2Analysis of in vivo patterns of caspase 3 gene expression in primary hepatocellular carcinoma and its relationship to p21^(WAF1) expression and hepatic apoptosis显示文摘AIM To detect the expression of caspase 3gene in primary human hepatocellular carcinoma(HCC)and investigate its relationship to p21WAF1gene expression and HCC apoptosis.METHODS In situ hybridization was employedto determine caspase 3 and p21WAF1expression inHCC.In situ end-labeling was used to detecthepatocytic apoptosis in HCC.RESULTS Twenty-one of 39(53.8%)cases ofHCC were found to express caspase 3transcripts,while 45.2% of HCC failed toexpress caspase 3.Non-cancerous adjacent livertissues showed more positive caspase 3(87.5%,7/8)as compared with HCC(P<0.05).The expression of caspase 3 is correlated withHCC differentiation,72.2%(13/18)ofmoderately to highly differentiated HCC showedcaspase 3 transcripts positive,while only 38.1%of poorly differentiated HCC harbored caspase 3transcripts(P<0.05).No relationship wasfound between caspase 3 expression and tumorsize or grade or metastasis,although 52.5%(5/8)of HCC with metastasis were caspase 3positive and a little higher than that with nometastasis(51.6%,P>0.05).Expression of caspase 3 alone did not affect the apoptosisindex(AI)of HCC.The AI was 7.12%o in caspase3-positive tumors(n=21),while in caspase 3-negative cases(n=18)6.59%0(P>0.05).Expression of caspase 3 clearly segregated withp21WAF1positive tumors as compared withp21WAF1-negative cases(16 of 23,69.6% versus5 of 16,31.3%)with statistical significance(P=0.017).In the cases with positive caspase 3and negative p21WAF1,the Al was found slightlyhigher,but with no statistical significance,thanthat with expression of p21WAF1and caspase 3(7.21‰ vs 6.98‰,P>0.05).CONCLUSION Loss of caspase 3 expressionmay contribute to HCC carcinogenesis,althoughthe expression of caspase 3 does not correlatewell with cell apoptosis in HCC.p21WAF1may bemerely one of the inhibitors which can reducecaspase 3 mediated cell apoptosis in HCCs.Bao Hua Sun Jun Zhang Bao JǜWang Xi Ping Zhao You Kun Wang Zhi Qun Yu Dong Liang Yang Lian Jie Hao Department of Clinical Immunology,Tongji Hospital,Tongji Medical University,Wuhan 430030,Hubei Province,China 2000World Journal of Gastroenterology2000,6,3:65
3Expression of nuclear factor-KB in hepatocellular carcinoma and its relation with the X protein of hepatitis B virus显示文摘AIM In this study we investigated therelationship of the X protein of HBV and nuclearfactor-KB (NF-κB) and the expression of NF-KB inhuman hepatocellular carcinoma tissues.METHODS Immunohistochemistry SP methodwas used to detect the expression of NF-κB and the X protein of HBV in human hepatocellularcarcinoma tissues of 52 cases. NF-κB is abnormally activated inhepatocellular carcinoma, which is probablyrelated to the X protein of HBV. 4A novel advanced oxidation process to degrade organic pollutants in wastewater:Microwave-activated persulfate oxidation显示文摘This article,for the first time,provides a novel advanced oxidation process based on sulfate radical(SO4. -) to degrade organic pollutants in wastewater:microwave(MW)-activated persulfate oxidation(APO)with or without active carbon(AC).Azo dye acid Orange 7(AO7)is used as a model compound to investigate the high reactivity of MW-APO.It is found that AO7(up to 1000 mg/L)is completely decolorized within 5-7 min under an 800 W MW furnace assisted-APO.In the presence of chloride ion(up to 0.50 mol/L),the decolorization is still 100%completed,though delayed for about 1-2 min.Experiments are made to examine the enhancement by AC.It is exciting to find that the 100%decolorization of AO7(500 mg/L)is achieved within 3 min by MW-APO using 1.0 g/L AC as catalyst,while the degradation effciency maintains at 50%by MW energy without persulfate after about 5 min.Besides the destruction of visible light chromophore band of AO7(484 nm),during MW-APO,two bands in the ultraviolet region(228 nm and 310 nm)are rapidly broken down.The removal of COD is about 83%-95%for 500 mg/L AO7.SO4. -is identified with quenching studies using specific alcohols.Both SO4. -and.OH could degrade AO7,but SO 4 .-plays the dominant role.In a word,MW-APO AC is a new catalytic combustion technology for destruction of organic contamination even for high concentration.YANG Shiying WANG Ping YANG Xin WEI Guang ZHANG Wenyi SHAN Liang 2009Journal of Environmental Sciences2009,21,9:41
