1Differential regulation of survivin by p53 contributes to cell cycle dependent apoptosis显示文摘Recent studies indicate that cell-cycle checkpoints are tightly correlated with the regulation of apoptosis, in which p53 plays an important role. Our present works show that the expression of E6/E7 oncogenes of human papillomavirus in HeLa cells is inhibited in the presence of anti-tumor reagent tripchlorolide (TC), which results in the up-regulation of p53 in HeLa cells. Interestingly, under the same TC-treatment, the cells at the early S-phase are more susceptible to apoptosis than those at the middle S-phase although p53 protein is stabilized to the same level in both situations. Significant difference is exhibited between the two specified expression profiles. Further analysis demonstrates that anti-apoptotic gene survivin is up-regulated by p53 in the TC-treated middle-S cells, whereas it is down-regulated by p53 in the TC-treated early-S cells. Taken together, the present study indicates that the differential p53-regulated expres- sion of survivin at different stages of the cell cycle results in different cellular outputs under the same apoptosis-inducer.YanJIN YongWEI LeiXIONG YingYANG JiaRuiWU 2005Cell Research2005,15,5:21
2Combined effects of Cantide and chemotherapeutic drugs on inhibition of tumor cells' growth in vitro and in vivo显示文摘AIM: To investigate the combination effect of hTERT antisense oligonucleotide 'Cantide' and three chemotherapeutic drugs (cisplatin, 5-fluorouracil (5-FU) and adriamycin (ADM)) on inhibiting the proliferation of HepG2, BGC and A549 cell lines in vitro, and to investigate the efficacy of Cantide used in combination with cisplatin (DDP) in vivo.METHODS: Cantide was transfected into these tumor cells by Lipofectin, and cell growth activity was calculated by microcytotoxicity assay. In vivo study, cells of HepG2 were implanted in Balb/c nude mice for 4 d. Then Cantide, DDP and Cantide+DDP were given intraperitoneally for 24 d respectively. The body weights of the tumor-bearing animals and their tumor mass were measured later to assess the effect of combination therapy in the nude mice.To evaluate the interaction of Cantide and these chemotherapeutic drugs, SAS software and Jin Zhengjun method were used.RESULTS: Combination treatments with 0.1 μmol/L Cantide reduced the IC50 of DDP, 5-FU and ADM from 1.07, 4.15and 0.29 μg/mL to 0.25, 1.52 and 0.12 μg/mL respectively.The inhibition ability of DDP, 5-FU and ADM respectively in combination with Cantide in these tumor cells was higher than that of these drugs alone (P<0.0001). And synergism (Q≥1.15) was observed at the lower concentration of DDP (≤1 μg/mL), 5-FU (≤10 μg/mL) and ADM (≤0.1 μg/mL)with combination of Cantide. In vivo, combination treatment with Cantide and DDP produced the greater growth inhibition of human liver carcinoma cells HepG2 in nude mice (0.65±0.19 g tumor) compared with that when only one of these drugs was used (Cantide group: 1.05±0.16 g tumor, P= 0.0009<0.001; DDP group: 1.13±0.09 g tumor,P= 0.0001<0.001).CONCLUSION: These findings indicate that Cantide may enhance therapeutic effectiveness of chemotherapeutic drugs over a wide range of tumor cells in vitro, and the combination use of Cantide and DDP can produce much higher inhibition rates, as compared with when either of these drugs was used only in vivo.YingYang Qiu-JunLv Qing-YouDu Bing-HuYang Ru-XianLin Sheng-QiWang 2005World Journal of Gastroenterology2005,11,16:9
3Effective siRNA targets screening for human telomerase reverse transcriptase显示文摘AIM: To study the inhibitory effects of siRNAs targeting different hTERT sequences and to screen the effective siRNA sequence.METHODS: Five double-stranded siRNAs targeting coding and non-coding regions of hTERT gene were designed and synthesized by T7 transcription system in vitro. siRNA4sequence was screened by full length gene targeting technique and the rest of the siRNA sequences were selected randomly. After being purified by ethanol precipitation, the siRNAs were transfected to the human hepatocellular carcinoma cell (HepG2) by Lipofectamine 2000TM. At 48-72 h after siRNAs transfection, MTT assay,RT-PCR and Western-blot were applied to evaluate the effects of siRNAs on cell growth, mRNA and protein expression level of hTERT gene, respectively.RESULTS: Compared to the control cells, the cells treated with the five double-stranded siRNAs exhibited different degrees of inhibition of cell proliferation in a dose-dependent manner. siRNA2 and siRNA4, exhibited obvious effects of inhibiting hTERT mRNA and protein expression in HepG2cells.CONCLUSION: siRNAs targeting different hTERT sequences have significantly various inhibitory effects on hTERT gene expression. The siRNA sequence screened by full length gene targeting technique has comparable inhibitory effect with the rest siRNA sequences screened by random selection, suggesting that siRNAs and antisense oligonucleic acids may have the same effective target sites. 