1The apricot(Prunus armeniaca L.)genome elucidates Rosaceae evolution and beta-carotenoid synthesis显示文摘Apricots,scientifically known as Prunus armeniaca L,are drupes that resemble and are closely related to peaches or plums.As one of the top consumed fruits,apricots are widely grown worldwide except in Antarctica.A high-quality reference genome for apricot is still unavailable,which has become a handicap that has dramatically limited the elucidation of the associations of phenotypes with the genetic background,evolutionary diversity,and population diversity in apricot.DNA from P.armeniaca was used to generate a standard,size-selected library with an average DNA fragment size of~20 kb.The library was run on Sequel SMRT Cells,generating a total of 16.54 Gb of PacBio subreads(N50=13.55 kb).The high-quality P.armeniaca reference genome presented here was assembled using long-read single-molecule sequencing at approximately 70×coverage and 171×Illumina reads(40.46 Gb),combined with a genetic map for chromosome scaffolding.The assembled genome size was 221.9 Mb,with a contig NG50 size of 1.02 Mb.Scaffolds covering 92.88%of the assembled genome were anchored on eight chromosomes.Benchmarking Universal Single-Copy Orthologs analysis showed 98.0%complete genes.We predicted 30,436 protein-coding genes,and 38.28%of the genome was predicted to be repetitive.We found 981 contracted gene families,1324 expanded gene families and 2300 apricot-specific genes.The differentially expressed gene(DEG)analysis indicated that a change in the expression of the 9-cis-epoxycarotenoid dioxygenase(NCED)gene but not lycopene beta-cyclase(LcyB)gene results in a lowβ-carotenoid content in the white cultivar“Dabaixing”.This complete and highly contiguous P.armeniaca reference genome will be of help for future studies of resistance to plum pox virus(PPV)and the identification and characterization of important agronomic genes and breeding strategies in apricot.Fengchao Jiang Junhuan Zhang Sen Wang Li Yang Yingfeng Luo Shenghan Gao Meiling Zhang Shuangyang Wu Songnian Hu Haoyuan Sun Yuzhu Wang 2019Horticulture Research2019,6,1:9
2紫杉醇脂质体与紫杉醇联合铂类同步放化疗治疗宫颈癌的随机对照研究(英文)显示文摘Objective:The aim of the study was to compare the efficacy,side effect and influence on the survival rate of two chemotherapy regimens,paclitaxel liposome combined with platinum and paclitaxel combined with platinum,in concurrent chemoradiotherapy for cervical carcinoma.Methods:The 162 cases with primary cervical carcinoma diagnosed between January 2008 and November 2009 in Jiangxi Maternal and Child Health Hospital(China) were enrolled in this randomized controlled trial.Seventy-one cases were included in paclitaxel group and 91 in paclitaxel liposome group.And the chemotherapy doses were as follows:paclitaxel liposome and paclitaxel 135 mg/m2;cisplatin 80 mg/m2 or carboplatin AUC 4-6;then repeated every 21 days for two or three times.Radical radiotherapy was given to both groups at the same time.Efficacy was evaluated according to the tumor regression six months later and follow-up was done consistently.Results:The overall response rates of paclitaxel group and paclitaxel liposome group were 90.1% and 89 % respectively(P > 0.05).The one year cumulative survival was 91.4% for paclitaxel group and 89.2% for paclitaxel liposome group(P > 0.05).The incidence rates of adverse effects such as rash,gastrointestinal toxicity,bone marrow suppression and muscle/joint pain in paclitaxel liposome group were much lower than those in paclitaxel group(P < 0.05),while there was no difference regarding hair loss,hepatic function damage,peripheral neuritis and other aspects(P > 0.05).Conclusion:Paclitaxel liposome plus platinum is a safe and effective method for staging IIa-IV cervical carcinomas.While the long-term efficacy should be further observed.Siyuan Zeng Ling Li Meiling Zhong Wei Jiang Yun Xiao 2012The Chinese-German Journal of Clinical Oncology2012,11,2:3
3Frequency-domain diagonal extension imaging显示文摘The pixel size of a charge-coupled device(CCD)camera plays a major role in the image resolution,and the square pixels are attributed to the physical anisotropy of the sampling frequency.We synthesize the high sampling frequency directions from multiple frames acquired with different angles to enhance the resolution by 1.4×over conventional CCD orthogonal sampling.To directly demonstrate the improvement of frequency-domain diagonal extension(FDDE)microscopy,lens-free microscopy is used,as its resolution is dominantly determined by the pixel size.We demonstrate the resolution enhancement with a mouse skin histological specimen and a clinical blood smear sample.Further,FDDE is extended to lens-based photography with an ISO 12233 resolution target.This method paves a new way for enhancing the image resolution for a variety of imaging techniques in which the resolution is primarily limited by the sampling pixel size,for example,microscopy,photography,and spectroscopy.Shan Jiang Meiling Guan Jiamin Wu Guocheng Fang Xinzhu Xu Dayong Jin Zhen Liu Kebin Shi Fan Bai Shu Wang Peng Xi 2020Advanced Photonics2020,2,3:3