5Analysis of Heavy Metal Sources for Vegetable Soils from Shandong Province, China显示文摘Heavy metal pollution in agricultural soils has serious negative influence on human health. Concentrations of Cd, Hg, As, Pb, Cr, Cu, Zn, and Ni in top soils (0-20 cm) of greenhouses and farmlands from four main vegetable production areas Shouguang, Laiyang, Jinxiang, and Zhangqiu in Shandong Province, one of the most rapidly developing regions in China, were measured in this study. Shouguang is mainly occupied by greenhouse vegetables and the other three areas are mainly open field culture. Total of 149 soil samples were collected. The average concentrations of the eight heavy metals of the tested 149 soil samples were all below the threshold values according to 'Farmland environmental quality evaluation standards for edible agricultural products (HJ332-2006)'of China. However, most of the studied heavy metals were present at higher concentrations than those of the natural background levels in local agricultural soils. Among the total 149 soil samples, 22 samples were contaminated by Cd, Ni, Cu, or Hg. Comparisons showed that the main pollution element in greenhouse vegetable soils was Cd, while that of open field vegetable soils was Cu. The results of principal components analysis (PCA) suggested that concentrations of Cr, As, and Ni were mainly controlled by parent rocks; Hg and Pb were affected by anthropogenic activities such as vehicle and industrial fumes and waste water irrigation. Meanwhile, concentrations of Cd, Cu, and Zn were affected mainly by the use of agrochemicals. Most of the heavy metals were positively correlated with each other in concentration. 7Recent Advances on the Technologies to Increase Fertilizer Use Efficiency显示文摘To increase fertilizer use efficiency (FUE) and to minimize its negative impact on environment have been the focal points in the world for a long time. It is very important to increase FUE in China for its relatively low FUE and serious losses of nutrients. Recent advances of the technologies to increase FUE are reviewed in this article. These include site-specific and real-time nitrogen management, non-destructive quick test of the nitrogen status of plants, new types of slow release and controlled release fertilizers, site-specific nutrient management, and use of urease inhibitor and nitrification inhibitor to decrease nitrogen losses. 9The Long-term Variation of Extreme Heavy Precipitation and Its Link to Urbanization Effects in Shanghai during 1916–2014显示文摘Using the hourly precipitation records of meteorological stations in Shanghai, covering a period of almost a century(1916–2014), the long-term variation of extreme heavy precipitation in Shanghai on multiple spatial and temporal scales is analyzed, and the effects of urbanization on hourly rainstorms studied. Results show that:(1) Over the last century, extreme hourly precipitation events enhanced significantly. During the recent urbanization period from 1981 to 2014, the frequency of heavy precipitation increased significantly, with a distinct localized and abrupt characteristic.(2) The spatial distribution of long-term trends for the occurrence frequency and total precipitation intensity of hourly heavy precipitation in Shanghai shows a distinct urban rain-island feature; namely, heavy precipitation was increasingly focused in urban and suburban areas.Attribution analysis shows that urbanization in Shanghai contributed greatly to the increase in both frequency and intensity of heavy rainfall events in the city, thus leading to an increasing total precipitation amount of heavy rainfall events. In addition,the diurnal variation of rainfall intensity also shows distinctive urban–rural differences, especially during late afternoon and early nighttime in the city area.(3) Regional warming, with subsequent enhancement of water vapor content, convergence of moisture flux and atmospheric instability, provided favorable physical backgrounds for the formation of extreme precipitation.This accounts for the consistent increase in hourly heavy precipitation over the whole Shanghai area during recent times.Ping LIANG Yihui DING 2017Advances in Atmospheric Sciences2017,34,3:38