4REAL-TIME THREE-DIMENSIONAL ECHOCARDIOGRAPHY FOR QUANTIFYING LEFT VENTRICULAR MASS显示文摘To test the accuracy of real-time three-dimensional echocardiography (RT3DE) imaging system for evaluating left ventricular mass (LVM) in phantom and excised canine heart. Methods Ten left ventricular (LV) wall phantoms made of two rubber-bursas, ten excised canine hearts underwent RT3DE and two-dimensional echocardiography (2DE). In RT3DE 'full volume' imaging, the myocardial volume was mea-sured using 2,4,8, and 16-plane method with the analysis software of RT3DE. Mass was then calculated by multiplying the resulting myocardial volume by specific density of myocardial tissue. In 2DE the masses were measured by area-length meth-od. The true LV wall phantom mass was measured by water displacement and the canine LVM was weighed by anatomy, which served as a reference standard. We compared RT3DE or 2DE with true mass. Results In LV wall phantoms, RT3DE correlated with true masses strongly (r = 0.813-0.994) and weakly correlated between 2DE and true masses (r = 0.628). In excised canine hearts, there is an excellent correlation between RT3DE and true masses (r = 0.764-0.991), while 2DE value showed a lesser correlation (r = 0.514). There are no difference between RT-3DE and true masses (P > 0.05) but different between 2DE and true masses (P < 0.05). In different planes, there was no difference between 8-plane and 16-plane (P > 0.05) but different between 8-plane and 2, 4-plane (P < 0.05). Conclusion RT3DE can accurately quantify LVM and provide a new tool to evaluate LV function. 5Mathematical Simulation of Lithium Bromide Solution Laminar Falling Film Evaporation in Vertical Tube显示文摘For utilization of the residual heat of flue gas to drive the absorption chillers,a lithium-bromide falling film in vertical tube type generator is presented.A mathematical model was developed to simulate the heat and mass coupled problem of laminar falling film evaporation in vertical tube.In the model,the factor of mass transfer was taken into account in heat transfer performance calculation.The temperature and concentration fields were calculated.Some tests were conducted for the factors such as Re number,heating flux,the inlet concentration and operating pressure which can affect the heat and mass transfer performance in laminar falling film evaporation.The heat transfer performance is enhanced with the increasing of heat flux.An increasing inlet concentration can weaken the heat transfer performance.The operating pressure hardly affects on heat and mass transfer.The bigger inlet Re number means weaker heat transfer effects and stronger mass transfer.The mass transfer obviously restrains the heat transfer in the falling film solution.The relation between dimensionless heat transfer coefficient and the inlet Re number is obtained.Chengming Shi YangWang Huili Hu YingYang 2010Journal of Thermal Science2010,19,3:3
6Anti-tumor Effects of pNEgr-mlL-12 Recombinant Piasmid Induced by X-irradiation and Its Mechanisms显示文摘Objeelive To study the effect of gene radiotherapy combining injection of recombinant plasmid pNEgr-mlL-12 with local X-irradiation on cancer growth and to elucidate the mechanisms of tumor inhibition. Methods Alkaline lysis was used to extract the plasmid and polyethylene glycol 8000 (PEG 8000) was applied for further purification of plasmids. