| 2 | On the modification of hypereutectic Al-Si alloys using rare earth Er显示文摘Hypereutectic Al-17.5Si (wt pct) and Al-25Si (wt pct) alloys with various content of rare earth Er were prepared by conventional casting technique. The effect of Er on the microstructure and properties of the hypereutectic Al-Si alloys was investigated using optical microscopy, scanning electron microscopy as well as friction and wear tests. It was found that optimal amount of Er exists in modifying the hypereutectic Al-Si alloys. An appropriate addition of Er improved the anti-wear properties, and reduced the friction coefficient of the hypereutectic Al-Si alloys studied. The primary silicon crystals in the hypereutectic Al-Si alloys were refined with the suitable addition of Er element. | 3 | Application of ~1H NMR-based metabolomics for discrimination of different parts and development of a new processing workflow for Cistanche deserticola显示文摘Cistanche deserticola(CD) is one of the two authoritative source plants of Cistanches Herba, a well-known medicinal plant. Herein,~1H NMR spectroscopy was employed to characterize the chemical profile and to distinguish the different parts, as well as to propose a new processing workflow for CD.Signal assignment was achieved by multiple one and two dimensional NMR spectroscopic techniques in combination with available databases and authentic compounds. The upper parts of the plant were distinguished from the lower parts by combining ~1H NMR spectroscopic dataset with multivariate statistical analysis. A new processing method that hyphenated steaming with freeze-drying, was demonstrated to be superior to either steaming coupled with oven-drying or direct freeze-drying via holistic ~1H NMR-based metabolomic characterization. Phenylethanoid glycosides, mainly echinacoside and acteoside, were screened out and confirmed as the chemical markers responsible for exhibiting the superiority of the new processing workflow, whereas serial primary metabolites, especially carbohydrates and tricarboxylic acid cycle metabolites, were found as the primary molecules governing the discrimination between the upper and lower parts of the plant. | 4 | Authentic compound-free strategy for simultaneous determination of primary coumarins in Peucedani Radix using offline high performance liquid chromatography–nuclear magnetic resonance spectroscopy–tandem mass spectrometry显示文摘Herein, a strategy is proposed for the simultaneous determination of primary coumarins in Peucedani Radix(Chinese name: Qianhu). The methodology consists of three consecutive steps: 1) Semi-preparative LC in combination with a home-made automated fraction collection module to fragment the universal metabolome standard into ten fractions(Frs. I–X); 2) LC–accurate MS/MS and quantitative1 H NMR spectroscopy conducted in parallel to acquire the qualitative and quantitative data of each fraction; 3) Robust identification and quantification of components by use of LC coupled to multiple reaction monitoring. In this final step, the most significant fractions(Frs. III–X) were pooled to serve as the pseudo-mixed standard solution. Meticulous online parameter optimization was performed to obtain the optimal parameters, including ion transitions and collision energies. Concerns were particularly paid onto pursuing the parameters being capable of monitoring regiospecific isomers, notably praeruptorin E vs. 3′-isovaleryl-4′-angeloylkhellactone. The quantitative performance of the method was validated according to diverse assays. Eleven primary coumarins(1–11) were unambiguously identified and absolutely quantified, even though no external reference compound was used. | 5 | Schisandrol A protects AGEs-induced neuronal cells death by allosterically targeting ATP6V0d1 subunit of V-ATPase显示文摘Diabetes have been shown to cause progressive neuronal injury with pain and numbness via advanced glycation end-products(AGEs)-induced neuronal cell apoptosis;however, the valuable drug targets for diabetic neuropathy have been poorly reported so far. In this study, we discovered a natural small-molecule schisandrol A(SolA) with significant protective effect against AGEs-induced neuronal cell apoptosis. ATP6V0D1, a major subunit of vacuolar-type ATPase(V-ATPase) in lysosome was identified as a crucial cellular target of SolA. Moreover, SolA allosterically mediated ATP6V0D1 conformation via targeting a unique cysteine 335 residue to activate V-ATPase-dependent lysosomal acidification.Interestingly, SolA-induced lysosome pH downregulation resulted in a mitochondrial-lysosomal crosstalk by selectively promoting mitochondrial BH3-only protein BIM degradation, thereby preserving mitochondrial homeostasis and neuronal cells survival. | 9 | Neutralization of SARS-CoV-2 pseudovirus using ACE2-engineered extracellular vesicles显示文摘The spread of coronavirus disease 2019(COVID-19)throughout the world has resulted in stressful healthcare burdens and global health crises.Developing an effective measure to protect people from infection is an urgent need.The blockage of interaction between angiotensin-converting enzyme 2(ACE2)and S protein is considered an essential target for anti-severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)drugs.A full-length ACE2 protein could be