3Usefulness of human epididymis protein 4 in predicting cytoreductive surgical outcomes for advanced ovarian tubal and peritoneal carcinoma显示文摘Objective: Human epididymis protein 4(HE4) is a promising biomarker of epithelial ovarian cancer(EOC). But its role in assessing the primary optimal debulking(OD) of EOC remains unknown. The purpose of this study is to elucidate the ability of preoperative HE4 in predicting the primary cytoreductive outcomes in advanced EOC, tubal or peritoneal carcinoma.Methods: We reviewed the records of 90 patients with advanced ovarian, tubal or peritoneal carcinoma who underwent primary cytoreduction at the Department of Obstetrics and Gynecology of Peking University People's Hospital between November 2005 and October 2010. Preoperative serum HE4 and CA125 levels were detected with EIA kit. A receiver operating characteristic(ROC) curve was used to determine the most useful HE4 cut-off value. Logistic regression analysis was performed to identify significant preoperative clinical characteristics to predict optimal primary cytoreduction.Results: OD was achieved in 47.7%(43/48) of patients. The median preoperative HE4 level for patients with OD vs. suboptimal debulking was 423 and 820 pmol/L, respectively(P<0.001). The areas under the ROC curve for HE4 and CA125 were 0.716 and 0.599, respectively(P=0.080). The most useful HE4 cut-off value was 473 pmol/L. Suboptimal cytoreduction was obtained in 66.7%(38/57) of cases with HE4 ≥473 pmol/L compared with only 27.3%(9/33) of cases with HE4 <473 pmol/L. At this threshold, the sensitivity, specificity, positive predictive value(PPV) and negative predictive value(NPV) for diagnosing suboptimal debulking were 81%, 56%, 67%, and 73%, respectively. Logistic regression analysis showed that the patients with HE4 ≥473 pmol/L were less likely to achieve OD(odds ratio =5.044, P=0.002).Conclusions: Preoperative serum HE4 may be helpful to predict whether optimal cytoreductive surgery could be obtained or whether extended cytoreduction would be needed by an interdisciplinary team.Zhijian Tang Xiaohong Chang Xue Ye Yi Li Hongyan Cheng Heng Cui 2015Chinese Journal of Cancer Research2015,27,3:10
4Clinical features of seizures after cerebral venous sinus thrombosis and its effect on outcome among Chinese Han population显示文摘background and purpose Clinical features of epileptic seizures after cerebral venous sinus thrombosis(CVST)among Chinese patients are not known,and it is still controversial whether seizures would affect the outcome of CVST.Methods In a Chinese hospital-based study of consecutive patients with CVST between 2003 and 2015,we described the clinical features of seizures and determined the predictors of seizure onset using multivariable logistic regression analysis.We also compared the in-hospital case-fatality and short-term functional outcome(modified Rankin Scale(mRS)at discharge)in patients with versus without seizures using ordinal regression analysis.results Among 151 patients with CVST,52(34.4%)presented seizures,of which 42(80.8%)were generalised seizures.Male gender(OR 6.32,95% CI 2.06 to 19.35,p=0.001),motor deficits(OR 4.89,95% CI 1.52 to 15.68,p=0.008),intracerebral haemorrhage(OR 3.93,95% CI 1.16 to 13.26,p=0.027),cerebral infarction(OR 3.78,95% CI 1.15 to 12.36,p=0.029)and superior sagittal sinus thrombosis(OR 3.38,95% CI 91.16 to 9.86,p=0.026)were independent predictors for seizures.The overall in-hospital case-fatality rate was 2.0%(3/151),and 21(13.9%)had mRS>2 at discharge.Compared with patients without seizures,patients with seizures were more likely to have a worse outcome(p=0.02)at discharge,independent of age,gender,clinical presentation,clot burden and presence of parenchymal lesions.conclusions In Chinese Han patients,compared with patients without seizures,patients with seizures after CVST had a worse outcome.Risk factors such as male gender,paresis,parenchymal lesion and superior sagittal sinus thrombosis were independently associated with seizure onset after CVST.Generalised seizure was the main form of seizures after CVST,which was obviously different to seizures after strokes of arterial origin.Hongyan Ding Yanan Xie Linxin Li Heling Chu Yuping Tang Qiang Dong Mei Cui 2017Stroke & Vascular Neurology2017,2,4:8
