| 1 | Ketamine suppresses intestinal NF-kappa B activation and proinflammatory cytokine in endotoxic rats显示文摘AIM: To investigate the protective effect of ketamine on the endotoxin-induced proinflammatory cytokines and NFkappa B activation in the intestine. ETHODS: Adult male Wistar rats were randomly divided into 6 groups: (a) normal saline control, (b) challenged with endotoxin (5 mg/kg) and treated by saline, (c) challenged with endotoxin (5 mg/kg) and treated by ketamine (0.5 mg/kg), (d) challenged with endotoxin (5 mg/kg) and treated by ketamine (5 mg/kg ), (e) challenged with endotoxin (5 mg/kg) and treated by ketamine (50 mg/kg), and (f) saline injected and treated by ketamine (50 mg/kg). After 1, 4 or 6 h, TNF-α and IL-6 mRNA were investigated in the tissues of the intestine (jejunum) by RT-PCR. TNF-α and IL-6 were measured by ELISA. We used electrophoretic mobility shift assay (EMSA) to investigate NF-kappa B activity in the intestine. RESULTS: NF-kappa B activity, the expression of TNF-α and IL-6 were enhanced in the intestine by endotoxin. Ketamine at a dose of 0.5 mg/kg could suppress endotoxininduced TNF-α mRNA and protein elevation and inhibit NFkappa B activation in the intestine. However the least dosage of ketamine to inhibit IL-6 was 5 mg/kg in our experiment. CONCLUSION: Ketamine can suppress endotoxin-induced production of proinflammatory cytokines such as TNF-α and IL-6 production in the intestine. | 3 | Synthesis of Branched Chains with Actuation Redundancy for Eliminating Interior Singularities of 3T1R Parallel Mechanisms显示文摘Although it is common to eliminate the singularity of parallel mechanism by adding the branched chain with actuation redundancy, there is no theory and method for the configuration synthesis of the branched chain with actuation redundancy in parallel mechanism. Branched chains with actuation redundancy are synthesized for eliminating interior singularity of 3-translational and 1-rotational(3T1R) parallel mechanisms. Guided by the discriminance method of hybrid screw group according to Grassmann line geometry, all the possibilities are listed for the occurrence of interior singularities in 3T1R parallel mechanism. Based on the linear relevance of screw system and the principles of eliminating parallel mechanism singularity with actuation redundancy, different types of branched chains with actuation redundancy are synthesized systematically to indicate the layout and the number of the branched chainsinterior with actuation redundancy. | 4 | Submarine Landslides on the North Continental Slope of the South China Sea显示文摘Recent and paleo-submarine landslides are widely distributed within strata in deep-water areas along continental slopes, uplifts, and carbonate platforms on the north continental margin of the South China Sea(SCS). In this paper, high-resolution 3D seismic data and multibeam data based on seismic sedimentology and geomorphology are employed to assist in identifying submarine landslides. In addition, deposition models are proposed that are based on specific geological structures and features, and which illustrate the local stress field over entire submarine landslides in deep-water areas of the SCS. The SCS is one of the largest fluvial sediment sinks in enclosed or semi-enclosed marginal seas worldwide. It therefore provides a set of preconditions for the formation of submarine landslides, including rapid sediment accumulation, formation of gas hydrates, and fluid overpressure. A new concept involving temporal and spatial analyses is tested to construct a relationship between submarine landslides and different time scale trigger mechanisms, and three mechanisms are discussed in the context of spatial scale and temporal frequency: evolution of slope gradient and overpressure, global environmental changes, and tectonic events. Submarine landslides that are triggered by tectonic events are the largest but occur less frequently, while submarine landslides triggered by the combination of