1Effects of temperature and organic and inorganic nutrients on the growth of Chattonella marina (Raphidophyceae)from the Daya Bay,South China Sea显示文摘The effects of temperature and different forms of nutrients on Chattonella marina growth have been investigated in strains isolated from the Daya Bay,the South China Sea.The strain of C.marina preferred high temperatures, with an optimal temperature of 25°C,and 18 °C was the minimum for its survival.Higher cell number and growth rate were obtained in high nitrogen and phosphorus concentrations (500 μ g/L, 74 μ g/L) than under nutrient limitation.Nitrogen influenced the growth most, as the specific growth rate and maximum cell density were lower in nitrogen-limited cultures than noted under phosphorus limitation or under limitation from both.C.marina was capable of using many kinds of organic nitrogen sources including L-serine (L-Ser),glycine (Gly),alanine (Ala),L-threonine (L-Thr), glutamic acid (Glu) and urea, but could not utilize uric acid.Various forms of organic phosphorus compound such as glucose-6-phosphate (G6P),sodium glycerophosphate (GYP),adenosine triphosphate (ATP),adenosine monophosphate (AMP),cytidine monophosphate (CMP),guanosine monophosphate (GMP),uridine monophosphate (UMP),4-nitrophenylphosphate (NPP) and triethyl phosphate (TEP) supported the growth as well. Algal cells had the ability to sustain growth under nitrogen-and/or phosphorus-free conditions particularly under phosphorus depleted condition.These results led to the hypothesis that high loading of nitrogen has played an important role in frequent C.marina blooms in the past decade,and its capability for utilization of diverse forms of organic nutrients and growth in low nutrient conditions make this species a likely recurrent dominant in the Daya Bay phytoplankton assemblages,visibleWANG Zhaohui YUAN Meiling LIANG Yu LU Sollghui 2011Acta Oceanologica Sinica2011,30,3:9
2Spatial-resolved metabolomics reveals tissue-specific metabolic reprogramming in diabetic nephropathy by using mass spectrometry imaging显示文摘Detailed knowledge on tissue-specific metabolic reprogramming in diabetic nephropathy(DN)is vital for more accurate understanding the molecular pathological signature and developing novel therapeutic strategies.In the present study,a spatial-resolved metabolomics approach based on air flowassisted desorption electrospray ionization(AFADESI)and matrix-assisted laser desorption ionization(MALDI)integrated mass spectrometry imaging(MSI)was proposed to investigate tissue-specific metabolic alterations in the kidneys of high-fat diet-fed and streptozotocin(STZ)-treated DN rats and the therapeutic effect of astragalosideⅣ,a potential anti-diabetic drug,against DN.As a result,a wide range of functional metabolites including sugars,amino acids,nucleotides and their derivatives,fatty acids,phospholipids,sphingolipids,glycerides,carnitine and its derivatives,vitamins,peptides,and metal ions associated with DN were identified and their unique distribution patterns in the rat kidney were visualized with high chemical specificity and high spatial resolution.These region-specific metabolic disturbances were ameliorated by repeated oral administration of astragaloside Ⅳ(100 mg/kg)for 12 weeks.This study provided more comprehensive and detailed information about the tissue-specific metabolic reprogramming and molecular pathological signature in the kidney of diabetic rats.These findings highlighted the promising potential of AFADESI and MALDI integrated MSI based metabolomics approach for application in metabolic kidney diseases.Zhonghua Wang Wenqing Fu Meiling Huo Bingshu He Yaqi Liu Lu Tian Wanfang Li Zhi Zhou Baili Wang Jianzhen Xia Yanhua Chen Jinfeng Wei Zeper Abliz 2021Acta Pharmaceutica Sinica B2021,11,11:7

4Wheat breeding history reveals synergistic selection of pleiotropic genomic sites for plant architecture and grain yield显示文摘Diversity surveys of crop germplasm are important for gaining insights into the genomic basis for plant architecture and grain yield improvement,which is still poorly understood in wheat.In this study,we exome sequenced 287 wheat accessions that were collected in the past 100 years.Population genetics analysis identified that 6.7%of the wheat genome falls within the selective sweeps between landraces and cultivars,which harbors the genes known for yield improvement.These regions were asymmetrically distributed on the A and B subgenomes with regulatory genes being favorably selected.Genome-wide association study(GWAS)identified genomic loci associated with traits for yield potential,and two underlying genes,TaARF12 encoding an auxin response factor and TaDEP1 encoding the G-proteinγ-subunit,were located and characterized to pleiotropically regulate both plant height and grain weight.Elite single-nucleotide haplotypes with increased allele frequency in cultivars relative to the landraces were identified and found to have accumulated over the course of breeding.Interestingly,we found that TaARF12 and TaDEP1 function in epistasis with the classical plant height Rht-1 locus,leading to propose a“Green Revolution”-based working model for historical wheat breeding.Collectively,our study identifies selection signatures that fine-tune the gibberellin pathway during modern wheat breeding and provides a wealth of genomic diversity resources for the wheat research community.Aili Li Chenyang Hao Zhenyu Wang Shuaifeng Geng Meiling Jia Fang Wang Xiang Han Xingchen Kong Lingjie Yin Shu Tao Zhongyin Deng Ruyi Liao Guoliang Sun Ke Wang Xingguo Ye Chengzhi Jiao Hongfeng Lu Yun Zhou Dengcai Liu Xiangdong Fu Xueyong Zhang Long Mao 2022Molecular Plant2022,15,3:4
