Role of live microbial feed supplements with reference to anaerobic fungi in ruminant productivity: A review显示文摘To keep the concept of a safe food supply to the consumers, animal feed industries world over are showing an increasing interest in the direct-fed microbials(DFM) for improved animal performance in terms of growth or productivity. This becomes all the more essential in a situation, where a number of the residues of antibiotics and/or other growth stimulants reach in milk and meat with a number of associated potential risks for the consumers. Hence, in the absence of growth stimulants, a positive manipulation of the rumen microbial ecosystem to enhance the feedstuff utilization for improved production efficiency by ruminants has become of much interest to the researchers and entrepreneurs. A few genera of live microbes(i.e., bacteria, fungi and yeasts in different types of formulations from paste to powder) are infrequently used as DFM for the domestic ruminants. These DFM products are live microbial feed supplements containing naturally occurring microbes in the rumen. Among different DFM possibilities, anaerobic rumen fungi(ARF) based additives have been found to improve ruminant productivity consistently during feeding trials. Administration of ARF during the few trials conducted, led to the increased weight gain, milk production, and total tract digestibility of feed components in ruminants. Anaerobic fungi in the rumen display very strong cell-wall degrading cellulolytic and xylanolytic activities through rhizoid development, resulting in the physical disruption of feed structure paving the way for bacterial action. Significant improvements in the fiber digestibility were found to coincide with increases in ARF in the rumen indicating their role. Most of the researches based on DFM have indicated a positive response in nutrient digestion and methane reducing potential during in vivo and/or in vitro supplementation of ARF as DFM. Therefore, DFM especially ARF will gain popularity but it is necessary that all the strains are thoroughly studied for their beneficial properties to have a confirmed ‘generally regarded as safe' status for ruminants. | Anil K Puniya Abdelfattah Z M Salem Sanjay Kumar Sumit S Dagar Gareth W Griffith Monica Puniya Sreenivas R Ravella Nikhil Kumar Tejpal Dhewa Ravinder Kumar | 2015 | Journal of Integrative Agriculture2015,14,3: | 17
Neuroprotective potential of Quercetin in combination with piperine against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced neurotoxicity显示文摘1-Methy-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)is a neurotoxin that selectively damages dopaminergic neurons in the substantia nigra pars compacta and induces Parkinson’s like symptoms in rodents.Quercetin(QC)is a natural polyphenolic bioflavonoid with potent antioxidant and anti-inflammatory properties but lacks of clinical attraction due to low oral bioavailability.Piperine is a well established bioavailability enhancer used pre-clinically to improve the bioavailability of antioxidants(e.g.,Quercetin).Therefore,the present study was designed to evaluate the neuroprotective potential of QC together with piperine against MPTP-induced neurotoxicity in rats.MPTP(100μg/μL/rat,bilaterally)was injected intranigrally on days 1,4 and 7 using a digital stereotaxic apparatus.QC(25 and 50 mg/kg,intragastrically)and QC(25 mg/kg,intragastrically)in combination with piperine(2.5 mg/kg,intragastrically)were administered daily for 14 days starting from day 8 after the 3^(rd) injection of MPTP.On day 22,animals were sacrificed and the striatum was isolated for oxidative stress parameter(thiobarbituric acid reactive substances, nitrite and glutathione),neuroinflammatory cytokine(interleukin-1β,interleukin-6,and tumor necrosis factor-α)and neurotransmitter(dopamine,norepinephrine,serotonin,gamma-aminobutyric acid,glutamate,3,4-dihydroxyphenylacetic acid, homovanillic acid, and 5-hydroxyindoleacetic acid)evaluations.Bilateral infusion of MPTP into substantia nigra pars compacta led to significant motor deficits as evidenced by impairments in locomotor activity and rotarod performance in open field test and grip strength and narrow beam walk performance. Both QC(25 and 50 mg/kg)and QC(25 mg/kg)in combination with piperine(2.5 mg/kg),in particular the combination therapy, significantly improved MPTP-induced behavioral abnormalities in rats, reversed the abnormal alterations of neurotransmitters in the striatum, and alleviated oxidative stress and inflammatory response in the striatum. These findings indicate that piperine can enhance the antioxidant and anti-inflammatory properties of QC,and QC in combination with piperine exhibits strong neuroprotective effects against MPTP-induced neurotoxicity. | Shamsher Singh Sumit Jamwal Puneet Kumar | 2017 | Neural Regeneration Research2017,12,7: | 6