slope gradient and overpressure evolution are the smallest but most frequently occurring events. | 5 | Inhibitory effect of adeno-associated virus-mediated gene transfer of human endostatin on hepatocellular carcinoma显示文摘AIM: To investigate the effect of adeno-associated virusmediated gene transfer of human endostatin on the growth of hepatocellular carcinoma (HCC).METHODS: HCC cell line Hep3B was infected with recombinantadeno-associated virus containing human endostatin gene (rAAV2-hEndo). The results of transfection were detected by RT-PCR and SDS-PAGE assay. MTT assay was used to observe the effects of supernatant of transfected cells on ECV304 cell proliferation. An animal model of HCC was established by injecting Hep3B cells subcutaneously into the back of nude mice. Intratumoral injection of rAAV2hEndo, empty virus and phosphate-buffered saline were given sequentially. Serum endostatin was determined byELISA, the inhibitory effect of endostatin on the growth of xenograft was assessed in 3 wk.RESULTS: The results of RT-PCR and SDS-PAGE assay confirmed that rAAV2-hEndo successfully transfected Hep3B cells, and endostatin was secreted from Hep3B cells to medium. The supernatant of transfected cells markedly inhibited the proliferation of ECV304 cells (P<0.01). Intratumoral injection of rAAV2-hEndo (2×1010v.g.) led to a sustained serum endostatin level ofapproximately (86.71±5.19) ng/mL. | 6 | Radiosensitivity of human colon cancer cell enhanced by immunoliposomal docetaxel显示文摘AIM: To enhance the radiosensitivity of human colon cancer cells by docetaxel.METHODS: Immunoliposomal docetaxel was prepared by coupling monodonal antibody against carcinoembryonic antigen to cyanuric chloride at the PEG terminus of liposome. LoVo adenocarcinoma cell line was treated with immunoliposomal docetaxel or/and irradiation. MTT colorimetric assay was used to estimate cytotoxicity of immunoliposomal docetaxel and radiotoxicity. Cell cycle redistribution and apoptosis were determined with flow cytometry. Survivin expression in LoVo cells was verified by immunohistochemistry. D801 morphologic analysis system was used to semi-quantify immunohistochemical staining of survivin.RESULTS: Cytotoxicity was induced by immunoliposomal docetaxel alone in a dose-dependent manner. Immunoliposomal docetaxel yielded a cytotoxicity effect at a low dose of 2 nmol/L. With a single dose irradiation, the relative surviving fraction of LoVo cells showed a dosedependent response, but there were no significant changes as radiation delivered from 4 to 8 Gy. Compared with liposomal docetaxel or single dose irradiation,strongly radiopotentiating effects of immunoliposomal docetaxel on LoVo cells were observed. A low dose of immunoliposomal docetaxel could yield sufficient radiosensitivity. Immunoliposomal docetaxel were achieved both specificity of the conjugated antibody and drug radiosensitization. Combined with radiation,immunoliposomal docetaxel significantly increased the percentage of G2/M cells and induced apoptosis, but significantly decreased the percentage of cells in G2/G1 and S phase by comparison with liposomal docetaxel.Immunohistochemical analysis showed that the brown stained survivin was mainly in cytoplasm of LoVo cells.Semi-quantitative analysis of the survivin immunostaining showed that the expression of survivin in LoVo cells under irradiation with immunoliposomal docetaxel was significantly decreased.CONCLUSION: Immunoliposomal docetaxel is strongly effective for target radiosensitation in LoVo colon carcinoma cells, and may offer the potential to improve local radiotherapy. | Qing-weiwang Hui-LanLǖ Chang-ChengSong HongLiu Cong-GaoXu | 2005 | World Journal of Gastroenterology2005,11,26: | 10 |
