3A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. The medium transforms the wavefront to realize sophisticated computing tasks such as image recognition. At the output, the optical energy is concentrated in well-defined locations, which, for example, can be interpreted as the identity of the object in the image.These computing media can be as small as tens of wavelengths and offer ultra-high computing density. 5On the Key Dynamical Processes Supporting the 21.7 Zhengzhou Record-breaking Hourly Rainfall in China显示文摘An extremely heavy rainfall event occurred in Zhengzhou,China,on 20 July 2021 and produced an hourly rainfall rate of 201.9 mm,which broke the station record for China's Mainland.Based on radar observations and a convection-permitting simulation using the WRF-ARW model,this paper investigates the multiscale processes,especially those at the mesoscale,that support the extreme observed hourly rainfall.Results show that the extreme rainfall occurred in an environment characteristic of warm-sector heavy rainfall,with abundant warm moist air transported from the ocean by an abnormally northward-displaced western Pacific subtropical high and Typhoon In-Fa(2021).However,rather than through back building and echo training of convective cells often found in warm-sector heavy rainfall events,this extreme hourly rainfall event was caused by a single,quasi-stationary storm in Zhengzhou.Scale separation analysis reveals that the extreme-rainproducing storm was supported and maintained by the dynamic lifting of low-level converging flows from the north,south,and east of the storm.The low-level northerly flow originated from a mesoscale barrier jet on the eastern slope of the Taihang Mountain due to terrain blocking of large-scale easterly flows,which reached an overall balance with the southerly winds in association with a low-level meso-β-scale vortex located to the west of Zhengzhou.The large-scale easterly inflows that fed the deep convection via transport of thermodynamically unstable air into the storm prevented the eastward propagation of the weak,shallow cold pool.As a result,the convective storm was nearly stationary over Zhengzhou,resulting in record-breaking hourly precipitation.Peng WEI Xin XU Ming XUE Chenyue ZHANG Yuan WANG Kun ZHAO Ang ZHOU Shushi ZHANG Kefeng ZHU 2023Advances in Atmospheric Sciences2023,40,3:9
6Optical coherence tomography angiography in diabetic retinopathy: a review of current applications显示文摘Background:Diabetic retinopathy(DR)is a leading cause of vision loss in adults.Currently,the standard imaging technique to monitor and prognosticate DR and diabetic maculopathy is dye-based angiography.With the introduction of optical coherence tomography angiography(OCTA),it may serve as a potential rapid,non-invasive imaging modality as an adjunct.Main text:Recent studies on the role of OCTA in DR include the use of vascular parameters e.g.,vessel density,intercapillary spacing,vessel diameter index,length of vessels based on skeletonised OCTA,the total length of vessels,vascular architecture and area of the foveal avascular zone.These quantitative measures may be able to detect changes with the severity and progress of DR for clinical research.OCTA may also serve as a non-invasive imaging method to detect diabetic macula ischemia,which may help predict visual prognosis.However,there are many limitations of OCTA in DR,such as difficulty in segmentation between superficial and deep capillary plexus;and its use in diabetic macula edema where the presence of cystic spaces may affect image results.Future applications of OCTA in the anterior segment include detection of anterior segment ischemia and iris neovascularisation associated with proliferative DR and risk of neovascular glaucoma.Conclusion:OCTA may potentially serve as a useful non-invasive imaging tool in the diagnosis and monitoring of diabetic retinopathy and maculopathy in the future.Future studies may demonstrate how quantitative OCTA measures may have a role in detecting early retinal changes in patients with diabetes.Kai Yuan Tey Kelvin Teo Anna C.S.Tan Kavya Devarajan Bingyao Tan Jacqueline Tan Leopold Schmetterer Marcus Ang 2019Eye and Vision2019,6,1:6
