2Serum arylesterase and paraoxonase activity in patients with chronic hepatitis显示文摘AIM: To investigate the relationship between serum paraoxonase (PON1), AST, ALT, GGT, and arylesterase (AE) activity alterations and the degree of liver damage in patients with chronic hepatitis.METHODS: We studied 34 chronic hepatitis patients and 32 control subjects, aged between 35 and 65 years,in the Department of Infection and Clinical Microbiology at the Firat University School of Medicine. Blood samples were collected from subjects between 8:00 and 10:00 a.m. following a 12-h fast. Baseline and salt-stimulated PON1 activities were measured by the hydrolysis of paraoxon. Phenyl acetate was used as the substrate and formed phenol was measured spectrophotometrically at 270 nm after the addition of a 10-fold diluted serum sample in AE activity measurements.RESULTS: The results of this investigation revealed that the levels of AE activity decreased from 132±52 to 94±36 (29%), baseline PON1 activity from 452±112 to 164±67 (64%), salt-stimulated PON1 activity from 746±394 to 294±220 (61%), HDL from 58.4±5.1 to 47.2±5.6(20%), triglyceride from 133±51.2 to 86±34.0 (35%),while a slight increase in the level of LDL (from 163±54.1 to 177.3±56.0; 9%) and significant increases in the levels of AST (from 29±9.3 to 98±44), ALP (from 57.2±13.1 to 91±38.1), ALT (from 27.9±3.32 to 89±19.1), GGT (from 24.3±2.10 to 94±48.2), total bilirubin (from 0.74±0.02 to 1.36±0.06; 84%) and direct bilirubin (from 0.18±0.01 to 0.42±0.04; 133%) were detected.However, the levels of albumin, total protein, cholesterol,and uric acid were almost the same in chronic hepatitis and the control subjects.CONCLUSION: Low PON1 and AE activity may contribute to the increased liver dysfunction in chronic hepatitis patients by reducing the ability of HDL to retard LDL oxidation and might be clinically useful for monitoring the disease of chronic hepatitis.Suleyman Sirri Kilic Suleyman Aydin Nermin Kilic Fazilet Erman Suna Aydin Ilhami Celik 2005World Journal of Gastroenterology2005,11,46:4
3Incorporating travel time reliability in predicting the likelihood of severe crashes on arterial highways using non-parametric random-effect regression显示文摘Travel time reliability(TTR) modeling has gain attention among researchers’ due to its ability to represent road user satisfaction as well as providing a predictability of a trip travel time.Despite this significant effort,its impact on the severity of a crash is not well explored.This study analyzes the effect of TTR and other variables on the probability of the crash severity occurring on arterial roads.To address the unobserved heterogeneity problem,two random-effect regressions were applied;the Dirichlet random-effect(DRE)and the traditional random-effect(TRE) logistic regression.The difference between the two models is that the random-effect in the DRE is non-parametrically specified while in the TRE model is parametrically specified.The Markov Chain Monte Carlo simulations were adopted to infer the parameters’ posterior distributions of the two developed models.Using four-year police-reported crash data and travel speeds from Northeast Florida,the analysis of goodness-of-fit found the DRE model to best fit the data.Hence,it was used in studying the influence of TTR and other variables on crash severity.The DRE model findings suggest that TTR is statistically significant,at 95 percent credible intervals,influencing the severity level of a crash.A unit increases in TTR reduces the likelihood of a severe crash occurrence by 25 percent.Moreover,among the significant variables,alcohol/drug impairment was found to have the highest impact in influencing the occurrence of severe crashes.Other significant factors included traffic volume,weekends,speed,work-zone,land use,visibility,seatbelt usage,segment length,undivided/divided highway,and age.Emmanuel Kidando Ren Moses Eren Erman Ozguven Thobias Sando 2019Journal of Traffic and Transportation Engineering(English Edition)2019,6,5:4
4Cytoprotective effects of amifostine,ascorbic acid and N-acetylcysteine against methotrexate-induced hepatotoxicity in rats显示文摘AIM:To investigate the potential role of oxidative stress and the possible therapeutic effects of N-acetyl cysteine(NAC),amifostine(AMF)and ascorbic acid(ASC)in methotrexate(MTX)-induced hepatotoxicity.METHODS:An MTX-induced hepatotoxicity model was established in 44 male Sprague Dawley rats by administration of a single intraperitoneal injection of20 mg/kg MTX.Eleven of the rats were left untreated(Model group;n=11),and the remaining rats were treated with a 7-d course of 50 mg/kg per day NAC (MTX+NAC group;n=11),50 mg/kg per single dose AMF(MTX+AMF group;n=11),or 10 mg/kg per day ASC(MTX+ASC group;n=11).Eleven rats that received no MTX and no treatments served as the negative control group.Structural and functional changes related to MTX-and the various treatments were assessed by histopathological analysis of liver tissues and biochemical assays of malondialdehyde(MDA),superoxide dismutase(SOD),catalase,glutathione(GSH)and xanthine oxidase activities and of serum levels of aspartate aminotransferase,alanine aminotransferase,alkaline phosphatase and total bilirubin.RESULTS:Exposure to MTX caused structural and functional hepatotoxicity,as evidenced by significantly worse histopathological scores[median(range)injury score:control group:1(0-3)vs 7(6-9),P=0.001]and significantly higher MDA activity[409(352-466)nmol/g