2Full-thickness tissue engineered skin constructed with autogenic bone marrow mesenchymal stem cells显示文摘To explore the feasibility of repairing clinical cutaneous deficiency, autogenic bone marrow mesen-chymal stem cells (BMSCs) were isolated and differentiated into epidermal cells and fibroblasts in vitro supplemented with different inducing factors and biomaterials to construct functional tissue- engineered skin. The results showed that after 72 h induction, BMSCs displayed morphologic changes such as typical epidermal cell arrangement, from spindle shape to round or oval; tonofibrils, melano-somes and keratohyaline granules were observed under a transmission electronic microscope. The differentiated cells expressed epidermal stem cell surface marker CK19 (59.66% ± 4.2%) and epidermal cells differentiation marker CK10. In addition, the induced epidermal cells acquired the anti-radiation capacity featured by lowered apoptosis following exposure to UVB. On the other hand, the collagen microfibrils deposition was noticed under a transmission electronic microscope after differentiating into dermis fibroblasts; RT-PCR identified collagen type I mRNA expression in differentiated cells; radioimmunoassay detected the secretion of interleukin-6 (IL-6) and interleukin-8 (IL-8) (up to 115.06 pg/mL and 0.84 ng/mL, respectively). Further in vivo implanting BMSCs with scaffold material short-ened skin wound repair significantly. 3Resequencing of 145 Landmark Cultivars Reveals Asymmetric Sub-genome Selection and Strong Founder Genotype Effects on Wheat Breeding in China显示文摘Controlled pedigrees and the multi-decade timescale of national crop plant breeding programs offer a unique experimental context for examining how selection affects plant genomes.More than 3000 wheat cultivars have been registered,released,and documented since 1949 in China.In this study,a set of 145 elite cultivars selected from historical points of wheat breeding in China were re-sequenced.A total of 43.75 Tb of sequence data were generated with an average read depth of 17.94x for each cultivar,and more than 60.92 million SNPs and 2.54 million InDels were captured,based on the Chinese Spring RefSeq genome v1.0.Seventy years of breeder-driven selection led to dramatic changes in grain yield and related phenotypes,with distinct genomic regions and phenotypes tar-geted by different breeders across the decades.There are very clear instances illustrating how introduced Italian and otherforeign germplasm was integrated into Chinese wheat programs and reshaped the genomic landscape of local modern cultivars.Importantly,the resequencing data also highlighted significant asymmetric breeding selec-tion among the three sub-genomes:this was evident in both the collinear blocks for homeologous chromosomes and among sets of three homeologous genes.Accumulation of more newly assembled genes in newer cultivars implied the potential value of these genes in breeding.Conserved and extended sharing of linkage disequilibrium(LD)blocks was highlighted among pedigree-related cultivars,in which fewer haplotype differences were detected.Fixation or replacement of haplotypes from founder genotypes after generations of breeding was related to their breeding value.Based on the haplotype frequency changes in LD blocks of pedigree-related cultivars,we propose a strategy for evaluating the breeding value of any given line on the basis of the accumulation(pyramiding)of bene-ficial haplotypes.Collectively,our study demonstrates the influence of 'founder genotypes' on the output of breeding efforts over many decades and also suggests that founder genotype perspectives are in fact more dy-namic when applied in the context of modern genomics-informed breeding.Chenyang Hao Chengzhi Jiao Jian Hou Tian Li Hongxia Liu Yuquan Wang Jun Zheng Hong Liu Zhihong Bi Fengfeng Xu Jing Zhao Lin Ma Yamei Wang Uzma Majeed Xu Liu Rudi Appels Marco Maccaferri Roberto Tuberosa Hongfeng Lu Xueyong Zhang 2020Molecular Plant2020,13,12:9
4Elemental sulfur in northern South China Sea sediments and its significance显示文摘Elemental sulfur(ES) is one of the intermediates in the inorganic sulfur cycle and thus plays a key role in the fractionation of stable sulfur isotopes in different reservoirs and the marine environment. In this study, solid ES is discovered in sediments near the Jiulong Methane Reef in the northern South China Sea by scanning electron microscopy and Raman spectroscopy. Combining the morphology and distribution of ES, pyrite concentrations, and sulfur isotopes, we conclude that:(1) solid ES coexists with pyrite microcrystals and sulfide(oxyhydr)oxides as well as clay minerals, and they are mainly distributed on the surface of mineral aggregates;(2) ES mainly occurs within and near the sulfate-methane transition zone(SMTZ) despite little morphological diversity;(3) ES formation might be related to hydrogen sulfide oxidation and is therefore linked with fluctuations in the SMTZ. Within the SMTZ, hydrogen sulfide is produced and pyrite precipitates because of enhanced anaerobic oxidation of methane coupled with dissimilatory sulfate reduction. This enhances the efficiency of the inorganic sulfur cycle and provides favorable conditions for ES formation. 