4PECTATE LYASE-LIKE10 is associated with pollen wall development in Brassica campestris显示文摘PECTATE LYASE‐LIKE10(PLL10) was previously identified as one of the differentially expressed genes both in microspores during the late pollen developmental stages and in pistils during the fertilization process in Chinese cabbage(Brassica campestris ssp. chinensis). Here, antisense‐RNA was used to study the functions of BcPLL10 in Chinese cabbage. Abnormal pollen was identified in the transgenic lines(bcpll10‐4, ‐5, and ‐6). In fertilization experiments, fewer seeds were harvested when the antisense‐RNA lines were used as pollen donor. In vivo and in vitro pollen germination assays less germinated pollen tubes were observed in bcpll10 lines. Scanning electron microscopy observation verified that the tryphine materials were over accumulated around the pollen surface and sticked them together in bcpll10.Moreover, transmission electron microscopy observation revealed that the internal endintine was overdeveloped and predominantly occupied the intine, and disturbed thenormal proportional distribution of the two layers in the non‐germinal furrow region; and no obvious demarcation existed between them in the germinal furrow region in the bcpll10 pollen. 6All-Climate Stretchable Dendrite-Free Zn-Ion Hybrid Supercapacitors Enabled by Hydrogel Electrolyte Engineering显示文摘Hybrid supercapacitors have shown great potentials to fulfill the demand of future diverse applications such as electric vehicles and portable/wearable electronics.In particular,aqueous zinc-ion hybrid supercapacitors(ZHSCs)have gained much attention due to their low-cost,high energy density,and environmental friendliness.Nevertheless,typical ZHSCs use Zn metal anode and normal liquid electrolyte,causing the dendrite issue,restricted working temperature,and inferior device flexibility.Herein,a novel flexible Zn-ion hybrid supercapacitor(FZHSC)is developed by using activated carbon(AC)anode,δ-MnO_(2) cathode,and innovative PVA-based gel electrolyte.In this design,heavy Zn anode and its dendrite issue are avoided and layered cathode with large interlayer spacing is employed.In addition,flexible electrodes are prepared and integrated with an anti-freezing,stretchable,and compressible hydrogel electrolyte,which is attained by simultaneously using glycerol additive and freezing/thawing technique to regulate the hydrogen bond and microstructure.The resulting FZHSC exhibits good rate capability,high energy density(47.86 Wh kg^(−1);3.94 mWh cm^(−3)),high power density(5.81 kW kg^(−1);480 mW cm^(−3)),and excellent cycling stability(~91%capacity retention after 30000 cycles).Furthermore,our FZHSC demonstrates outstanding flexibility with capacitance almost unchanged even after various continuous shape deformations.The hydrogel electrolyte still maintains high ionic conductivity at ultralow temperatures(≤−30℃),enabling the FZHSC cycled well,and powering electronic timer robustly within an all-climate temperature range of−30~80℃.This work highlights that the promising Zn metal-free aqueous ZHSCs can be designed with great multifunctionality for more practical application scenarios.Yuqi Jiang Kun Ma Meiling Sun Yuanyuan Li Jinping Liu 2023Energy & Environmental Materials2023,6,2:2
7Microstructure and heat treatment for Ni3Al-based single crystal alloy with different crystal orientations显示文摘Ni3Al-based single crystal alloy IC6SX with different crystal orientations were prepared by seed crystal method.The microstructure and heat treatment of the alloy were investigated.The results showed that the microstructure of the alloy was in dendrite structure,and the crystal orientation had significant effect on the dendrite morphology of this alloy.The precipitated phases of(MoNi)6C and NiMo appeared in the microstructure of the three alloys with different crystal orientations during solidification process.Compared with other two alloys,the volume fractions of precipitated phases of both(MoNi)6C and NiMo was the most in the alloy with[111]orientation and the least in the alloy with[001]orientation.The solidus and liquidus temperatures of the alloy IC6SX tested by differential scanning calorimetry(DSC)were 1356℃and 1387℃,respectively.Meanwhile,the effect of different solution temperatures on the microstructure of the alloy with different orientations was studied.The results showed that the precipitated phases of(MoNi)6C and NiMo were eliminated with the solid solution treatment under the condition of 1300℃/10h.However,the incipient melting of the alloys occurred due to the dissolution of low melting point phases.As the temperature dropped to 1280℃,the area of incipient melting in the alloy with different orientations decreased gradually.However,there was no incipient melting appearing in the three alloys with different orientations when the solution treatment temperature dropped to 1260℃.Liwu Jiang Xuezheng Dou Meiling Wu Min Cai 2020Progress in Natural Science:Materials International2020,30,4:2
