1Hepatitis B and C virus co-infections in human immunodeficiency virus positive North Indian patients显示文摘AIM: To determine the prevalence of hepatitis B and C virus infections in human immunodeficiency virus (HIV) -positive patients at a tertiary care hospital in New Delhi, India. METHODS: Serum samples from 451 HIV positive patients were analyzed for HBsAg and HCV antibodies during three years (Jan 2003-Dec 2005). The control group comprised of apparently healthy bone-marrow and renal donors. RESULTS: The study population comprised essentially of heterosexually transmitted HIV infection. The prevalence rate of HBsAg in this population was 5.3% as compared to 1.4% in apparently healthy donors (P < 0.001). Though prevalence of HCV co-infection (2.43%) was lower than HBV in this group of HIV positive patients, the prevalence was significantly higher (P < 0.05) than controls (0.7%). Triple infection of HIV, HBV and HCV was not detected in any patient. CONCLUSION: Our study shows a significantly high prevalence of hepatitis virus infections in HIV infected patients. Hepatitis viruses in HIV may lead to faster progression to liver cirrhosis and a higher risk of antiretroviral therapy induced hepatotoxicity. 2Simultaneous quantification of prodrug oseltamivir and its metabolite oseltamivir carboxylate in human plasma by LC-MS/MS to support a bioequivalence study显示文摘A simple,precise and rapid liquid chromatography-tandem mass spectrometry(LC-MS/MS) method has been developed and validated for the simultaneous determination of oseltamivir and oseltamivir carboxylate,a neuraminidase inhibitor,using their deuterated analogs as internal standards(ISs).The method involved solid phase extraction of the analytes and ISs from 200 μL human plasma with no reconstitution and drying steps.The chromatographic separation was achieved on a Symmetry C18(100 mm 4.6 mm,5 μm) column using 10 mM ammonium formate and acetonitrile(30:70,v/v) as the mobile phase in a run time of 2.0 min.Quantitation of analytes and ISs were done by multiple reaction monitoring on a triple quadrupole mass spectrometer in the positive ionization mode.The linearity of the method was established in the concentration range of 0.5-200 ng/mL and 2.0-800 ng/mL for oseltamivir and oseltamivir carboxylate respectively.The mean extraction recovery for oseltamivir(94.4%) and oseltamivir carboxylate(92.7%) from spiked plasma samples was consistent and reproducible.The application of this method was demonstratedby a bioequivalence study in 42 healthy Indian subjects with 75 mg oseltamivir phosphate capsules.The assay reproducibility was established by reanalysis of 151 incurred subject samples.Ajay Gupta Swati Guttikar Pranav S. 3Deep Learning Based Automated Detection of Diseases from Apple Leaf Images显示文摘In Agriculture Sciences, detection of diseases is one of the mostchallenging tasks. The mis-interpretations of plant diseases often lead towrong pesticide selection, resulting in damage of crops. Hence, the automaticrecognition of the diseases at earlier stages is important as well as economicalfor better quality and quantity of fruits. Computer aided detection (CAD)has proven as a supportive tool for disease detection and classification, thusallowing the identification of diseases and reducing the rate of degradationof fruit quality. In this research work, a model based on convolutional neuralnetwork with 19 convolutional layers has been proposed for effective andaccurate classification of Marsonina Coronaria and Apple Scab diseases fromapple leaves. For this, a database of 50,000 images has been acquired bycollecting images of leaves from apple farms of Himachal Pradesh (H.P)and Uttarakhand (India). An augmentation technique has been performedon the dataset to increase the number of images for increasing the accuracy.The performance analysis of the proposed model has been compared with thenew two Convolutional Neural Network (CNN) models having 8 and 9 layersrespectively. The proposed model has also been compared with the standardmachine learning classifiers like support vector machine, k-Nearest Neighbour, Random Forest and Logistic Regression models. 