| 7 | Effects of Guiyuanfang and autologous transplantation of bone marrow stem cells on rats with liver fibrosis显示文摘AIM: To investigate the therapeutic effects of Guiyuanfang and bone marrow stem cells (BMSCs) on rats with liver fibrosis.METHODS: Liver fibrosis model was induced by carbon tetrachloride, ethanol, high lipid and assessed biochemically and histologically. Liver function and hydroxyproline contents of liver tissue were determined.Serum hyaluronic acid (HA) level and procollagen Ⅲ level were performed by radioimmunoassay. The VG staining was used to evaluate the collagen deposit in the liver.Immunohistochemical SABC methods were used to detect transplanted BMSCs and expression of urokinase plasminogen activator (uPA).RESULTS: Serum transaminase level and liver fibrosis in rats were markedly reduced by Guiyuanfang and BMSCs. HA level and procollagen Ⅲ level were also reduced obviously,compared to model rats (HA: 47.18±10.97 ng/mL,48.96±14.79 ng/mL; PCⅢ: 22.48±5.46 ng/mL, 26.90±3.35ng/mL; P<0.05).Hydroxyproline contents of liver tissue in both BMSCs group and Guiyuanfang group were far lower than that of model group (1 227.2±43.1 μg/g liver tissue, 1390.8±156.3 μg/g liver tissue; P<0.01). After treatment fibrosis scores were also reduced. Both Guiyuanfang and BMSCs could increase the expression of uPA. The transplanted BMSCs could engraft, survive, and proliferate in the liver.CONCLUSION: Guiyuanfang protects against liver fibrosis.Transplanted BMSCs may engraft, survive, and proliferate in the fibrosis livers indefinitely. | 8 | Antigenic and immunogenic changes due to mutation of sgene of HBV显示文摘AIM: To investigate the change of immunological characteristics of HBsAg caused by the mutation at codon 145 of HBsAg using DNA-based immunization.METHODS: Plasmids expressing mutant and wild type envelope antigens were transfected into human hepatocellular carcinoma cells via electrotransformation. The antigenicity of HBsAg was studied with EIA and immunocytochemical staining. Then plasmids were used to immunize 5 C57BL/6 mice. Sera of mice were detected for anti-HBs and anti-preS2with ELISA. RESULTS: The mutant HBsAg could be detected by native antibody in EIA and immunocytochemical study. But the A(450nm) value of the mutant HBsAg in the supernatant was apparently lower than that of the wild-type. Both mutant and native HBsAg expression plasmid could stimulate a strong humoral immune response to HBsAg and preS2 antigen in mice. Protective antibodies against HBsAg elicited by the native HBsAg occurred earlier than that elicited by the mutant HBsAg about one to two weeks. | 9 | Immunocytochemical detection of HoxD9 and Pbx1 homeodomain protein expression in Chinese esophageal squamous cell carcinomas显示文摘AIM: To evaluate the expression pattern of two novel oncofetal antigens, the HoxD9 and Pbx1 homeoproteins in esophageal squamous cell carcinomas (ESCCs) to determine what role they would play in the carcinogenesis of ESCC.METHODS: We obtained tissue samples of ESCC from 56 patients who underwent esophagectomy but not preoperative chemotherapy or radiotherapy. The diagnosis of ESCC was established and confirmed by staff pathologists. We used a highly sensitive, indirect,immunocytochemical method to detect HoxD9 and PbX1 proteins. We qualitatively and quantitively evaluated cells that exhibited and staining using a light microscope.RESULTS: In all observed carcinoma tissue samples, more than 60% of neoplastic cells stained lightly or strongly for HoxD9, and more than 50% of neoplastic cells stained lightly or strongly for Pbx1.CONCLUSION: Our data suggest that HoxD9 and Pbx1 are inappropriately expressed in most human esophageal squamous cell carcinoma. | 10 | Research of a novel biodegradable surgical staple made of high purity magnesium显示文摘Surgical staples made of pure titanium and titanium alloys are widely used in gastrointestinal anastomosis.However the Ti staple cannot be absorbed in human body and produce artifacts on computed tomography(CT)and other imaging examination,and cause the risk of incorrect diagnosis.The bioabsorbable staple made from polymers that can degrade in human body environment,is an alternative.In the present study,biodegradable high purity magnesium staples were developed