2One ion to catch them all: Targeted high-precision Boltzmann thermometry over a wide temperature range with Gd^(3+)显示文摘Ratiometric luminescence thermometry with trivalent lanthanide ions and their 4f^(n) energy levels is an emerging technique for non-invasive remote temperature sensing with high spatial and temporal resolution.Conventional ratiometric luminescence thermometry often relies on thermal coupling between two closely lying energy levels governed by Boltzmann’s law.Despite its simplicity,Boltzmann thermometry with two excited levels allows precise temperature sensing,but only within a limited temperature range.While low temperatures slow down the nonradiative transitions required to generate a measurable population in the higher excitation level,temperatures that are too high favour equalized populations of the two excited levels,at the expense of low relative thermal sensitivity.In this work,we extend the concept of Boltzmann thermometry to more than two excited levels and provide quantitative guidelines that link the choice of energy gaps between multiple excited states to the performance in different temperature windows.By this approach,it is possible to retain the high relative sensitivity and precision of the temperature measurement over a wide temperature range within the same system.We demonstrate this concept using YAl_(3)(BO_(3))_(4)(YAB):Pr^(3+),Gd^(3+)with an excited 6 PJ crystal field and spin-orbit split levels of Gd^(3+)in the UV range to avoid a thermal black body background even at the highest temperatures.This phosphor is easily excitable with inexpensive and powerful blue LEDs at 450 nm.Zero-background luminescence thermometry is realized by using blue-to-UV energy transfer upconversion with the Pr^(3+)−Gd^(3+)couple upon excitation in the visible range.This method allows us to cover a temperature window between 30 and 800 K.Dechao Yu Huaiyong Li Dawei Zhang Qinyuan Zhang Andries Meijerink Markus Suta 2021Light(Science & Applications)2021,10,12:4
3A novel cancer vaccine with the ability to simultaneously produce anti-PD-1 antibody and GM-CSF in cancer cells and enhance Th1-biased antitumor immunity显示文摘Tumor escape from immune-mediated destruction has been associated with immunosuppressive mechanisms that inhibit T-cell activation.A promising strategy for cancer immunotherapy is to disrupt key pathways regulating immune tolerance,such as program death-1(PD-1/PD-L1)pathway in the tumor environment.However,the determinants of response to anti-PD-1 monoclonal antibodies(mAbs)treatment remain incompletely understood.In murine models,PD-1 blockade alone fails to induce effective immune responses to poorly immunogenic tumors,but is successful when combined with additional interventions,such as cancer vaccines.Novel cancer vaccines combined with antibody may offer promising control of cancer development and progression.In this investigation,we generated a novel tumor cell vaccine simultaneously expressing anti-PD-1 mAbs and granulocyte-macrophage colony stimulating factor(GM-CSF)in CT26 colon cancer and B16-F10 melanoma.The antitumor effect of the vaccine was verified by therapeutic and adoptive animal experiments in vivo.The antitumor mechanism was analyzed using Flow cytometry,Elispot and in vivo intervention approaches.The results showed that tumor cell vaccine secreting PD-1 neutralizing antibodies and GM-CSF induced remarkable antitumor immune effects and prolonged the survival of tumor-bearing animals compared with animals treated with either PD-1 mAbs or GM-CSF alone.Antitumor effects and prolonged survival correlated with strong antigen-specific T-cell responses by analyzing CD11c^(+)CD86^(+)DC,CD11b^(+)F4/80^(+)MΦcells,increased ratio of Teff/Treg in the tumor microenvironment,and higher secretion levels of Th1 proinflammatory cytokines in serum.Furthermore,the results of ELISPOT and in vivo blocking strategies further confirmed that the antitumor immune response is acquired by CD4 and CD8 T immune responses,primarily dependent on CD4 Th1 immune response,not NK innate immune response.The combination of PD-1 blockade with GM-CSF secretion potency creates a novel tumor cell vaccine immunotherapy,affording significantly improved antitumor responses by releasing the state of immunosuppressive microenvironment and augmenting the tumor-reactive T-cell responses.Hongwei Tian Gang Shi Qin Wang Yiming Li Qianmei Yang Chunlei Li Guoyou Yang Min Wu Qian Xie Shuang Zhang Yang Yang Rong Xiang Dechao Yu Yuquan Wei Hongxin Deng 2016Signal Transduction and Targeted Therapy2016,1,1:3
