Human umbilical cord mesenchymal stem cell-loaded amniotic membrane for the repair of radial nerve injury显示文摘In this study,we loaded human umbilical cord mesenchymal stem cells onto human amniotic membrane with epithelial cells to prepare nerve conduits,i.e.,a relatively closed nerve regeneration chamber. After neurolysis, the injured radial nerve was enwrapped with the prepared nerve conduit, which was fixed to the epineurium by sutures, with the cell on the inner surface of the conduit.Simultaneously,a 1.0 mL aliquot of human umbilical cord mesenchymal stem cell suspension was injected into the distal and proximal ends of the injured radial nerve with 1.0 cm intervals.A total of1.75×107cells were seeded on the amniotic membrane. In the control group, patients received only neurolysis. At 12 weeks after cell transplantation, more than 80%of patients exhibited obvious improvements in muscular strength, and touch and pain sensations. In contrast, these improvements were observed only in 55–65%of control patients. At 8 and 12 weeks, muscular electrophysiological function in the region dominated by the injured radial nerve was significantly better in the transplantation group than the control group. After cell transplantation,no immunological rejections were observed. These findings suggest that human umbilical cord mesenchymal stem cell-loaded amniotic membrane can be used for the repair of radial nerve injury.Zhi Li Hanjiao Qin Zishan Feng Wei Liu Ye Zhou Lifeng Yang Wei Zhao Youjun Li 2013Neural Regeneration Research2013,8,36:13
Characterization of disinfection byproduct formation potential in 13 source waters in China显示文摘The formation potential of four trihalomethanes (THMFP) and seven haloacetic acids (HAA 7 FP) in 13 source waters taken from four major water basin areas in China was evaluated using the simulated distribution system (SDS) chlorination method. The specific ultraviolet absorbance (SUVA 254:the ratio of UV 254 to dissolved organic carbon (DOC)),which ranged between 0.9 and 5.0 L/(mg·m),showed that the organic compounds in different source waters exhibited different reactivities with chlorine. The HAA 7 FP of source waters ranged from 20 to 448 μg/L and the THMFP ranged from 29 to 259 μg/L.The HAA 7 FP concentrations were higher than the THMFP concentrations in all but one of the samples. Therefore, the risks of haloacetic acids (HAAs) should be of concern in some source waters.TCM (chloroform) and BDCM (bromodichloromethane) were the major THM constituents, while TCAA (trichloroacetic acid) and DCAA (dichloroacetic acid) were the major HAA species.Br-THM (brominated THM species) were much higher than BrHAA (brominated HAA species),and the formation of Br-DBP (Br-THM and Br-HAA) should be of concern when the bromide concentration is over 100 μg/L.Junzhi Zhang Jianwei Yu Wei An Juan Liu Yongjing Wang Youjun Chen Jia Tai Min Yang 2011Journal of Environmental Sciences2011,23,2:13
Effects of different forms of yeast Saccharomyces cerevisiae on growth performance, intestinal development, and systemic immunity in early-weaned piglets显示文摘The present study was conducted to determine effects of different forms of yeast(Saccharomyces cerevisiae, strain Y200007) on the growth performance, intestinal development, and systemic immunity in early-weaned piglets.A total of 96 piglets(14-d old, initial average body weight of 4.5 kg) were assigned to 4 dietary treatments:(1) basal diet without yeast(Control);(2) basal diet supplemented with 3.00 g/kg live yeast(LY);(3) basal diet supplemented with 2.66 g/kg heat-killed whole yeast(HKY);and(4) basal diet supplemented with 3.00 g/kg superfine yeast powders(SFY).Diets and water were provided ad libitum to the piglets during 3-week experiment.Growth performance of piglets was measured weekly.Samples of blood and small intestine were collected at days 7 and 21 of experiment.Dietary supplementation with LY and SFY improved G:F of piglets at days 1-21 of the experiment(P<0.05) compared to Control group.Serum concentrations of growth hormone(GH),triiodothyronine(T_3),tetraiodothyronine(T_4),and insulin growth factor 1(IGF-1) in piglets at day 21 of the experiment were higher when fed diets supplemented with LY and SFY than those in Control group(P < 0.05).Compared to Control group, contents of serum urea nitrogen of piglets were reduced by the 3 yeast-supplemented diets(P< 0.05).Diets supplemented with