2Exotic origin of the Chinese continental shelf: new insights into the tectonic evolution of the western Pacific and eastern China since the Mesozoic显示文摘The effect of paleo-Pacific subduction on the geological evolution of the western Pacific and continental China is likely complex. Nevertheless, our analysis of the distribution of Mesozoic granitoids in the eastern continental China in space and time has led us to an interesting conclusion: The basement of the continental shelf beneath East and South China Seas may actually be of exotic origin geologically unrelated to the continental lithosphere of eastern China. By accepting the notion that the Jurassic–Cretaceous granitoids in the region are genetically associated with western Pacific subduction and the concept that subduction may cease to continue only if the trench is being jammed, then the termination of the granitoidmagmatism throughout the vast region at *88 ± 2 Ma manifests the likelihood of ‘‘sudden'', or shortly beforehand(*100 Ma), trench jam of the Mesozoic western Pacific subduction. Trench jam happens if the incoming‘‘plate'' or portion of the plate contains a sizeable mass that is too buoyant to subduct. The best candidate for such a buoyant and unsubductable mass is either an oceanic plateau or a micro-continent. We hypothesize that the basement of the Chinese continental shelf represents such an exotic, buoyant and unsubductable mass, rather than seaward extension of the continental lithosphere of eastern China. The locus of the jammed trench(i.e., the suture) is predictably located on the shelf in the vicinity of, and parallel to, the arc-curved coastal line of the southeast continental China. It is not straightforward to locate the locus in the northern section of the East China Sea shelf because of the more recent(\20 Ma) tectonic re-organization associated with the opening of the Sea of Japan. We predict that the trench jam at *100 Ma led to the re-orientation of the Pacific plate motion in the course of NNW direction as inferred from the age-progressive Emperor Seamount Chain of Hawaiian hotspot origin(its oldest unsubdued Meiji and Detroit seamounts are *82 Ma),making the boundary between the Pacific plate and the newly accreted plate of eastern Asia a transform fault at the location east of the continental shelf of exotic origin. This explains the apparent *40 Myr magmatic gap from *88to *50 Ma prior to present-day western Pacific subduction initiation. We propose that basement penetration drilling on well-chosen sites is needed to test the hypothesis in order to reveal the true nature of the Chinese continental shelf basement. This testing becomes critical and cannot longer be neglected in order to genuinely understand the tectonic evolution of the western Pacific and its effect on the geology of eastern China since the Mesozoic, includingthe cratonic lithosphere thinning, related magmatism/mineralization, and the mechanism of the subsequent South China Sea opening, while also offering novel perspectives on aspects of the plate tectonics theory. We also suggest the importance of future plate tectonic reconstruction of the western Pacific to consider the nature and histories of the Chinese continental shelf of exotic origin as well as the probable transform plate boundary from *100 to*50 Ma. 4Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis显示文摘Objective PERK/eI F2α/CHOP is a major signaling pathway mediating endoplasmic reticulum(ER) stress related with atherosclerosis.Oxidized LDL(ox-LDL) also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis.The present study was conducted to explore the regulatory effect of ox-LDL on PERK/e IF2α/CHOP signaling pathway in vascular endothelial cells.Methods The effects of ox-LDL on PERK and p-e IF2α protein expression of primary human umbilical vein endothelial cells(HUVECs) were investigated by Western blot analysis.PERK gene silencing and selective eI F2α phosphatase inhibitor,salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis,caspase-3 activity,and CHOP mR NA level.Results Ox-LDL treatment significantly increased the expression of PERK,PERK-mediated inactivation of e IF2α phosphorylation,and the expression of CHOP,as well as the caspase-3 activity and apoptosis.The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral sh RNA or by selective eI F2α phosphatase inhibitor,salubrinal.Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/eI F2α/CHOP ER-stress pathway.It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis.TAO Yong Kang YU Pu Lin BAI Yong Ping YAN Sheng Tao ZHAO Shui Ping ZHANG Guo Qiang 2016Biomedical and Environmental Sciences2016,29,12:21

