| 3 | Mechanism underlying effect of Chaihu Shugan San(柴胡疏肝散)on major depressive disorder:a network pharmacology-based study显示文摘OBJECTIVE:To investigate the mechanism underpinning the effect of Chaihu Shugan San(柴胡疏肝散,CHSGS)on major depressive disorder(MDD).METHODS:We searched the compound components of from seven herbal ingredients of CHSGS from TCMSP,SymMap,ETCM,NPASS databases,and the chemical structure of the compound from PubChem,and collected the compound targets from TCMSP and TargetNet databases,and MDD-related targets from DiseaseGene Network.We established protein-protein interaction in the STRING database.Through gene mapping,topology analysis and enrichment analysis,the core targets and pathways of CHSGS for MDD,and the involved biological processes(BP),cell components(CC),and molecular functions(MF)were predicted.RESULTS:We collected a total of 1135 CHSGS compounds.After screening by ADME standards and the five rules of Ribinski,we obtained 99 different chemical components with different chemical structures,and related targets of 183 different CHSGS compounds.In the DiseaseGene Network,a total of 740 relevant targets for MDD were collected.Through gene mapping and topological analysis,62 related targets of CHSGS for MDD,24 targets with topological Chinese herbal medicine were obtained.Through enrichment analysis,10 relevant pathways and 3 core pathways were obtained with the involvement of 127 BP,27 CC,and 43 MF.CONCLUSION:There are multiple targets and signaling pathways are involved in the action of CHSGS in the treatment of MDD. | MU Jie ZHANG Shuang TANG Feifei LIU Shuling ZHANG Qianyi DU Qianlei WANG Xueqian WANG Qingguo CHENG Fafeng | 2021 | Journal of Traditional Chinese Medicine2021,41,2: | 4 |

| 5 | Differentiation and Pro-Angiogenic Potential of Infantile Hemangioma Stem Cells显示文摘Objectives Infantile hemangioma(IH)is defined as a benign vascular tumor composed of immature vascular endothelial cells,with the unique characteristics of rich vascularization and self-regression into fibro-fatty tissue.CD133-positive hemangioma stem cells(HemSCs),present in the proliferating IH tissue,can be used to establish the IH animal model,which has highlighted the pathogenesis of IH in recent years.This study focused on the biological characteristics and differentiation capacity of HemSCs and aimed to provide a theoretical possibility for its application in tissue engineering.Methods To further confirm our hypothesis,we used fluorescence-activated cell sorting(FACS),in vitro multipotent induced differentiation,angiogenesis assay,and antibody array to identify the surface markers,multipotent differential potential,angiogenesis potential,and secreted factors of HemSCs,respectively,utilizing human adipose stem cells(hADSCs)as the control.Results We successfully isolated and cultured HemSCs.FACS indicated that,on average,more than 80%of HemSCs matched the criteria of mesenchymal stem cell(MSC)surface markers.Our results confirmed that HemSCs could differentiate into adipocytes,osteocytes,and chondrocytes.Additionally,compared with fibroblasts or hADSCs,HemSCs could promote angiogenesis through para-secretion.Conclusions HemSCs may originate from normal MSCs,and owing to their powerful proliferative and angiogenic abilities,they could be considered an IH pathogenic factor.These characteristics demonstratet heir potential as a candidate seed cell in tissue engineering. | Hanru YING Qianyi CHEN Xuanfeng CHEN Zhang YU MD Shih-Jen CHANG Lei CHANG Gang MA Yajing QIU Hui CHEN Xiaoxi LIN | 2020 | Chinese Journal Of Plastic and Reconstructive Surgery2020,2,1: | 2 |
| 6 | Tunable magneto-optical polarization device for terahertz waves based on InSb and its plasmonic structure显示文摘The nonreciprocal circular dichroism and Faraday rotation effect for terahertz(THz) waves in longitudinally magnetized InSb were investigated by theoretical and experimental studies in the THz regime, which indicated its ability for a THz circularly polarized isolator, THz circular polarizer, tunable polarization converter, and polarization modulator by manipulation of different magnetic fields. Furthermore, we demonstrated the InSb plasmonics based on its magneto-optical effects combined with artificial microstructure. We found the magnetooptical enhancement mechanisms in this magneto-plasmonic structure, achieving broadband near-perfect orthogonal linear polarization conversion modulated by the weak magnetic field in an experiment with an extinction ratio of 33 dB. Moreover, the magneto-optical modulation with an amplitude modulation depth of 95.8% can be achieved by this device under a weak magnetic field of 150 mT. | 12 | Imaging ultrafast evolution of subwavelengthsized topography using single-probe structured light microscopy显示文摘Imaging ultrafast processes in femtosecond(fs) laser–material interactions such as fs laser ablation is very important to understand the physical mechanisms involved. To achieve this goal with high resolutions in both spatial and temporal domains, a combination of optical pump–probe microscopy and structured illumination microscopy