5Optical observations of LIGO source GW 170817 by the Antarctic Survey Telescopes at Dome A,Antarctica显示文摘The LIGO detection of gravitational waves(GW) from merging black holes in 2015 marked the beginning of a new era in observational astronomy. The detection of an electromagnetic signal from a GW source is the critical next step to explore in detail the physics involved. The Antarctic Survey Telescopes(AST3),located at Dome A, Antarctica, is uniquely situated for rapid response time-domain astronomy with its continuous night-time coverage during the austral winter. We report optical observations of the GW source(GW 170817) in the nearby galaxy NGC 4993 using AST3. The data show a rapidly fading transient at around 1 day after the GW trigger, with the i-band magnitude declining from 17:23 ± 0:13 magnitude to 17:72 ± 0:09 magnitude in ~1:8 h. The brightness and time evolution of the optical transient associated with GW 170817 are broadly consistent with the predictions of models involving merging binary neutron stars. 6IGHG1 promotes motility likely through epithelial-mesenchymal transition in ovarian cancer显示文摘Objective: Ovarian cancer(OC) is one of the leading causes of death for female cancer patients. COC166-9 is an OC-specific monoclonal antibody and we have identified immunoglobulin γ-1 heavy chain constant region(IGHG1) as its antigen. We explore the function of IGHG1 in proliferation, apoptosis and motility of OC cells further in this research.Methods: IGHG1 expression in OC specimens was detected through immunohistochemistry. Real-time quantitative polymerase chain reaction(RT-q PCR) or western blotting assay was used to test IGHG1 expression in OC cells. Viability of OC cells was tested by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay. Flow cytometry or western blotting assay was used to detect cell cycle and apoptosis. Cellular motility was analyzed by using transwell assay and the markers of epithelial-mesenchymal transition(EMT) were tested through immunoblots.Results: Although it exerts negligible effect on the viability and apoptosis of OC cells, IGHG1 could promote migration and invasion of malignant cells in vitro. Mechanistically, IGHG1 increases the expression of N-cadherin and Vimentin while decreases E-cadherin expression. Additionally, IGHG1 expression in OC specimens is higher relative to the paired normal counterparts. Further analysis demonstrates that the increased IGHG1 expression correlates positively with the lymph node metastasis of OC.Conclusions: IGHG1 promotes the motility of OC cells likely through executing the EMT program. 7Causes for different spatial distributions of minimum Arctic sea-ice extent in 2007 and 2012显示文摘Satellite records show the minimum Arctic sea ice extents(SIEs) were observed in the Septembers of 2007 and2012, but the spatial distributions of sea ice concentration reduction in these two years were quite different.Atmospheric circulation pattern and the upper-ocean state in summer were investigated to explain the difference.By employing the ice-temperature and ice-specific humidity(SH) positive feedbacks in the Arctic Ocean, this paper shows that in 2007 and 2012 the higher surface air temperature(SAT) and sea level pressure(SLP)accompanied by more surface SH and higher sea surface temperature(SST), as a consequence, the strengthened poleward wind was favorable for melting summer Arctic sea ice in different regions in these two years. SAT was the dominant factor influencing the distribution of Arctic sea ice melting. The correlation coefficient is –0.84 between SAT anomalies in summer and the Arctic SIE anomalies in autumn. The increase SAT in different regions in the summers of 2007 and 2012 corresponded to a quicker melting of sea ice in the Arctic. The SLP and related wind were promoting factors connected with SAT. Strengthening poleward winds brought warm moist air to the Arctic and accelerated the melting of sea ice in different regions in the summers of 2007 and 2012. 8Spatio-temporal measures of electrophysiological correlates for behavioral multisensory enhancement during visual,auditory and somatosensory stimulation:A behavioral and ERP study显示文摘Multisensory enhancement,as a facilitation phenomenon,is responsible for superior behavioral performance when an individual is responding to cross-modal versus modality-specific stimuli.However,the event-related potential(ERP)counterparts of behavioral multisensory enhancement are not well known.We recorded ERPs and behavioral data from 14 healthy volunteers with three types of target stimuli(modality-specific,bimodal,and trimodal)to examine the spatio-temporal electrophysiological characteristics of multisensory enhancement by comparing behavioral data with ERPs.We found a strong correlation between P3latency and behavioral performance in terms of reaction time(RT)(R=0.98,P<0.001),suggesting that P3 latency constitutes a temporal measure of behavioral multisensory enhancement.In addition,a fast RT and short P3 latency were found when comparing the modality-specific visual target with the modality-specific auditory and somatosensory targets.Our results indicate that behavioral multisensory enhancement can be identified by the latency and source distribution of the P3 component.These findings may advance our understanding of the neuronal mechanisms of multisensory enhancement.Wuyi Wang Li Hu Hongyan Cui Xiaobo Xie Yong Hu 2013Neuroscience Bulletin2013,29,6:4
