| 1 | Structure and deformation around the Gyirong basin, north Himalaya, and onset of the south Tibetan detachment system显示文摘Gyirong basin and its adjacent area are located at a special position in the Himalayan orogen, where the south Tibetan detachment system (STDS) and N-S trending rift converged. The north Himalayan orogen here can be divided into five petrologic-tectonic units successively from south to north: 1) the Greater Himalayan crystalline complex (GHC); 2) the STDS shear zone; 3) the Tethyan Himalayan sedimentary sequence (THS); 4) the late Cenozoic sedimentary basins, such as Gyirong and Oma basins; and 5) the Malashan gneiss dome. Structural studies show that this area experienced four stages of deformation: 1) the earlier south-directed thrusting, preserved both in the GHC and THS; 2) top-down-to-north slip along the STDS, normal faults related to this slip formed the early controlling structures of the Cenozoic basins,and the tilted pattern of the blocks between the basins indicated a north-directed slip; 3) east-west extension, the resultant N-S trending normal fault formed the eastern boundary of the basins; and 4) late gravitational collapse. | 2 | A novel cancer vaccine with the ability to simultaneously produce anti-PD-1 antibody and GM-CSF in cancer cells and enhance Th1-biased antitumor immunity显示文摘Tumor escape from immune-mediated destruction has been associated with immunosuppressive mechanisms that inhibit T-cell activation.A promising strategy for cancer immunotherapy is to disrupt key pathways regulating immune tolerance,such as program death-1(PD-1/PD-L1)pathway in the tumor environment.However,the determinants of response to anti-PD-1 monoclonal antibodies(mAbs)treatment remain incompletely understood.In murine models,PD-1 blockade alone fails to induce effective immune responses to poorly immunogenic tumors,but is successful when combined with additional interventions,such as cancer vaccines.Novel cancer vaccines combined with antibody may offer promising control of cancer development and progression.In this investigation,we generated a novel tumor cell vaccine simultaneously expressing anti-PD-1 mAbs and granulocyte-macrophage colony stimulating factor(GM-CSF)in CT26 colon cancer and B16-F10 melanoma.The antitumor effect of the vaccine was verified by therapeutic and adoptive animal experiments in vivo.The antitumor mechanism was analyzed using Flow cytometry,Elispot and in vivo intervention approaches.The results showed that tumor cell vaccine secreting PD-1 neutralizing antibodies and GM-CSF induced remarkable antitumor immune effects and prolonged the survival of tumor-bearing animals compared with animals treated with either PD-1 mAbs or GM-CSF alone.Antitumor effects and prolonged survival correlated with strong antigen-specific T-cell responses by analyzing CD11c^(+)CD86^(+)DC,CD11b^(+)F4/80^(+)MΦcells,increased ratio of Teff/Treg in the tumor microenvironment,and higher secretion levels of Th1 proinflammatory cytokines in serum.Furthermore,the results of ELISPOT and in vivo blocking strategies further confirmed that the antitumor immune response is acquired by CD4 and CD8 T immune responses,primarily dependent on CD4 Th1 immune response,not NK innate immune response.The combination of PD-1 blockade with GM-CSF secretion potency creates a novel tumor cell vaccine immunotherapy,affording significantly improved antitumor responses by releasing the state of immunosuppressive microenvironment and augmenting the tumor-reactive T-cell responses. | Hongwei Tian Gang Shi Qin Wang Yiming Li Qianmei Yang Chunlei Li Guoyou Yang Min Wu Qian Xie Shuang Zhang Yang Yang Rong Xiang Dechao Yu Yuquan Wei Hongxin Deng | 2016 | Signal Transduction and Targeted Therapy2016,1,1: | 3 |

This paper discusses the mineralogical features of the hypoxenoliths. It shows that the xenoliths are characterized by the upper mantle rocks modified to certain extent by the enriched mantle fluid metasomatism in the mantle environment, with the enriched mantle property of Iow-degree partial melting. | 6 | A generalizable strategy based on the rule of“like dissolves like”to construct porous liquids with low viscosity for CO_(2)capture显示文摘Porous liquids(PLs),an emerging porous material with permanent cavities,have attracted extensive attention in recent years.However,the current construction methods are complicated and resulting PLs possess high viscosity values,which cannot meet the requirements of practical industrial applications.Herein,we demonstrate a generalizable and simple strategy to prepare type III PLs with low viscosity based on the rule of“like dissolves like”.Specifically,the monoglycidyl ether terminated polydimethylsiloxane(denoted by E-PDMS)is