5Comparative transcriptomics revealed enhanced light responses, energy transport and storage in domestication of cassava(Manihot esculenta)显示文摘Cassava is a staple food, feed and bioenergy crop important to the world especially in the tropics.Domesticated cassava is characterized by powerful carbohydrate accumulation but its wild progenitor is not.Here, we investigated the transcriptional differences of eight c DNA libraries derived from developing leaf, stem and storage root of cassava cv. Arg7 and an ancestor line,W14, using next generation sequencing system. A total of41302 assembled transcripts were obtained and from these,25961 transcripts with FPKM≥3 in at least one library were named the expressed genes. A total of 2117, 1963 and3584 transcripts were found to be differentially expressed in leaf, stem and storage root(150 d after planting),respectively, between Arg7 and W14 and ascribed to 103,93 and 119 important pathways in leaf, stem and storage root, respectively. The highlight of this work is that the genes involved in light response, such as those for photosystem I(PSA) and photosystem II(PSB), other genes involved in light harvesting, and some of the genes in the Calvin cycle of carbon fixation were specially upregulated in leaf. Genes for transport and also for key rate-limiting enzymes(PFK, PGK and PK, GAPDH)coupling ATP consumption in glycolysis pathway were predominantly expressed in stem, and genes for sucrose degradation(INVs), amylose synthesis(GBSS) and hydrolysis(RCP1, AMYs), the three key steps of starch metabolism, and transport associated with energy translocation(ABC, AVPs and ATPase) and their upstream transcription factors had enhanced expression in storage root in domesticated cassava. 6Cycle number of neoadjuvant chemotherapy might influence survival of patients with T1-4N2-3M0 nasopharyngeal carcinoma显示文摘Objective: Stage N2-3 nasopharyngeal carcinoma(NPC) shows a high risk of distant metastasis, which will finally cause death. This study aimed to evaluate the impact of neoadjuvant chemotherapy(NACT) of various cycles before radical radiotherapy on distant metastasis and survival of patients with stage N2-3 diseases.Methods: In this study, a total of 1,164 consecutive patients with non-metastatic N2-3 NPC were recruited and prospectively observed. Then 231 patients who received NACT of 4 cycles(NACT=4 group) were matched 1:2:1 to 462 patients treated with NACT of 2 cycles(NACT=2 group) and 231 patients treated without NACT(NACT=0 group), according to age, histological subtype, N stage and NACT regimen. Five candidate variables(sex, T stage, concurrent chemotherapy, intensity-modulated radiation therapy and cycle number of NACT) were analyzed for their association with patients' survival.Results: After matching, the overall survival(OS), disease-free survival(DFS), local-recurrence-free survival(RFS) and distant-metastasis-free survival(MFS) of the NACT=4 group(89.2%, 81.0%, 83.3% and 84.8%,respectively) were better than those of the NACT=2 group(83.3%, 72.5%, 81.2% and 77.9%, respectively) and the NACT=0 group(74.0%, 63.2%, 74.0% and 68.8%, respectively). 8Characteristics of oral methicillin-resistant Staphylococcus epidermidis isolated from dental plaque显示文摘The oral microbial community is widely regarded as a latent reservoir of antibiotic resistance genes. This study assessed the molecular epidemiology, susceptibility profile, and resistance mechanisms of 35 methicillin-resistant Staphylococcus epidermidis(MRSE) strains isolated from the dental plaque of a healthy human population. Broth microdilution minimum inhibitory concentrations(MICs) revealed that all the isolates were nonsusceptible to oxacillin and penicillin G. Most of them were also resistant to trimethoprim(65.7%) and erythromycin(54.3%). The resistance to multiple antibiotics was found to be largely due to the acquisition of plasmid-borne genes. The mec A and dfr A genes were found in all the isolates, mostly dfr G(80%), aac A-aph D(20%), aad D(28.6%), aph A3(22.9%), msr A(5.7%), and the erm C gene(14.3%). Classical mutational mechanisms found in these isolates were mainly efflux pumps such as qac A(31.4%), qac C(25.7%), tet K(17.1%), and nor A(8.6%). Multilocus sequence type analysis revealed that sequence type 59(ST59) strains comprised 71.43% of the typed isolates, and the e BURST algorithm clustered STs into the clonal complex 2-II(CC2-II). The staphyloccoccal cassette chromosome mec(SCCmec) type results showed that 25(71.43%) were assigned to type IV. Moreover, 88.66% of the isolates were found to harbor six or more biofilm-associated genes. The aap, atl E, embp, sdr F, and IS256 genes were detected in all 35 isolates. 