9Evaluation of COC183B2 antibody targeting ovarian cancer by near-infrared fluorescence imaging显示文摘Objective: To evaluate the imaging potential of a novel near-infrared(NIR) probe conjugated to COC183 B2 monoclonal antibodies(MAb) in ovarian cancer(OC).Methods: The expression of OC183 B2 antigen in OC was determined by immunohistochemical(IHC) staining using tissue microarrays with the H-score system and immunofluorescence(IF) staining of tumor cell lines.Imaging probes with the NIR fluorescent dye cyanine 7(Cy7) conjugated to COC183 B2 Mab were chemically engineered. OC183 B2-positive human OC cells(SKOV3-Luc) were injected subcutaneously into BALB/c nude mice. Bioluminescent imaging(BLI) was performed to detect tumor location and growth. COC183 B2-Cy7 at 1.1,3.3, 10, or 30 μg were used for in vivo fluorescence imaging, and phosphate-buffered saline(PBS), free Cy7 dye and mouse isotype immunoglobulin G(IgG)-Cy7(delivered at the same doses as COC183 B2-Cy7) were used as controls.Results: The expression of OC183 B2 with a high H-score was more prevalent in OC tissue than fallopian tube(FT) tissue. Among 417 OC patients, the expression of OC183 B2 was significantly correlated with the histological subtype, histological grade, residual tumor size, relapse state and survival status. IF staining demonstrated that COC183 B2 specifically expressed in SKOV3 cells but not HeLa cells. In vivo NIR fluorescence imaging indicated that COC183 B2-Cy7 was mainly distributed in the xenograft and liver with optimal tumor-to-background(T/B)ratios in the xenograft at 30 μg dose. The highest fluorescent signals in the tumor were observed at 96 h postinjection(hpi). Ex vivo fluorescence imaging revealed the fluorescent signals mainly from the tumor and liver. 10Impacts of Park Landscape Structure on Thermal Environment Using QuickBird and Landsat Images显示文摘Urban parks composed mostly of vegetation and water bodies can effectively mitigate the urban heat island effect. Many studies have investigated the cooling effects of urban parks; however, little attention has been given to park landscape structure. Based on landscape metrics, this study has explored the influences of the park landscape structure on its inner thermal environment, taking heavily urbanized Beijing Municipality in China as the study area. Three indices, including the percentage of landscape(PLAND), landscape shape index(LSI) and aggregation index(AI), were used to measure the composition and configuration characteristics of the landscape components inside the parks. The indices were calculated for five landscape types being interpreted from Quickbird images. Urban thermal conditions were measured using the land surface temperature(LST) derived from Landsat TM images. The results showed that the park LST had a negative relationship with the park size, but no significant relationship was found with park shape. For the park's interior landscape, however, the configuration and composition characteristics of the landscape components inside the park explained 70% of the park LST variance. The area percentage of water bodies and the aggregation index of woodland were identified as the key influencing characteristics. In addition, when the composition and configuration characteristics of the park landscape components were separately considered, the configuration characteristics(LSI and AI) explained approximately 54% of the variance in park LST, which was comparable with that explained by the composition characteristics(PLAND). 11Nano-needle strontium-substituted apatite coating enhances osteoporotic osseointegration through promoting osteogenesis and inhibiting osteoclastogenesis显示文摘Implant loosening remains a major clinical challenge for osteoporotic patients.This is because osteoclastic bone resorption rate is higher than osteoblastic bone formation rate in the case of osteoporosis,which results in poor bone repair.Strontium(Sr)has been widely accepted as an anti-osteoporosis element.In this study,we fabricated a series of apatite and Sr-substituted apatite coatings via electrochemical deposition under different acidic conditions.The results showed that Ca and Sr exhibited different mineralization behaviors.The main mineralization products for Ca were CaHPO4⋅2H2O and Ca3(PO4)2 with the structure changed from porous to spherical as the pH values increased.The main mineralization products for Sr were SrHPO4 and Sr5(PO4)3OH with the structure changed from flake to needle as the pH values increased.The in vitro experiment demonstrated that coatings fabricated at high pH condition with the presence of Sr were favorable to MSCs adhesion,spreading,proliferation,and osteogenic differentiation.In addition,Sr-substituted apatite coatings could evidently inhibit osteoclast differentiation and fusion.Moreover,the in vivo study indicated that nano-needle like Sr-substituted apatite coating could suppress osteoclastic activity,improve new bone formation,and enhance bone-implant integration.This study provided a new theoretical guidance for implant coating design and the fabricated Sr-substituted coating might have potential applications for osteoporotic patients.Zhen Geng Luli Ji Zhaoyang Li Jing Wang Hongyan He Zhenduo Cui Xianjin Yang Changsheng Liu 2021Bioactive Materials2021,6,4:4