LY increased villus height and villus-to-crypt ratio in duodenum and jejunum of piglets(P < 0.05) compared to other two groups at day 7 of the experiment.Feeding diets supplemented with LY and SFY increased(P< 0.05) serum concentrations of IgA,IL-2,and IL-6 levels in piglets compared to Control. The CD4^+/CD8^+ ratio and proliferation of T-lymphocytes in piglets fed diets supplemented with LY were increased compared to that of Control group at day 7 of the experiment(P < 0.05).In conclusion, dietary supplementation with both LY and SFY enhanced feed conversion, small intestinal development, and systemic immunity in early-weaned piglets, with better improvement in feed conversion by dietary supplementation with LY,while dietary supplementation with SFY was more effective in increasing systemic immune functions in early-weaned piglets.Zongyong Jiang Shaoyong Wei Zhilin Wang Cui Zhu Shenglan Hu Chuntian Zheng Zhuang Chen Youjun Hu Li Wang Xianyong Ma Xuefen Yang 2016Journal of Animal Science and Biotechnology2016,7,1:13
The influence of pore structure on P-& S-wave velocities in complex carbonate reservoirs with secondary storage space显示文摘Secondary storage spaces with very complex geometries are well developed in Ordovician carbonate reservoirs in the Tarim Basin, which is taken as a study case in this paper. It is still not clear how the secondary storage space shape influences the P-& S-wave velocities (or elastic properties) in complex carbonate reservoirs. In this paper, three classical rock physics models (Wyllie timeaverage equation,Gassmann equation and the Kuster-Toks z model) are comparably analyzed for their construction principles and actual velocity prediction results, aiming at determining the most favourable rock physics model to consider the influence of secondary storage space shape. Then relationships between the P-& S-wave velocities in carbonate reservoirs and geometric shapes of secondary storage spaces are discussed from different aspects based on actual well data by employing the favourable rock physics model. To explain the influence of secondary storage space shape on V P-V S relationship,it is analyzed for the differences of S-wave velocities between derived from common empirical relationships (including Castagna's mud rock line and Greenberg-Castagna V P-V S relationship) and predicted by the rock physics model. We advocate that V P-V S relationship for complex carbonate reservoirs should be built for different storage space types. For the carbonate reservoirs in the Tarim Basin, the V P-V S relationships for fractured,fractured-cavernous,and fractured-hole-vuggy reservoirs are respectively built on the basis of velocity prediction and secondary storage space type determination.Through the discussion above,it is expected that the velocity prediction and the V P-V S relationships for complex carbonate reservoirs should fully consider the influence of secondary storage space shape, thus providing more reasonable constraints for prestack inversion, further building a foundation for realizing carbonate reservoir prediction and fluid prediction.Wang Haiyang Sam Zandong Sun Yang Haijun Gao Hongliang Xiao Youjun Hu Hongru 2011Petroleum Science2011,8,4:10

A new method of formation evaluation for fractured and caved carbonate reservoirs:A case study from the Lundong area,Tarim Basin,China显示文摘The carbonate reservoirs in the Tarim Basin are characterized by low matrix-porosity,heterogeneity and anisotropy, which make it difficult to predict and evaluate these reservoirs. The reservoir formations in Lundong area experienced a series of diagenesis and tectonic evolution stages. And secondary storage spaces such as fractures and dissolution caves were developed while nearly all the primary pores have disappeared.Based on a summary of different types of storage spaces and their responses in conventional logs,FMI and full waveform sonic logs which are sensitive to different reservoirs, the comprehensive probability index (CPI) method is applied to evaluating the reservoirs and a standard of reservoir classification is established.By comparing the evaluation results with actual welllogging results, the method has proven to be practical for formation evaluation of carbonate reservoirs, especially for the fractured carbonate reservoirs. In reservoir fluid identification, the multivariate stepwise discriminant analysis (MSDA) method is introduced.Combining the CPI method and MSDA method, comprehensive formation evaluation has been performed for fractured and caved carbonate reservoirs in the Tarim Basin.Additionally,on the basis of secondary pore inversion results, another new method of formation evaluation is also proposed in the discussion part of this paper.Through detailed application result analysis, the method shows a promising capability for formation evaluation of complex carbonate reservoirs dominated by various secondary pores such as holes, caves, and cracks.Yang Haijun Sam Zandong Sun Cai Lulu Xiao Youjun WangHaiyang Luo Chunshu Hu Hongru 2011Petroleum Science2011,8,4:7
