Relationship between T-lymphocyte cytokine levels and sero-response to hepatitis B vaccines显示文摘AIM: To investigate the cellular defects by analyzing the (Th1/Th2) cytokine levels in vaccine responders and non-responders. METHODS: Peripheral blood mononuclear cell (PBMC) from responders and non-responders were stimulated with or with out recombinant HBsAg or PHA. Broad spectrum of cytokines viz (Th1) IFN-γ, IL-2, TNF-α, IL-12 and (Th2) IL-10, IL-4 were measured after in vitro stimulation with recombinant HBsAg and were compared with respective antibody titers. RESULTS: A significant decrease (P = 0.001) in Th1 and Th2 cytokines namely, IL-2, INF-γ, TNF-α and IL-10in non-responders was observed. The level of IL-4 was not significant between the three groups. Furthermore, despite a strong Th1 and Th2 cytokine response, the level of IL-12 was elevated in high-responders compared to other groups (P = 0.001) and demonstrated a positive correlation with anti-HBs titers and Th1 cytokine response. CONCLUSION: Our findings suggest that unrespon-siveness to recombinant hepatitis B vaccines (rHB) is multifactorial, including specific failure of antigen presentation or the lack of both T helper Th1 and Th2 response.Vijayakumar Velu Shanmugam Saravanan Subhadra Nandakumar Esaki Muthu Shankar Appasamy Vengatesan Suresh Sakharam Jadhav Prasad Suryakant Kulkarni Sadras Panchatcharam Thyagarajan 2008World Journal of Gastroenterology2008,14,22:22
Role of essential oils in food safety: Antimicrobial and antioxidant applications显示文摘Essential oils(EOs)are more complex and comprise a number of volatile and natural bioactive compounds, which often used in food industries as the best alternatives. This review focuses on the impact of EOs and the roles of their major components in food manufacturing as natural preservatives with the related mechanisms of action. In addition, the major bioactive molecules of different types of EOs and their pharmacological activities such as antioxidant, antifungal and antimicrobial effects on crop protection were also discussed. The major compounds of EOs represent potential antioxidant, antimicrobial and antifungal activities through various mechanisms.Different types of EOs such as tea tree oil, lemon oil, clove oil, cinnamon oil and thyme oil from various traditional plants, have significantly showed better antimicrobial and antioxidant activities, and also effectively increased the shelf lives of the cereal products and increased the quality of food safety. The major groups of EOs such as terpenes and aromatic volatile compounds, play a key role in food safety without affecting the quality.Due to their various activities including antioxidant and antimicrobial activities,EOs could be used as alternative preservatives to increase the shelf lives of cereals and crops.Sundaresan Bhavaniramya Selvaraju Vishnupriya Mohammad Saleh Al-Aboody Rajendran Vijayakumar Dharmar Baskaran 2019Grain & Oil Science and Technology2019,2,2:8
Trigonelline protects the cardiocyte from hydrogen peroxide induced apoptosis in H9c2 cells显示文摘Objective:To elucidate the key parameters associated with hydrogen peroxide induced oxidative stress and investigates the mechanism of trigonelline(TG)for reducing the H_2O_2induced toxicity in H9c2 cells. Methods:Cytotoxicity and antioxidant activity of TG was assessed by EZ-CYTOX kit.RNA extraction and cDNA synthesized according to the kit manufacture protocol.Apoptosis was measured by the Flowcytometry, general PCR and qPCR.Results:It was found that the TG significantly rescued the morphology of the H9c2 cells.Treatment of cells with TG attenuated H_2O_2 induced cell deaths and improved the antioxidant activity. In addition,TG regulated the apoptotic gene caspase-3,caspase-9 and anti-apoptotic gene Bcl-2.Bcl-XL during H_2O_2 induced oxidative stress in H9c2 cells. These results were comparable with quercetin treatment. For evident, flow cytometer results also confirmed the TG significantly reduced the H_2O_2 induced necrosis and apoptosis in H9c2 cells. However, further increment of TG concentration against H_2O_2 could induce the necrosis and apoptosis along with H_2O_2.Conclusions:It is suggested that less than 125μM of TG could protect the cells from H_2O_2 induced cell damage by down regulating the caspases and up regulating the Bcl-2 and Bcl-XL expression. Therefore,we suggest the trigonelline could be useful for treatment of oxidative stress mediated cardiovascular diseases in future.Soundharrajan Ilavenil Da Hye Kim Young-Il Jeong Mariadhas Valan Arasu Mayakrishnan Vijayakumar Ponnuraj Nagendra Prabhu Srisesharam Srigopalram Ki Choon Choi 2015Asian Pacific Journal of Tropical Medicine2015,8,4:6
