1The Reference Genome of Tea Plant and Resequencing of 81 Diverse Accessions Provide Insights into Its Genome Evolution and Adaptation显示文摘Tea plant is an important economic crop,which is used to produce the world's oldest and most widely consumed tea beverages.Here,we present a high-quality reference genome assembly of the tea plant(Camellia sinensis var.sinensis)consisting of 15 pseudo-chromosomes.LTR retrotransposons(LTR-RTs)account for 70.38%of the genome,and we present evidence that LTR-RTS play critical roles in genome size expansion and the transcriptional diversification of tea plant genes through preferential insertion in promoter regions and introns.Genes,particularly those coding for terpene biosynthesis pro-teins,associated with tea aroma and stress resistance were significantly amplified through recent tandem duplications and exist as gene clusters in tea plant genome.Phylogenetic analysis of the sequences of 81 tea plant accessions with diverse origins revealed three well-differentiated tea plant populations,support-ing the proposition for the southwest origin of the Chinese cultivated tea plant and its later spread to western Asia through introduction.Domestication and modern breeding left significant signatures on hundreds of genes in the tea plant genome,particularly those associated with tea quality and stress resis-tance.The genomic sequences of the reported reference and resequenced tea plant accessions provide valuable resources for future functional genomics study and molecular breeding of improved cul-tivars of tea plants.Enhua Xia Wei Tong Yan Hou Yanlin An Linbo Chen Qiong Wu Yunlong Liu Jie Yu Fangdong Li Ruopei Li Penghui Li Huijuan Zhao Ruoheng Ge Jin Huang Ali Inayat Mallano Yanrui Zhang Shengrui Liu Weiwei Deng Chuankui Song Zhaoliang Zhang Jian Zhao Shu Wei Zhengzhu Zhang Tao Xia Chaoling Wei Xiaochun Wan 2020Molecular Plant2020,13,7:51
2Inconsistent seasonal variation of antibiotics between surface water and groundwater in the Jianghan Plain:Risks and linkage to land uses显示文摘Antibiotics are widely used in humans and animals,but their transformation from surface water to groundwater and the impact of land uses on them remain unclear.In this study,14 antibioticswere systematically surveyed in a complex agricultural area in Central China.Results indicated that the selected antibiotic concentrations in surface waters were higher in winter(average:32.7 ng/L)than in summer(average:17.9 ng/L),while the seasonal variation in groundwaters showed an opposite trend(2.2 ng/L in dry winter vs.8.0 ng/L in summer).Macrolides were the predominant antibiotics in this area,with a detected frequency of over 90%.A significant correlation between surface water and groundwater antibiotics was only observed in winter(R^(2)=0.58).This study further confirmed the impact of land uses on these contaminants,with optimal buffer radii of 2500 m in winter and 500 m in summer.Risk assessment indicated that clarithromycin posed high risks in this area.Overall,this study identified the spatiotemporal variability of antibiotics in a typical agricultural area in Central China and revealed the impact of land uses on antibiotic pollution in aquatic environments.Xi Liu Zhi Wang Lu Zhang Weiying Fan Chao Yang Enhua Li Yun Du Xuelei Wang 2021Journal of Environmental Sciences2021,33,11:9
3Heparanase Expression Correlates with Angiogenesis and Lymphangiogenesis in Human Lung Cancer显示文摘Background and objective Heparanase has been thought to be a good molecular marker of tumor,and the heparanase expression level was correlated closely with tumor metastasis. In this study,we investigate the effects of heparanase on angiogenesis and lymphangiogenesis of lung cancer and the relationship between heparanase expression and vascular endothelial growth factor (VEGF),vascular endothelial growth factor-C (VEGF-C). Methods Immunohistochemistry was used to detect the expression of heparanase,VEGF,VEGF-C protein and microvascular density (MVD),lymphatic vessel density (LVD) in 115 cases of non-small cell lung cancer (NSCLC) and 45 cases of adjacent normal tissue samples. Results Our results showed that heparanase expression was significantly increased in 91 (79.13%) of the 115 cases and correlated with lymph node metastasis (node positive rate 87.0%; node negative rate 36.8%; P=0.003). Heparanase positive expression cases have significantly higher concentration of microvascular density (MVD) and lymphatic vessel density (LVD) as compared with heparanase negative expression cases (P<0.01,P<0.01,respectively),heparanase expression was significantly correlated with VEGF,VEGF-C expression in NSCLC. 