8Termination of Transcription of LAT Increases the Amounts of ICP0 mRNA but Does Not Alter the Course of HSV-1 Infection in Latently Infected Murine Ganglia显示文摘On entering sensory ganglia,herpes simplex viruses 1(HSV-1)establishes a latent infection with the synthesis of a latency associated transcript(LAT)or initiates productive infection with expression of a set of immediate early viral proteins.The precise mechanisms how expression of a genes is suppressed during the latency are unknown.One mechanism that has been proposed is illustrated in the case of ICP0,a key immediate early viral regulatory protein.Specifically,the 2 kb LAT intron is complementary to the 30 terminal portion of ICP0 m RNA.To test the hypothesis that accumulation of LAT negatively affects the accumulation of ICP0 m RNA,we inserted a DNA fragment encoding two poly(A)sequences into LAT to early terminate LAT transcript without interrupting the complementary sequence of ICP0 transcript(named as SR1603).Comparisons of the parent(SR1601)and mutant(SR1603)HSV-1 viruses showed the following:Neurons harboring latent SR1603 virus accumulated equivalent amounts of viral DNA but higher amounts of ICP0 m RNA and lower amounts of LAT,when compared to neurons harboring the SR1601 virus.One notable difference between the two viruses is that viral RNA accumulation in explanted ganglia harboring SR1603 virus initiated significantly sooner than that in neurons harboring SR1601 virus,suggesting that ICP0 may act as an activator of viral gene expression in permissive cells.Collectively,these data suggest that increased ICP0 m RNA by suppressed LAT did not affect the establishment of latency in latently infected murine ganglia.Haifang Jiang Jiaming Wu Xianjie Liu Ruitao Lu Manling Zhou Meiling Chen Yonghong Liu Grace Guoying Zhou Wenmin Fu 2021Virologica Sinica2021,36,2:2

14Tangeretin inhibits fungal ferroptosis to suppress rice blast显示文摘Flavonoids are polyphenolic secondary metabolites that function as signaling molecules,allopathic compounds,phytoalexins,detoxifying agents and antimicrobial defensive compounds in plants.Blast caused by the fungus Magnaporthe oryzae is a serious disease affecting rice cultivation.In this study,we revealed that a natural flavonoid,tangeretin,substantially delays the formation of M.oryzae appressoria and blocks the development of blast lesions on rice plants.Our data suggest that tangeretin has antioxidant activity that interferes with conidial cell death/ferroptosis,which is critical for M.oryzae pathogenicity.Tangeretin showed a ferroptosis inhibition efficacy comparable to the well-established liproxstatin-1.Furthermore,overexpression of the NADPH oxidases NOX1 or NOX2 significantly decreased sensitivity toward tangeretin treatment,suggesting Nox-mediated lipid peroxidation as a possible target for tangeretin in regulating redox signaling and ferroptosis in M.oryzae.Our nursery and field tests showed that application of tangeretin can effectively mitigate overall disease symptoms and prevent leaf blast.Our study reveals the plant-derived fungal ferroptosis inhibitor tangeretin as a potential and novel antifungal agrochemical for the sustainable prevention of the devastating blast disease in important cereal crops.Meiling Liang Huijuan Ye Qing Shen Xianya Jiang Guobing Cui Wenxiang Gu Lian-Hui Zhang Naweed I.Naqvi Yi Zhen Deng 2021Journal of Integrative Plant Biology2021,63,12:1
15Neurodegeneration-associated FUS is a novel regulator of circadian gene expression显示文摘Background:Circadian rhythms are oscillating physiological and behavioral changes governed by an internal molecular clock,and dysfunctions in circadian rhythms have been associated with ageing and various neurodegenerative diseases.However,the evidence directly connecting the neurodegeneration-associated proteins to circadian control at the molecular level remains sparse.Methods:Using meta-analysis,synchronized animals and cell lines,cells and tissues from FUS R521C knock-in rats,we examined the role of FUS in circadian gene expression regulation.Results:We found that FUS,an oscillating expressed nuclear protein implicated in the pathogenesis of amyotrophic lateral sclerosis(ALS)and frontotemporal dementia(FTD),exerted a novel feedback route to regulate circadian gene