4IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. 8Solid polymer electrolytes in all-solid-state lithium metal batteries:From microstructures to properties显示文摘All-solid-state lithium(Li)metal batteries(ASSLMBs)are considered one of the most promising secondary batteries due to their high theoretical capacity and high safety performance.However,low room-temperature ionic conductivity and poor interfacial stability are two key factors affecting the practical application of ASSLMBs,and our understanding of the mechanisms behind these key problems from microscopic perspective is still limited.In this review,the mechanisms and advanced characterization techniques of ASSLMBs are summarized to correlate the microstructures and properties.Firstly,we summarize the challenges faced by solid polymer electrolytes(SPEs)in ASSLMBs,such as the low roomtemperature ionic conductivity and the poor interfacial stability.Secondly,several typical improvement methods of polymer ASSLMBs are discussed,including composite SPEs,ultra-thin SPEs,SPEs surface modification and Li anode surface modification.Finally,we conclude the characterizations for correlating the microstructures and the properties of SPEs,with emphasis on the use of emerging advanced techniques(e.g.,cryo-transmission electron microscopy)for in-depth analyzing ASSLMBs.The influence of the microstructures on the properties is very important.Until now,it has been difficult for us to understand the microstructures of batteries.However,some recent studies have demonstrated that we have a better understanding of the microstructures of batteries.Then we suggest that in situ characterization,nondestructive characterization and sub-angstrom resolution are the key technologies to help us further understand the batteries'microstructures and promote the development of batteries.And potential investigations to understand the microstructures evolution and the batteries behaviors are also prospected to expect further reasonable theoretical guidance for the design of ASSLMBs with ideal performance.Zongxi Lin Ouwei Sheng Xiaohan Cai Dan Duan Ke Yue Jianwei Nai Yao Wang Tiefeng Liu Xinyong Tao Yujing Liu 2023Journal of Energy Chemistry2023,,6:1

10GSH-induced chemotaxis nanomotors for cancer treatment by ferroptosis strategy显示文摘Overexpression of glutathione(GSH) in tumor cells greatly inhibits the therapy effect of traditional ferroptosis inducers;thus,control of the GSH level is an important way to improve the efficacy of ferroptosis.Herein,a kind of nanomotor based on metal organic framework material NH_(2)-MIL-101 is constructed,in which polyethylene glycol(PEG) and glutathione hydrolase γ-glutamyltransferase(GGT) are asymmetrically modified to obtain mPEG@MIL-101@GGT nanomotors(PMG NMs).The nanomotor proposed in this article can be induced by overexpressing GSH in tumors to form chemotactic effects through the specific affinity between enzymes and substrates.Results indicate that the tail structure provided by PEG and the affinity between GGT and GSH can enable the stable chemotaxis behavior of nanomotors in a complex environment,thus enriching and penetrating deeply at the tumor site.In addition,after loading the ferroptosis inducer Erastin,the system shows a highly effective induction effect of tumor ferroptosis.Erastin in the system can effectively inhibit the synthesis of GSH,and PMG NMS can react with GSH through Fe^(3+)and GGT to promote GSH depletion.The produced Fe^(2+)can generate excessive reactive oxygen species through Fenton reaction,which further promotes the death of tumor cells.Meantime,the chemotaxis behavior of the nanomotors based on the endogenous biochemical reaction of GGT-catalyzed GSH hydrolysis can endow nanomotors with the enhanced delivery and penetration ability in tumors,thus collaboratively enhancing the ferroptosis effect.This strategy designed according to the physiological characteristics of tumors has good biosafety and treatment effect,providing new perspectives for micro/nanomotor and tumor treatment.Zhiyong Liu Ting Li Nan Li Yujing Wang Lin Chen Xueting Tang Mimi Wan Chun Mao 2022Science China Chemistry2022,65,5:1

13Gut microbiome,cognitive function and brain structure:a multi-omics integration analysis显示文摘Background:Microbiome-gut-brain axis may be involved in the progression of age-related cognitive impairment and relevant brain structure changes,but evidence from large human cohorts is lacking.This study was aimed to investigate the associations of gut microbiome with cognitive impairment and brain structure based on multi-omics from three independent populations.Methods:We included 1430 participants from the Guangzhou Nutrition and Health Study(GNHS)with both gut microbiome and cognitive assessment data available as a discovery cohort,of whom 272 individuals provided fecal samples twice before cognitive assessment.We selected 208 individuals with baseline microbiome data for brain magnetic resonance imaging during the follow-up visit.Fecal 16S rRNA and shotgun metagenomic sequencing,tar-geted serum metabolomics,and cytokine measurements were performed in the GNHS.The validation analyses were conducted in an Alzheimer’s disease case-control study(replication study 1,n=90)and another community-based cohort(replication study 2,n=1300)with cross-sectional dataset.Results:We found protective associations of specific gut microbial genera(Odoribacter,Butyricimonas,and Bac-teroides)with cognitive impairment in both the discovery cohort and the replication study 1.Result of Bacteroides was further validated in the replication study 2.Odoribacter was positively associated with hippocampal volume(β,0.16;95%CI 0.06-0.26,P=0.002),which might be mediated by acetic acids.Increased intra-individual alterations in gut microbial composition were found in participants with cognitive impairment.We also identified several serum metabolites and inflammation-associated metagenomic species and pathways linked to impaired cognition.Conclusions:Our findings reveal that specific gut microbial features are closely associated with cognitive impair-ment and decreased hippocampal volume,which may play an important role in dementia development.Xinxiu Liang Yuanqing Fu Wen-ting Cao Zhihong Wang Ke Zhang Zengliang Jiang Xiaofang Jia Chun-ying Liu Hong-rou Lin Haili Zhong Zelei Miao Wanglong Gou Menglei Shuai Yujing Huang Shengdi Chen Bing Zhang Yu-ming Chen Ju-Sheng Zheng 2022Translational Neurodegeneration2022,11,1:1

