1First SHRIMP zircon U-Pb ages for Hutuo Group in Wutaishan:Further evidence for Palaeoproterozoic amalgamation of North China Craton显示文摘A felsic tuffaceous rock, obtained from a metamorphosed sequence of volcanics and sediments of the Hutuo Group, 8 km south of Taihuai in Wutaishan, contains two zircon populations. These record SHRIMP ^207Pb/^206Pb weighted mean ages of 2180 ± 5 Ma and 2087 ± 9 Ma, respectively. The older date is within error of the age of the Dawaliang Granite in Wutaishan and is considered to be derived from a similar crustal magmatic source. The younger date is within error of reported ages from metasediments and meta-volcanics of the Wanzi supracrustal rocks and the Nanying granitic gneisses in the adjacent Fuping Complex and is interpreted to be the age of volcanism in the Hutuo Group. These data establish that: (1) the Hutuo Group is Paleoproterozoic and not Archean in age and (2) the volcanism and sedimentation were coeval in the Wutai and Fuping complexes. Sedimentation was therefore widespread at this time and possibly reflects deposition along an evolving continental margin, most likely the western margin of the Eastern Block of the North China Craton. The age of 2087±9 Ma for volcanism in the Hutuo Group means that it must have been deformed and metamorphosed after this time.This further supports the evidence, obtained from other recent studies, that the main tectonism in the Wutaishan areaoccurred at ~1.8 Ga during the Lüliang orogeny. 2Anti-Obesity Effects of Chang-Chul-Eui-Ee-ln-Tang(苍术薏苡仁汤) in Female Rats with Diet-Induced Obesity显示文摘Objective:To investigate the effects of Chang-Chul-Eui-Ee-In-Tang(苍术薏苡仁汤,CCEET), modified CCEET(MCCEET),and Semen Coicis(SC,a major component of CCEET) on energy and glucose homeostasis.The possible mechanism of action of CCEET was also determined.Methods:A total of 100 Sprague Dawley female rats were randomly assigned to 5 groups,with 20 in each group.Rats in 4 groups were fed with a high fat diet supplementation(2 g/kg body weight),and water extracts of CCEET,MCCEET,SC,and cellulose(negative control),respectively.The last group was fed with a low-fat diet as a positive control.Results: CCEET and MCCEET decreased body weight and body fat(mesenteric and retroperitoneal fat) more than SC. This decrease was due to decreased energy intake and increased energy expenditure and fat oxidation.The improvement in energy homeostasis was associated with the enhancement of the hypothalamic leptin signalling pathway involving potentiating the phosphorylation of the signal transducer and activator of transcription-3,as well as attenuating the phosphorylation of 5’ adenosine monophosphate-activated protein kinase(AMPK).Both CCEET and MCCEET improved glucose tolerance without changing serum insulin levels during an oral glucose tolerance test but MCCEET had a better effect than CCEET.Conclusions:Both CCEET and MCCEET safely exerted anti-obesity effects by enhancing energy balance in female rats with diet-induced obesity;MCCEET showed a better effect on glucose homeostasis.Sunmin Park Jung Bok Lee James W.Daily 2011Chinese Journal of Integrative Medicine2011,17,12:3
3A novel secretagogin/ATF4 pathway is involved in oxidized LDL-induced endoplasmic reticulum stress and islet β-cell apoptosis显示文摘Excessive accumulation of cholesterol inβcells initiates endoplasmic reticulum(ER)stress and associated apoptosis.We have reported that excessive uptake of cholesterol by MIN6 cells decreases the expression of secretagogin(SCGN)and then attenuates insulin secretion.Here,we aimed to determine whether cholesterol-induced SCGN decrease is involved in the modulation of ER stress and apoptosis in pancreaticβcells.In this study,MIN6 cells were treated with oxidized low-density lipoprotein(ox-LDL)for 24 h,and then intracellular lipid droplets and cell apoptosis were quantified,and SCGN and ER stress markers were identified by western blot analysis.Furthermore,small interfer RNA(siRNA)-mediated SCGN knockdown and recombinant plasmid-mediated SCGN restoration experiments were performed to confirm the role of SCGN in ER stress and associated cell apoptosis.Finally,the interaction of SCGN with ATF4 was computationally predicted and then validated by a co-immunoprecipitation assay.We found that ox-LDL treatment increased the levels of ER stress markers,such as phosphorylated protein kinase-like endoplasmic reticulum kinase,phosphorylated eukaryotic initiation factor 2 alpha,activating transcription factor 4(ATF4),and transcription factor CCAAT-enhancer-binding protein homologous protein,and promoted MIN6 cell apoptosis;in addition,the expression of SCGN was downregulated.siRNA-mediated SCGN knockdown exacerbatedβ-cell ER stress by increasing ATF4 expression.Pretreatment of MIN6 cells with the recombinant SCGN partly antagonized ox-LDL-induced ER stress and apoptosis.Furthermore,a co-immunoprecipitation assay revealed an interaction between SCGN and ATF4 in MIN6 cells.Taken together,these results demonstrated that pancreaticβ-cell apoptosis induced by ox-LDL treatment can be attributed,in part,to an SCGN/ATF4-dependent ER stress response.Li Wu Yuncheng Lv Ying Lv Sunmin Xiang Zhibo Zhao Ziqing Tang Linling Ou Bin Yan Xinhua Xiao Gebo Wen Renxian Cao Jing Yang 2021Acta Biochimica et Biophysica Sinica2021,53,1:2
4Preparation, optimization, and characterization of chitosancoated solid lipid nanoparticles for ocular drug delivery显示文摘The present study aimed to develop and optimize chitosan coated solid lipid nanoparticles(chitosan-SLNs)encapsulated with methazolamide. Chitosan-SLNs were successfully prepared by a modified oil-in-water emulsification-solvent evaporation method with glyceryl monostearate as the solid lipid and phospholipid as the surfactant. Systematic screening of formulation factors was carried out. The optimized formula for preparation was screened by orthogonal design as well as Box-Behnken design with entrapment efficiency, particle size and zeta potential as the indexes. The entrapment efficiency of the optimized formulation(methazolamide-chitosan-SLNs)prepared was(58.5± 4.5)%,Particle size(247.7 ± 17.3) nm and zeta potential(33.5 ±3.9) mV. Transmission electron microscopy showed homogeneous spherical particles in the nanometer range. A prolonged methazolamide in vitro release profile was obtained in the optimized chitosan-SLNs suspension compared with methazolamide solution. No ocular damages were observed in the susceptibility test on albino rabbits.