Comparison of QTLs for rice seedling morphology under different water supply conditions显示文摘The variation of seedling characteristics under different water supply conditions is strongly associated with drought resistance in rice (Oryza sativa L.) and a better elucidation of its genetics is helpful for improving rice drought resistance. Ninety-six doubled-haploid (DH) rice lines of an indica and japonica cross were grown in both flooding and upland conditions and QTLs for morphological traits at seedling stage were examined using 208 restriction fragment length polymorphism (RFLP) and 76 microsatellite (SSR) markers. A total of 32 putative QTLs were associated with the four seedling traits: average of three adventitious root lengths (ARL), shoot height (SH), shoot biomass (SW), and root to shoot dry weight ratio (RSR). Five QTLs detected were the same under control and upland conditions. The ratio between the mean value of the seedling trait under upland and flooding conditions was used for assessing drought tolerance. A total of six QTLs for drought tolerance were detected. Comparative analysis was performed for the QTLs detected in this case and those reported from two other populations with the same upland rice variety Azucena as parent. Several identical QTLs for seedling elongation across the three populations with the positive alleles from the upland rice Azucena were detected, which suggests that the alleles of Azucena might be involved in water stress-accelerated elongation of rice under different genetic backgrounds. Five cell wall-related candidate genes for OsEXP1, OsEXP2, OsEXP4, EXT, and EGase were mapped on the intervals carrying the QTLs for seedling traits.Bingsong Zheng Ling Yang Chuanzao Mao Youjun Huang Ping Wu 2008Journal of Genetics and Genomics2008,35,8:7
Methylation of RAR-β2, RASSF1A, and CDKN2A Genes Induced by Nickel Subsulfide and Nickel-carcinogenesis in Rats显示文摘Objective To investigate the expression variation of RAR‐β2, RASSF1A, and CDKN2A gene in the process of nickel‐induced carcinogenesis. Methods Nickel subsulfide (Ni 3 S 2 ) at dose of 10 mg was given to Wistar rats by intramuscular injection. The mRNA expression of the three genes in induced tumors and their lung metastasis were examined by Real‐time PCR. The methylation status of the 5' region of these genes were detected by Quantitative Real‐time methylation specific PCR. Results The mRNA expressions of the three genes both in muscle and lung tumor were decreased distinctly in comparison with normal tissue. But hypermethylation was found only in muscle tumor. Conclusion These findings suggest that loss of function or decrease of RAR‐β2, RASSF1A, and CDKN2A, as well as the hypermethylation of 5' region of these genes, are related with nickel exposure.ZHANG Jun ZHANG Jing LI MengJie Wu YouJun FAN YingYing ZHOU Yang TAN LiLi SHAO ZhiHua SHI HongJun 2011Biomedical and Environmental Sciences2011,24,2:4
RNA interference-mediated silencing of vATPase subunits A and E affect survival and development of the 28-spotted ladybeetle,Henosepilachna vigintioctopunctata显示文摘RNA interference(RNAi)has emerged as a powerful tool for developing novel management strategies for controlling insect pests. The 28-spotted ladybeetle,Henosepilachna vigintioctopunctata is one of the most important pests attacking solanaceous plants in Asia.In this study, the potential of dietary RNAi to manage H.vigintioctopunctata was investigated using both in vitro synthesized and bacterially expressed double-stranded RNAs(dsRNAs)of HvvATPase A and HvvATPase E.The expression levels of HvvATPase A and HvvATPase E were higher in Malpighian tubules than in other tissue types. The silencing of HvvATPase A and HvvATPase E led to significant mortality in