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression of IL-12 protein. C57BL/6J mice were subcutaneously inoculated with B16 melanoma cells and the plasmid was injected directly into the tumor. Gene-radiotherapy combining pNEgr-mlL-12 recombinant plasmid with X-irradiation was given three times to C57BL/6J mice beating B16 melanoma. Changes in immunologic parameters of tumor-bearing mice were detected with relevant immunologic assays. Results Results showed a significant decrease in tumor growth rate (P<0.05-0.001) after 3 times of gene-radiotherapy with IL-12 and X-irradiation. Immunologic studies showed a significant increase in CTL and NK cytolytic activity (P<0.05-0.001) and an up-regulated secretion of IFN-γ and TNF-α (P<0.01-0.001). Moreover,the expression of mlL-12 in B16 melanoma cells of the treated tumor-beating mice was found to be higher than that of control. 7AN MAC PROTOCOL SUPPORTING MULTIPLE TRAFFIC OVER MOBILE AD HOC NETWORKS显示文摘This letter presents the design and performance of a multi-channel MAC protocol that supports multiple traffics for IEEE 802.11 mobile ad-hoc networks. The dynamic channel selection scheme by receiver decision is implemented and the number of the data channel is independent of the network topology. The priority for real-time traffic is assured by the proposed adaptive back off algorithm and different IFS. 16Secure Performance Analysis and Optimization for FD-NOMA Vehicular Communications显示文摘Vehicle-to-vehicle(V2V)communication appeals to increasing research interest as a result of its applications to provide safety information as well as infotainment services.The increasing demand of transmit rates and various requirements of quality of services(QoS)in vehicular communication scenarios call for the integration of V2V communication systems and potential techniques in the future wireless communications,such as full duplex(FD)and non-orthogonal multiple access(NOMA)which enhance spectral efficiency and provide massive connectivity.However,the large amount of data transmission and user connectivity give rise to the concern of security issues and personal privacy.In order to analyze the security performance of V2V communications,we introduce a cooperative NOMA V2V system model with an FD relay.This paper focuses on the security performance of the FD-NOMA based V2V system on the physical layer perspective.We first derive several analytical results of the ergodic secrecy capacity.Then,we propose a secrecy sum rate optimization scheme utilizing the instantaneous channel state information(CSI),which is formulated as a non-convex optimization problem.Based on the differential structure of the non-convex constraints,the original problem is approximated and solved by a series of convex optimization problems.Simulation results validate the analytical results and the effectiveness of the secrecy sum rate optimization algorithm.Lai Wei Yingyang Chen Dongsheng Zheng Bingli Jiao 2020China Communications2020,17,11:0
17Diagnostic accuracy and safety of Dermatophagoides pteronyssinus extracts used for skin prick test显示文摘Background:Dermatophagoides pteronyssinus is a common allergen causing allergic diseases in China.The aim of this study was to evaluate the efficacy and safety of D.pteronyssinus extracts produced by Peking Union Medical College Hospital(PUMCH)for the skin prick test(SPT)in the diagnosis of D.pteronyssinus allergy.Methods:A total of 910 subjects with allergic diseases were prescribed D.pteronyssinus SPT and specific sIgE(sIgE)test among the Outpatients of Department of Allergy,PUMCH from August 10,2015 to August 30,2017.Receiver operating characteristic curve(ROC)analysis was performed according to the results of D.pteronyssinus-sIgE detection.The accuracy of D.pteronyssinus extracts used for SPT in the diagnosis of D.pteronyssinus allergy was evaluated under different cutoff values.Adverse events after SPT were recorded to evaluate safety.Results:There were 796 and 618 subjects in the full analysis set(FAS)and the per protocol set(PPS),respectively.The areas under the curve of FAS and PPS were 0.871 and 0.873,respectively.According to the ROC of PPS,the optimal and 95%specificity diagnostic cutoff values of D.pteronyssinus SPT mean wheal diameter were 3.25 and 3.75 mm,respectively.No adverse events occurred.Conclusion:The extracts of D.pteronyssinus for SPT were simple,highly accurate,and safe and should be considered for recommendation in the clinical diagnosis of D.pteronyssinus allergy.Rui Tang Xiaohong Lyu Yuxi Liu Ruiqi Wang Lianglu Wang Hong Li Jinlyu Sun Yuxiang Zhi Jianqing Gu Kai Guan Liping Wen Zixi Wang Lisha Li Le Cui Yingyang Xu Junxiong Zhou Tao Xu Jia Yin 2022Chinese Medical Journal2022,135,21:0