4Electroacupuncture at Tianshu (ST 25) for diarrhea-predominant irritable bowel syndrome using positron emission tomography Changes in visceral sensation center显示文摘Previous studies have demonstrated that electroacupuncture therapy is effective in the treatment of irritable bowel syndrome. However, the precise mechanism of this therapy is unknown. The present study served to investigate the effects of electroacupuncture therapy on treatment of patients with diarrhea-predominant irritable bowel syndrome (IBS). We compared brain activation maps based on the changes of cerebral glucose metabolism obtained by 18-fluorodeoxyglucose positron emission tomography scanning under three conditions: resting, rectal balloon distension and rectal balloon distension plus electroacupuncture. Under the resting condition, compared with healthy controls, IBS patients displayed an increasing regional cerebral metabolic rate of glucose over a wide range: bilateral superior temporal gyrus, right middle occipital gyrus, superior frontal gyrus and bilateral middle frontal gyrus. However, there was no significant activity in the visceral pain center. Compared with the resting condition, under the rectal balloon distension condition, patients with IBS had a greater regional cerebral metabolic rate of glucose in the prefrontal cortex, left anterior cingulate cortex, postcentral gyrus, precentral gyrus and temporal gyrus. Under the rectal balloon distension plus electroacupuncture condition, stimulation by electroacupuncture at Tianshu (ST 25) manifested a decreased regional cerebral metabolic rate of glucose in the left cingulate gyrus, right insula, right caudate nucleus, fusiform gyrus and hippocampal gyrus. 5Development and experimental validation of a novel hybrid powertrain with dual planetary gear sets for transit bus applications显示文摘Given that energy conservation and environmental protection are two important goals for the automotive industry, the application of a hybrid electric powertrain can improve vehicle energy efficiency while decreasing fuel consumption and engine emissions. Planetary gear-based power-split hybrid powertrains have become widely used in passenger vehicles, but remain rarely employed on transit buses. This study proposes a novel hybrid powertrain based on two planetary gear sets(CHS) and presents its operating principles along with development of a control strategy for the powertrain. The CHS hybrid powertrain operates in electric mode when the driving power demand is low, and changes to a hybrid electric mode according to the power-split principle of the planetary gear set. To validate the feasibility of the designed CHS hybrid powertrain, a prototype transit bus equipped with the designed hybrid powertrain system was built, and the operating characteristics of the system were analyzed through a performance test conducted on a chassis dynamometer. Compared with a conventional powertrain, the CHS hybrid powertrain can reduce fuel consumption by 39%. 6Robust interactive image segmentation via graph-based manifold ranking显示文摘Interactive image segmentation aims at classifying the image pixels into foreground and background classes given some foreground and background markers. In this paper, we propose a novel framework for interactive image segmentation that builds upon graph-based manifold ranking model, a graph-based semi-supervised learning technique which can learn very smooth functions with respect to the intrinsic structure revealed by the input data. The final segmentation results are improved by overcoming two core problems of graph construction in traditional models: graph structure and graph edge weights. The user provided scribbles are treated as the must-link and must-not-link constraints. Then we model the graph as an approximatively k-regular sparse graph by integrating these constraints and our extended neighboring spatial relationships into graph structure modeling. The content and labels driven locally adaptive kernel parameter is proposed to tackle the insufficiency of previous models which usually employ a unified kernel parameter. After the graph construction,a novel three-stage strategy is proposed to get the final segmentation results. Due to the sparsity and extended neighboring relationships of our constructed graph and usage of superpixels, our model can provide nearly real-time, user scribble insensitive segmentations which are two core demands in interactive image segmentation. Last but not least, our framework is very easy to be extended to multi-label segmentation,and for some less complicated scenarios, it can even get the segmented object through single line interaction. 7A particle-based method for granular flow simulation显示文摘We present a new particle-based method for granular flow simulation.In the method,a new elastic stress term,which is derived from a modified form of the Hooke's law,is included in the momentum governing equation to handle the friction of granular materials.Viscosity force is also added to simulate the dynamic friction for the purpose of smoothing the velocity field and further maintaining the simulation stability.Benefiting from the Lagrangian nature of the SPH method,large flow deformation can be well handled easily and naturally.In addition,a signed distance field is also employed to enforce the solid boundary condition.The experimental results show that the proposed method is effective and efficient for handling the flow of granular materials,and different kinds of granular behaviors can be well simulated by adjusting just one parameter.CHANG YuanZhang BAO Kai ZHU Jian WU EnHua 2012Science China(Information Sciences)2012,55,5:4