4Understanding and tuning blue-to-near-infrared photon cutting by the Tm^(3+)/Yb^(3+) couple显示文摘Lanthanide-based photon-cutting phosphors absorb high-energy photons and‘cut’them into multiple smaller excitation quanta.These quanta are subsequently emitted,resulting in photon-conversion efficiencies exceeding unity.The photon-cutting process relies on energy transfer between optically active lanthanide ions doped in the phosphor.However,it is not always easy to determine,let alone predict,which energy-transfer mechanisms are operative in a particular phosphor.This makes the identification and design of new promising photon-cutting phosphors difficult.Here we unravel the possibility of using the Tm^(3+)/Yb^(3+) lanthanide couple for photon cutting.We compare the performance of this couple in four different host materials.Cooperative energy transfer from Tm^(3+)to Yb^(3+) would enable blue-to-near-infrared conversion with 200% efficiency.However,we identify phonon-assisted cross-relaxation as the dominant Tm^(3+)-to-Yb^(3+) energy-transfer mechanism in YBO_(3),YAG,and Y_(2)O_(3).In NaYF_(4),in contrast,the low maximum phonon energy renders phonon-assisted cross-relaxation impossible,making the desired cooperative mechanism the dominant energy-transfer pathway.Our work demonstrates that previous claims of high photon-cutting efficiencies obtained with the Tm^(3+)/Yb^(3+) couple must be interpreted with care.Nevertheless,the Tm^(3+)/Yb^(3+) couple is potentially promising,but the host material-more specifically,its maximum phonon energy-has a critical effect on the energy-transfer mechanisms and thereby on the photon-cutting performance.Dechao Yu Ting Yu Arnoldus Jvan Bunningen Qinyuan Zhang Andries Meijerink Freddy T.Rabouw 2020Light(Science & Applications)2020,9,1:2

12Genome-wide identification of cancer-related polyadenylated and non-polyadenylated RNAs in human breast and lung cell lines显示文摘Eukaryotic mRNAs consist of two forms of transcripts:poly(A)+ and poly(A),based on the presence or absence of poly(A) tails at the 3 end.Poly(A)+ mRNAs are mainly protein coding mRNAs,whereas the functions of poly(A) mRNA are largely unknown.Previous studies have shown that a significant proportion of gene transcripts are poly(A) or bimorphic(containing both poly(A)+ and poly(A) transcripts).We compared the expression levels of poly(A) and poly(A)+ RNA mRNAs in normal and cancer cell lines.We also investigated the potential functions of these RNA transcripts using an integrative workflow to explore poly(A)+ and poly(A) transcriptome sequences between a normal human mammary gland cell line(HMEC) and a breast cancer cell line(MCF-7),as well as between a normal human lung cell line(NHLF) and a lung cancer cell line(A549).The data showed that normal and cancer cell lines differentially express these two forms of mRNA.Gene ontology(GO) annotation analyses hinted at the functions of these two groups of transcripts and grouped the differentially expressed genes according to the form of their transcript.The data showed that cell cycle-,apoptosis-,and cell death-related functions corresponded to most of the differentially expressed genes in these two forms of transcripts,which were also associated with the cancers.Furthermore,translational elongation and translation functions were also found for the poly(A) protein-coding genes in cancer cell lines.We demonstrate that poly(A) transcripts play an important role in cancer development.ZHAO GuoGuang JIAO Fei LIAO Qi LUO HaiTao LI Hui SUN Liang BU DeChao YU KunTao ZHAO Yi CHEN RunSheng 2013Science China(Life Sciences)2013,56,6:0
13Pan-cancer analysis of RNA 5-methylcytosine reader (ALYREF)显示文摘The incre asing interest in RNA modifications has signifcantly advanced epigenomic and epitranscriptomic technologies.This study focuses on the immuno oncological impact of ALYREF in human cancer through a pan-cancer analysis,enhancing understanding of this gene's role in cancer.We observed differential ALYREF expression between tumor and normal samples,correl ating strongly with prognosis in various cancers,particularly kidney renal papillary cell carcinoma(KIRP)and liver hepatocellular carcinoma(LIHC).ALYREF showed a negative correlation with most tumor-infitrating cells in lung squamous cell carcinoma(LUSC)and lymphoid neoplasm difuse large B-cell lymphoma(DLBC),while positive correlations were noted in IIHC,kidney chromophobe(KICH),mesothelioma(MESO),KIRP,pheochromocytoma and paraganglioma(PARD),and glioma(GBMLGG).Aditionally,ALYREF expression was closely associated with tumor heterogeneity,stemness indices,and a high mutation rate in TP53 across these cancers.In conclusion,ALYREF may serve as an oncogenic biomarker in numerous cancers,meriting further research attention.XING YE ZHOUTING TUO KAI CHEN RUICHENG WU JIE WANG QINGXIN YU LUXIA YE AKIRA MIYAMOTO KOO HAN YOO CHI ZHANG WURAN WEI DENGXIONG LI DECHAO FENG 2024Oncology Research2024,32,3:0
