2Application of ~1H NMR-based metabolomics for discrimination of different parts and development of a new processing workflow for Cistanche deserticola显示文摘Cistanche deserticola(CD) is one of the two authoritative source plants of Cistanches Herba, a well-known medicinal plant. Herein,~1H NMR spectroscopy was employed to characterize the chemical profile and to distinguish the different parts, as well as to propose a new processing workflow for CD.Signal assignment was achieved by multiple one and two dimensional NMR spectroscopic techniques in combination with available databases and authentic compounds. The upper parts of the plant were distinguished from the lower parts by combining ~1H NMR spectroscopic dataset with multivariate statistical analysis. A new processing method that hyphenated steaming with freeze-drying, was demonstrated to be superior to either steaming coupled with oven-drying or direct freeze-drying via holistic ~1H NMR-based metabolomic characterization. Phenylethanoid glycosides, mainly echinacoside and acteoside, were screened out and confirmed as the chemical markers responsible for exhibiting the superiority of the new processing workflow, whereas serial primary metabolites, especially carbohydrates and tricarboxylic acid cycle metabolites, were found as the primary molecules governing the discrimination between the upper and lower parts of the plant. 5Au-catalysed free-standing wurtzite structured InAs nanosheets grown by molecular beam epitaxy显示文摘In this study,we report the growth of free-standing InAs nanosheets using Au catalysts in molecular beam epitaxy.Detailed structural characterizations suggest that wurtzite structured InAs nanosheets,with features of extensive{1120}surfaces,grown along the<1102>direction and adopted{0001}nanosheet/catalyst interfaces,are initiated from wurtzite structured[0001]nanowires as the inclined epitaxial growth due to relatively higher In concentrations in Au catalysts,and grown from these inclined nanostructures through catalyst-induced axial growth and their enhanced lateral growth under the high growth temperature.Based on the facts that the nanosheets contain large low energy{1120}surfaces and{0001}nanosheet/catalyst interfaces,the growth of our nanosheets is a thermodynamically driven process.This study provides new insights into fabricating free-standing Ⅲ-Ⅴ nanosheets for their applications in future nanoscale devices.Qiang Sun Han Gao Xiaomei Yao Kun Zheng Pingping Chen Wei Lu Jin Zou 2019Nano Research2019,12,11:2

9Efficient genome editing in Claviceps purpurea using a CRISPR/Cas9 ribonucleoprotein method显示文摘Claviceps purpurea produces many pharmacologically important ergot alkaloids(EAS),which are widely used to treat migraine and hypertension and to aid childbirth.Although an EAS biosynthetic cluster of C.purpurea has been discovered more than 20 years ago,the complete biosynthetic pathway of EAS has not been fully characterized until now.The main obstacle to elucidating this pathway and strain modification is the lack of efficient genome-editing tools for C.purpurea.The conventional gene manipulation method for C.purpurea relies on homologous recombination(HR),although the efficiency of HR in C.purpurea is very low(~1-5%).Consequently,the disruption of target genes is laborious and time-consuming.Although CRISPR/Cas9 genome-editing methods based on in vivo Cas9 expression and gRNA transcription have been reported recently,their gene-disruption efficiency is still very low.Here,we developed an efficient genome-editing system in C.purpurea based on in vitro assembled CRISPR/Cas9 gRNA ribonucleoprotein complexes.As proof of principle,three target genes were efficiently knocked out using this CRISPR/Cas9 ribonucleoprotein complex-mediated HR system,with editing efficiencies ranging from 50%to 100%.Inactivation of the three genes,which are closely related to uridine biosynthesis(ura5),hypha morphology(rac),and EAS production(easA),resulted in a uridine auxotrophic mutant,a mutant with a drastically different phenotype in axenic culture,and a mutant that did not produce EAS,respectively.Our ribonucleoprotein-based genome-editing system has a great advantage over conventional and in vivo CRISPR/Cas9 methods for genome editing in C.purpurea,which will greatly facilitate elucidation of the EAS biosynthetic pathway and other future basic and applied research on C.purpurea.Lu Yu Meili Xiao Zhihua Zhu Yinmei Wang Zhihua Zhou Pingping Wang Gen Zou 2022Synthetic and Systems Biotechnology2022,7,2:1

11Axiotaxy driven growth of belt-shaped InAs nanowires in molecular beam epitaxy显示文摘In this study,we demonstrate the axiotaxy driven growth of belt-shaped InAs nanowires using Au catalysts by molecular beam epitaxy.It is found that,the zinc-blende structured InAs nanowires,with the features of[113]growth direction and extensive{110}side-surfaces,are induced by catalysts in Au–In α phase through the axiotaxy growth,in which the lattice mismatch between the projections of atomic planes onto nanowire/catalyst interfaces is minimized by forming extraordinary tilted interfaces.Our atomic-resolution in situ TEM heating experiments show that the catalysts remained in the solid state of Au–In α phase during the axiotaxy growth,by which the vapor–solid–solid growth mechanism can be confirmed.Through manipulating the growth direction,this unusual growth mechanism can provide a practical pathway to control the morphology of the low-dimensional nanomaterials,from conventional nanowires to belt-shaped nanowires utilizing a significant lateral growth,simply using nanoparticles as catalyst.Qiang Sun Dong Pan Xutao Zhang Jianhua Zhao Pingping Chen Wei Lu Jin Zou 2021Nano Research2021,14,7:0

13Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 15Re-examination of the impact of some non-pharmaceutical interventions and media coverage on the COVID-19 outbreak in Wuhan显示文摘In this paper,based on the classic Kermack-McKendrick SIR model,we propose an ordinary differential equation model to re-examine the COVID-19 epidemics in Wuhan where this disease initially broke out.The focus is on the impact of all those major nonpharmaceutical interventions(NPIs)implemented by the local public healthy authorities and government during the epidemics.We use the data publicly available and the nonlinear least-squares solver lsqnonlin built in MATLAB to estimate the model parameters.Then we explore the impact of those NPIs,particularly the timings of these interventions,on the epidemics.The results can help people review the responses to the outbreak of the COVID-19 inWuhan,while the proposed model also offers a framework for studying epidemics of COVID-19 and/or other similar diseases in other places,and accordingly helping people better prepare for possible future outbreaks of similar diseases.Ao Li Yang Wang Pingping Cong Xingfu Zou 2021Infectious Disease Modelling2021,6,1:0