11Does Helicobacter pylori infection play a role in iron deficiency anemia? 12Gd-EOB-DTPA-enhanced magnetic resonance imaging for focal liver lesions in Chinese patients:a multicenter,open-label,phase Ⅲ study显示文摘BACKGROUND:Contrast agents help to improve visibility in magnetic resonance(MR)imaging.However,owing to the large interstitial spaces of the liver,there is a reduction in the natural contrast gradient between lesions and healthy tissue.This study was undertaken to evaluate the efficacy and safety of the liverspecific MR imaging contrast agent gadoxetate disodium(GdEOB-DTPA)in Chinese patients.METHODS:This was a single-arm,open-label,multicenter study in patients with known or suspected focal liver lesions referred for contrast-enhanced MR imaging.MR imaging was performed in 234 patients before and after a single intravenous bolus of Gd-EOB-DTPA(0.025 mmol/kg body weight).Images were evaluated by clinical study investigators and three independent,blinded radiologists.The primary efficacy endpoint was sensitivity in lesion detection.RESULTS:Gd-EOB-DTPA improved sensitivity in lesion detection by 9.46%compared with pre-contrast imaging for the average of the three blinded readers(94.78%vs 85.32%for Gd-EOB-DTPA vs pre-contrast,respectively).Improvements in detection were more pronounced in lesions less than 1cm.Gd-EOB-DTPA improved diagnostic accuracy in lesion classification.CONCLUSIONS:This open-label study demonstrated that Gd-EOB-DTPA improves diagnostic sensitivity in liver lesions,particularly in those smaller than 1 cm.Gd-EOB-DTPA also significantly improves the diagnostic accuracy in lesion classification,and furthermore,Gd-EOB-DTPA is safe in Chinese patients with liver lesions.Meng-Su Zeng Hui-Yi Ye Liang Guo Wei-Jun Peng Jian-Ping Lu Gao-Jun Teng Josy Breuer Yi Huan Ping Li Jian-Rong Xu Chang-Hong Liang 2013Hepatobiliary & Pancreatic Diseases International2013,12,6:31
13Effect of hydrogen on the stress corrosion cracking behavior of X80 pipeline steel in Ku'erle soil simulated solution显示文摘Hydrogen was a key factor resulting in stress corrosion cracking (SCC) of X80 pipeline steel in Ku'erle soil simulated solution. In this article, the effect of hydrogen on the SCC susceptibility of X80 steel was investigated further by slow strain rate tensile test, the surface fractures were observed using scanning electron microscopy (SEM), and the fracture mechanism of SCC was discussed. The results indicate that hydrogen increases the SCC susceptibility. The SEM micrographs of hydrogen precharged samples presents a brittle quasi-cleavage feature, and pits facilitate the transgranular crack initiation. In the electrochemical impedance spectroscopy (EIS) measurement, the decreased polarization resistance and the pitting resistance of samples with hydrogen indicate that hydrogen increases the dissolution rate and deteriorates the pitting corrosion resistance. 15Efficacy and safety of Xinfeng capsule in patients with rheumatoid arthritis:a multi-center parallel-group double-blind randomized controlled trial显示文摘OBJECTIVE:To evaluate the efficacy and safety of Xinfeng capsule in patients suffering rheumatoid arthritis(RA).METHODS:A multi-center parallel-group designed,double-blind,randomized,controlled trial was conducted.Totally 304 RA patients were assigned to two groups:one group was administered Xinfeng capsule(XFC) plus the placebo of leflunomide and the other given leflunomide(LEF) plus the placebo of XFC for twelve weeks.The clinical and laboratory parameters were compared at baseline and fourth,eighth,and twelfth weeks.RESULTS:After twelve-week treatment,patients in two groups all showed some trend of effectiveness when compared in terms of American Rheumatism Association(ACR) recommended 20%,50%,70%improvement criteria,but it was insignificant.The validity in ameliorate modified disease activity score(DAS28) and laboratory indexes as erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),rheumatoid factor(RF) were also found no difference.The score of health assessment questionnaire(HAQ),self-rating anxiety scale(SAS),self-rating depression scale(SDS) and quality of life questionnaire with rheumatoid arthritis(RAQOL)both lower than the first week and the