H.vigintioctopunctata larvae. In addition, the ingestion of HvvATPase A and HvvATPase E significantly deterred feeding behavior and subsequently arrested the development of H.vigintioctopunctata.Notably,the bacterially expressed dsRNAs consistently caused higher mortality in larvae and adults. Finally, the nontarget effects of the dsRNAs of H.vigintioctopunctata on the predatory ladybeetle Propylaea japonica were evaluated.P.japonica 1st instar larvae were administered vATPase A and vATPase E dsRNAs from H.vigintioctopunctata and P.japonica under the worst-case scenario,in which dsGFP served as negative control. There were significant effects of dsHvvATPase A on P.japonica at the transcriptional level but not at the organismal level, whereas dsHvvATPase E did not effect P.japonica at either the transcriptional or the organismal level.Collectively, the results of the study suggest that HvvATPase A and HvvATPase E can act as novel molecular targets for the control of H.vigintioctopunctata.Wei Guo Mujuan Guo Chunxiao Yang Zhuoqi Liu Shimin Chen Jing Lu Baoli Qiu Youjun Zhang Xuguo Zhou Huipeng Pan 2021Insect Science2021,28,6:3
Spatiotemporal distributions and environmental drivers of diversity and community structure of nosZ-type denitrifiers and anammox bacteria in sediments of the Bohai Sea and North Yellow Sea, China显示文摘Denitrification and anammox processes are major nitrogen removal processes in coastal ecosystems. However, the spatiotemporal dynamics and driving factors of the diversity and community structure of involved functional bacteria have not been well illustrated in coastal environments, especially in human-dominated ecosystems. In this study, we investigated the distributions of denitrifiers and anammox bacteria in the eutrophic Bohai Sea and the northern Yellow Sea of China in May and November of 2012 by constructing clone libraries employing nosZ and 16S rRNA gene biomarkers. The diversity of nosZ-denitrifier was much higher at the coastal sites compared with the central sites, but not significant among basins or seasons. Alphaproteobacteria were predominant and prevalent in the sediments, whereas Betaproteobacteria primarily occurred at the site near the Huanghe (Yellow) River estuary. Anammox bacteria Candidatus Scalindua was predominant in the sediments, and besides, Candidatus Brocadia and Candidatus Kuenenia were also detected at the site near the Huanghe River estuary that received strong riverine and anthropogenic impacts. Salinity was the most important in structuring communities of nosZ-denitrifier and anammox bacteria. Additionally, anthropogenic perturbations (e.g. nitrogen overloading and consequent high primary productivity, and heavy metal discharges) contributed significantly to shaping community structures of denitrifier and anammox bacteria, suggesting that anthropogenic activities would influence and even change the ecological function of coastal ecosystems.CAI Youjun ZHANG Xiaoli LI Guihao DONG Jun YANG Anjing WANG Guangyu ZHOU Xiaojian 2019Journal of Oceanology and Limnology2019,37,4:2
The Gravitational-wave physics Ⅱ: Progress显示文摘It has been a half-decade since the first direct detection of gravitational waves, which signifies the coming of the era of the gravitational-wave astronomy and gravitational-wave cosmology. The increasing number of the detected gravitational-wave events has revealed the promising capability of constraining various aspects of cosmology, astronomy, and gravity. Due to the limited space in this review article, we will briefly summarize the recent progress over the past five years, but with a special focus on some of our own work for the Key Project 'Physics associated with the gravitational waves' supported by the National Natural Science Foundation of China. In particular,(1) we have presented the mechanism of the gravitational-wave production during some physical processes of the early Universe, such as inflation, preheating and phase transition, and the cosmological implications of gravitational-wave measurements;(2) we have put constraints on the neutron star maximum mass according to GW170817 observations;(3) we have developed a numerical relativity algorithm based on the finite element method and a waveform model for the binary black hole coalescence along an eccentric orbit.Ligong Bian Rong-Gen Cai Shuo Cao Zhoujian Cao He Gao Zong-Kuan Guo Kejia Lee Di Li Jing Liu Youjun Lu Shi Pi Jian-Min Wang Shao-Jiang Wang Yan Wang Tao Yang Xing-Yu Yang Shenghua Yu Xin Zhang 2021Science China(Physics,Mechanics & Astronomy)2021,64,12:2
