7Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29

9Correlation between anti-fibrotic effect of baicalin and serum cytokines in rat hepatic fibrosis显示文摘AIM: To investigate the correlation between the antifibrotic effect of baicalin and serum cytokine production in rat hepatic fibrosis. METHODS: Forty male Sprague-Dawley rats were divided randomly into four groups: normal control group, model group, baicalin-treated group, and colchicine-treated group. Except for the normal control group, all rats in the other groups were administered with carbon tetrachloride to induce hepatic fibrosis. At the same time, the last two groups were also treated with baicalin or colchicine. At the end of the 8 wk, all animals were sacrif iced. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), transforming growth factor (TGF)-β1, tumor necrosis factor (TNF)-α, interleukin (IL)-6 and IL-10 were measured. Liver index, hepatic hydroxyproline content and the degree of liver fibrosis were also evaluated. RESULTS: The levels of ALT, AST and liver index in the baicalin-treated group were markedly lower than those in the model group (ALT: 143.88 ± 14.55 U/L vs 193.58 ± 24.35 U/L; AST: 263.66 ± 44.23 U/L vs 404.37 ± 68.29 U/L; liver index: 0.033 ± 0.005 vs 0.049 ± 0.009, P < 0.01). Baicalin therapy also significantly attenuated the degree of hepatic fibrosis, collagen area and collagen area percentage in liver tissue (P < 0.01). Furthermore, the levels of serum TGFβ1, TNF-α and IL-6 were strikingly reduced in the baicalin-treated group compared with the model group, while the production of IL-10 was up-regulated: (TGF-β1: 260.21 ± 31.01 pg/mL vs 375.49 ± 57.47 pg/mL; TNF-α: 193.40 ± 15.18 pg/mL vs 260.04 ± 37.70 pg/mL; IL-6: 339.87 ± 72.95 pg/mL vs 606.47 ± 130.73 pg/mL; IL-10: 506.22 ± 112.07 pg/mL vs 316.95 ± 62.74 pg/mL, P < 0.01). 11Chinese Trauma Surgeon Association for management guidelines of vacuum sealing drainage application in abdominal surgeries-Update and systematic review显示文摘Vacuum sealing drainage (VSD) is frequently used in abdominal surgeries. However, relevant guidelines are rare. Chin ese Trauma Surge on Associati on orga nized a committee composed of 28 experts across China in July 2017, aiming to provide an evidence-based recommendation for the application of VSD in abdominal surgeries.Eleven questions regarding the use of VSD in abdominal surgeries were addressed:(1) which type of materials should be respectively chosen for the intraperitoneal cavity, retroperitoneal cavity and superficial incisions?(2) Can VSD be preventively used for a high-risk abdominal incision w让h primary suture?(3) Can VSD be used in severely contaminated/infected abdominal surgical sites?(4) Can VSD be used for temporary abdominal cavity closure under some special conditions such as severe abdominal trauma, infection, liver transplantation and intra-abdominal volume increment in abdominal compartment syndrome?(5) Can VSD be used in abdominal organ inflammation, injury, or postoperative drainage?(6) Can VSD be used in the treatment of intestinal fistula and pancreatic fistula?(7) Can VSD be used in the treatment of intra-abdominal and extra-peritoneal abscess?(8) Can VSD be used in the treatment of abdominal wall wounds, wound cavity, and defects?(9) Does VSD in crease the risk of bleeding?(10) Does VSD increase the risk of intestinal wail injury?(11) Does VSD increase the risk of peritoneal adhesion? Focusing on these questions, evidence-based recommendations were given accordingly. VSD was strongly recommended regarding the questions 2-4. Weak recommendations were made regarding questions 1 and 5-11. 12(La0.2Ce0.2Nd0.2Sm0.2Eu0.2)2Zr2O7:A novel high-entropy ceramic with low thermal conductivity and sluggish grain growth rate显示文摘Fine grains and slow grain growth rate are beneficial to preventing the thermal stress-induced cracking and thermal conductivity increase of thermal barrier coatings.Inspired by the sluggish diffusion effect of high-entropy materials,a novel high-entropy(HE)rare-earth zirconate solid solution(La0.2Ce0.2Nd0.2Sm0.2Eu0.2)2 Zr2 O7 was designed and successfully synthesized in this work.The as-synthesized(La0.2Ce0.2Nd0.2Sm0.2Eu0.2)2 Zr2 O7 is phase-pure with homogeneous rare-earth element distribution.The thermal conductivity of as-synthesized(La0.2Ce0.2Nd0.2Sm0.2Eu0.2)2 Zr2 O7 at room temperature is as low as 0.76 W m^-1 K^-1.Moreover,after being heated at 1500℃for 1-18 h,the average grain size of(La0.2Ce0.2Nd0.2Sm0.2Eu0.2)2 Zr2 O7 only increases from 1.69μm to 3.92μm,while the average grain size of La2Zr2O7 increases from 1.96μm to 8.89μm.Low thermal conductivity and sluggish grain growth rate indicate that high-entropy(La0.2Ce0.2Nd0.2Sm0.2Eu0.2)2Zr2O7 is suitable for application as a thermal barrier coating material and it may possess good thermal stress-induced cracking resistance.Zifan Zhao Huimin Xiang Fu-Zhi Dai Zhijian Peng Yanchun Zhou 2019Journal of Materials Science & Technology2019,35,11:25
13Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. 15Hepatitis C virus infections and genotypes in China显示文摘Objective: To define the conditions of hepatitis C vi-rus (HCV) infections, and geographic and demo-graphic distributions of genotypes in China.Methods: HCV infected patients were selected fromindividuals with different patterns of liver diseasesand high risk populations in different parts of China.Genotypes of HCV in some isolates were further ana-lyzed, based on the data from our laboratory studiesand some carefully selected published literatures.Results: The anti-HCV positive rates were 9.7% inpatients with acute hepatitis, 13.3% in patients withchronic hepatitis, 18.3% in patients with hepatocel-lular carcinoma, 33.0% in patients with liver cirrho-sis, and 43.2% in patients with posttransfusionalhepatitis (average, 16.2% in patients with liver di-seases). The anti-HCV positive rates in the high riskpopulations were 36.4% in leukemic patients,43.0% in hemodialysis patients, 12.7% in blood do-nors, 64.1% in drug abusers, 13.1% in prostitutes,and 2.57% in naturally healthy people. At least 4clades (clades 1, 2, 3 and 6) of HCV were found inChina with different geographic and demographicdistributions. 16A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide.Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen 2019Molecular Plant2019,12,5:18