5Geochemical and microbial characters of sediment from the gas hydrate area in the Taixinan Basin, South China Sea显示文摘The Taixinan Basin is one of the most potential gas hydrate bearing areas in the South China Sea and abundant gas hydrates have been discovered during expedition in 2013. In this study, geochemical and microbial methods are combinedly used to characterize the sediments from a shallow piston Core DH_CL_11(gas hydrate free) and a gas hydrate-bearing drilling Core GMGS2-16 in this basin. Geochemical analyses indicate that anaerobic oxidation of methane(AOM) which is speculated to be linked to the ongoing gas hydrate dissociation is taking place in Core DH_CL_11 at deep. For Core GMGS2-16, AOM related to past episodes of methane seepage are suggested to dominate during its diagenetic process; while the relatively enriched δ18O bulk-sediment values indicate that methane involved in AOM might be released from the 'episodic dissociation' of gas hydrate.Microbial analyses indicate that the predominant phyla in the bacterial communities are Firmicutes and Proteobacteria(Gammaproteobacteria and Epsilonproteobacteria), while the dominant taxa in the archaeal communities are Marine_Benthic_Group_B(MBGB), Halobacteria, Thermoplasmata, Methanobacteria,Methanomicrobia, Group C3 and MCG. Under parallel experimental operations, comparable dominant members(Firmicutes and MBGB) are found in the piston Core DH_CL_11 and the near surface layer of the long drilling Core GMGS2-16. Moreover, these members have been found predominant in other known gas hydrate bearing cores, and the dominant of MBGB has even been found significantly related to gas hydrate occurrence. Therefore,a high possibility for the existing of gas hydrate underlying Core DH_CL_11 is inferred, which is consistent with the geochemical analyses. 13Mechanism of progressive broad deformation from oceanic transform valley to off-transform faulting and rifting显示文摘Oceanic transform faults(TFs)are commonly viewed as single,narrow strikeslip seismic faults that offset two mid-ocean ridge segments.However,broad zones of complex deformation are ubiquitous at TFs.Here,we propose a new conceptual model for the progressive deformation within broad zones at oceanic TFs through detailed morphological,seismic,and stress analyses.We argue that,under across-transform extension due to a change in plate motion,plate deformation occurs first along high-angle transtensional faults(TTFs)within the transform valleys.Off-transform normal faults(ONFs)form when across-transform deviatoric extensional stresses exceed the yield strength of the adjacent oceanic lithosphere.With further extension,these normal faults can develop into off-transform rift zones(ORZs),some of which can further develop into transform plate boundaries.We illustrate that such progressive complex deformation is an inherent feature of oceanic TFs.The new conceptual model provides a unifying theory to explain the observed broad deformation at global transform systems.Fan Zhang Jian Lin Zhiyuan Zhou Hongfeng Yang Jason P.Morgan 2022The Innovation2022,3,1:0

15A systematic assessment of hydroxyapatite nanoparticles used in the treatment of melanoma显示文摘Nano Research volume 13,pages2106–2117(2020)Cite this article 106 Accesses 1 Altmetric Metrics details Abstract Melanoma is a highly malignant skin tumor which is prone to recurrence and metastasis.Hydroxyapatite nanoparticles(nHAPs)were reported to possess a suppressive effect on proliferation of various tumor cells in vitro.This study aimed to assess in vitro and in vivo anti-tumor ability and biosafety of the nHAPs used in the treatment of melanoma.Three types of nHAPs with different morphology and crystallinity were synthesized.In vitro cell viability and proliferation studies demonstrated that all three types of nHAPs can inhibit viability and proliferation of A375 and SK-MEL-28 melanoma cells in a concentration-dependent manner.In addition,the rod-shape nHAPs with a crystallinity of 45.60%had the most prominent suppressive effect on the two melanoma cells tested.An important positive regulator of G1/S phase transition in cell cycle,Cyclin D1 protein,was reduced by nHAPs treatment in vivo.We further discovered that the migration ability of the nHAPs treated melanoma cells was greatly decreased.RNA sequencing result revealed that melanoma metastasis related genes were down-regulated by nHAPs,including MMP2,MMP14,ITGA9,ITGB3,ITGB4 and S100B.High concentration of nHAPs treatment in melanoma-bearing nude mice showed a strong inhibitory effect on tumor size and weight.Most importantly,hemolysis,electrolyte disturbance or inflammation response was not discovered in the experimental animals from nHAPs treated groups.We proved that the nHAPs synthesized in the current study has a selective effect to suppress melanoma tumor proliferation and was safe with regard to normal cells and tissue.Zhongtao Li Jiaoqing Tang Hongfeng Wu Zhixin Ling Siyu Chen Yong Zhou Bo Guo Xiao Yang Xiangdong Zhu Lin Wang Chongqi Tu Xingdong Zhang 2020Nano Research2020,13,8:0