Simulation of blast induced crater in jointed rock mass by discontinuous deformation analysis method显示文摘Rock blasting is a dynamic process accom panied with the propagations of shock waves and the dispersion of the explosion gas. This paper adopts the discontinuous deformation analysis(DDA)method to simulate the rock blasting process.A dynamic parameter adjustment and the non-reflecting boundary condition are implemented in the DDA method. The sub-block DDA method to simulate fracture problems is used. The blasting process in jointed rock mass is simulated by application of the explosion gas pressure on the expanding borehole walls and induced connected fracture surfaces around the boreholes. The blast craters with different overburdens are derived. The whole process including the explosion gas dispersion, borehole expansion, rock mass failure and cast, and the formation of the final blasting piles in rock blasting are well reproduced numerically.Parametric study for different overburdens is carried out,and the results are analyzed and discussed.Youjun NING Jun YANG Xinmei AN Guowei MA 2010Frontiers of Structural and Civil Engineering2010,4,2:2
Ring-restricted N-nitrosated rhodamine as a green-light triggered,orange-emission calibrated and fast-releasing nitric oxide donor显示文摘Nitric oxide(NO) donors are versatile tools for nitric oxide biology. The biological response of NO is dependent on the transient concentration and the sustained duration. N-Nitrosated rhodamines are photo-triggered and photo-calibrated NO donors. We recently discovered that suppression of the dihedral angle between the N-nitroso fragment with the rhodamine scaffold facilitates NO release.Inspired by this discovery, we developed a fast-releasing NO donor (NOD575) suitable for biological applications, e.g., the pulmonary arterial smooth muscle cells(PASMCs).Haihong He Tingting He Ziqian Zhang Xiu Xu Huibin Yang Xuhong Qian Youjun Yang 2018Chinese Chemical Letters2018,29,10:2
A general approach to the design of high-performance near-infrared (NIR) D-π-A type fluorescent dyes显示文摘Near infrared(NIR) absorbing and emitting dyes are sought after for their potentials in bioimaging and theranostic applications. They are typically not as stable as dyes absorbing and emitting in the visible spectral range, as the result of a reduced HOMO-LUMO band-gap. Also, they are not as efficient fluorescence emitters due to accelerated internal conversion kinetics. In addition, their large conjugative backbone render them high tendency to form aggregate and low aqueous solubility. In this tutorial, we have described a four-step approach for rational design of organic dyes with an overall high-performance to meet the rigorous requirements of biological applications. Also, some background regarding 'NIR' is provided along with some recent breakthroughs of the field.Xiao Luo Jin Li Jie Zhao Luyan Gu Xuhong Qian Youjun Yang 2019Chinese Chemical Letters2019,30,4:2

Ultra-robust stretchable electrode for e-skin:In situ assembly using a nanofiber scaffold and liquid metal to mimic water-to-net interaction显示文摘The development of stretchable electronics could enhance novel interface structures to solve the stretchability-conductivity dilemma, which remains a major challenge.Herein,we report a nano-liquid metal(LM)-based highly robust stretchable electrode(NHSE)with a self-adaptable interface that mimics water-tonet interaction.Based on the in situ assembly of electrospun elastic nanofiber scaffolds and electrosprayed LM nanoparticles, the NHSE exhibits an extremely low sheet resistance of 52 mΩsq^(-1).It is not only insensitive to a large degree of mechanical stretching(i.e.,350%electrical resistance change upon 570%elongation)but also immune to cyclic deformation(i.e.,5%electrical resistance increases after 330000 stretching cycles with 100%elongation).These key properties are far superior to those of the state-of-the-art reports. Its robustness and stability are verified under diverse circumstances, including long-term exposure to air(420 days),cyclic submersion(30000 times),and resilience against mechanical damages. The combination of conductivity, stretchability, and durability makes the NHSE a promising conductor/electrode solution for flexible/stretchable electronics for applications such as wearable on-body physiological signal detection,human-machine interaction, and heating e-skin.Jinwei Cao Fei Liang Huayang Li Xin Li Youjun Fan Chao Hu Jing Yu Jin Xu Yiming Yin Fali Li Dan Xu Hanfang Feng Huali Yang Yiwei Liu Xiaodong Chen Guang Zhu Run-Wei Li 2022InfoMat2022,4,4:1