changes showed no difference.However,the score of SDS dropped more in XFC group than in the other.A total of 147 adverse reaction cases were reported,which shows no difference between the two groups.The most common adverse reactions were hepatic impairment,anemia,leukocytopenia,epigastric discomfort and phalacrosis.CONCLUSION:XFC demonstrated better improvement in the scores of SDS and compared with those of LEF group.Liu Jian Wang Yuan Huang Chuanbing Xu Jianhua Li Zhijun Xu Liang He Liyun Sun Yue Wang Yali Xu Shengqian Zhao Ping Mao Tongjun Tan Bin Zhu Fubing Zhang Pingheng Fang Li 2015Journal of Traditional Chinese Medicine2015,35,5:27

17Construction, expression and characterization of the engineered antibody against tumor surface antigen, p185^(c-erbB-2)显示文摘The c-erbB-2 proto-oncogene encodes a 185kDa protein p185, which belongs to epidermal growth factorreceptor family. Amplification of this gene has been shown to correlate with poor clinical prognosis forcertain cancer patients. The monoclonal antibody A21 which directed against p185 specifically inhibitsproliferation of tumor cells overexpressing p185, hence allows it to be a candidate for targeted therapy. Inorder to overcome several drawbacks of murine MAb, we cloned its VH and VL genes and constructed thesingle-chain Fv (scFv) through a peptide linker. The recombinant scFvA21 was expressed in Escherichiacoli and purified by the affinity column. Subsequently it was characterized by ELISA, Western blot, cellimmunohistochemistry and FACS. All these assays showed the binding activity to extracellular domain(ECD) of p185. Based on those properties of scFvA21, we further constructed the scFv-Fc fusion moleculewith a homodimer form and the recombinant product was expressed in mammalian cells. In a series ofsubsequent analysis this fusion protein showed identical antigen binding site and activity with the parentantibody. 20Non-alcoholic fatty liver disease:An early mediator predicting metabolic syndrome in obese children?显示文摘AIM:To investigate if non-alcoholic fatty liver disease (NAFLD) is an early mediator for prediction of metabolic syndrome,and if liver B-ultrasound can be used for its diagnosis.METHODS:We classified 861 obese children (6-16 years old) into three subgroups:group 0 (normal liver in ultrasound and normal transaminases);group 1 (fatty liver in ultrasound and normal transaminases);and group 2 (fatty liver in ultrasound and elevated transaminases).We measured the body mass index,waist and hip circumference,blood pressure,fasting blood glucose,insulin,homeostasis model assessment of insulin resistance (HOMA-IR),whole-body insulin sensitivity index (WBISI),lipid profile and transaminases in all the participants.The risk of developing metabolic syndrome (MS) was assessed according to the degree of liver fatty infiltration based on the B-ultrasound examination.RESULTS:Among the 861 obese children,587 (68.18%) were classified as having NAFLD,and 221 (25.67%) as having MS.The prevalence of MS in NAFLD children (groups 1 and 2) was 37.64% (221/587),which was much higher than that in non-NAFLD group (group 0,12.04%) (P < 0.01).There were significantly higher incidences concerning every component of MS in group 2 compared with group 0 (P < 0.05).The incidence of NAFLD in MS patients was 84.61% (187/221),which was significantly higher than that of hypertension (57.46%,127/221) and glucose metabolic anomalies (22.62%,50/221),and almost equal to the prevalence of dyslipidemia (89.14%,197/221).Based on the B-ultrasound scales,the presence of moderate and severe liver fatty infiltration carried a high risk of hypertension [odds ratio (OR):2.18,95% confidence interval (95% CI):1.27-3.75],dyslipidemia (OR:7.99,95% CI:4.34-14.73),impaired fasting glucose (OR:3.65,95% CI:1.04-12.85),and whole MS (OR:3.77;95% CI:1.90-7.47,P < 0.01).The state of insulin resistance (calculated by HOMA-IR and WBISI) deteriorated as the degree of fatty infiltration increased.CONCLUSION:NAFLD is not only a liver disease,but also an early mediator that reflects metabolic disorder,and liver B-ultrasound can be a useful tool for MS screening.Jun-Fen Fu Hong-Bo Shi Li-Rui Liu Ping Jiang Li Liang Chun-Lin Wang Hong-Bo Shi Ping Jiang Xi-Yong Liu 2011World Journal of Gastroenterology2011,17,6:20