8Effects of electroacupuncture on c-Fos expression in the spinal cord and brain of rats with chronic visceral hypersensitivity显示文摘BACKGROUND:Visceral hypersensitivity is the main cause of irritable bowel syndrome.c-Fos is a marker of visceral hypersensitivity in the central nervous system.Electroacupuncture can relieve chronic visceral hypersensitivity in rats,but the mechanism is still unknown. OBJECTIVE:To identify c-Fos expression in the spinal cord and cerebral cortex of rats with chronic visceral hypersensitivity,and to test the effects of electroacupuncture on pain sensitivity in rats with chronic visceral hypersensitivity. DESIGN,TIME AND SETTING:A randomized controlled animal experiment was performed at the Animal Experimental Center,Shanghai University of Traditional Chinese Medicine,from January to April,2007. MATERIALS:A total of 24 neonatal,male,Sprague Dawley rats,aged five days old,were equally and randomly assigned into a normal group,a model group,and an electroacupuncture group. Rabbit anti-rat c-Fos antibody and Evision secondary antibody kits(Sigma,USA),diaminobenzidine kit(Dako,Denmark),and an LD202H electroacupuncture apparatus(Huawei,Beijing,China) were used in this study. METHODS:Neonatal rats from the model and electroacupuncture groups were used to establish rat models of chronic visceral hypersensitivity by the saccule stimulation method.After model establishment,0.25 mm diameter electric needles were inserted into Tianshu(ST 25) and Shangjuxu(ST37) at a depth of approximately 0.5 cm,with an square wave(alternating current frequency at 100/20 Hz,amplitude ranged 0.2-0.6 ms,intensity at 1 mA) once for 20 minutes,once a day,for seven days.Rats in the normal and model groups were not treated. MAIN OUTCOME MEASURES:Following 7 days of treatment,c-Fos expression in the spinal cord and cerebral cortex was detected by immunohistochemistry.After the first electroacupuncture treatment,abdominal withdrawal reflex scores were investigated to evaluate the pain threshold for chronic visceral hypersensitivity in rats. RESULTS:Visceral hypersensitivity increased c-Fos staining(P<0.05),and electroacupuncture significantly decreased the number of these cells to near normal levels(P>0.05).Abdominal withdrawal reflex scores were significantly lower in the electroacupuncture and normal groups than in the model group(P<0.05) and were similar between the electroacupuncture and normal groups (P>0.05). 9Breathable,antifreezing,mechanically skin-like hydrogel textile wound dressings with dual antibacterial mechanisms显示文摘Hydrogels are emerging as the most promising dressings due to their excellent biocompatibility,extracellular matrix mimicking structure,and drug loading ability.However,existing hydrogel dressings exhibit limited breathability,poor environmental adaptability,potential drug resistance,and limited drug options,which extremely restrict their therapeutic effect and working scenarios.Here,the current research introduces the first paradigm of hydrogel textile dressings based on novel gelatin glycerin hydrogel(glyhydrogel)fibers fabricated by the Hofmeister effect based wet spinning.Benefiting from the unique knitted structure,the textile dressing features excellent breathability(1800 times that of the commercially available 3 M dressing)and stretchability(535.51±38.66%).Furthermore,the glyhydrogel textile dressing can also withstand the extreme temperature of-80℃,showing the potential for application in subzero environments.Moreover,the introduction of glycerin endows the textile dressing with remarkable antibacterial property and expands the selection of loaded drugs(e.g.,clindamycin).The prepared glyhydrogel textile dressing shows an excellent infected wound healing effect with a complete rat skin closure within 14 days.All these functions have not been achievable by traditional hydrogel dressings and provide a new approach for the development of hydrogel dressings.Sihan Jiang Jiajia Deng Yuhui Jin Bo Qian Wanqi Lv Qiangqiang Zhou Enhua Mei Rasoul Esmaeely Neisiany Yuehua Liu Zhengwei You Jie Pan 2023Bioactive Materials2023,,3:3