7Oral microbiome and pancreatic cancer显示文摘BACKGROUND Microbiota profiles differ between patients with pancreatic cancer and healthy people,and understanding these differences may help in early detection of pancreatic cancer.Saliva sampling is an easy and cost-effective way to determine microbiota profiles compared to fecal and tissue sample collection.AIM To investigate the saliva microbiome distribution in patients with pancreatic adenocarcinoma(PDAC)and the role of oral microbiota profiles in detection and risk prediction of pancreatic cancer.METHODS We conducted a prospective study of patients with pancreatic cancer(n=41)and healthy individuals(n=69).Bacterial taxa were identified by 16S ribosomal ribonucleic acid gene sequencing,and a linear discriminant analysis effect size algorithm was used to identify differences in taxa.Operational taxonomic unit values of all selected taxa were converted into a normalized Z-score,and logistic regressions were used to calculate risk prediction of pancreatic cancer.RESULTS Compared with the healthy control group,carriage of Streptococcus and Leptotrichina(z-score)was associated with a higher risk of PDAC[odds ratio(OR)=5.344,95%confidence interval(CI):1.282-22.282,P=0.021 and OR=6.886,95%CI:1.423-33.337,P=0.016,respectively].Veillonella and Neisseria(z-score)were considered a protective microbe that decreased the risk of PDAC(OR=0.187,95%CI:0.055-0.631,P=0.007 and OR=0.309,95%CI:0.100-0.952,P=0.041,respectively).Among the patients with PDAC,patients reporting bloating have a higher abundance of Porphyromonas(P=0.039),Fusobacterium(P=0.024),and Alloprevotella(P=0.041);while patients reporting jaundice had a higher amount of Prevotella(P=0.008);patients reporting dark brown urine had a higher amount of Veillonella(P=0.035).Patients reporting diarrhea had a lower amount of Neisseria and Campylobacter(P=0.024 and P=0.034),and patients reporting vomiting had decreased Alloprevotella(P=0.036).CONCLUSION Saliva microbiome was able to distinguish patients with pancreatic cancer and healthy individuals.Leptotrichia may be specific for patients living in Sichuan Province,southwest China.Symptomatic patients had different bacteria profiles than asymptomatic patients.Combined symptom and microbiome evaluation may help in the early detection of pancreatic cancer.Ai-Lin Wei Mao Li Guo-Qing Li Xuan Wang Wei-Ming Hu Zhen-Lu Li Jue Yuan Hong-Ying Liu Li-Li Zhou Ka Li Ang Li Mei Rosemary Fu 2020World Journal of Gastroenterology2020,26,48:4

9A Dose-response Meta-analysis of Egg Intake and Breast Cancer Risk显示文摘Aim Eggs are one of the most nutritious foods in nature,but there is no unified conclusion about the association between egg intake and breast cancer risk.Methods The PubMed and Web of Science databases for the literature on egg intake and breast cancer risk were searched for papers published during the last 10 years.These were then filtered according to the inclusion and exclusion criteria.Stata16.0 software was applied to perform a meta-analysis,the generalized least squares method and constrained cubic spline model were used to assess the dose-response trends between egg intake and breast cancer risk.Results A total of 9 articles were included:6 case control studies and 3 cohort studies.The Newcastle-Ottawa scale(NOS)values of the included articles were all≥6 points.The pooled relative risks(RR)of egg intake and breast cancer risk was 0.91(95%CI:0.69-1.19).The dose-response analysis showed a linear trend for egg intake and breast cancer risk(P=0.689).With every 10 g/day increase in egg intake,the incidence of breast cancer increased by 2%(RR=1.02,95%CI:0.99-1.05).However,these results were not statistically significant.Conclusion This meta-analysis found no significant association between egg intake and breast cancer.Peng Wei Ma Fei Fei Feng Zi Ang Shi Ke Di Xu Yuan Lin Zou Yan Li Wang Si Qi Ning Chun Hua Song 2021Journal of Nutritional Oncology2021,6,2:3