Neuroprotective Effect of Chrysophanol as a PI3K/AKT/mTOR Signaling Inhibitor in an Experimental Model of Autologous Blood-induced Intracerebral Hemorrhage显示文摘Objective Intracerebral hemorrhage(ICH)refers to predominant, sporadic, and non-traumatic bleeding in the brain parenchyma. The PI3K/AKT/mTOR signaling pathway is an important signal transduction pathway regulated by enzyme-linked receptors and has many biological functions in mammals. It plays a key role in neuronal metabolism, gene expression regulation, and tissue homeostasis in the healthy and diseased brain. Methods In the present study, the role of the PI3K/AKT/mTOR pathway inhibitor chrysophanol(CPH)(10 mg/kg and 20 mg/kg,orally)in the improvement of ICH-associated neurological defects in rats was investigated.Autologous blood(20µL/5 min/unilateral/intracerebroventricular)mimics ICH-like defects involving cellular and molecular dysfunction and neurotransmitter imbalance. The current study also included various behavioral assessments to examine cognition, memory, and motor and neuromuscular coordination. The protein expression levels of PI3K,AKT,and mTOR as well as myelin basic protein and apoptotic markers, such as Bax,Bcl-2,and caspase-3,were examined using ELISA kits.Furthermore, the levels of various neuroinflammatory cytokines and oxidative stress markers were assessed.Additionally, the neurological severity score, brain water content, gross brain pathology, and hematoma size were used to indicate neurological function and brain edema. Results CPH was found to be neuroprotective by restoring neurobehavioral alterations and significantly reducing the elevated PI3K,AKT,and mTOR protein levels, and modulating the apoptotic markers such as Bax,Bcl-2,and caspase-3 in rat brain homogenate.CPH substantially reduced the inflammatory cytokines like interleukin(IL)-1β,IL-6,and tumor necrosis factor-α.CPH administration restored the neurotransmitters GABA,glutamate, acetylcholine, dopamine, and various oxidative stress markers. Conclusion Our results show that CPH may be a promising therapeutic approach for overcoming neuronal damage caused by the overexpression of the PI3K/AKT/mTOR signaling pathway in ICH-induced neurological dysfunctions in rats. | Kuldeep Singh JADAUN Sidharth MEHAN Aarti SHARMA Ehraz Mehmood SIDDIQUI Sumit KUMAR NaifALSUHAYMI | 2022 | Current Medical Science2022,42,2: | 5
Protease inhibitors:recent advancement in its usage as a potential biocontrol agent for insect pest management显示文摘Plant derived protease inhibitors(PIs)are a promising defensin for crop im-provement and insect pest management.Although agronomist made significant efforts in utilizing PIs for managing insect pests.the potentials of PIs are still obscured.Insect ability to compensate nutrient starvation induced by dietary PI feeding using different strategies, that is,overexpression of PI-sensitive protease, expression of PI-insensitive proteases, degradation of PI,has made this innumerable collection of PIs worthless.A practical challenge for agronomist is to identify potent PI candidates,to limit insect compensatory responses and to elucidate insect compensatory and resistance mechanisms activated upon herbivory. This knowledge could be further efficiently utilized to identify potential targets for RNAi-mediated pest control. These vital genes of insects could be functionally anno-tated using the advanced gene-editing technique,CRISPR/Cas9.Contemporary research is exploiting different in silico and modern molecular biology techniques to utilize PIs in controlling insect pests efficiently. This review is structured to update recent advancements in this field, along with is chronological background. | Sujata Singh Archana Singh Sumit Kumar Pooja Mittal Indrakant K.Singh | 2020 | Insect Science2020,27,2: | 4

Adaptive Linear Quadratic Regulator for Continuous-Time Systems With Uncertain Dynamics显示文摘In this paper, adaptive linear quadratic regulator(LQR) is proposed for continuous-time systems with uncertain dynamics. The dynamic state-feedback controller uses inputoutput data along the system trajectory to continuously adapt and converge to the optimal controller. The result differs from previous results in that the adaptive optimal controller is designed without the knowledge of the system dynamics and an initial stabilizing policy. Further, the controller is updated continuously using input-output data, as opposed to the commonly used switched/intermittent updates which can potentially lead to stability issues. An online state derivative estimator facilitates the design of a model-free controller. Gradient-based update laws are developed for online estimation of the optimal gain. Uniform exponential stability of the closed-loop system is established using the Lyapunov-based analysis, and a simulation example is provided to validate the theoretical contribution. | Sumit Kumar Jha Shubhendu Bhasin | 2020 | IEEE/CAA Journal of Automatica Sinica2020,7,3: | 3