Challenges in animal modelling of mesenchymal stromal cell therapy for inflammatory bowel disease显示文摘Utilization of mesenchymal stromal cells(MSCs) for the treatment of Crohn's disease and ulcerative colitis is of translational interest.Safety of MSC therapy has been well demonstrated in early phase clinical trials but efficacy in randomized clinical trials needs to be demonstrated.Understanding MSC mechanisms of action to reduce gut injury and inflammation is necessary to improve current ongoing and future clinical trials. However, two major hurdles impede the direct translation of data derived from animal experiments to the clinical situation:(1) limitations of the currently available animal models of colitis that reflect human inflammatory bowel diseases(IBD).The etiology and progression of human IBD are multifactorial and hence a challenge to mimic in animal models; and(2) Species specific differences in the functionality of MSCs derived from mice versus humans.MSCs derived from mice and humans are not identical in their mechanisms of action in suppressing inflammation. Thus, preclinical animal studies with murine derived MSCs cannot be considered as an exact replica of human MSC based clinical trials. In the present review, we discuss the therapeutic properties of MSCs in preclinical and clinical studies of IBD.We also discuss the challenges and approaches of using appropriate animal models of colitis, not only to study putative MSC therapeutic efficacy and their mechanisms of action, but also the suitability of translating findings derived from such studies to the clinic.Raghavan Chinnadurai Spencer Ng Vijayakumar Velu Jacques Galipeau 2015World Journal of Gastroenterology2015,21,16:6
Coinfection of hepatitis B and hepatitis C virus in HIV-infectedpatients in south India显示文摘AIM: To screen for the co-infection of hepatitis B (HBV) and hepatitis C virus (HCV) in human immunodeficiency virus (HIV) infected patients in southern India. METHODS: Five hundred consecutive HIV infected patients were screened for Hepatitis B Virus (HBsAg and HBV-DNA) and Hepatitis C virus (anti-HCV and HCV-RNA) using commercially available ELISA kits; HBsAg, HBeAg/ anti-HBe (Biorad laboratories, USA) and anti-HCV (Murex Diagnostics, UK). The HBV-DNA PCR was performed to detect the surface antigen region (pre S-S). HCV-RNA was detected by RT-PCR for the detection of the constant 5' putative non-coding region of HCV. RESULTS: HBV co-infection was detected in 45/500 (9%) patients and HCV co-infection in 11/500 (2.2%) subjects. Among the 45 co-infected patients only 40 patients could be studied, where the detection rates of HBe was 55% (22/40), antiHBe was 45% (18/40) and HBV-DNA was 56% (23/40). Among 11 HCV co-infected subjects, 6 (54.5%) were anti-HCV and HCV RNA positive, while 3 (27.2%) were positive for anti-HCV alone and 2 (18%) were positive for HCV RNA alone. CONCLUSION: Since the principal routes for HIV transmission are similar to that followed by the hepatotropic viruses, as a consequence, infections with HBV and HCV are expected in HIV infected patients. Therefore, it would be advisable to screen for these viruses in all the HIV infected individuals and their sexual partners at the earliest.Shanmugam Saravanan Vijayakumar Velu Nagalingeswaran Kumarasamy Subhadra Nandakumar Kailapuri Gangatharan Murugavel Pachamuthu Balakrishnan Solomon Suniti Sadras Panchatcharam Thyagarajan 2007World Journal of Gastroenterology2007,13,37:6
Double-Blockchain Assisted Secure and Anonymous Data Aggregation for Fog-Enabled Smart Grid显示文摘As a future energy system, the smart grid is designed to improve the efficiency of traditional power systems while providing more stable and reliable services. However, this efficient and reliable service relies on collecting and analyzing users’electricity consumption data frequently, which induces various security and privacy threats. To address these challenges,we propose a double-blockchain assisted secure and anonymous data aggregation scheme for fog-enabled smart grid named DA-SADA.Specifically,we design a three-tier architecture-based data aggregation framework by integrating fog computing and the blockchain, which provides strong support for achieving efficient and secure data collection in smart grids.Subsequently,we develop a secure and anonymous data aggregation mechanism with low computational overhead by jointly leveraging the Paillier encryption, batch aggregation signature and anonymous authentication. In particular, the system achieves fine-grained data aggregation and provides effective support for power dispatching and price adjustment by the designed double-blockchain and two-level data aggregation. Finally, the superiority of the proposed scheme is illustrated by a series of security and computation cost analyses.Siguang Chen Li Yang Chuanxin Zhao Vijayakumar Varadarajan Kun Wang 2022Engineering2022,8,1:5