10Cobalt nanoparticle decorated N-doped carbons derived from a cobalt covalent organic framework for oxygen electrochemistry显示文摘The low cost and highly efficient construction of electrocatalysts has attracted significant attention owing to the use of clean and sustainable energy technologies.In this work,cobalt nanoparticle decorated N-doped carbons(Co@NC)are synthesized by the pyrolysis of a cobalt covalent organic framework under an inert atmosphere.The Co@NC demonstrates improved electrocatalytic capabilities compared to N-doped carbon without the addition of Co nanoparticles,indicating the important role of cobalt.The well-dispersed active sites(Co-Nx)and the synergistic effect between the carbon matrix and Co nanoparticles greatly enhance the electrocatalytic activity for the oxygen reduction reaction.In addition,the Co content has a significant effect on the catalytic activity.The resulting Co@NC-0.86 exhibits a superb electrocatalytic activity for the oxygen reduction reaction in an alkaline electrolyte in terms of the onset potential(0.90 V),half-wave potential(0.80 V)and the limiting current density(4.84 mA·cm^(−2)),and a high selectivity,as well as a strong methanol tolerance and superior durability,these results are comparable to those of the Pt/C catalyst.Furthermore,the superior bifunctional activity of Co@NC-0.86 was also confirmed in a home-built Zn-air battery,signifying the possibility for application in electrode materials and in current energy conversion and storage devices.Rui-Qi Zhang Ang Ma Xiang Liang Li-Min Zhao Hui Zhao Zhong-Yong Yuan 2021Frontiers of Chemical Science and Engineering2021,15,6:2

14Status of CAS global ionospheric maps after the maximum of solar cycle 24显示文摘As a new Ionosphere Associate Analysis Center(IAAC)of the International GNSS Service(IGS),Chinese Academy of Sciences(CAS)started the routine computation of the real-time,rapid,and final Global Ionospheric Maps(GIMs)in 2015.The method for the generation of CAS rapid and final GIMs and recent updates are presented in the paper.The quality of CAS post-processed GIMs is assessed during 2015-2018 after the maximum of solar cycle 24.To perform an independent and fair assessment,Jason-2/3 Vertical Total Electron Contents(VTEC)are first used as the references over the ocean.GPS differential Slant TECs(dSTEC)generated from 55 Multi-GNSS Experimental(MGEX)stations of the IGS are also employed,which provides a complementing way to evaluate the ability of electron content models to reproduce the spatial and temporal gradients in the ionosphere.During the test period,Jet Propulsion Laboratory(JPL)GIMs present significantly positive deviations compared to the Jason VTEC and GPS dSTEC.Technical University of Catalonia(UPC)rapid GIM UQRG exhibits the best performance in both Jason VTEC and GPS dSTEC analysis.The CAS GIMs show comparable performance with the results of the first four IAACs of the IGS.As expected,the poor performance of all GIMs is in equatorial regions and the high latitudes of the southern hemisphere.The consideration of generating multi-layer or three-dimensional ionospheric maps is emphasized to mitigate the inadequacy of ionospheric single-layer assumption in the presence of pronounced latitudinal gradients.The use of ionospheric observations from the new GNSS constellations and other space-or ground-based observation techniques is also suggested in the generation of future GIMs,given the sparse GPS/GLONASS stations in the southern hemisphere.Zishen Li Ningbo Wang Ang Liu Yunbin Yuan Liang Wang Manuel Hernández-Pajares Andrzej Krankowski Hong Yuan 2021Satellite Navigation2021,2,1:1

16Dose,Plasma Level,and Treatment Outcome Among Methadone Patients in Shanghai,China显示文摘The purpose of this study was to investigate the blood levels of methadone in participants receiving methadone for the treatment of opioid dependence.After stabilization on methadone for four weeks,blood samples from 95 participants were collected between treatment weeks 4 and 12,before and after receiving doses of methadone,and its blood levels were measured.A multiple linear regression model was used to examine me association between methadone blood levels and the outcomes of methadone maintenance treatment(MMT).Outcome differences between participants who had high(>2) or low(<2) peak-to-trough ratios were also compared using an independent sample /-test.The blood level of methadone was not correlated with the clinical outcome of MMT with the moderate range of doses given.However,the retention of patients who had a free peak-to-trough ratio >2 was significantly poorer than those whose ratio was <2.Thus,monitoring plasma methadone levels is unlikely to be effective for guiding dosing decisions in situations where compliance with MMT is already very high or when the methadone dose is no longer the dominant factor in determining the clinical outcome.However,monitoring plasma methadone levels is still helpful for guiding the dosage for patients with a rapid metabolism.Haifeng Jiang Maureen Hillhouse Jiang Du Shujun Pan Ang Alfonso Jun Wang Zhirong Zhou Weijun Yuan Walter Ling Min Zhao 2016Neuroscience Bulletin2016,32,6:1