9CHRT:Clustering-Based Hybrid Re-Routing System for Traffic Congestion Avoidance显示文摘Re-routing system has become an important technology to improve traffic efficiency.The traditional re-routing schemes do not consider the dynamic characteristics of urban traffic,making the planned routes unable to cope with the changing traf-fic conditions.Based on real-time traffic information,it is challenging to dynamically re-route connected vehicles to alleviate traffic congestion.Moreover,how to obtain global traffic information while reducing communication costs and improving travel efficiency poses a challenge to the re-routing system.To deal with these challenges,this paper proposes CHRT,a clustering-based hybrid re-routing system for traffic congestion avoidance.CHRT develops a multi-layer hybrid architecture.The central server accesses the global view of traffic,and the distributed part is composed of vehicles divided into clusters to reduce latency and communication overhead.Then,a clustering-based priority mechanism is proposed,which sets priorities for clusters based on realtime traffic information to avoid secondary congestion.Furthermore,to plan the optimal routes for vehicles while alleviating global traffic congestion,this paper presents a multi-metric re-routing algorithm.Through extensive simulations based on the SUMO traffic simulator,CHRT reduces vehicle traveling time,fuel consumption,and CO2 emissions compared to other systems.In addition,CHRT globally alleviates traffic congestion and improves traffic efficiency.Jie Huo Xiangming Wen Luning Liu Luhan Wang Meiling Li Zhaoming Lu 2021China Communications2021,18,7:2

11Termination of Transcription of LAT Increases the Amounts of ICP0 mRNA but Does Not Alter the Course of HSV-1 Infection in Latently Infected Murine Ganglia显示文摘On entering sensory ganglia,herpes simplex viruses 1(HSV-1)establishes a latent infection with the synthesis of a latency associated transcript(LAT)or initiates productive infection with expression of a set of immediate early viral proteins.The precise mechanisms how expression of a genes is suppressed during the latency are unknown.One mechanism that has been proposed is illustrated in the case of ICP0,a key immediate early viral regulatory protein.Specifically,the 2 kb LAT intron is complementary to the 30 terminal portion of ICP0 m RNA.To test the hypothesis that accumulation of LAT negatively affects the accumulation of ICP0 m RNA,we inserted a DNA fragment encoding two poly(A)sequences into LAT to early terminate LAT transcript without interrupting the complementary sequence of ICP0 transcript(named as SR1603).Comparisons of the parent(SR1601)and mutant(SR1603)HSV-1 viruses showed the following:Neurons harboring latent SR1603 virus accumulated equivalent amounts of viral DNA but higher amounts of ICP0 m RNA and lower amounts of LAT,when compared to neurons harboring the SR1601 virus.One notable difference between the two viruses is that viral RNA accumulation in explanted ganglia harboring SR1603 virus initiated significantly sooner than that in neurons harboring SR1601 virus,suggesting that ICP0 may act as an activator of viral gene expression in permissive cells.Collectively,these data suggest that increased ICP0 m RNA by suppressed LAT did not affect the establishment of latency in latently infected murine ganglia.Haifang Jiang Jiaming Wu Xianjie Liu Ruitao Lu Manling Zhou Meiling Chen Yonghong Liu Grace Guoying Zhou Wenmin Fu 2021Virologica Sinica2021,36,2:2

19Surface decoration of development-inspired synthetic N-cadherin motif via Ac-BP promotes osseointegration of metal implants显示文摘Research works on the synergistic effect of surface modified bioactive molecules and bone metal implants have been highlighted.N-cadherin is regarded as a key factor in directing cell-cell interactions during the mesenchymal condensation preceding the osteogenesis in the musculoskeletal system.In this study,the N-cadherin mimetic peptide(Cad)was biofunctionalized on the titanium metal surface via the acryloyl bisphosphonate(Ac-BP).To learn the synergistic effect of N-cadherin mimetic peptide,when tethered with titanium substrates,on promoting osteogenic differentiation of the seeded human mesenchymal stem cells(hMSCs)and the osseointegration at the bone-implant interfaces.Results show that the conjugation of N-cadherin mimetic peptide with Ac-BP promoted the osteogenic gene markers expression in the hMSCs.The biofunctionalized biomaterial surfaces promote the expression of the Wnt/β-catenin downstream axis in the attached hMSCs,and then enhance the in-situ bone formation and osseointegration at the bone-implant interfaces.We conclude that this N-cadherin mimetic peptide tethered on Ti surface promote osteogenic differentiation of hMSCs and osseointegration of biomaterial implants in vitro and in vivo.These findings demonstrate the importance of the development-inspired surface bioactivation of metal implants and shed light on the possible cellular mechanisms of the enhanced osseointegration.Meiling Zhu Kunyu Zhang Lu Feng Sien Lin Qi Pan Liming Bian Gang Li 2021Bioactive Materials2021,6,5:1