6Isolation and Characterization of an Algicidal Bacterium Indigenous to Lake Taihu with a Red Pigment able to Lyse Microcystis aeruginosa显示文摘Objective To isolate and characterize indigenous algicidal bacteria and their algae-lysing compounds active against Microcystis aeruginosa, strains TH1, TH2, and FACHB 905. Methods The bacteria were identified using the Biolog automated microbial identification system and 16S rDNA sequence analysis. The algae-lysing compounds were isolated and purified by silica gel column chromatography and reverse-phase high performance liquid chromatography. Their structures were confirmed by Nuclear Magnetic Resonance (NMR) and Fourier Transform Infrared (FT-IR) spectroscopy. Algae-lysing activity was observed using microscopy. Results The algae-lysing bacterium LTH-2 isolated from Lake Taihu was identified as Serratia marcescens. Strain LTH-2 secreted a red pigment identified as prodigiosin (C 20 H 25 N 3 O), which showed strong lytic activity with algal strains M. aeruginosa TH1, TH2, and FACHB 905 in a concentration-dependent manner. The 50% inhibitory concentration (IC 50 ) of prodigiosin with the algal strains was 4.8 (±0.4)×10 -2 μg/mL, 8.9 (±1.1)×10-2 μg/mL, and 1.7 (±0.1)×10 -1 μg/mL in 24 h, respectively. Conclusion The bacterium LTH-2 and its pigment had strong Microcystis-lysing activity probably related to damage of cell membranes. 7A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide.Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen 2019Molecular Plant2019,12,5:18
8Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 9Sequential invasive-noninvasive mechanical ventilation weaning strategy for patients after tracheostomy显示文摘BACKGROUND: Because the continuity and integrity of the trachea are likely damaged to some extent after tracheostomy, the implementation of sequential ventilation has certain difficulties, and sequential invasive-noninvasive ventilation on patients after tracheostomy is less common in practice. The present study aimed to investigate the feasibility of invasive-noninvasive sequential weaning strategy in patients after tracheostomy.METHODS: Fifty patients including 24 patients with withdrawal of mechanical ventilation(conventional group) and 26 patients with sequential invasive-noninvasive weaning by directly plugging of tracheostomy(sequential group) were analyzed retrospectively after appearance of pulmonary infection control(PIC) window. The analysis of arterial blood gases, ventilator-associated pneumonia(VAP) incidence, the total duration of mechanical ventilation, the success rate of weaning and total cost of hospitalization were compared between the two groups.RESULTS: Arterial blood gas analysis showed that the sequential weaning group was better than the conventional weaning group 1 and 24 hours after invasive ventilation. 11Neuroprotection of Cyperus esculentus L. orientin against cerebral ischemia/reperfusion induced brain injury显示文摘Orientin is a flavonoid monomer.In recent years,its importance as a source of pharmacological active substance is growing rapidly due to its properties such as anti-myocardial ischemia,anti-apoptosis,anti-radiation,anti-tumor,and anti-aging.However,the neuroprotective effects of Orientin on stroke injury have not been comprehensively evaluated.The aim of the present study was thus to investigate the neuroprotective capacity and the potential mechanisms of Cyperus esculentus L.orientin(CLO)from Cyperus esculentus L.leaves against ischemia/reperfusion(I/R)injury using standard orientin as control.For in vitro studies,we treated HT22 cells with CoCl2 as an in vitro ischemic injury model.HT22 cells in the control group were treated with CoCl2.For in vivo studies,we used rat models of middle cerebral artery occlusion,and animals that received sham surgery were used as controls.We found that CLO protected CoCl2-induced HT22 cells against ischemia/reperfusion injury by lowering lipid peroxidation and reactive oxygen species formation as well as decreasing protein oxidation.However,CLO did not reduce the release of lactate dehydrogenase nor increase the activity of superoxide dismutase.Results showed that CLO could decrease neurological deficit score,attenuate brain water content,and reduce cerebral infarct volume,leading to neuroprotection during cerebral ischemia-reperfusion injury.Our studies indicate that CLO flavonoids can be taken as a natural antioxidant and bacteriostastic substance in food and pharmaceutical industry.The molecular mechanisms of CLO could be at least partially attributed to the antioxidant properties and subsequently inhibiting activation of casepase-3.All experimental procedures and protocols were approved on May 16,2016 by the Experimental Animal Ethics Committee of Xinjiang Medical University of China(approval No.IACUC20160516-57).Si-Qun Jing Sai-Sai Wang Rui-Min Zhong Jun-Yan Zhang Jin-Zi Wu Yi-Xian Tu Yan Pu Liang-Jun Yan 2020Neural Regeneration Research2020,15,3:16