8Andrographolide inhibits chikungunya virus infection by up-regulating host innate immune pathways显示文摘Objective: To investigate the therapeutic efficacy of andrographolide, a plant derived compound, against chikungunya virus(CHIKV) infection. Methods: Using flow cytometry and immunoblotting assay, in vitro viral protein expression was studied in THP-1 cells line. In Balb/c mouse neonates, viral RNA copy number was determined by real time PCR. Results:The results showed reduced CHIKV protein expression on andrographolide treatment in CHIKV-infected human peripheral blood mononuclear cells, Vero cells and THP-I cell line.In vivo, andrographolide treatment to CHIKV-infected neonates reduced viral RNA copy number. Further. andrographolide also increased cytotoxic T lymphocytes both in vitro and in vivo. Andrographolide also activated host innate immune pathways, viz., protein kinase R.phosphorylated eukaryotic initiation factor 2 α, retinoic acid inducible gene-Ⅰ and interferon regulatory factor 3/7, thereby increasing IFN-a secretion. CHIKV-induced nuclear factor κlight chain enhancer of activated B cells and tumor necrosis factor-a was also reduced on andrographolide treatment. 9Sexual Dimorphism of Foramen Magnum between Two Different Groups of Indian Population: A Cross‑Sectional Cone‑Beam Computed Tomography Study显示文摘The foramen magnum (FM), being a vital landmark at the base of the skull, includes interests to many fields of medicine. Many authors haverecounted the usefulness of the FM in gender determination. The emphasis of this study is to evaluate the shape and dimensions of the FM among males and females and to establish its role in sexual dimorphism using cone-beam computed tomography (CBCT). Furthermore, to evaluate the area and FM index (FMI) among males and females and finally to evaluate the differences in FM measurements if any in twodifferent regions of an Indian population. One hundred and ten CBCT scans (55 males and 55 females;age range, 20–80 years) were selectedfor this study. The sagittal diameter, transverse diameter, area, FMI, and circumference of FM were measured, and data were subjected to discriminant analysis for the evaluation of sexual dimorphism. The area of FM was the best discriminant parameter which was used to study the sexual dimorphism with an overall accuracy of 90.9%. 13Down-conversion materials for organic solar cells:Progress,challenges,and perspectives显示文摘Organic solar cells(OSCs)in terms of power conversion efficiency(PCE)and operational lifetime have made remarkable progress during the last decade by improving the active layer materials and introducing new interlayers.The newly developed wide bandgap organic donor and low bandgap acceptor molecules covered the absorption from the visible to the near-infrared region.Whereas the incident high energy region(UV)is not in favor of OSCs.Its absorption causes thermalization losses and photoinduced degradation,which hinders the PCE and lifetime of OSCs.Recently,lanthanide and non-lanthanide-based down-conversion(DC)materials have been introduced,which can effectively convert the high-energy photons(UV)to low-energy photons(visible)and resolve the spectral mismatch losses that limit the absorption of OSCs in high energy incident spectrum.Furthermore,the DC materials also protect the OSCs from UV-induced degradation.The DC materials were also proposed to cross the Shockley-Queisser efficiency limit of the solar cell.In this review,the need for DC materials and their processing method for OSCs have been thoroughly discussed.However,the main emphasis has been given to developing lanthanides and non-lanthanides-based DC materials for OSCs,their applications,and their impact on photovoltaic device performance,stability,and future perspectives.Ram Datt Swati Bishnoi Harrison Ka Hin Lee Sandeep Arya Sonal Gupta Vinay Gupta Wing Chung Tsoi 2022Aggregate2022,3,3:0