for gastric anastomosis.U-shape staples with two different interior angles,namely original 90and modified 100,were designed.Finite element analysis(FEA)showed that the residual stress concentrated on the arc part when the original staple was closed to B-shape,while it concentrated on the feet for the modified staple after closure.The in vitro tests indicated that the arc part of the original staple ruptured firstly after 7 days immersion,whereas the modified one kept intact,demonstrating residual stress greatly affected the corrosion behavior of the HP-Mg staples.The in vivo implantation showed good biocompatibility of the modified Mg staples,without inflammatory reaction 9 weeks post-operation.The Mg staples kept good closure to the Anastomosis,no leaking and bleeding were found,and the staples exhibited no fracture or severe corrosion cracks during the degradation. | Hongliu Wu Changli Zhao Jiahua Ni Shaoxiang Zhang Jingyi Liu Jun Yan Yigang Chen Xiaonong Zhang | 2016 | Bioactive Materials2016,1,2: | 3 |
| 11 | An investigation on a novel PDLC film's fabrication and its electro-optical properties显示文摘A polymer dispersed liquid crystal (PDLC) film that has good electro-optical properties is produced by the method of polymerized-induced phase separation. Based on the application foreground, its capability parameters, such as contrast ratio, work voltage, and visual angle, are characterized for the first time by a white light but not a fixed wavelength light. The results show the PDLC film has a low work-voltage of 20 V, more than 150° visual angle, high stability, and long lifetime. The differences between plastic and glass ITO-coated substrates of PDLC films are also studied in this paper. The plastic substrate has better property and will have a wider perspective especially in the portable, tender and folded display devices. | 12 | Thl Bias in PBMC Induced by Large Scale Auto-CIK Infusion in Malignant Solid Tumor Patients in China显示文摘OBJECTIVE This article is to verify feasibility and validity of autologous cytokine-induced killer cell (Auto-CIK) treatment in solid malignancypatients.METHODS Amplification, phenotypic characteristics, cytokine secretion,antitumor cytotoxicity and clinical response to Auto-CIK derived from 65cases of solid tumor patients with different pathological types and clinicalstages were compared with LAKs in a large-scale clinical trial,RESUL'r$ We found that seriousness of disease and metastatic status hadno influence on effective components and antitumor immunological activityof Auto-ClK. Comparing cytotoxicity against various tumor cells with LAKsat various effector to target ratios, ClKs showed more effective cytotoxicityagainst NK sensitive or non-sensitive solid tumor cell lines at a low E/T ratio(6:1) which suggests indirectly that Auto-CIK had a longer effective time invivo than LAKs. These results suggest that CIKs are more suitable forimmunotherapy for those solid malignancy patients at high risk of relapse orrecurrence.CONCLUSIONS Our experimental data were consistent ~ith the reportedconclusion that the potent antitumor activity of Auto-ClK mainly rooted in theCD4- part of CIKs, including CD3~CD56~ cells and CD8 ~ CTLs. The CD4~part of ClKs seemed to have no direct tumor lytic activity. | 13 | Study of Cell Behaviors on Anodized TiO_2 Nanotube Arrays with Coexisting Multi-Size Diameters显示文摘It has been revealed that the different morphologies of anodized TiO_2 nanotubes, especially nanotube diameters, triggered different cell behaviors. However, the influence of TiO_2 nanotubes with coexisting multi-size diameters on cell behaviors is seldom reported. In this work, coexisting four-diameter TiO_2 nanotube samples, namely,one single substrate with the integration of four different nanotube diameters(60, 150, 250, and 350 nm), were prepared by repeated anodization. The boundaries between two different diameter regions show well-organized structure without obvious difference in height. The adhesion behaviors