| 1 | A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. | 2 | Factors Affecting Sensory Quality of Cooked japonica Rice显示文摘The sensory quality of cooked rice is an important factor in determining its market price, as well as consumer acceptance and breeding efforts aimed at improvement of rice grain quality. In this study,the sensory quality and physicochemical properties of three japonica rice varieties harvested in two different growing locations(Xiangshui and Hangzhou of China) were compared to determine the most important factors affecting the sensory quality. All the three varieties had higher scores for overall sensory quality in Xiangshui than in Hangzhou, indicating that the growing location is a key factor in determining the sensory quality of cooked japonica rice. In addition to growing location, variety(genotype) also had an important effect. Longdao 18 scored the highest for overall sensory quality in the two locations, whereas Longdao 30 had the lowest score in Xiangshui, and both Longdao 20 and Longdao 30 had the lowest scores in Hangzhou. Many physicochemical properties, such as apparent amylose content, protein content, thermal properties and free amino acid contents, showed significant differences between the two locations. Correlation analysis showed that apparent amylose content and protein content had contrasting effects on all the sensory attributes. The overall sensory quality was negatively correlated with protein content(r =-0.89, P < 0.01) and positively correlated with gel hardness(r = 0.91, P < 0.01),indicating that the protein content and hardness are important physicochemical properties for predicting the sensory quality of japonica rice. | 3 | Hyperbaric oxygen therapy improves cognitive functioning after brain injury显示文摘Hyperbaric oxygen therapy has been widely applied and recognized in the treatment of brain injury;however,the correlation between the protective effect of hyperbaric oxygen therapy and changes of metabolites in the brain remains unclear.To investigate the effect and potential mechanism of hyperbaric oxygen therapy on cognitive functioning in rats,we established traumatic brain injury models using Feeney’s free falling method.We treated rat models with hyperbaric oxygen therapy at 0.2 MPa for 60 minutes per day.The Morris water maze test for spatial navigation showed that the average escape latency was significantly prolonged and cognitive function decreased in rats with brain injury.After treatment with hyperbaric oxygen therapy for 1 and 2 weeks,the rats’spatial learning and memory abilities were improved.Hydrogen proton magnetic resonance spectroscopy analysis showed that the N-acetylaspartate/creatine ratio in the hippocampal CA3 region was significantly increased at 1 week,and the N-acetylaspartate/choline ratio was significantly increased at2 weeks after hyperbaric oxygen therapy.Nissl staining and immunohistochemical staining showed that the number of nerve cells and Nissl bodies in the hippocampal CA3 region was significantly increased,and glial fibrillary acidic protein positive cells were decreased after a 2-week hyperbaric oxygen therapy treatment.Our findings indicate that hyperbaric oxygen therapy significantly improves cognitive functioning in rats with traumatic brain injury,and the potential mechanism is mediated by metabolic changes and nerve cell restoration in the hippocampal CA3 region. | Su Liu Guangyu Shen Shukun Deng Xiubin Wang Qinfeng Wu Aisong Guo | 2013 | Neural Regeneration Research2013,8,35: | 16 |

| 7 | ESR Dating of the Evolution of the Shuanghu Basin in the Northern Tibetan Plateau显示文摘The Shuanghu basin is a NE-trending rift basin bounded by NE-striking normal faults and NW-striking shear-extensional faults of the northern Tibetan Plateau. Four samples from calcite veins in marginal faults and one sample from mudstone (S-3) were collected for dating the evolution of the Shuanghu basin by using the ESR spectrograph of EXM-type. Ages were calculated according to the close-equilibrium model on the basis of the measured ESR signal spectra of samples, providing good chronological information. | 11 | 控制气管插管套囊压力与术后并发症关系的多中心研究显示文摘背景与气管插管相关的术后呼吸系统并发症通常表现为咳嗽、咽喉痛、声嘶、痰中带血。该研究中,我们研究了短时间(数小时)测量和控制气管插管套囊(endotracheal tube cuff,ETrc)压力对术后并发症的影响。方法选择中国上海4所大学附属三级医院拟在全身麻醉下行择期手术的患者509例,分为不测量ETTc压力的对照组和测量并调整ETTc压力的研究组,记录手术时间和插管时间。每组选择插管时间在120—180分钟的20例患者,拔管后即刻行纤维支气管镜检查。拔管后24小时记录咳嗽、咽喉痛、声嘶、痰中带血等气管插管相关并发症。结果两组间性别、年龄、身高、体重、手术时间和插管时问均无显著性差异。研究组调整前通过触摸指示气囊估计的平均ETTc压力为43±23.3mmHg(最高210mmHg),调整后为20±3.1mmHg(P〈0.001)。对照组术后咽喉痛、声嘶、痰中带血的发生率明显高于研究组。随着插管时间的增加,对照组中咽喉痛和痰中带血的发生率增加。研究组中咽喉痛的发生率也随插管时间增加而增加。对40例患者的纤维支气管镜检查结果显示支气管黏膜均有不同程度的损伤,对照组的损伤比研究组更严重。结论依据个人经验通过触捏估计ETTc压力明显高于测量结果或最适压力。即使在短时间(1—3小时)手术中,通过压力计合理控制ETTc压也有助于减少E]丌相关的咳嗽、咽喉痛、声嘶、痰中带血等术后呼吸系统并发症。 | Jianhui Liu Xiaoqing Zhang Wei Gong Shitong Li Fen Wang Shukun Fu Mazhong Zhang Yannan Hang 安博(译) 杜金(校) | 2013 | 麻醉与镇痛2013,,1: | 3 |

| 16 | 来源于人膀胱癌细胞株EJ的HSP70负载的DC所激发的特异性CTL细胞的体外应答效应(英文)显示文摘Objective: To investigate whether human dendritic cells (DC) derived from peripheral blood mononuclear cells (PBMC), which were pulsed by heat shock protein 70 (HSP70) isolated from human bladder tumor cell lines of EJ, were able to induce peptide specific cytotoxic T-lymphocytes (CTL) response in vitro and give the experimental foundation for the future clinical trials of immunotherapy in bladder tumor. Methods: The EJ-derived HSP70 co-cultured with DC from the healthy volunteers' PBMC, along with the crude lysate (the supernatant before HSP70 purification) from EJ cells were used as the experimental groups and DC not pulsed by any tumor cells antigen were the blank control. The autologous T-lymphocytes were added into the above various DC groups, and after incubation, the stimulation indexes (SI) and interferon-γ (IFN-γ) were detected to evaluate the immune activities of various DC groups. The killing effects of CTL to target cells, EJ and Hela cells, were determined with 51Cr releasing test. Results: Both DC/HSP70 and DC/the crude lysate could effectively activate CTL in vitro and kill target cells EJ. The killing effect of DC/HSP70 to EJ was much stronger than DC/the crude lysate (the supernatant before HSP70 purification) (P < 0.05). DC without any tumor cell antigens had a lower killing power to EJ. Meanwhile, DC/ HSP70 had little killing power to Hela non-relevant to bladder tumor histopathologically as compared with EJ cells (P < 0.05).