12Synthesis and evaluation of methionine and folate co-decorated chitosan self-assembly polymeric micelles as a potential hydrophobic drug-delivery system显示文摘In this study, an amphiphilic copolymer folate-succinyl-methionine-chitosan-octyl (FSMCO) was successfully synthesized step by step for self-assembling polymeric micelles. The copolymers formed micelle-like nanoparticles by their amphiphilic characteristics and structures were examined by UV-Vis absorption and Fourier transform spectroscopy. The sizes of blank and ICG derivativeloaded micelles measured by dynamic light scattering were about 170 and 140 nm, respectively, which were spherical in shape with an average zeta potential of 10 mV. Further studies on the stability showed that the micellar solutions maintain their sizes at room temperature for 1 month without distinct aggregation or dissociation. ICG derivative was much better photostable after being entrapped by the new carrier. The prepared FSMCO micelles displayed a good drug loading content (11.7%), entrapment efficiency (66.5%) and sustained release rate for the model drug fluorescein. The copolymers demonstrated weeny cytotoxicity toward Bel-7402, L02 and A549 cells when incubated for 2 d. Ligands modified micelles endowed preferable cell targeting capability and beautiful cell inhibition of HCPT-FSMCO on Bel-7402 tumor cells. 13Combined detection and subclass characteristics analysis of CTCs and CTECs by SE-iFISH in ovarian cancer显示文摘Objective:Hematogenous metastasis is essential for the progression of ovarian cancer(OC),and circulating tumor cells(CTCs)are part of the metastatic cascade.However,the detection rate of CTC is low due to the use of less sensitive detection methods.Therefore,this study aimed to detect CTCs and circulating tumorigenic endothelial cells(CTECs)in patients with OC using subtraction enrichment and immunostaining and fluorescence in situ hybridization(SE-iFISH).Methods:We enrolled a total of 56 subjects,including 20 OC patients and 36 ovarian benign tumor patients.CTCs and CTECs were captured by subtraction enrichment(SE)and counted and classified according to immunofluorescence staining of tumor markers(TMs)carbohydrate antigen 125(CA125)and human epididymis protein 4(HE4)combined with fluorescence in situ hybridization(iFISH)of chromosome 8(Chr8)aneuploidy.The diagnostic value and subtype characteristics of CTCs and CTECs were investigated.Results:The detection rate of CTCs by SE-iFISH was high.Compared with CA125 and HE4,Chr8 aneuploidy was the major identification feature of CTC.CTC counts in OC were statistically higher than those in benign groups.CTC and CTEC with≥pentaploidy were detected in both groups,illustrating the poor diagnostic value of CTC or CTEC.Distributions of triploid and tetraploid CTC subtypes were significantly different,and combined detection of triploid and tetraploid CTCs showed the best diagnostic value.In contrast,the distribution of CTECs in the OC and benign groups had no statistically significant difference.Small CTCs accounted for over 1/3 of the total CTC count.We also found that small CTCs and CTECs primarily comprised triploid cells,while large CTCs and CTECs mainly comprised pentaploidy and beyond.Conclusions:The application of SE-iFISH offered a more comprehensive understanding of heterogeneous CTCs and CTECs in OC.Analysis of subclass characteristics of the CTCs and CTECs according to Chr8 aneuploidy and cell size may broaden their potential clinical utility and deepen mechanistic studies in OC.Hongyan Cheng Shang Wang Wenqing Luan Xue Ye Sha Dou Zhijian Tang Honglan Zhu Peter Ping Lin Yi Li Heng Cui Xiaohong Chang 2021Chinese Journal of Cancer Research2021,33,2:3
14Atacamite and Nantokite in Kaerqueka Copper Deposit of Qimantag Area:Evidence for Cenozoic Climate Evolution of the Qaidam Basin显示文摘Using a mineralogy method to reflect climate evolution is a new approach to research Cenozoic environmental progression of the Qaidam Basin. In this paper, we present the chemical composition and crystallographic parameter of atacamite, nantokite, and tenorite in cryptocrystalline aggregates from the Kaerqueka copper deposit of Qimantag metallogenic belt by means of electron microprobe and in-situ X-ray diffraction analyses. Atacamite and nantokite occur in an intimate intergrowth as the secondary precipitation of chalcopyrite and bornite filling in the interstitial space of andradite in the Kaerqueka copper deposit, with an average composition of Cl: 12.38 wt.%, Cu: 63.76 wt.%, O: 21.46 wt.