Puerarin improves diabetic wound healing via regulation of macrophage M2 polarization phenotype显示文摘Background:Skin wound healing depends on the progress of different but overlapping stages of healing, including hemostasis, inflammatory, proliferative and remodeling.Failure of these stages to occur in a timely and gradual manner may result in non-healing pathological wounds.Macrophages and neutrophils have been shown to play an essential role in the inflammatory responses of wound tissue, and their active plasticity allows them to modulate tissue damage and repair functions. The ability of macrophages and neutrophils to regulate the occurrence and resolution of inflammatory processes is essential for the treatment of pathological wound healing. Methods:Mice were categorized into negative control, streptozotocin, streptozotocin+puerarin and puerarin groups. The traditional Chinese medicine extract puerarin was selected to treat different groups of mice with a full-thickness skin defect wound.Cells of the RAW264.7 cell line were stimulated under different puerarin conditions. Then, real time quantitative polymerase chain reaction(RT-qPCR),western blot, immunofluorescence and other assays were carried out to explore the effect of puerarin on wound healing and its molecular mechanism. Results:Animal experiments found that the wound healing of diabetic mice treated with puerarin was significantly accelerated, and histological analysis found that puerarin treatment markedly decreased the infiltration of macrophages and neutrophils in wound tissue.Through western blot,RT-qPCR and immunofluorescence experiments,it was observed that puerarin treatment remarkably inhibited nuclear factor kinase B(NF-κB)and mitogen-activated protein kinase(MAPK)signaling pathways, downregulated the expression of inflammatory cytokines and induced the M2 polarization of macrophages. At the cellular level,we also observed that puerarin improved M2 macrophage polarization and inhibited inflammatory pathway activation in a high-glucose culture. Conclusion:Puerarin has a significant therapeutic effect on wound healing in diabetic mice. The therapeutic effect is achieved by regulating macrophage polarization through suppressing NF-κB and MAPK signaling cascades.Shiyan Li Ping Yang Xiaofeng Ding Hao Zhang Youjun Ding Qian Tan 2022Burns & Trauma2022,10,1:1

An ischemic area-targeting,peroxynitrite-responsive,biomimetic carbon monoxide nanogenerator for preventing myocardial ischemia-reperfusion injury显示文摘Myocardial ischemia-reperfusion (MI/R) injury is common in patients who undergo revascularization therapy for myocardial infarction, often leading to cardiac dysfunction. Carbon monoxide (CO) has emerged as a therapeutic molecule due to its beneficial properties such as anti-inflammatory, anti-apoptotic, and mitochondrial biogenesis-promoting properties. However, its clinical application is limited due to uncontrolled release, potential toxicity, and poor targeting efficiency. To address these limitations, a peroxynitrite (ONOO )-triggered CO donor (PCOD585) is utilized to generate a poly (lactic-co-glycolic acid) (PLGA)-based, biomimetic CO nanogenerator (M/PCOD@PLGA) that is coated with the macrophage membrane, which could target to the ischemic area and neutralize proinflammatory cytokines. In the ischemic area, local produced ONOO triggers the continuous release of CO from M/PCOD@PLGA, which efficiently ameliorates MI/R injury by clearing harmful ONOO , attenuating the inflammatory response, inhibiting cardiomyocyte apoptosis, and promoting mitochondrial biogenesis. This study provides a novel insight into the safe therapeutic use of CO for MI/R injury by utilizing a novel CO donor combined with biomimetic technology. The M/PCOD@PLGA nanogenerator offers targeted delivery of CO to the ischemic area, minimizing potential toxicity and enhancing therapeutic efficacy.Jinyan Zhang Liwei Liu Zhen Dong Xicun Lu Wenxuan Hong Jin Liu Xiaoyi Zou Jinfeng Gao Hao Jiang Xiaolei Sun Kai Hu Youjun Yang Junbo Ge Xiao Luo Aijun Sun 2023Bioactive Materials2023,,10:1
