| 1 | Meso-damage cracking characteristics analysis for rock and soil aggregate with CT test显示文摘The aim of this paper is to investigate the damage cracking characteristics of rock and soil aggregate(RSA)by X-ray computed tomography(CT)under uniaxial compressive loading.The mean CT value for the region of interest(ROI)is used to analyze the cracking characteristics.Also,the mathematical morphology method based on the image threshold segmentation is used to obtain characteristic parameters of cracks to describe the cracking evolution of RSA.Results show that the elastic mismatch between rock blocks and soil matrix is the primary reason for RSA cracking.The mean CT value for the RSA specimen,rock block inclusions,and their adjacent soil regions decreases with the increasing stress level.However,it is more sensitive for block inclusions than soil regions.Using the image segmentation method,length,area and mean width of cracks obey to power function distribution.Crack statistical characteristics are closely related to the rock block’s distribution and morphology.These results may be useful to reveal the mesoscopic cracking mechanism,establish meso-damage evolution equation,and constitutive relation for RSA. | WANG Yu LI Xiao ZHANG Bo WU YanFang | 2014 | Science China(Technological Sciences)2014,57,7: | 21 |
| 2 | Enhancing the antihepatitis B virus immune response by adefovir dipivoxil and entecavir therapies显示文摘Chronicity of hepatitis B(CHB)infection is characterized by a weak immune response to the virus.Entecavir(ETV)and adefovir dipivoxil(ADV)are effective in suppressing hepatitis B virus(HBV)replication.However,the underlying immune mechanism in the antiviral response of patients treated with nucleoside or nucleotide analogs is not clearly understood.In this study,regulatory T cells(Tregs)and intracellular cytokines,including IL-2,interferon(IFN)-γ,tumor-necrosis factor(TNF)-α and IL-4,were measured prior to and at 12,24,36 and 48 weeks after treatment with ETV or ADV.The cytokines were increased from 24 to 48 weeks after treatment.Higher levels of Th1 cytokines were observed with ETV(n=29)versus ADV(n=28)treatment.By contrast,the numbers of Tregs in both groups were decreased.The altered cytokine profile and cellular component was accompanied by a decrease in HBV DNA levels in both groups,which may contribute to their therapeutic effect in CHB infection.Our findings suggest that the antiviral effect of the drugs may be attributed not only to their direct effect on virus suppression but also to their immunoregulatory capabilities. | Yanfang Jiang Wanyu Li Lei Yu Jingjing Liu Guijie Xin Hongqing Yan Pinghui Sun Hong Zhang Damo Xu Junqi Niu | 2011 | Cellular & Molecular Immunology2011,8,1: | 16 |
| 3 | One-step base editing in multiple genes by direct embryo injection for pig trait improvement显示文摘The precise and simultaneous acquisition of multiple beneficial alleles in the genome is in great demand for the development of elite pig breeders. Cytidine base editors(CBEs) that convert C:G to T:A have emerged as powerful tools for single-nucleotide replacement. Whether CBEs can effectively mediate C-to-T substitution at multiple sites/loci for trait improvement by direct zygote injection has not been verified in large animals. Here, we determined the editing efficiency of four CBE variants in porcine embryonic fibroblast cells and embryos. The findings showed that hA3A-BE3-Y130F and hA3A-eBE-Y130F consistently resulted in increased base-editing efficiency and low toxic effects in embryonic development. Further, we verified that using a one-step approach, direct zygote microinjection of the CBE system can generate pigs harboring multiple point mutations.Our process resulted in a stop codon in CD163 and myostatin(MSTN) and introduced a beneficial allele in insulin-like growth factor-2(IGF2). The pigs showed disrupted expression of CD163 and MSTN and increased expression of IGF2, which significantly improved growth performance and infectious disease resistance. | 4 | Targeted delivery of chlorogenic acid by mannosylated liposomes to effectively promote the polarization of TAMs for the treatment of glioblastoma显示文摘Tumor-associated macrophages(TAMs)generally display an immunosuppressive M2 phenotype and promote tumor progression and metastasis,suggesting their potential value as a target in cancer immunotherapy.Chlorogenic acid(CHA)has been identified as a potent immunomodulator that promotes the polarization of TAMs from an M2 to an M1 phenotype.However,rapid clearance in vivo and low tumor accumulation have compromised the immunotherapeutic efficacy of CHA in clinical trials.In this study,mannosylated liposomes are developed for targeted delivery of CHA to TAMs.The immunoregulatory effects of CHA,along with the overall antitumor efficacy of CHA-encapsulated mannosylated liposomes,are investigated through in vitro and in vivo experiments.The prepared CHA-encapsulated mannosylated liposomes exhibit an ideal particle size,favorable stability,and preferential accumulation in tumors via the mannose receptor-mediated TAMs-targeting effects.Further,CHA-encapsulated mannosylated liposomes inhibit G422 glioma tumor growth by efficiently promoting the polarization of the pro-tumorigenic M2 phenotype to the anti-tumorigenic M1 phenotype.Overall,these findings indicate that CHA-encapsulated mannosylated liposomes have great potential to enhance the immunotherapeutic efficacy of CHA by inducing a shift from the M2 to the M1 phenotype. | Jun Ye Yanfang Yang Jing Jin Ming Ji Yue Gao Yu Feng Hongliang Wang Xiaoguang Chen Yuling Liu | 2020 | Bioactive Materials2020,5,3: | 9 |