%. X-ray microdiffraction shows that the intergrowth contains nantokite with a cubic unit cell a=5.403(2) ? and atacamite with an orthorhombic unit cell a=6.030(3), b=6.883(2), c=9.114(1) ?. XRD quantitative calculation shows that the nanometric aggregate contains 36.07 wt.% tenorite, 18.41 wt.% atacamite, and 45.52 wt.% nantokite. The presence of nantokite and atacamite requires saline solutions for their formation and hyper-arid climate conditions for their preservation. Combined with the data of salt lakes and the pollen sequence of western China, we suggest that during the uplift of the Tibetan Plateau and the retreat of the Paratethys, saline water was forced to the surface through a basal fracture zone. In the hyper-arid climate of the Qaidam Basin, the recharge of groundwater by direct precipitation is negligible, and groundwater is derived from inflow from the salt lakes. Thus, atacamite is preserved. 15Routing Loop Audit Mechanism Based on SDN显示文摘Routing loops can cause packet loss and long delay problems in the traditional network. Even in future networks – Software-Defined Networks, with loop-free implementation mechanism, it still suffers loop problems. In this paper, we propose an architecture to solve the loop problem in SDN. Unlike the existing passive routing loop detection algorithm, this algorithm processes based on the SDN loop-free characteristic, by auditing third-party network forwarding rules’ modification requests to avoids loop generation, thus avoid the network loop problems. Testbed is built to simulate our proposed algorithm. 17Major and Trace Elements of Magnetite from the Qimantag Metallogenic Belt: Insights into Evolution of Ore–forming Fluids显示文摘Magnetite, as a genetic indicator of ores, has been studied in various deposits in the world. In this paper, we present textural and compositional data of magnetite from the Qimantag metallogenic belt of the Kunlun Orogenic Belt in China, to provide a better understanding of the formation mechanism and genesis of the metallogenic belt and to shed light on analytical protocols for the in situ chemical analysis of magnetite. Magnetite samples from various occurrences, including the ore–related granitoid pluton, mineralised endoskarn and vein–type iron ores hosted in marine carbonate intruded by the pluton, were examined using scanning electron microscopy and analysed for major and trace elements using electron microprobe and laser ablation–inductively coupled plasma–mass spectrometry. The field and microscope observation reveals that early–stage magnetite from the Hutouya and Kendekeke deposits occurs as massive or banded assemblages, whereas late–stage magnetite is disseminated or scattered in the ores. Early–stage magnetite contains high contents of Ti, V, Ga, Al and low in Mg and Mn. In contrast, late–stage magnetite is high in Mg, Mn and low in Ti, V, Ga, Al. Most magnetite grains from the Qimantag metallogenic belt deposits except the Kendekeke deposit plot in the ' Skarn ' field in the Ca+Al+Mn vs Ti+V diagram, far from typical magmatic Fe deposits such as the Damiao and Panzhihua deposits. According to the(Mg O+Mn O)–Ti O2–Al2O3 diagram, magnetite grains from the Kaerqueka and Galingge deposits and the No.7 ore body of the Hutouya deposit show typical characteristics of skarn magnetite, whereas magnetite grains from the Kendekeke deposit and the No.2 ore body of the Hutouya deposit show continuous elemental variation from magmatic type to skarn type. This compositional contrast indicates that chemical composition of magnetite is largely controlled by the compositions of magmatic fluids and host rocks of the ores that have reacted with the fluids. 18An SDN Oriented Loop Detection Mechanism Based on TTL Statistics显示文摘The rise of the network has injected new impetus into the development of traditional networks.Due to the complexity of the network itself and its network programmability,there is a risk of routing loops occurring in the SDN network.This paper proposes a loop detection mechanism.According to the Time To Live(TTL)value of the loop packet,there is approximately periodicity in the same loop.We use sFlow to count the number of packets corresponding to each TTL value of a switch in the loop over a period of time,and perform discrete Fourier transform on the obtained finite-length sequence to observe its frequency domain performance and determine whether there are periodic features.By doing so,it is determined whether there is a routing loop,and the purpose of passively detecting the routing loop is achieved.Compared to existing algorithms,it has advantages in real-time,scalability and false positive rate.The experimental results show that the routing loop detection algorithm based on TTL statistics in this paper still maintains high judgment accuracy under the scenarios of lower stream sampling rate and smaller detection period.Tao Yu Longfei Yu Diyue Chen Hongyan Cui Jilong Wang 2020China Communications2020,17,6:2