a potential drug to block early entry of SARS-Co V-2 into host cells.In this study,a therapeutic strategy was developed by using extracellular vesicles(EVs)with decoy receptor ACE2 for neutralization of SARS-Co V-2.The EVs embedded with engineered ACE2(EVs-ACE2)were prepared;the EVs-ACE2 were derived from an engineered cell line with stable ACE2 expression.The potential effect of the EVs-ACE2 on anti-SARS-Co V-2 was demonstrated by both in vitro and in vivo neutralization experiments using the pseudovirus with the S protein(S-pseudovirus).EVs-ACE2 can inhibit the infection of S-pseudovirus in various cells,and importantly,the mice treated with intranasal administration of EVs-ACE2 can suppress the entry of S-pseudovirus into the mucosal epithelium.Therefore,the intranasal EVs-ACE2 could be a preventive medicine to protect from SARS-Co V-2 infection.This EVs-based strategy offers a potential route to COVID-19 drug development. | Canhao Wu Qin Xu Huiyuan Wang Bin Tu Jiaxin Zeng Pengfei Zhao Mingjie Shi Hong Qiu Yongzhuo Huang | 2022 | Acta Pharmaceutica Sinica B2022,12,3: | 2 |
| 10 | Uncertainties in stormwater runoff data collection from a small urban catchment, Southeast China显示文摘Monitoring data are often used to identify stormwater runoff characteristics and in stormwater runoff modelling without consideration of their inherent uncertainties. Integrated with discrete sample analysis and error propagation analysis, this study attempted to quantify the uncertainties of discrete chemical oxygen demand (COD), total suspended solids (TSS) concentration, stormwater flowrate, stormwater event volumes, COD event mean concentration (EMC), and COD event loads in terms of flow measurement, sample collection, storage and laboratory analysis. The results showed that the uncertainties due to sample collection, storage and laboratory analysis of COD from stormwater runoff are 13.99%, 19.48% and 12.28%. Meanwhile, flow measurement uncertainty was 12.82%, and the sample collection uncertainty of TSS from stormwater runoff was 31.63%. | 13 | Effects of parasitic plant Cistanche deserticola on chlorophyll a fluorescence and nutrient accumulation of host plant Haloxylon ammodendron in the Taklimakan Desert显示文摘The parasitic plant Cistanche deserticola attaches to Haloxylon ammodendron, a perennial shrub with high tolerance to salinity and drought. However, little was known about the parasite-host relation between the two species. Effects of the parasite on chlorophyll a fluorescence and nutrient accumulation in the host plant (H. ammodendron) were investigated in the Taklimakan Desert. Some photosynthetic parameters of both host and non-host H. ammodendron plants were measured by in vivo chlorophyll a fluorescence technology in the field. The assimilating branches of host and non-host plants were collected and nutrient and inorganic ion contents were analyzed. The results from field experiments showed that the infection of C. deserticola reduced the non-photochemical quenching of the variable chlorophyll fluorescence (NPQ) and the potential maximum quantum yield for primary photochemistry (F v /F m ) of the host. Compared with non-host plants, the host H. ammodendron had low nutrient, low inorganic ion contents (Na + and K + ) and low K + /Na + ratios in the assimilating branches. It suggested that C. deserticola infection reduced the nutrient acquisition and caused damage to the photoprotection through thermal dissipation of the energy of the photosystem II in the host, resulting in a decrease in the tolerance to salinity and high radiation. | 14 | Probing the active sites of 2D nanosheets with Fe-N-C carbon shell encapsulated Fe_(x)C/Fe species for boosting sodium-ion storage performances显示文摘Developing stable but high active metal-nitrogen-carbon(M-N-C)-based hard carbon anode is a promising way to be the alternatives to graphene and blank hard carbon for sodium-ion batteries(SIBs),requiring the precise tailoring of the electronic structure for optimizing the Na+intercalation behavior,yet is greatly challenging.Herein,Fe-N-C graphitic layer-encapsulating Fe3C species within hard carbon nanosheets(Fe-N-C/Fe3C@HCNs)are rationally engineered by pyrolysis of self-assembled polymer.Impressively,the Fe-N-C/Fe3C@HCNs exhibit outstanding rate capacity(242 mAh·g^(−1)at 2,000 mA·g^(−1)),which is 2.1 and 4.2 times higher than that of Fe-N-C and N-doped carbon(N-C),respectively,and prolonged cycling stability(176 mAh·g^(−1)at 2,000 mA·g^(−1)after 2,000 cycles).Theoretical calculations unveil that the Fe3C species enhance the electronic transfer from Na to Fe-N-C,resulting in the charge redistribution between the interfaces of Fe3C and Fe-N-C.Thus,the optimized adsorption behavior towards Na+reduces the thermodynamic energy barriers.The synergistic effect of Fe3C and Fe-N-C species maintains the structural integrity of electrode materials during the sodiation/desodiation process.The in-depth insight into the advanced Na+storage mechanisms of Fe3C@Fe-N-C offers precise guidance for the rational establishment of confinement heterostructures in SIBs. | Huicong Xia Pengfei Yuan Lingxing Zan Gan Qu Yunchuan Tu Kaixin Zhu Yifan Wei Zeyu WeiFangying Zheng Mo Zhang Yongfeng Hu Dehui Deng Jianan Zhang | 2022 | Nano Research2022,15,8: | 1 |