expression.Nr1d1-encoded core circadian protein REV-ERBαbound the Fus promoter and regulated the expression of Fus.Meanwhile,FUS was in the same complex as PER/CRY,and repressed the expression of E box-containing core circadian genes,such as Per2,by mediating the promoter occupancy of PSF-HDAC1.Remarkably,a common pathogenic mutant FUS(R521C)showed increased binding to PSF,and caused decreased expression of Per2.Conclusions:Therefore,we have demonstrated FUS as a modulator of circadian gene expression,and provided novel mechanistic insights into the mutual influence between circadian control and neurodegeneration-associated proteins.Xin Jiang Tao Zhang Haifang Wang Tao Wang Meiling Qin Puhua Bao Ruiqi Wang Yuwei Liu Hung-Chun Chang Jun Yan Jin Xu 2018Translational Neurodegeneration2018,7,1:1
16DNA Extraction from Formalin-fixed and Paraffin-embedded Tissues by Triton X-100 for Effective Amplification of EGFR Gene by Polymerase Chain Reaction显示文摘For first-line non-small-cell lung cancer(NSCLC) therapy,detecting mutation status of the epidermal growth factor receptor(EGFR) gene constitutes a prudent test to identify patients who are most likely to benefit from EGFR-tyrosine kinase inhibitor(TKI) therapy.Now,the material for detecting EGFR gene mutation status mainly comes from formalin-fixed and paraffin-embedded(FFPE) tissues.DNA extraction from FFPE and the amplification of EGFR gene by polymerase chain reaction(PCR) are two key steps for detecting EGFR gene mutation.We showed a simple method of DNA extraction from FFPE tissues for the effective amplification of EGFR gene.Extracting DNA from the FFPE tissues of NSCLC patients with 1% Triton X-100(pH=10.0) was performed by heating at 95 °C for 30 min.Meanwhile,a commercial kit was used to extract DNA from the same FFPE tissues of NSCLC patients for comparison.DNA extracted products were used as template for amplifying the exons 18,19,20 and 21 of EGFR by PCR for different amplified fragments.Results show that DNA fragment size extracted from FFPE tissues with 1% Triton X was about 250―500 base pairs(bp).However,DNA fragment size extracted from FFPE tissues via commercial kit was about from several hundreds to several thousands bp.The DNA yield extracted from FFPE tissues with 1% Triton X was larger than that via commercial kit.For about 500 bp fragment,four exons of EGFR could not be amplified more efficiently from extracted DNA with 1% Triton X than with commercial kit.However,for about 200 bp fragment.This simple and non-laborious protocol could successfully be used to extract DNA from FFPE tissue for the amplification of EGFR gene by PCR,further screening of EGFR gene mutation and facilitating the molecular analysis of a large number of FFPE tissues from NSCLC patients.WANG Xiao-feng DU Zhen-wu WU Meil ZHANG Yu-cheng JIANG Yang ZHANG Gui-zhen 2012Chemical Research in Chinese Universities2012,28,4:1

20A petunia transcription factor, PhOBF1, regulates flower senescence by modulating gibberellin biosynthesis显示文摘Flower senescence is commonly enhanced by the endogenous hormone ethylene and suppressed by the gibberellins(GAs)in plants.However,the detailed mechanisms for the antagonism of these hormones during f lower senescence remain elusive.In this study,we characterized one up-regulated gene PhOBF1,belonging to the basic leucine zipper transcription factor family,in senescing petals of petunia(Petunia hybrida).Exogenous treatments with ethylene and GA3 provoked a dramatic increase in PhOBF1 transcripts.Compared with wild-type plants,PhOBF1-RNAi transgenic petunia plants exhibited shortened f lower longevity,while overexpression of PhOBF1 resulted in delayed flower senescence.Transcript abundances of two senescence-related genes PhSAG12 and PhSAG29 were higher in PhOBF1-silenced plants but lower in PhOBF1-overexpressing plants.Silencing and overexpression of PhOBF1 affected expression levels of a few genes involved in the GA biosynthesis and signaling pathways,as well as accumulation levels of bioactive GAs GA1 and GA3.Application of GA3 restored the accelerated petal senescence to normal levels in PhOBF1-RNAi transgenic petunia lines,and reduced ethylene release and transcription of three ethylene biosynthetic genes PhACO1,PhACS1,and PhACS2.Moreover,PhOBF1 was observed to specifically bind to the PhGA20ox3 promoter containing a G-box motif.Transient silencing of PhGA20ox3 in petunia plants through tobacco rattle virus-based virus-induced gene silencing method led to accelerated corolla senescence.Our results suggest that PhOBF1 functions as a negative regulator of ethylene-mediated f lower senescence by modulating the GA production.Xiaotong Ji Ziwei Xin Yanping Yuan Meiling Wang Xinyi Lu Jiaqi Li Yanlong Zhang Lixin Niu Cai-Zhong Jiang Daoyang Sun 2023Horticulture Research2023,10,4:0