18Diferences and similarities of high-resolution computed tomography features between pneumocystis pneumonia and cytomegalovirus pneumonia in AIDS patients显示文摘Background:Accurately diferentiating pneumocystis from cytomegalovirus pneumonia is crucial for correct therapy selection in AIDS patients.Hence,the goal of this study was to compare the computerized tomography(CT)features of pneumocystis pneumonia and cytomegalovirus pneumonia in AIDS patients and identify clinical hallmarks to accurately distinguish these two pathologies.Methods:A total of 112 AIDS patients(78 with pneumocystis pneumonia and 34 cytomegalovirus pneumonia)at Beijing Ditan Hospital from January 2017 to May 2019 were included in this study.Two experienced chest radiologists retrospectively reviewed CT images for 17 features including ground-glass opacity,consolidation,nodules,and halo sign.Binary logistic regression analyses were conducted to identify the signifcant parameters that distinguished pneumocystis pneumonia from cytomegalovirus pneumonia.Correlations were analyzed by Pearson or Spearman correlation analyses.Result were considered signifcant if P<0.05.Results:The presence of consolidation,halo signs,and nodules(all P<0.05)were signifcantly more frequent in patients with cytomegalovirus pneumonia than in those with pneumocystis pneumonia.Small nodules(32.5%in cytomegalovirus pneumonia,6.41%in pneumocystis pneumonia,P<0.001)without perilymphatic distribution were particularly common in patients with cytomegalovirus pneumonia.Large nodules were not found in any of patients with cytomegalovirus pneumonia.The presence of ground-glass opacity,reticulation,and bronchial wall thickening(all P>0.05)were common in both groups.Conclusions:Analysis of consolidation,nodules,and halo signs may contribute to the diferential diagnosis of pneumocystis pneumonia or cytomegalovirus pneumonia.However,some CT features considered typical in one or other diseases appear with similar frequency in both cohorts of AIDS patients.CT features are potentially useful for the diferential diagnosis of pneumocystis pneumonia and cytomegalovirus pneumonia in AIDS patients.Chun‑Jing Du Jing‑Yuan Liu Hui Chen Shuo Yan Lin Pu Hao‑Feng Xiong Pan Xiang Chuan‑Sheng Li Ming Zhang Ru‑Ming Xie Bu‑Dong Chen Ang Li 2020Infectious Diseases of Poverty2020,9,5:1

20Arabidopsis Profilin Isoforms,PRF1 and PRF2 Show Distinctive Binding Activities and Subcellular Distributions显示文摘Profilin is an actin-binding protein that shows complex effects on the dynamics of the actin cytoskeleton. There are five profilin isoforms in Arabidopsis thaliana L. However, it is still an open question whether these isoforms are functionally different. In the present study, two profilin isoforms from Arabidopsis, PRF1 and PRF2 were fused with green fluorescent protein (GFP) tag and expressed in Escherichia coli and A. thaliana in order to compare their biochemical properties in vitro and their cellular distributions in vivo. Biochemical analysis revealed that fusion proteins of GFP-PRF1 and GFP-PRF2 can bind to poly-L-proline and G-actin showing remarkable differences. GFP-PRF1 has much higher affinities for both poly-L-proline and G-actin compared with GFP-PRF2. Observations of living cells in stable transgenic A. thaliana lines revealed that 35S::GFP-PRF1 formed a filamentous network, while 35S::GFP-PRF2 formed polygonal meshes. Results from the treatment with latrunculin A and a subsequent recovery experiment indicated that filamentous alignment of GFP-PRF1 was likely associated with actin filaments. However, GFP-PRF2 localized to polygonal meshes resembling the endoplasmic reticulum. Our results provide evidence that Arabidopsis profilin isoforms PRF1 and PRF2 have different biochemical affinities for poly-L-proline and G-actin, and show distinctive localizations in living cells.