10Image completion with perspective constraint based on a single image显示文摘Although image completion has been used in many image filling and editing problems for a long time, it is rarely applied in the scenario regarding the perspective effect which is demonstrated largely in the design of urban architectures. Thus this paper proposes a method to capture the perspective information automatically based on a single image and employs it to image completion tasks effectively. Actually it is difficult to automatize the extraction from only a single image, but it has been observed that architectures usually contain plenty of repetitions and symmetries which could be described through periodicity. Therefore,our approach first determines the initial vanishing points through the cascaded hough transformation. Then, a refinement scheme based on SIFT feature matching is proposed to obtain the accurate vanishing points. At last,a good perspective mesh is generated from the geometry calculation. 11Texture synthesis via the matching compatibility between patches显示文摘A new patch-based texture synthesis method is presented in this paper. By the method, a set of patches that can be matched with a sampled patch for growing textures effectively, called the matching compat- ibility between patches, is generated first for each patch, and the set is further optimized by culling the patches that may cause synthesis confficts. In this way, similarity measurement calculation for selecting suitable patches in texture synthesis can be greatly saved, and synthesis confficts between neighbouring patches are substantially reduced. Furthermore, retrace computation is integrated in the synthesis process to improve the texture quality. As a result, the new method can produce high quality textures as texture optimization, the best method to date for producing good textures, and run in a time complexity linear to the size of the output texture. 12Coherence-enhancing line drawing for color images显示文摘Line drawing plays an important role in many image-based non-photorealistic applications.However,most existing approaches use a grayscale edge detector for line extraction,so that only luminance diferences between nearby image pixels is taken into account,but the chrominance diferences is ignored.This leads to the undesirable consequence that visually signifcant edges in adjacent regions with diferent colors of similar luminance cannot be detected.To address this limitation,we present a novel enhanced line drawing method based on a flow-based diference-of-Gaussians(FDoG)flter.Because of an inherent property of the thresholded DoG edge model,captured lines may appear dislodged from the true edges in the image.To this end,we provide a gradient-guided warping technique so that smooth and coherent lines can be extracted in the correct location.The GPU implementation of the proposed algorithms allows real-time performance,and experimental examples with various color images demonstrate the method’s superior qualitative performance over previous approaches.WANG ShanDong MA ZiYang LIU XueHui CHEN YanYun WU EnHua 2013Science China(Information Sciences)2013,56,11:2
13Duplication and transcriptional divergence of three Kunitz protease inhibitor genes that modulate insect and pathogen defenses in tea plant(Camellia sinensis)显示文摘Kunitz protease inhibitors(KPIs)are ubiquitous in plants and act as crucial compounds in defense responses against insect attack and pathogen infection.However,the influence of gene duplication on the postdivergence of the CsKPI genes involved in biotic stresses in tea plant is not well known.Here,we identified three CsKPI genes from tea plant(Camellia sinensis)and characterized their expression and evolutionary patterns among plant species.We found that CsKPI1,CsKPI2,and CsKPI3 diverged from their common ancestor 72.94 million years ago(MYA),and the tandem duplication of CsKPI2 and CsKPI3 occurred 26.78 MYA.An in vitro protein assay showed that the three CsKPI proteins were functional and inhibited the production of p-nitroanilide(PNA)from an artificial substrate.The three CsKPI-GFP fusion proteins localized to the cytoplasm.We showed that salicylic acid(SA)and transcripts of CsKPI2 and CsKPI3 significantly accumulated after infection with Glomerella cingulata.The application of exogenous SA stimulated the high expression of both CsKPI2 and CsKPI3 by activating cis-elements within their promoters.Under Ectropis oblique attack,CsKPI1 expression and jasmonic acid(JA)levels were more abundant in both insect-damaged leaf tissues and undamaged neighboring leaves.The application of jasmonic acid methyl ester elicited high expression levels of CsKPI1,suggesting that CsKPI1 accumulation requires JA production in tea plant.The overall findings suggest that the transcriptional divergence of KPI genes after duplication led to the specialized role of CsKPI1 in the physiological response to insect stress;the functional conservation between CsKPI2 and CsKPI3 confers resistance to pathogen infection in tea plant.Junyan Zhu Yaxian He Xiaomei Yan Lu Liu Rui Guo Xiaobo Xia Daojie Cheng Xiaozeng Mi Lidiia Samarina Shenrui Liu Enhua Xia Chaoling Wei 2019Horticulture Research2019,6,1:2

16Circular dichroism spectroscopic studies on structures formed by telomeric DNA sequences in vitro显示文摘Telomere plays an important role in cellular processes, such as cell aging, death and carcinogenisis. Having special sequences, it can form quadrupiex structure in vitro. Circular dichroism (CD) spectroscopic studies show that TTAGGG, (TTAGGG)2 and (TTAGGG)4 can all form quadrupiex in vitro and exist mainly as parallel quadrupiex without metal ions. Both K+ and Na+ can stabilize the tetrameric structure and facilitate the forming of anti-parallel conformation.