vs 455.5(419-516)nmol/g,P<0.05].The extent of MTX-induced perturbation of both parameters was reduced by all three cytoprotective agents,but only the reduction in hepatotoxicity scores reached statistical significance[4(3-6)for NAC,4.5(3-5)for AMF and 6(5-6)for ASC;P=0.001,P=0.001 and P<0.005vs model group respectively].Exposure to MTX also caused a significant reduction in the activities of GSH and SOD antioxidants in liver tissues[control group:3.02(2.85-3.43)μmol/g and 71.78(61.88-97.81)U/g vs model group:2.52(2.07-3.34)μmol/g and 61.46(58.27-67.75)U/g,P<0.05];however,only the NAC treatment provided significant increases in these antioxidant enzyme activities[3.22(2.54-3.62)μmol/g and 69.22(61.13-100.88)U/g,P<0.05 and P<0.01vs model group respectively].CONCLUSION:MTX-induced structural and functional damage to hepatic tissues in rats may involve oxidative stress,and cytoprotective agents(NAC>AMF>ASC)may alleviate MTX hepatotoxicity.Sami Akbulut Hulya Elbe Cengiz Eris Zumrut Dogan Gulten Toprak Emrah Otan Erman Erdemli Yusuf Turkoz 2014World Journal of Gastroenterology2014,20,29:3
5Assessment of traffic performance measures and safety based on driver age and experience:A microsimulation based analysis for an unsignalized T-intersection显示文摘Traffic safety and performance measures such as crash risk and queue lengths or travel times are influenced by several important factors including those related to environment,human,and roadway design,especially at intersections.Previous research has studied different aspects related to these factors,yet these characteristics are not fully investigated with a focus on age and experience of drivers.In this paper,we investigate this issue by using a two-phase approach via a case study application on a critical T-intersection in the City of Tallahassee,Florida.The first phase includes a scenario-based microsimulation analysis through the use of a microscopic simulation software,namely VISSIM,to illustrate the variations in traffic performance measures with respect to driver compositions of different age groups in the traffic stream.A variety of scenarios is created where the driving characteristics are provided as inputs to these scenarios in terms of decision making and risk taking.This is also supported by a sensitivity analysis conducted based on the driver composition in the traffic.The second phase includes the analysis of microsimulation outputs via a tool developed by Federal Highway Administration tool,namely the Surrogate Safety Assessment Model(SSAM),in order to determine the traffic conflicts that occur in each scenario.These conflicts are also compared with real-life crash data for validation purposes.Results show that(a) the differences in risk perception that affect driving behavior might be significant in influencing traffic safety and performance measures,and(b) the proposed approach is considerably successful in simulating the actual crash conflict points.Mehmet Baran Ulak Eren Erman Ozguven Ren Moses Thobias Sando Walter Boot Yassir AbdelRazig John Olusegun Sobanjo 2019Journal of Traffic and Transportation Engineering(English Edition)2019,6,5:2

8Open reduction of a total talar dislocation: A case report and review of the literature显示文摘BACKGROUND Total talar dislocation (TTD) is very uncommon for many orthopedic surgeons and emergency/trauma specialists. Scarce cases of TTD have been reported, mainly in the form of open fracture-dislocation injury. CASE SUMMARY We report a very rare injury of closed TTD with a follow-up period of 36 mo. Initial closed reduction was not successful because of a fractured highly unstable medial malleolus displaced into the ankle mortise, blocking the relocation of the talus. The patient was able to walk pain-free after the 3rd month of surgery. At the 36-mo follow-up, there were 10 degrees of flexion loss and 10 degrees of extension loss in the tibiotalar joint. Furthermore, 5 degrees of subtalar joint inversion-eversion loss was present. 9Reversing the imbalance in bone homeostasis via sustained release of SIRT-1 agonist to promote bone healing under osteoporotic condition显示文摘The imbalance of bone homeostasis is the root cause of osteoporosis.However current therapeutic approaches mainly focus on either anabolic or catabolic pathways,which often fail to turn the imbalanced bone metabolism around.Herein we reported that a SIRT-1 agonist mediated molecular therapeutic strategy to reverse the imbalance in bone homeostasis by simultaneously regulating osteogenesis and osteoclastogenesis via locally sustained release of SRT2104 from mineral coated acellular matrix microparticles.Immobilization of SRT2104 on mineral coating(MAM/SRT)harnessing their electrostatic interactions resulted in sustained release of SIRT-1 agonist for over 30 days.MAM/SRT not only enhanced osteogenic differentiation and mineralization,but also attenuated the formation and function of excessive osteoclasts via integrating multiple vital upstream signals(β-catenin,FoxOs,Runx2,NFATc1,etc.)in vitro.Osteoporosis animal model also validated that it accelerated osteoporotic bone healing and improved osseointegration of the surrounding bone.Overall,our work proposes a promising strategy to treat osteoporotic bone defects by reversing the imbalance in bone homeostasis using designated small molecule drug delivery systems.Wei Zhang Xingzhi Zhou Weiduo Hou Erman Chen Chenyi Ye Mo Chen Qian Lu Xiaohua Yu Weixu Li 2023Bioactive Materials2023,,1:2