14肠结核与克罗恩病差异表达基因的全基因分析及FOXP3 mRNA表达的鉴别价值显示文摘背景:克罗恩病(CD)和肠结核(ITB)是有着相似表型的慢性肉芽肿性疾病,因此,找到一种生物标志物来鉴别这两种疾病是必要的。本研究旨在对ITB和CD的结肠标本进行全基因表达分析。同时,为了评价调节性T细胞的作用,我们对血清标本及结肠标本进行FOXP3 mRNA定量检测。方法:收集33名研究对象(CD患者、ITB患者及正常对照)血清和结肠组织配对样本,提取标本中的总RNA。采用Illumina HumanWG-6 BeadChip试剂盒对3组研究对象中的6种总RNA样本进行人类全基因组表达芯片分析。采用real-time PCR对具有合格RNA样本的病例(CD 4例,ITB 5例,正常对照4例)血清和组织标本进行FOXP3 mRNA表达水平检测。结果:在CD和ITB患者中,分别有92和382个基因表达上调1.5倍,91和256个基因表达下调1.5倍。过氧化物酶体增殖物介导的PPARγ信号通路在CD中下调最为显著(P<0.005);此外,IL4/5/6信号通路和Toll样受体信号通路也存在显著差异表达,上调2倍以上。而在ITB患者上调最为显著是补体活化通路尤其是经典通路。ITB患者结肠组织标本中FOXP3 mRNA表达水平明显高于CD患者(4.7062.21 vs 1.4860.31,P=0.016)。结论:结肠黏膜FOXP3 mRNA表达可作为鉴别ITB与CD的一项标志物。ITB患者补体激活通路的上调提示ITB的发病机制与肺结核相似。CD患者中PPARγ通路的下调提示PPARγ依赖的微生物屏障功能可能成为新的治疗靶点。Vineet Ahuja Swati Subodh Amit Tuteja Veena Mishra Sushil Kumar Garg Neha Gupta Govind Makharia SK Acharya 2016Gastroenterology Report2016,4,1:0
15Detection of Behavioral Patterns Employing a Hybrid Approach of Computational Techniques显示文摘As far as the present state is concerned in detecting the behavioral pattern of humans(subject)using morphological image processing,a considerable portion of the study has been conducted utilizing frontal vision data of human faces.The present research work had used a side vision of human-face data to develop a theoretical framework via a hybrid analytical model approach.In this example,hybridization includes an artificial neural network(ANN)with a genetic algorithm(GA).We researched the geometrical properties extracted from side-vision human-face data.An additional study was conducted to determine the ideal number of geometrical characteristics to pick while clustering.The close vicinity ofminimum distance measurements is done for these clusters,mapped for proper classification and decision process of behavioral pattern.To identify the data acquired,support vector machines and artificial neural networks are utilized.A method known as an adaptiveunidirectional associative memory(AUTAM)was used to map one side of a human face to the other side of the same subject.The behavioral pattern has been detected based on two-class problem classification,and the decision process has been done using a genetic algorithm with best-fit measurements.The developed algorithm in the present work has been tested by considering a dataset of 100 subjects and tested using standard databases like FERET,Multi-PIE,Yale Face database,RTR,CASIA,etc.The complexity measures have also been calculated under worst-case and best-case situations.Rohit Raja Chetan Swarup Abhishek Kumar Kamred Udham Singh Teekam Singh Dinesh Gupta Neeraj Varshney Swati Jain 2022Computers, Materials & Continua2022,,7:0
16Selective and rapid determination of raltegravir in human plasma by liquid chromatography–tandem mass spectrometry in the negative ionization mode显示文摘A selective and rapid high-performance liquid chromatography–tandem mass spectrometry method was developed and validated for the quantification of raltegravir using raltegravir-d3 as an internal standard(IS). The analyte and IS were extracted with methylene chloride and n-hexane solvent mixture from 100 mL human plasma. The chromatographic separation was achieved on a Chromolith RP-18 e endcapped C18(100 mm 4.6 mm) column in a run time of 2.0 min. Quantitation was performed in the negative ionization mode using the transitions of m/z 443.1-316.1 for raltegravir and m/z 446.1-319.0 for IS. The linearity of the method was established in the concentration range of 2.0–6000 ng/m L.The mean extraction recovery for raltegravir and IS was 92.6% and 91.8%, respectively, and the IS-normalized matrix factors for raltegravir ranged from 0.992 to 0.999. 