| 1 | Discrimination of False Negative Results in RT-PCR Detection of SARS-CoV-2 RNAs in Clinical Specimens by Using an Internal Reference显示文摘Reverse transcription-polymerase chain reaction(RT-PCR)is an essential method for specific diagnosis of SARS-CoV-2 infection.Unfortunately,false negative test results are often reported.In this study,we attempted to determine the principal causes leading to false negative results of RT-PCR detection of SARS-CoV-2 RNAs in respiratory tract specimens.Multiple sputum and throat swab specimens from 161 confirmed COVID-19 patients were tested with a commercialfluorescent RT-PCR kit targeting the ORF1 ab and N regions of SARS-CoV-2 genome.The RNA level of a cellular housekeeping gene ribonuclease P/MRP subunit p30(RPP30)in these specimens was also assessed by RT-PCR.Data for a total of 1052 samples were retrospectively re-analyzed and a strong association between positive results in SARS-CoV-2 RNA tests and high level of RPP30 RNA in respiratory tract specimens was revealed.By using the ROC-AUC analysis,we identified Ct cutoff values for RPP30 RT-PCR which predicted false negative results for SARS-CoV-2 RT-PCR with high sensitivity(95.03%–95.26%)and specificity(83.72%–98.55%)for respective combination of specimen type and ampli-fication reaction.Using these Ct cutoff values,false negative results could be reliably identified.Therefore,the presence of cellular materials,likely infected host cells,are essential for correct SARS-CoV-2 RNA detection by RT-PCR in patient specimens.RPP30 could serve as an indicator for cellular content,or a surrogate indicator for specimen quality.In addition,our results demonstrated that false negativity accounted for a vast majority of contradicting results in SARS-CoV-2 RNA test by RT-PCR. | Yafei Zhang Changtai Wang Mingfeng Han Jun Ye Yong Gao Zhongping Liu Tengfei He Tuantuan Li Mengyuan Xu Luping Zhou Guizhou Zou Mengji Lu Zhenhua Zhang | 2020 | Virologica Sinica2020,35,6: | 5 |
| 2 | Japanese Encephalitis Virus NS1' Protein Antagonizes Interferon Beta Production显示文摘Japanese encephalitis virus(JEV) is a mosquito-borne virus and the major cause of viral encephalitis in Asia. NS1', a52-amino acid C-terminal extension of NS1, is generated with a-1 programmed ribosomal frameshift and is only present in members of the Japanese encephalitis serogroup of flaviviruses. Previous studies demonstrated that NS1' plays a vital role in virulence, but the mechanism is unclear. In this study, an NS1' defected(rG66A) virus was generated. We found that rG66A virus was less virulent than its parent virus(pSA14) in wild-type mice. However, similar mortality caused by the two viruses was observed in an IFNAR knockout mouse model. Moreover, we found that rG66A virus induced a greater type Ⅰ interferon(IFN) response than that by pSA14, and JEV NS1' significantly inhibited the production of IFN-b and IFN-stimulated genes. | 3 | Variation analysis of growth traits of four poplar clones under different water and fertilizer management显示文摘To determine appropriate quantities of water and fertilizer required for early growth of hybrid poplar cuttings,we recorded the growth traits of four clones grown under four factors(irrigation and nitrogen,phosphorus,and potassium fertilizers),each with four levels,using an orthogonal experimental design.A logistic model was used to estimate growth in height.The growth curves for tree height were sigmoid,and the model R^2 values were greater than 0.9,which indicated that the fit was highly significant.ANOVA results for tree height and basal diameter indicated that all sources of variance showed significant differences(p<0.001).The average tree height and basal diameter for all the four clones under the different treatments ranged from 155.39 to 235.04 cm,and from 13.71 to 17.42 mm,respectively.A highly positive correlation between the extreme k value and tree height was observed,suggesting that the k value was an accurate estimation of tree height.For model parameters,the earliest average time point for the onset of the rapid growth period of poplar clones was 131 d,and the highest average increment in tree height during the rapid growth period was 138.78 cm.The highest average tree height for all clones under each factor was 219,210.51,200,and 201 cm when treated with either 1200 mL of water applied every third day,3 g of nitrogen,0 g of phosphorus,or 0 g of potassium,respectively.The most suitable treatment for the early growth of hybrid poplar cuttings,as suggested by the developed logistic model,was 1200 mL of water applied every third day and three applications of 1 g nitrogen(in the form of CH4N2O). | Ammar Khalil Mohamed Ahmed Luping Jiang Fang Wang Song Chen Xueyan Zhou Xiaona Pei Xiyang Zhao Guanzheng Qu | 2020 | Journal of Forestry Research2020,31,1: | 2 |

| 19 | Comparison of the effectiveness and pregnancy outcomes of labor induction with dinoprostone or single-balloon catheter in term nulliparous women with borderline oligohydramnios显示文摘Backgrounds:At present,there is no consensus on the induction methods in term pregnancy with borderline oligohydramnios.This study aimed to compare the effectiveness and pregnancy outcomes of labor induction with dinoprostone or single-balloon catheter(SBC)in term nulliparous women with borderline oligohydramnios.Methods:We conducted a retrospective cohort study from January 2016 to November 2018.During the study period,a total of 244 cases were enrolled.Of these,103 cases were selected for induction using dinoprostone and 141 cases were selected for induction with SBC.The pregnancy outcomes between the two groups were compared.Primary outcomes were successful vaginal delivery rates.Secondary outcomes were maternal and neonatal adverse events.Multivariate logistic regression was used to assess the risk factors for vaginal delivery failure in the two groups.Results:The successful vaginal delivery rates were similar between the dinoprostone group and the SBC group(64.1%[66/103]vs.59.6%,[84/141]P=0.475),even after adjustment for potential confounding factors(adjusted odds ratio[aOR]:1.07,95%confidence interval[CI]:0.57-2.00,P=0.835).The incidence of intra-amniotic infection was lower in the dinoprostone group than in the SBC group(1.9%[2/103]vs.7.8%[11/141],P<0.001),but the presence of non-reassuring fetal heart rate was higher in the dinoprostone group than in the SBC group(12.6%[13/103]vs.0.7%,[1/141]P<0.001).Multivariate logistic regression showed that nuchal cord was a risk factor for vaginal delivery failure after induction with dinoprostone(aOR:6.71,95%CI:1.96-22.95).There were three factors related to vaginal delivery failure after induction with SBC,namely gestational age(aOR:1.51,95%CI:1.07-2.14),body mass index(BMI)>30 kg/m^(2)(aOR:2.98,95%CI:1.10-8.02),and fetal weight>3500 g(aOR:2.49,95%CI:1.12-5.50).Conclusions:Term nulliparous women with borderline oligohydramnios have similar successful vaginal delivery rates after induction with dinoprostone or SBC,with their advantages and disadvantages.In women with nuchal cord,the risk of vaginal delivery failure is increased if dinoprostone is used in the induction of labor.BMI>30 kg/m^(2),large gestational age,and estimated fetal weight>3500 g are risk factors for vaginal delivery failure after induction with SBC. | Yongqing Zhang Luping Chen Guohui Yan Menglin Zhou Zhengyun Chen Zhaoxia Liang Danqing Chen | 2022 | Chinese Medical Journal2022,,6: | 0 |