attached to the surface of Universitetet i Oslo(UiO)-66-NH_(2)via covalent linkage,constructing the pore generator(UiO-66-NH_(2)-E-PDMS,denoted by P-UiO-66).Then,P-UiO-66 is dispersed into different types and amounts of sterically hindered solvents(PDMS400 or PDMS6000),obtaining a series of type III PLs(denoted by P-UiO-66-PLs)with permanent cavities and low viscosities.The gas sorption-desorption test shows that P-UiO-66-PLs have an enormous potential for CO_(2)/N_(2) selective separation.Besides,the porosity of P-UiO-66-PLs and the CO_(2)sorption mechanism are demonstrated by molecular simulation.Furthermore,the generality of the synthesis strategy is confirmed by the successful construction of PLs using two other amino-metal-organic frameworks(MOFs)(MIL-53(Al)-NH_(2)and MIL-88B(Fe)-NH_(2)).Importantly,it’s worth noting that the strategy based on the rule of“like dissolves like”sheds light on the preparation of other types of PLs for task-specific applications. | Yangyang Xin Hailong Ning Dechao Wang(✉) Xiaoqian Li Wendi Fan Xiaoqian Ju Hongni Wang Yichi Zhang Zhiyuan Yang Dongdong Yao Yaping Zheng | 2023 | Nano Research2023,16,7: | 1 |

| 16 | Single-cell Long Non-coding RNA Landscape of T Cells in Human Cancer Immunity显示文摘The development of new biomarkers or therapeutic targets for cancer immunotherapies requires deep understanding of Tcells.To date,the complete landscape and systematic characterization of long noncoding RNAs(lncRNAs)in T cells in cancer immunity are lacking.Here,by systematically analyzing full-length single-cell RNA sequencing(scRNA-seq)data of more than 20,000 libraries of T cells across three cancer types,we provided the first comprehensive catalog and the functional repertoires of lncRNAs in human T cells.Specifically,we developed a custom pipeline for de novo transcriptome assembly and obtained a novel lncRNA catalog containing 9433 genes.This increased the number of current human lncRNA catalog by 16%and nearly doubled the number of lncRNAs expressed in T cells.We found that a portion of expressed genes in single T cells were lncRNAs which had been overlooked by the majority of previous studies.Based on metacell maps constructed by the MetaCell algorithm that partitions scRNA-seq datasets into disjointed and homogenous groups of cells(metacells),154 signature lncRNA genes were identified.They were associated with effector,exhausted,and regulatory T cell states.Moreover,84 of them were functionally annotated based on the co-expression networks,indicating that lncRNAs might broadly participate in the regulation of T cell functions.Our findings provide a new point of view and resource for investigating the mechanisms of T cell regulation in cancer immunity as well as for novel cancer-immune biomarker development and cancer immunotherapies. | Haitao Luo Dechao Bu Lijuan Shao Yang Li Liang Sun Ce Wang Jing Wang Wei Yang Xiaofei Yang Jun Dong Yi Zhao Furong Li | 2021 | Genomics, Proteomics & Bioinformatics2021,19,3: | 0 |
| 17 | Unveiling clinical significance and tumor immune landscape of CXCL12 in bladder cancer: Insights from multiple omics analysis显示文摘Objective: The interplay between chemokine C-X-C motif ligand 12(CXCL12) and its specific receptors is known to trigger various signaling pathways, contributing to tumor proliferation and metastasis. Consequently,targeting this signaling axis has emerged as a potential strategy in cancer therapy. However, the precise role of CXCL12 in clinical therapy, especially in immunotherapy for bladder cancer(BCa), remains poorly elucidated.Methods: We gathered multiple omics data from public databases to unveil the clinical relevance and tumor immune landscape associated with CXCL12 in BCa patients. Univariate and multivariate Cox regression analyses were employed to assess the independent prognostic significance of CXCL12 expression and formulate a nomogram. The expression of CXCL12 in BCa cell lines and clinical tissue samples was validated using enzymelinked immunosorbent assays(ELISA) and immunohistochemistry(IHC).Results: While transcriptional expression of CXCL12 exhibited a decrease in nearly all tumor tissues, CXCL12 methylation expression was notably increased in BCa tissues. Single-cell RNA analysis highlighted tissue stem cells and endothelial cells as the primary sources expressing CXCL12. Abnormal CXCL12 expression, based on transcriptional and methylation levels, correlated with various clinical characteristics in BCa patients. Functional analysis indicated enrichment of CXCL12 and its co-expression genes in immune regulation and cell adhesion. The immune landscape analysis unveiled a significant association between CXCL12 expression and M2 macrophages(CD163~+ cells) in BCa tissues.