17Prolonged existence of SARS-CoV-2 RNAs in the extracellular vesicles of respiratory specimens from patients with negative reverse transcription-polymerase chain reaction显示文摘Background and aim:Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is primarily in the respiratory tract,particularly in patients with underlying comorbidities.This study aimed to investigate the presence of the virus inside the extracellular vesicles(EVs)in patients with and without chronic liver disease(CLD).Methods:Eighty patients with positive SARS-CoV-2,including twenty-four patients with CLD and fiftysix patients without CLD,and five healthy controls with negative SARS-CoV-2 were enrolled.Nasal swab specimens were tested for the detection of SARS-CoV-2 using reverse transcription-polymerase chain reaction(RT-PCR).Patients with coronavirus disease 2019(COVID-19)were followed up on days 7 and 14.Nasal swab,collected in viral transport media(VTM),and plasma samples were investigated at each time point.EVs were isolated from the nasal swabs(collected in VTM)and plasma using differential ultracentrifugation and estimated at each time point.The transmission or replication by the EVs was assessed in Vero E6 cells.Results:In patients with baseline RT-PCR positive,SARS-CoV-2 RNAs inside the EVs were found in 68/80(85%)patients with higher viral load in the nasal swabs than in the EVs(cycle threshold(Ct)value,23.4±5.7 vs.30.3±5.0,P<0.001).On follow-up at day 7,of the 32 patients negative for COVID-19,15(46.9%)had virus persistence in the EVs(Ct value,30.7±2.7),and on day 14,of the 56 patients with negative SARS-CoV-2,16 patients(28.6%)had positive SARS-CoV-2 RNAs in the EVs(Ct value,31.4±3.0).The mean viral load decreased on days 7 and 14 compared to baseline in the nasal swabs(P<0.001)but not in the EVs.Additionally,SARS-CoV-2 RNAs were undetectable in the plasma,but 12.5% of patients were positive in the plasma EVs.Significantly prolonged and high viral load was found in the EVs on day 14 in COVID-19 patients combined with CLD compared with COVID-19 patients(P?0.0004).We found significant higher levels of EV-associated with endothelial cells and hepatocytes in the COVID-19 t CLD group than COVID-19 group(P?0.032 and P?0.002,respectively),suggesting more endothelial cells and hepatocytes cellular injury in liver disease patients with COVID-19.Interestingly,we also found EVs could transmit SARS-CoV-2 RNAs into Vero E6 cells at 24 h post-infection.Conclusions:The identification of SARS-CoV-2 RNAs in the EVs in patients with negative RT-PCR indicates the persistence of infection and likely recurrence of the infection.It is suggestive of another route of transmission as EVs harbor SARS-CoV-2 RNAs.EV-associated RNAs may determine the ongoing inflammation and clinical course of subjects with undetectable SARS-CoV-2 virus and this may have relevance to better management of patients with CLD.P.Debishree Subudhi Sheetalnath Rooge Chhagan Bihari Swati Thangariyal Sivang Goswami Reshu Agarwal Savneet Kaur Ekta Gupta Sukriti Baweja 2023Liver Research2023,7,3:0
18Marchiafava-Bignami disease:A case report显示文摘Rationale:Clinicians encounter multiple alcohol-related illnesses in practice,and Marchiafava-Bignami disease is a rare and devastating entity among them.It is a toxic-demyelinating disease and seen in chronic alcoholics,although it may be occasionally observed in chronically malnourished teetotalers.The clinical presentations are diverse.The symptoms and signs are non-specific,and the onset can be acute or chronic.Patient’s concerns:A 45-year-old right-handed patient suffered from alcohol use disorder with multiple non-specific neuropsychiatric manifestations.Diagnosis:Marchiafava-Bignami disease.Interventions:Thiamine,folate,vitamin B12,and steroid therapy.Outcome:The patient’s behaviour was significantly improved but dysarthria and pyramidal signs persisted.He was left with permanent cognitive impairment.Lessons:Though prompt therapy may halt the demyelinating process in this disease,the treatment remains a challenge in clinical practice.The recognition of the neuro-radiologic features is crucial to establish an early diagnosis.Monica Gupta Swati Garg Saurabh Gaba Rekha Gupta 2021Journal of Acute Disease2021,10,3:0