18Isoliquiritigenin inhibits TGF-β1-induced fibrogenesis through activating autophagy via PI3K/AKT/mTOR pathway in MRC-5 cells显示文摘Isoliquiritigenin(ISL),a natural flavonoid derived from the root of liquorice,has been reported to possess anti-inflammatory and antioxidant activities.Previous studies have found that ISL plays a crucial role in anti-fibrosis of adipose tissue and renal tissue;however,its effect on pulmonary fibrogenesis has not been demonstrated.In this study,we aimed to explore the roles and the underlying mechanisms of ISL in TGF-β1-induced fibrogenesis using human lung fibroblast-derived MRC-5 cells.Cell proliferation and migration were determined by MTT and wound healing assay,respectively.The expression levels of alpha-smooth muscle actin(α-SMA),collagen type I alpha 1(COLIA1)and fibronectin(FN),microtubule-associated protein light chain 3(LC3)and related signaling molecules were detected by quantitative real-time PCR,western blot and immunofluorescence assay,correspondingly.EGFP-LC3 transfection was used for autophagy analysis.The results showed that ISL inhibited the TGF-β1-induced proliferation and migration,and down-regulated the expressions ofα-SMA,COLIA1 and FN.ISL treatment led to up-regulation of LC3 in TGF-β1-treated MRC-5 cells,accompanied by significant decrease in the phosphorylation levels of phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT),and mammalian target of rapamycin(mTOR).In addition,the inhibitory effects of ISL on TGF-β1-induced fibrogenic features in MRC-5 cells were enhanced by pretreatment with autophagy activator Rapmycin and PI3K/AKT inhibitor LY294002 and reversed by autophagy inhibitor 3-methyladenine and PI3K/AKT activator IGF-1.Taken together,our results demonstrated that ISL could attenuate the fibrogenesis of TGF-β1-treated MRC-5 cells by activating autophagy via suppressing the PI3K/AKT/mTOR pathway.Therefore,ISL holds a great potential to be developed as a novel therapeutic agent for the treatment of pulmonary fibrosis.Jinjuan He Hao Peng Meifang Wang Ying Liu Xingrong Guo Bin Wang Longjun Dai Xueqin Cheng Zhongji Meng Leyong Yuan Fenglin Cai Yijun Tang 2020Acta Biochimica et Biophysica Sinica2020,52,8:16

20Geographical characteristics of China’s wetlands derived from remotely sensed data显示文摘In this paper, we report the first wetland mapping of the entire China using Landsat enhanced thematic mapper plus (ETM+) data. These data were obtained from the Global Land Cover Facility at the University of Maryland spanning from 1999 to 2002. A total of 597 scenes of Landsat images were georeferenced and mosaiced. Manual image interpretation of satellite images was aided with elevation data, soil data, land cover/land use data and Google Earth. The minimum mapping unit is 10 pixel × 10 pixel, equivalent to 9 ha. The aim of our first round of mapping was only targeted at the boundary delineation of any type of wetland except those wetlands that are under agricultural use (i.e., paddy fields), which has already been well mapped by others. Our interpretation results indicate that a total of 359478 km2 of wetlands are of non-agricultural use. Among our preliminarily mapped wetland, 339353 km2 are inland wetland, 2786 km2 are non-agricultural artificial wetland, and 17609 km2 are coastal wetland. Because low-tide is rarely captured in satellite images, an under-estimation of coastal wetland is inevitable. We conducted some statistics based on our mapped wetlands and compared them with those previously obtained from a number of sources including a land cover/land use map made with satellite images during the late 1990s and early 2000s, a marshland map developed in approximately the same period, survey data of coastal wetland in early 1980s, and area data for approximately 400 larger patches of marshland in China compiled in 1996. Because some inconsistencies exist in the guidelines of those different wetland surveys, difference in area is expected. Some further comparison indicates that the wetland distributions derived from the preliminary wetland map are reasonable and more objective than other sources. The mapping process also indicated that the method adopted by us was efficient and cost-effective. We also found that in order to ensure comparability of the wetland maps developed at different times, a set of standard guidelines on the wetland categories to be mapped, and the mapping methods to be used must be well conceived, developed and effectively employed.