can be a promising approach, but suffers from the multiple-frame method with a phase shift that is inapplicable to irreversible ultrafast processes such as ablation. Here, we propose and build a wide-field singleprobe structured light microscopy(SPSLM) to image the ultrafast three-dimensional topography evolution induced by fs lasers, where only a single imaging frame with a single structured probe pulse is required for topography reconstruction, benefiting from Fourier transform profilometry. The second harmonic of the fs laser is used as the structured probe light to improve spatial lateral resolution into the subwavelength region of ~478 nm, and the spatial axial and temporal resolutions are estimated to be ~22 nm and ~256 fs, respectively. With SPSLM, we successfully image the ultrafast topography evolution of a silicon wafer surface impacted by single and multiple fs pulses. The variable formation and evolution of the laser induced periodic surface structures during an ultrashort time are visualized and analyzed. | 19 | Relationship between PaO_(2)/FiO_(2) and delirium in intensive care:A cross-sectional study显示文摘Background::To investigate the relationship between partial pressure of oxygen(PaO_(2))/fraction of inspired oxygen(FiO_(2))and the probability of delirium in intensive care units(ICUs).Methods::The investigation was a cross-sectional study that involved the collection of data from patients admitted to the Xiang Ya Hospital Cardiothoracic Surgical Care Unit and Comprehensive Intensive Care Unit from 01 September 2016 to 10 December 2016.Delirium was diagnosed using the simplified version of the Chinese Confusion Assessment Method(CAM)for the ICU.Demographic and medical data were obtained within 24 h of each patient admitted in the ICU.The PaO_(2)/FiO_(2) of each patient was recorded 24 h after admission in the ICU.The patients were divided into three groups according to PaO_(2)/FiO_(2) data:normal(PaO_(2)/FiO_(2)≥300 mmHg),slightly low(200≥PaO_(2)/FiO_(2)<300 mmHg),and severely low(PaO_(2)/FiO_(2)<200 mmHg).Baseline characteristics were compared in the three groups.Results of the unadjusted model,minimally adjusted model,and fully adjusted model are presented.Results::A total of 403 participants were included in the study,of which 184(45.7%)developed delirium.Age(P<0.001),Sequential Organ Failure Assessment(SOFA)score(P<0.001),Acute Physiology and Chronic Health Evaluation(APACHE)II score(P<0.001),mechanical ventilation time(P<0.001),history of hypertension(P=0.040),heart disease(P=0.040),sedation(P=0.001),and PaO_(2)/FiO_(2)(P=0.006)were significantly associated with delirium in univariate analysis.Multivariate regression analysis models were used to further analyze the associations between PaO_(2)/FiO_(2) and delirium.In the crude model,for 1 standard deviation(SD)increase in PaO_(2)/FiO_(2),the odds ratio(OR)of delirium was 0.8(95%confidence interval[CI]:0.6-0.9),but there was no significant correlation in the fully adjusted model.There was a non-linear relationship between the PaO_(2)/FiO_(2) and delirium in a generalized additive model.A two-piecewise linear regression model was used to calculate a PaO_(2)/FiO_(2) threshold of 243 mmHg.On the left side of the threshold,the OR was 0.9 and the 95%CI was 0.9-1.0(P=0.013)when PaO_(2)/FiO_(2) increased by 1 SD.Conclusions::PaO_(2)/FiO_(2) was negatively associated with delirium when PaO_(2)/FiO_(2) was below the identified threshold.As a readily available laboratory indicator,PaO_(2)/FiO_(2) has potential value in the clinical evaluation of risk of delirium in ICU patients. | Fang Gong Yuhang Ai Lina Zhang Qianyi Peng Quan Zhou Chunmei Gui | 2023 | Journal of Intensive Medicine2023,3,1: | 0 |
| 20 | Correction of a CADASIL point mutation using adenine base editors in hiPsCsandbloodvessel organoids显示文摘Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) is a monogenic small vessel disease caused by mutations in the NOTCH3 gene. However, the pathogenesis of CADASIL remains unclear, and patients have limited treatment options. Here, we use human induced pluripotent stem cells (hiPSCs) generated from the peripheral blood mononuclear cells of a patient with CADASIL carrying a heterozygous NOTCH3 mutation (c.1261C>T, p.R421C) to develop a disease model. The correction efficiency of different adenine base editors (ABEs) is tested using the HEK293T-NOTCH3 reporter cell line. ABEmax is selected based on its higher efficiency and minimization of predicted off-target effects. Vascular smooth muscle cells (VSMCs) differentiated from CADASIL hiPSCs show NOTCH3 deposition and abnormal actin cytoskeleton structure, and the abnormalities are recovered in corrected hiPSC-derived VSMCs. Furthermore, CADASIL blood vessel organoids generated for in vivo modeling show altered expression of genes related to disease phenotypes, including the downregulation of cell adhesion, extracellular matrix organization, and vessel development. The dual adeno-associated virus (AAV) split-ABEmax system is applied to the genome editing of vascular organoids with an average editing efficiency of 8.82%.
