| 1 | Evolving management of colorectal cancer显示文摘This article reviews recent advances in surgical techniques and adjuvant therapies for colorectal cancer, including total mesorectal excision, the resection of liver and lung metastasis and advances in chemoradiation and foreshadows some interventions that may lie just beyond the frontier. In particular, little is known about the intracellular and extracellular cascades that may influence colorectal cancer cell adhesion and metastasis. Although the phosphorylation of focal adhesion kinases and focal adhesion associated proteins in response to integrin-mediated cell matrix binding ('outside in integrin signaling') is well described, the stimulation of cell adhesion by intracellular signals activated by pressure prior to adhesion represents a different signal paradigm. However, several studies have suggested that increased pressure and shear stress activate cancer cell adhesion. | 2 | High-resolution multimodal flexible coherent Raman endoscope显示文摘Coherent Raman scattering microscopy is a fast,label-free,and chemically specific imaging technique that shows high potential for future in vivo optical histology.However,the imaging depth in tissues is limited to the sub-millimeter range because of absorption and scattering.Realization of coherent Raman imaging using a fiber endoscope system is a crucial step towards imaging deep inside living tissues and providing information that is inaccessible with current microscopy tools.Until now,the development of coherent Raman endoscopy has been hampered by several issues,mainly related to the fiber delivery of the excitation pulses and signal collection.Here,we present a flexible,compact,coherent Raman,and multimodal nonlinear endoscope(4.2mm outer diameter,71mm rigid length)based on a resonantly scanned hollow-core Kagomé-lattice double-clad fiber.The fiber design enables distortion-less,background-free delivery of femtosecond excitation pulses and back-collection of nonlinear signals through the same fiber.Sub-micrometer spatial resolution over a large field of view is obtained by combination of a miniature objective lens with a silica microsphere lens inserted into the fiber core.We demonstrate high-resolution,high-contrast coherent anti-Stokes Raman scattering,and second harmonic generation endoscopic imaging of biological tissues over a field of view of 320μm at a rate of 0.8 frames per second.These results pave the way for intraoperative label-free imaging applied to real-time histopathology diagnosis and surgery guidance. | Alberto Lombardini Vasyl Mytskaniuk Siddharth Sivankutty Esben Ravn Andresen Xueqin Chen Jérôme Wenger Marc Fabert Nicolas Joly Frédéric Louradour Alexandre Kudlinski Hervé Rigneault | 2018 | Light(Science & Applications)2018,7,1: | 6 |

| 16 | First characterization of endophytic Corynespora cassiicola isolates with variant cassiicolin genes recovered from rubber trees in Brazil显示文摘Corynespora cassiicola is the causal agent of Corynespora Leaf Fall(CLF)disease.CLF is one of the most important fungal diseases of rubber trees in Asia and Africa but disease outbreaks have not been reported in South America.Cassiicolin,a small cysteine-rich glycoprotein secreted by the pathogenic C.cassiicola isolate CCP,was previously identified as a potential disease effector in rubber tree.Recently,the cassiicolin-encoding gene(Cas1)was characterized and shown to be expressed in the early phase of infection.In this study,we investigated whether previously undetected strains of C.cassiicola are present in South American rubber plantations by examining the fungal endophyte population found in asymptomatic rubber tree leaves.Four isolates were identified as C.cassiicola.Genes encoding new forms of the cassiicolin precursor protein(Cas3 and Cas4)were identified from these isolates.Three of four isolates were able to induce symptoms on the cultivar they were isolated from in a detached leaf assay,with different kinetics and intensities.One isolate had the same pathogenicity profile as the pathogenic isolate CCP;the other two isolates developed symptoms late during the course of infection,suggesting saprotrophic capabilities.However,no Cas3 or Cas4 transcripts could be detected upon inoculation with the endophytic isolates,whereas the reference gene Cas1 was expressed upon inoculation with the CCP isolate.This work demonstrated that C.cassiicola is present in South America in an endophytic form and that it may evolve from an endophytic to a saprophytic or even potentially pathogenic life style. | Marine Déon Ana Scomparin Aude Tixier Carlos R.R.Mattos Thierry Leroy Marc Seguin Patricia Roeckel-Drevet Valérie Pujade-Renaud | 2012 | Fungal Diversity2012,,3: | 2 |

| 20 | Gut homeostasis, injury, and healing: New therapeutic targets显示文摘The integrity of the gastrointestinal mucosa plays a crucial role in gut homeostasis,which depends upon the balance between mucosal injury by destructive factors and healing via protective factors.The persistence of noxious agents such as acid,pepsin,nonsteroidal anti-inflammatory drugs,or Helicobacter pylori breaks down the mucosal barrier and injury occurs.Depending upon the size and site of the wound,it is healed by complex and overlapping processes involving membrane resealing,cell spreading,purse-string contraction,restitution,differentiation,angiogenesis,and vasculogenesis,each modulated by extracellular regulators.Unfortunately,the gut does not always heal,leading to such pathology as peptic ulcers or inflammatory bowel disease.Currently available therapeutics such as proton pump inhibitors,histamine-2 receptor antagonists,sucralfate,5-aminosalicylate,antibiotics,corticosteroids,and immunosuppressants all attempt to minimize or reduce injury to the gastrointestinal tract.More recent studies have focused on improving mucosal defense or directly promoting mucosal repair.Many investigations have sought to enhance mucosal defense by stimulating mucus secretion,mucosal blood flow,or tight junction function.Conversely,new attempts to directly promote mucosal repair target proteins that modulate cytoskeleton dynamics such as tubulin,talin,Ehm^(2),filamin-a,gelsolin,and flightless I or that proteins regulate focal adhesions dynamics such as focal adhesion kinase.This article summarizes the pathobiology of gastrointestinal mucosal healing and reviews potential new therapeutic targets. | Sema Oncel Marc D Basson | 2022 | World Journal of Gastroenterology2022,28,17: | 2 |