16The therapeutic effects of recombinant adenovirus RA538 on human gastric carcinoma cells in vitro and in vivo显示文摘AIM To evaluate the potential of RA-538 genetherapy for gastric carcinoma.METHODS Human gastric carcinoma cell lineSGC7901 treated with Ad-RA538 or Ad-LacZ wereanalysed by X-gal stain,MTT,DNA ladder,Tunel,flow cytometric analysis,PCR,andWestern Blot in vitro.The tumorigenicity andexperimental therapy in nude mice model wereassessed in vivo.RESULTS Ad-LacZ could efficiently transferthe LacZ gene into SGC7901 cells.X-gal-positivecells at MOI 25,50,100,and 200 were 90%,100%,100%,and 100% respectively.Ad-RA538could strongly inhibit cell growth and inducedapoptosis in SGC7901 cells.The proliferation ofthe Ad-RA538-infected SGC7901 cells wasreduced by 76.3%.The mechanism of killing ofgastric carcinoma cells by Ad-RA538 was foundto be apoptosis by DNA ladder,Tunel and flowcytometric analysis.The tumorigenicity in nudemice using Ad-RA538 showed that all three micefailed to form tumor from 7 to 30 days comparedwith Ad-LacZ and parent SGC7901 cells.Experimental therapy on the nude mice modelbearing subcutaneous tumor of SGC790| cells showed that intratumor instillation of Ad-RA538inhibited the growth of the tumors.Ad-RA538-treated tumors were inhibited by 60.66 %,compared with that of the tumor injected withAd-LacZ and mock.CONCLUSION The expression of Ad-RA538 can inhibit growth and induce apoptosis of gastric cancer cell in vitro and in vivo. 17Expression of Bmi-1 and PAI-1 in esophageal squamous cell carcinoma显示文摘AIM:To determine the correlation between invasiveness,migration and prognosis in esophageal squamous cell carcinoma(ESCC)and expression of the B-cellspecific Moloney leukemia virus insert site 1(Bmi-1)and plasminogen activator inhibitor-1(PAI-1).METHODS:Eighty previously untreated patients who underwent surgical excision of ESCC were included.The expression of Bmi-1 and PAI-1 was examined immunohistochemically in formalin-fixed paraffinembedded primary tissue specimens.The relationships between the expression of Bmi-1 and PAI-1,the clinicopathologic features of ESCC,and the survival rate of ESCC patients were also discussed.The correlation between Bmi-1 and PAI-1 protein expression in ESCC was analyzed.The relationship between Bmi-1 and PAI-1expression and ESCC prognosis was evaluated using a Cox regression model and Kaplan-Meier survival curve analysis.RESULTS:The rates of positive Bmi-1 and PAI-1 expression in ESCC were higher than those in normal esophageal tissue(P<0.05).The expression of Bmi-1and PAI-1 was correlated with depth of invasion and lymph node metastasis(P<0.05),but not with patient age,tumor size or nationality(P>0.05).The expression of Bmi-1 was positively correlated with that of PAI-1(P<0.05).The 10-year overall survival rate for all patients was 20%(16∕80).Univariate KaplanMeier survival analysis showed that patients with high expression of esophageal PAI-1 and Bmi-1 had lower survival,however,the difference was not statistically significant.Cox multivariate analysis showed that PAI-1and Bmi-1 were not independent factors for survival rate,while the depth of tumor invasion and metastasis were independent factors affecting patient survival.CONCLUSION:The expression of Bmi-1 and PAI-1plays a role in ESCC progression,and may be used as a prognostic marker in ESCC.Yan Zhang Ya-Lou Zhang Hong-Ming Chen Hong-Wei Pu Wen-Jing Ma Xiao-Mei Li Hong Ma Xiao Chen 2014World Journal of Gastroenterology2014,20,18:12
18Endovascular treatment with or without intravenous alteplase for acute ischaemic stroke due to basilar artery occlusion显示文摘Background and purpose It remains controversial if endovascular treatment(EVT)can improve the outcome of patients with acute basilar artery occlusion(BAO).This study aims to compare the functional outcomes between EVT with and without intravenous thrombolysis(IVT)first in patients who had acute ischaemic stroke(AIS)due to BAO.Methods Patients who had AIS with BAO who underwent EVT within 24 hours of onset were enrolled in this multicentre cohort study,and the efficacy and safety were compared between IVT+EVT and direct EVT.The primary outcome was 90-day functional independence.All outcomes were assessed with adjusted OR(aOR)from the multivariable logistic regression.In addition,a meta-analysis was performed on all recently published pivotal studies on functional independence after EVT in patients with BAO.Results Of 310 enrolled patients with BAO,241(78%)were treated with direct EVT and 69(22%)with IVT+EVT.Direct EVT was associated with a worse functional outcome(aOR,0.46(95%CI 0.24 to 0.85),p=0.01).IVT+EVT was associated with a lower percentage of patients who needed≥3 passes of stent retriever(10.14%vs 20.75%).The meta-analysis regression revealed a potential positive correlation between bridging with IVT first and functional independence(r=0.14(95%CI 0.05 to 0.24),p<0.01).Conclusions This study showed that compared with direct EVT,EVT with IVT first was associated with better functional outcomes in patients with BAO treated within 24 hours of onset.The meta-analysis demonstrated similar favourable efficacy of IVT first followed by EVT in patients with BAO.Ximing Nie David Wang Yuehua Pu Yufei Wei Qixuan Lu Hongyi Yan Xin Liu Lina Zheng Jingyi Liu Xinxuan Yang Yarong Ding Dacheng Liu Wanying Duan Zhe Zhang Zhonghua Yang Miao Wen Weibin Gu Xinyi Hou Xinyi Leng Yuesong Pan Zhongrong Miao Liping Liu 2022Stroke & Vascular Neurology2022,7,3:10
