro pores, two hierarchical porous carbons(HPCs) were prepared. With controlling of their mesopore length and the activation conditions, the complex system composed by HPCs and electrolyte was simplified and the effect of mesopore length on the performance of HPCs as electrodes in supercapacitors was investi gated. It is found that with the mesopore length getting smaller, the ordered area gets smaller and th aggregation occurs, which is caused by the high surface energy of small grains. HPC with long pore(HPCL) exhibits a donut-like morphology with well-defined ordered mesopores and a regular orientation while in HPC with short pores(HPCS), short mesopores are only orderly distributed in small regions Longer ordered channels form unobstructed ways for ions transport in the particles while shorter chan nels, only orderly distributed in small areas, results in blocked paths, which may hinder the electrolyt ions transport. Due to the unobstructed structure, HPCL exhibits good rate capability with a capacitanc retention rate over 86% as current density increasing from 50 m A/g to 1000 m A/g. 13The E Protein Is a Multifunctional Membrane Protein of SARS-CoV显示文摘The E (envelope) protein is the smallest structural protein in all coronaviruses andis the only viral structural protein in which no variation has been detected. Weconducted genome sequencing and phylogenetic analyses of SARS-CoV. Based ongenome sequencing, we predicted the E protein is a transmembrane (TM) pro-tein characterized by a TM region with strong hydrophobicity and α-helix con-formation. We identified a segment (NH2-_L-Cys-A-Y-Cys-Cys-N_-COOH) in thecarboxyl-terminal region of the E protein that appears to form three disulfide bondswith another segment of corresponding cysteines in the carboxyl-terminus of the S(spike) protein. These bonds point to a possible structural association between theE and S proteins. 17Inhibition effect of Chinese herbal medicine on transcription of hepatitis C virus structural gene in vitro显示文摘AIM: To investigate the inhibitory effect of Chinese herbal medicine on the transcription of hepatitis C virus (HCV) structural gene in Hela D cells.METHODS: Hela cell line was transfected with recombinant pBK-CMV-HCV containing HCV structural gene by Lipofectamine. RT-nested-PCR and Western blot assay were used to testify the HCV gene expression in Hela cells. The Hela cells expressing HCV structural protein were named Hela D cells. Prescriptions of Xiao chaihu Decoction (XCHD),Fufang Huangqi (FFHQ) and Bingganling (BGL) wererespectively added to Hela D cells in various concentrations. Semi-quantitative RT-nested-PCR product analysis was performed according to the fluorescent density between HCV DNA band and GAPDH DNA band in gel electrophoresisafter screened. RESULTS: Recombinant pBK-CMV-HCV could correctly express the HCV structural gene in Hela D cells. After coculture of Hela D cells with three prescriptional different concentrations for 48 h respectively, the transcription of HCVgene decreased with increasing of the concentration of each prescription. The lightness ratio of HCV product bands to GAPDH product bands was 0.24, 0.10 and 0.12 in Hela D cells incubated with 0.1 g/mL of XCHD, FFHQand BGL respectively and the lightness ratio HCV product bands to GAPDH product bands was 0.75, 0.67 and 0.61respectively in the control cells. 20Complete Genome Sequences of the SARS-CoV: the BJ Group (Isolates BJ01-BJ04)显示文摘Beijing has been one of the epicenters attacked most severely by the SARS-CoV (severe acute respiratory syndrome-associated coronavirus) since the first patient was diagnosed in one of the city's hospitals. We now report complete genome sequences of the BJ Group, including four isolates (Isolates B J01, B J02, B J03,and B J04) of the SARS-CoV. It is remarkable that all members of the BJ Group share a common haplotype, consisting of seven loci that differentiate the group from other isolates published to date. Among 42 substitutions uniquely identifled from the BJ group, 32 are non-synonymous changes at the amino acid level.Rooted phylogenetic trees, proposed on the basis of haplotypes and other sequence variations of SARS-CoV isolates from Canada, USA, Singapore, and China, gave rise to different paradigms but positioned the BJ Group, together with the newly discovered GD01 (GD-Ins29) in the same clade, followed by the H-U Group (from Hong Kong to USA) and the H-T Group (from Hong Kong to Toronto), leaving the SP Group (Singapore) more distant.
