2Liver-resident NK cells suppress autoimmune cholangitis and limit the proliferation of CD4^(+) T cells显示文摘Liver-resident NK cells are distinct from conventional NK cells and play an important role in the maintenance of liver homeostasis.How liver-resident NK cells participate in autoimmune cholangitis remains unclear.Here,we extensively investigated the impact of NK cells in the pathogenesis of autoimmune cholangitis utilizing the well-established dnTGFβRII cholangitis model,NK cell-deficient(Nfil3−/−)mice,adoptive transfer and in vivo antibody-mediated NK cell depletion.Our data demonstrated that disease progression was associated with a significantly reduced frequency of hepatic NK cells.Depletion of NK cells resulted in exacerbated autoimmune cholangitis in dnTGFβRII mice.We further confirmed that the DX5−CD11c^(hi) liver-resident NK cell subset colocalized with CD4^(+) T cells and inhibited CD4^(+) T cell proliferation.Gene expression microarray analysis demonstrated that liver-resident NK cells had a distinct gene expression pattern consisting of the increased expression of genes involved in negative regulatory functions in the context of the inflammatory microenvironment.Zhi-Bin Zhao Fang-Ting Lu Hong-Di Ma Yin-Hu Wang Wei Yang Jie Long Qi Miao Weici Zhang Zhigang Tian William M.Ridgway Jie Cao M.Eric Gershwin Zhe-Xiong Lian 2020Cellular & Molecular Immunology2020,17,2:4
3原发性胆汁性肝硬化的细胞免疫学发病机制显示文摘原发性胆汁性肝硬化(PBC)是一种器官特异性的进行性自身免疫疾病,常见于女性,以肝脏门脉周围的淋巴细胞浸润,胆管上皮细胞特异性损伤以及血清中高滴度抗线粒体抗体(AMA)为主要特征。最新的PBC研究采用了CD4启动子控制下的TGFβ受体2显性失活(dnTGFβRⅡ)小鼠模型,这种小鼠模型很好地模拟了PBC患者的典型特征。在这种小鼠的基因背景下,通过分别敲除Rag1,μ以及CD1d基因建立了多种双基因缺陷鼠,通过研究发现T细胞,B细胞以及CD1d限制的NKT细胞等对肝脏淋巴细胞浸润,胆管上皮细胞损伤以及AMA的产生发挥着重要的作用。一系列的细胞免疫学实验结果显示,是CD8+T细胞而非CD4+T细胞在肝脏损伤过程中起着决定性的作用,而B细胞除分泌抗自身抗体之外,还具有抑制PBC发生的免疫调节作用。这些机制的研究为揭示人类PBC疾病的细胞免疫学致病机理提供了有力的证据和依托。王寅虎 杨微 M.Eric Gershwin 廉哲雄 2011临床肝胆病杂志2011,27,6:4

Although PBC is a slowly progressive disease, with a natural history of 10–15 years leading to end-stage liver disease, highrisk cases may rapidly progress to decompensated cirrhosis or even death. As the most common autoimmune liver disease, the prevalence of PBC has been increasing globally, largely due to increased disease awareness, improved diagnosis, and treatment with ursodeoxycholic acid (UDCA). 7Treatment with a JAK1/2 inhibitor ameliorates murine autoimmune cholangitis induced by IFN overexpression显示文摘The interferon(IFN)signaling pathways are major immunological checkpoints with clinical significance in the pathogenesis of autoimmunity.We have generated a unique murine model named ARE-Del,with chronic overexpression of IFNγ,by altering IFNγmetabolism.Importantly,these mice develop an immunologic and clinical profile similar to patients with primary biliary cholangitis,including high titers of autoantibodies and portal inflammation.We hypothesized that the downregulation of IFN signaling pathways with a JAK1/2 inhibitor would inhibit the development and progression of cholangitis.To study this hypothesis,ARE-Del^(+/−)mice were treated with the JAK1/2 inhibitor ruxolitinib and serially studied.JAK inhibition resulted in a significant reduction in portal inflammation and bile duct damage,associated with a significant reduction in splenic and hepatic CD4^(+)T cells and CD8^(+)T cells.Functionally,ruxolitinib inhibited the secretion of the proinflammatory cytokines IFNγand TNF from splenic CD4^(+)T cells.Additionally,ruxolitinib treatment also decreased the frequencies of germinal center B(GC B)cells and T follicular helper(Tfh)cells and led to lower serological AMA levels.Of note,liver and peritoneal macrophages were sharply decreased and polarized from M1 to M2 with a higher level of IRF4 expression after ruxolitinib treatment.Mechanistically,ruxolitinib inhibited the secretion of IL-6,TNF and MCP1 and the expression of STAT1 but promoted the expression of STAT6 in macrophages in vitro,indicating that M1 macrophage polarization to M2 occurred through activation of the STAT6-IRF4 pathway.Our data highlight the significance,both immunologically and clinically,of the JAK/STAT signaling pathway in autoimmune cholangitis.Tihong Shao Patrick S.C.Leung Weici Zhang Koichi Tsuneyama William M.Ridgway Howard A.Young Zongwen Shuai Aftab A.Ansari M.Eric Gershwi 2022Cellular & Molecular Immunology2022,19,10:1

15The long and latent road to autoimmunity显示文摘Autoimmune diseases affect nearly 5%of the general population,yet etiology remains poorly understood.Genomic factors are clearly necessary but remain insufficient to explain the loss of tolerance;environmental and stochastic factors fill this gap.This paradigm is indicated by the concordance rates(ranging between 4 and 63%)for autoimmune diseases among monozygotic twins,who share an identical genome,compared to the invariably lower rates in dizygotic twins.The term“exposome”cumulatively refers to the non-hereditary(i.e.,environmental)factors that account for the remaining susceptibility and include endogenous factors such as hormones.Accumulating evidence suggests that exposure to infections,drugs,vaccines,and chemicals may contribute to the loss of tolerance.The mechanisms by which environmental factors can shape the immune system to generate autoimmunity include molecular mimicry,self-antigen modification,bystander activation,and immune reactivity modulation.In all cases,we should first consider the prolonged time between an environmental trigger and the appearance of autoreactivity and subsequent clinical disease.In this special issue,several excellent reviews discuss the potential mechanisms linking environmental factors and autoimmunity.Carlo Selmi Bin Gao M.Eric Gershwin 2018Cellular & Molecular Immunology2018,15,6:0