17Establishment of a prognostic scoring model for regional recurrent nasopharyngeal carcinoma after neck dissection显示文摘Objective:The main aim of this study was to establish a scoring model to predict risk of progression and survival in patients with regionally recurrent nasopharyngeal carcinoma(NPC).Methods:Three hundred and forty-eight patients subjected to neck dissection from 2003 to 2017 were included for study.Clinicopathologic information for each patient was analyzed.Independent prognostic factors were selected using the Cox proportional hazards model and incorporated into the scoring model.Concordance index(C-index)and calibration curves were used to verify discrimination and calibration,respectively and the results validated using bootstrap resampling.Results:Microscopic positive lymph node>2[hazard ratio(HR),2.19;95%confidence interval(CI),1.30–3.68;P=0.003],extranodal extension(HR,2.75;95%CI,1.69–4.47;P<0.001),and lower neck involvement(HR,1.78;95%CI,1.04–3.04;P=0.034)were identified from multivariate analysis as independent factors for overall survival(OS).A qualitative 4-point scale was generated to stratify patients into 4 risk groups for predicting OS and progression-free survival(PFS).The novel scoring model demonstrated enhanced discrimination(C-index=0.69;95%CI,0.62–0.76)relative to the original recurrent tumor-node-metastasis(rTNM)staging system(C-index=0.56;95%CI,0.50–0.62),and was internally validated with a bootstrap-adjusted C-index of 0.70.The calibration curve showed good agreement between predicted probabilities and actual observations.Conclusions:The scoring system established in this study based on a large regionally recurrent NPC cohort fills a gap regarding assessment of risk and prediction of survival outcomes after neck dissection in this population and could be further applied to identify high-risk patients who may benefit from more aggressive intervention.Xiaoyun Li Chao Lin Jinjie Yan Qiuyan Chen Xuesong Sun Sailan Liu Shanshan Guo Liting Liu Haojun Xie Qingnan Tang Yujing Liang Ling Guo Hao Li Xuekui Liu Xiang Guo Linquan Tang Haiqiang Mai 2020Cancer Biology & Medicine2020,17,1:0
18Preliminary analysis of cellular sociology of co-cultured glioma initiating cells and macrophages in vitro显示文摘Objective: Real-time monitoring of cytokine secretion at the single immunocyte level,based on the concept of immune cells, sociology has been recently reported. However,the relationships between glioma-initiating cells(GICs) and host immune cells and their mutual interactions in the tumor microenvironment have not been directly observed and remain unclear. Methods: The dual fluorescence tracing technique was applied to label the co-cultured GICs and host macrophages(M?), and the interactions between the two types of cells were observed using a live cell imaging system. Fusion cells in the co-culture system were monocloned and proliferated in vitro and their social interactions were observed and recorded. Results: Using real-time dynamic observation of target cells, 6 types of intercellular conjunction microtubes were found to function in the transfer of intercellular information between GICs and M?; GICs and host M? can fuse into hybrid cells after several rounds of mutual interactions, and then these fusion cells fused with each other; Fusion cells generated offspring cells through symmetrical and asymmetrical division or underwent apoptosis. A 'cell in cell' phenomenon was observed in the fusion cells, which was often followed by cell release, namely entosis. Conclusions: Preliminary studies revealed the patterns of cell conjunction via microtubes between GICs and host M? and the processes of cell fusion, division, and entosis. 20Efficacy and safety of Weichang’an pill(胃肠安丸)combined with Western Medicine on gastrointestinal diseases:a systematic review and Meta-analysis显示文摘OBJECTIVE:To systematically evaluate the efficacy and safety of Weichang’an pill(胃肠安丸,WCA)combined with Western Medicine(WM)for the treatment of gastrointestinal diseases.METHODS:Eight databases,including China National Knowledge Infrastructure Database,Wanfang Data,China Science and Technology Journal Database,SinoMed,PubMed,Web of Science,Cochrane Library,and Embase,were searched for randomized controlled trials(RCTs)of WCA from inception to 30 September 2021.We independently screened the literature,extracted data,and then evaluated the bias risk,effectiveness,safety,and other indicators of the included articles.RESULTS:A total of 33 RCTs were included in this study with 3368 patients.After analysis,it was found that WCA combined with WM could effectively prevent and treat antibiotic-associated gastrointestinal reaction,functional dyspepsia(FD),irritable bowel syndrome,rotavirus diarrhea(RVD),and ulcerative colitis(UC);no serious adverse reactions occurred.Moreover,compared with the control group,the experimental group showed significantly improved symptoms and some biochemical parameters.CONCLUSIONS:WCA combined with WM for the treatment of gastrointestinal diseases had better clinical efficacy than the control group,without serious adverse reactions.Notably,in the treatment of FD,RVD,and UC,WCA improved clinical symptoms and biochemical indicator expression.Nevertheless,owing to the restricted quality and quantity of the literature,the results need to be further studied using high-quality RCTs.WANG Jiabao ZHANG Lishuang NIU Baihan YU Yajun YANG Fengwen MIAO Lin CHAI Lijuan DING Xinya SUN Yingjie WANG Yujing WANG Lin ZHANG Han WANG Yi LI Lin 2023Journal of Traditional Chinese Medicine2023,43,6:0