| 6 | Estrogen and insulin synergistically promote endometrial cancer progression via crosstalk between their receptor signaling pathways显示文摘Objective: Despite evidence that estrogens and insulin are involved in the development and progression of many cancers, their synergistic role in endometrial carcinoma(EC) has not been analyzed yet.Methods: Here, we investigated how estrogens act synergistically with insulin to promote EC progression. Cell growth in vitro and in vivo, effects of estradiol and insulin on apoptosis and cell cycle distribution, and expression and activation of estrogen receptor(ER), insulin receptor(InsR), and key proteins in the PI3K and MAPK pathways were examined after combined stimulation with estradiol and insulin.Results: Compared to EC cells treated with estradiol or insulin alone, those treated with both estradiol and insulin exhibited stronger stimulation. Estradiol significantly induced phosphorylation of InsR-β and IRS-1, whereas insulin significantly induced phosphorylation of ER-α. In addition, treatment with both insulin and estradiol together significantly increased the expression and phosphorylation of Akt, MAPK, and ERK. Notably, InsR-β inhibition had a limited effect on estradiol-dependent proliferation,cell cycle, and apoptosis, whereas ER-α inhibition had a limited insulin-dependent effect, in EC cell lines. | 7 | Feasibility of Tea Saponin-Enhanced Soil Washing in a Soybean Oil-Water Solvent System to Extract PAHs/Cd/Ni Efficiently from a Coking Plant Site显示文摘Mixed contaminated brownfield sites have brought serious risks to human health and environmental safety. With the purpose of removing polycyclic aromatic hydrocarbons(PAHs) and heavy metals from a coking plant site, an innovative technology for ex-situ washing was developed in the present work. The combination of 15.0 mL L^(-1) soybean oil and 7.5 g L^(-1) tea saponin proved an effective method to extract co-pollutants from soil. After two consecutive washing cycles, the efficiency rates of removal for 3-, 4-,5(+6)-ring, and total PAHs, Cd, and Ni were approximately 98.2%, 96.4%, 92.3%, 96.3%, 94.1%, and 89.4%, respectively. Meanwhile,as evaluated by Tenax extraction method and metal stability indices, the residual PAHs and heavy metals after consecutive washing mainly existed in the form with extremely low bioaccessibility in the soil. Thus, in the soil after two washing cycles, there appeared limited environmental transfer risk of co-pollutants. Moreover, a subsequent precipitation method with alkaline solution and PAHdegrading strain Sphingobium sp. PHE9 inoculation effectively removed 84.6%–100% of Cd, 82.5%–91.7% of Ni, and 92.6%–98.4% of PAHs from the first and second washing solvents. The recovered solvents also exhibited a high recycling effectiveness. | 8 | Effect of artificially accelerated aging on the vigor of Metasequoia glyptostroboides seeds显示文摘Metasequoia glyptostroboides is an endangered relict plant native to China that has been widely introduced into many countries and areas around the world. However,its seeds germinate at a very low percentage. Consequently,population regeneration by seed is low under natural conditions, which probably contributes to the endangered status of this plant species. The present study aimed to describe the aging mechanism of M. glyptostroboides seeds. Our objective was to elucidate causes of the low germination rate in an effort to enhance potential for conservation of the species. We used germination tests,relative electrical conductivity and malondialdehyde content determination, ultrastructural observation of embryo cells, and analysis of superoxide dismutase, catalase,ascorbate peroxidase, and dehydroascorbate reductase activities during accelerated aging treatment. | 10 | Construction of chlorogenic acid-containing liposomes with prolonged antitumor immunity based on T cell regulation显示文摘As a potential cancer immunotherapeutic agent,chlorogenic acid(CHA)has entered phase II clinical trials in China as a lyophilized powder formulation for treating glioma.However,the in vivo instability of CHA necessitates daily intramuscular injections,resulting in patient noncompliance.In this study,CHA-phospholipid complex(PC)-containing PEGylated liposomes(CHA-PC PEG-Lipo,named as CPPL),with CHA-PC as the drug intermediate,were prepared to lower the administration frequency.CPPL demonstrated excellent physicochemical properties,enhanced tumor accumulation,and inhibited tumor growth even when the administration interval was prolonged to 4 days when compared to a CHA solution and CHA-PC loaded liposomes(CHA-PC Lipo,labeled as CPL),both of which only demonstrated antitumor efficacy with once-daily administration.Further evaluation of the in vivo antitumor immune mechanism suggested that the extended antitumor immune efficacy of CPPL could be attributed to its distinct immune-stimulating mechanism when compared with CHA solution and CPL,such as stimulating both CD4+and CD8+T cell infiltration,inhibiting myeloid-derived suppressor cell expression,reducing the expression of Th2 related factors,and notably,increasing the memory T cells in tumor tissues.This CHA-containing formulation could reduce the frequency of in vivo CHA administration during cancer treatment via T cells,especially memory T cell regulation. | Yun Zhang Yanfang Yang Jun Ye Yue Gao Hengfeng Liao Junzhuo Zhou Yu Feng Dongdong Liu Yingying Meng Xiaoguang Chen Lili Gao Yuling Liu | 2021 | Science China(Life Sciences)2021,64,7: | 2 |