Bio Caryota Fiber Reinforced Polymer Composites: Mechanical Properties and Vibration Behavior Analysis显示文摘Polymer composites reinforced with the biofibers are found to have improved applications in many fields and also they can reduce the environmental effect. In this work, caryota-a new natural fiber for polymer composites is fabricated and their tensile, flexural and impact properties are estimated to be used in economical and lightweight load carrying structures. Laminates at different weight fraction (wt%) of 10 wt%, 20 wt%, 25 wt%, 30 wt%, 35 wt%, 40 wt% and 45 wt% are manufactured using a compression molding technique. Results asserted that the unidirectional arrangement having 40 wt% fiber posses good properties. Also the dynamic characteristics such as natural frequency and damping study are carried out for different wt% of unidirectional caryota fiber reinforced composite laminates by ex? perimental modal analysis. From this study, it has been asserted that by using the caryota fiber reinforced polyester composites, the traditional synthetic fiber can be replaced especially in automobile sector. If the caryota fiber reinforced composite material is applied properly, then its application fields will be improved.Kayaroganam Palanikumar Vijayakumar Subbiah 2019Journal of Bionic Engineering2019,16,3:3
Evaluation of chemopreventive effect of Fumaria indica against N-nitrosodiethylamine and CCl_4-induced hepatocellular carcinoma in Wistar rats显示文摘Objective:To investigation the chemopreventive potential of Fumaria indica(F.indica) extract(FIE) on N-nitrosodiethylamine and CCl4-induced hepatocarcinogenesis in Wislar rats. Methods:The experimental animals were divided into six groups(n=6).Hepatocellular carcinoma was induced by single intraperitoneal injection of N-nitrosodiethylamine(NDEA) in normal saline at a dose of 200 mg/kg body weight followed by weekly subcutaneous injections of CCl4(3 mL/kg/week) for 6 weeks,as the promoter of carcinogenic effect. After administration of the carcinogen,200 and 400 mg/kg of FIE were administered orally once a day throughout the study. At the end of 20 weeks, the body weight, liver weight and relative liver weight were measured. The percentage of nodule incidence and liver cancer markers such as aspartate transaminase(AST),alanine transaminase(ALT),alkaline phosphatase(ALP),γ-glutamyl transferase(γ-GT),total bilirubin level(TBL),α-feto protein(AFP) and carcinoembryonic antigen were estimated along with histopathological investigation in experimental groups of rats. Results:Obtained results demonstrated that the cotreatment with FIE significantly prevented the decrease of the body weight and also increased in relative liver weight caused by NDEA. The treatment with FIE significantly reduced the nodule incidence and nodule multiplicity in the rats after NDEA administration. The levels of liver cancer markers such as AST,ALT,ALP,γ-glutamyl transferase,TBL,AFP and carcinoembryonic antigen were substantially increased by NDEA treatment. However,FIE treatment significantly reduced the liver injury and restored the entire liver cancer markers.Histological observations of liver tissues too correlated with the biochemical observations. Conclusions:These finding powerfully supports that F.indica exert chemopreventive effect by suppressing the tumor burden and restoring the activities of hepatic cancer marker enzymes on NDEA and CCl4-induced hepatocarcinogenesis in Wistar rats.Talib Hussain Hefazat H Siddiqui Sheeba Fareed M Vijayakumar Chandana V Rao 2012Asian Pacific Journal of Tropical Medicine2012,5,8:3
Review of Fresnel incoherent correlation holography with linear and non-linear correlations显示文摘Fresnel incoherent correlation holography(FINCH)is a well-established incoherent imaging technique. In FINCH,three selfinterference holograms are recorded with calculated phase differences between the two interfering, differently modulated object waves and projected into a complex hologram. The object is reconstructed without the twin image and bias terms by a numerical Fresnel back propagation of the complex hologram.A modified approach to implement FINCH by a single camera shot by pre-calibrating the system involving recording of the point spread function library and reconstruction by a nonlinear cross correlation has been introduced recently. The expression of the imaging characteristics from the modulation functions in original FINCH and the modified approach by pre-calibration in spatial and polarization multiplexing schemes are reviewed. The study reveals that a reconstructing function completely independent of the function of the phase mask is required for the faithful expression of the characteristics of the modulating function in image reconstruction. In the polarization multiplexing method by non-linear cross correlation,a partial expression was observed, while in the spatial multiplexing method by non-linear cross correlation, the imaging characteristics converged towards a uniform behavior.Vijayakumar Anand Tomas Katkus Soon Hock Ng Saulius Juodkazis 2021Chinese Optics Letters2021,19,2:3