Oxide ancillary ligand-based europium β-diketonate complexes and their enhanced luminosity显示文摘Ternary materials of europium complex with2,2,6,6-tetramethyl-3,5-heptanedione(tmhd) ligand and aqua ligand as ancillary ligands have been prepared and characterized for various optoelectronic characteristics.Reactions of hydrated complex [Eu(tmhd)_(3)(H_(2)O)_(2)] proceeded with triphenylphosphine oxide(TPPO) and pyridine-N-oxide(PNO) ancillary ligands were studied to develop novel complexes. The prepared complexes show good thermal stability. A comparative investigation of preparedmaterials[Eu(tmhd)_(3)(H_(2)O)_(2)],[Eu(tmhd)_(3)(TPPO)_(2)] and [Eu(tmhd)_(3)(PNO)_(2)] was conducted for their luminescent behaviors in order to obtain the role of ancillary ligand in the enhancement of illumination amount generated from europium(Eu^(3+)) ion. Color coordinates of prepared ternary complexes such as[Eu(tmhd)_(3)(H_(2)O)_(2)]with(x = 0.54,y = 0.32),[Eu(tmhd)_(3)(TPPO)_(2)] with(x = 0.56, y = 0.32) and[Eu(tmhd)_(3)(PNO)_(2)] with(x = 0.57, y = 0.33) indicated that these materials exhibited bright red emission in visible region spectrum. The complexes show a proficient energy transport pathway from the ligands to the innermost Eu^(3+)by means of an ancillary ligand-sensitized luminescence process. Interaction between the metal and ligand results in a distinguished effect on quantum efficiency(η) as well as on Judd–Ofelt intensity factor(Ω_(2)) of the prepared materials. | Devender Singh Shri Bhagwan Anuj Dalal Kapeesha Nehra Raman Kumar Saini Kapoor Singh Anura Priyajith Simantilleke Sumit Kumar Ishwar Singh | 2021 | Rare Metals2021,40,10: | 1

Induced structural modifications in ZnS nanowires via physical state of catalyst:Highlights of 15R crystal phase显示文摘Peculiar and unique growth mechanisms involved in semiconductor nanowires(NWs)pave the way to the achievement of new crystallographic phases and remarkable material properties, and hence, studying polytypism in semiconductor NWs arouses a strong interest for the next generation of electronic and photonic applications. In this context, the growth of ZnS nanowires has been investigated,as bulk ZnS compound exhibits numerous unstable polytypes at high temperatures, but their stable occurrence is highly anticipated in a nanowire due to its special quasi-dimensional shape and growth modes. In this work, the idea is to provide a change in the growth mechanism via the physical state of catalyst droplet(liquid or solid)and hence, study the induced structural modifications in ZnS nanowires. The HRTEM images of VLS(via liquid alloyed catalyst)grown ZnS NWs show periodic stacking faults, which is precisely identified as a stacking sequence of cubic or hexagonal individual planes leading to an astonishing 15R crystal polymorph. This crystallographic phase is observed for the first time in nanowires.Contrastingly,NWs grown with VSS(via solid catalyst)show crystal polytypes of zinc blende and wurtzite. We calculate and discuss the role of cohesive energies in the formation of such ZnS polytypes.Further,we present the selection rules for the crystallization of such 15R structure in NWs and discuss the involved VLS and VSS growth mechanisms leading to the formation of different crystal phases. | Sumit Kumar Frederic Fossard Gaelle Amiri Jean-Michel Chauveau Vincent Sallet | 2022 | Nano Research2022,15,1: | 1

Prospective use of bacteriocinogenic Pediococcus pentosaceus as direct-fed microbial having methane reducing potential显示文摘Direct-fed microbials(DFM), generally regarded as safe status, are successfully used in improving rumen ecology, gastro-intestinal health, feed efficiency, milk production and growth rate in ruminants. On the other hand, methanogenesis in rumen, which accounts for a significant loss of ruminant energy and increased greenhouse gas in environment, is of great concern, therefore, use of DFM for improving productivity without compromising the animal health and ecological sustainability is encouraged. The present study was conducted to investigate the methane reducing potential of bacteriocinogenic strain Pediococcus pentosaceus-34. Since, the culture showed no hemolysis on blood agar and DNase activity, hence, it was considered to be avirulent in nature, a prerequisite for any DFM. The culture also showed tolerance to pH 5.0 for 24 h with 0.5% organic acid mixture, whereas when given a shock for 2 h at different p H and organic acids concentrations, it showed growth at pH 3.0 and 4.0 with 0.1 and 1.0% organic acids, respectively, as having good animal probiotics attributes. The total gas production was significantly(P<0.05) higher in live pedicoccal culture(LPC) and dead pedicoccal culture(DPC) both with wheat straw, when compared to the control. In sugarcane bagasse, gas production was significantly lower(P<0.05) with LPC compared to the control and DPC both. Methane was reduced by the inclusion of LPC in sugarcane bagasse(0.07 mL CH4 mg–1 dry matter digestibility) with no effect on other rumen fermentation parameters. However, with wheat straw and LPC total gas, in vitro dry matter digestibility, total volatile fatty acids increased significantly but no reduction in methane production was observed in comparison to the control. Therefore, further research is warranted in this direction, if the bacteriocinogenic strains can be used as DFM for ruminants to improve the ruminant productivity. | Sanjay Kumar Sumit S Dagar Seyed H Ebrahimi Ravinder K Malik Ramesh C Upadhyay Anil K Puniya | 2015 | Journal of Integrative Agriculture2015,14,3: | 1