of MC3T3-E1 cells on the coexisting fourdiameter TiO_2 nanotube arrays were investigated. The results exhibit a significant difference of cell density between smaller diameters(60 and 150 nm) and larger diameters(250 and 350 nm) within 24 h incubation with the coexistence of different diameters, which is totally different from that on the single-diameter TiO_2 nanotube arrays. The coexistence of four different diameters does not change greatly the cell morphologies compared with the singlediameter nanotubes. | 14 | Replication and gene expression of mutant hepatitis B virus in a transgenic mouse containing the complete viral genome with mutant sgene显示文摘AIM:To establish the transgenic mouse line harbouring complete hepatitis B virus (HBV) genome with mutant s gene (adr subtype).METHODS: Transgenic mice were generated by microinjecting HBV genome into fertilized eggs. Integration, expression, replication of HBV gene and histological changes in transgenic mice were estimated by genomic DNA PCR, serum DNA PCR, Southern blot, ELISA, HE staining, immunohistochemistry and transmission electron microscopy. Transgenic mice with HBsAg positive in serum were bred and analyzed.RESULTS: A total of 288 eggs survived from microinjections were transplanted into the oviducts of 13 pseudopregnant mice and 49 pups were produced. Twenty-six mice were identified to have the integrated HBV gene. Serum HBsAg and HBeAg were detected in 2 of 43 mice. HBsAg and HBcAg in cytoplasm or nuclei of hepatocytes were detected in 10 mice. Founders with HBsAg in serum were named lineages G145R-15 and G145R-18. Of the 16 F1 offsprings generated by G145R-15 founder, 12 were positive for HBV genome with PCR, 10 were positive for HBsAg and HBcAg with immunohistochemistry and 7 were positive for HBsAg and HBeAg with ELISA. Only 1 of 8 F1 offsprings generated by G145R-18 founder was survived and it was detected positive for HBV genome, HBsAg, HBcAg and HBeAg. Both of the two lineages had some pathological characteristics of mild chronic hepatitis B in the liver, such as swelling of hepatocytes and focal hepatocellular necrosis and parenchymal lymphomononuclear cell infiltrate.CONCLUSION: Transgenic mice harbouring HBV with mutant s gene can be generated. The HBV genes are integrated in the transgenic mice genome and can be expressed, replicated, packaged and excreted. | 18 | Anti-Seizure and Neuronal Protective Effects of Irisin in Kainic Acid-Induced Chronic Epilepsy Model with Spontaneous Seizures显示文摘An increased level of reactive oxygen species is a key factor in neuronal apoptosis and epileptic seizures.Irisin reportedly attenuates the apoptosis and injury induced by oxidative stress.Therefore,we evaluated the effects of exogenous irisin in a kainic acid(KA)-induced chronic spontaneous epilepsy rat model.The results indicated that exogenous irisin significantly attenuated the KA-induced neuronal injury,learning and memory defects,and seizures.Irisin treatment also increased the levels of brain-derived neurotrophic factor(BDNF)and uncoupling protein 2(UCP2),which were initially reduced following KA administration.Furthermore,the specific inhibitor of UCP2(genipin)was administered to evaluate the possible protective mechanism of irisin.The reduced apoptosis,neurodegeneration,and spontaneous seizures in rats treated with irisin were significantly reversed by genipin administration.Our findings indicated that neuronal injury in KA-induced chronic epilepsy might be related to reduced levels of BDNF and UCP2.Moreover,our results confirmed the inhibition of neuronal injury and epileptic seizures by exogenous irisin.The protective effects of irisin may be mediated through the BDNF-mediated UCP2 level.Our results thus highlight irisin as a valuable therapeutic strategy against neuronal injury and epileptic seizures. | Jie Yu Yao Cheng Yaru Cui Yujie Zhai Wenshen Zhang Mengdi Zhang Wenyu Xin Jia Liang Xiaohong Pan Qiaoyun Wang Hongliu Sun | 2022 | Neuroscience Bulletin2022,38,11: | 1 |
