| 1 | Wheat powdery mildew resistance gene Pm64 derived from wild emmer (Triticum turgidum var.dicoccoides) is tightly linked in repulsion with stripe rust resistance gene Yr5显示文摘Stripe rust and powdery mildew are both devastating diseases for durum and common wheat.Pyramiding of genes conferring resistance to one or more diseases in a single cultivar is an important breeding approach to provide broader spectra of resistances in wheat improvement. A new powdery mildew resistance gene originating from wild emmer(Triticum turgidum var.dicoccoides) backcrossed into common wheat(T. aestivum) line WE35 was identified. It conferred an intermediate level of resistance to Blumeria graminis f. sp. tritici isolate E09 at the seedling stage and a high level of resistance at the adult plant stage. Genetic analysis showed that the powdery mildew resistance in WE35 was controlled by a dominant gene designated Pm64. Bulked segregant analysis(BSA) and molecular mapping indicated that Pm64 was located in chromosome bin 2 BL4-0.50–0.89. Polymorphic markers were developed from the corresponding genomic regions of Chinese Spring wheat and wild emmer accession Zavitan to delimit Pm64 to a 0.55 cM genetic interval between markers WGGBH1364 and WGGBH612, corresponding to a 15 Mb genomic region on Chinese Spring and Zavitan 2 BL, respectively. The genetic linkage map of Pm64 is critical for fine mapping and cloning. Pm64 was completely linked in repulsion with stripe rust resistance gene Yr5. | 2 | Formation condition of deep gas reservoirs in tight sandstones in Kuqa Foreland Basin显示文摘Due to the high expense of deep oil and gas exploration,prediction of gas-bearing properties before drilling is crucial for deep gas reservoir of tight sandstone.Deep tight sandstone gas fields in Kuqa Foreland Basin are characterized by high abundance,high gas saturation,high pressure,high and stable yield,which belong to high-efficiency tight gas reservoir.Based on theoretical analysis of controlling factors and mechanisms of gas-bearing properties for tight sandstone gas reservoir,and taking tight sandstone gas fields with high effectiveness such as Dibei,Keshen and Dibei gas fields in Kuqa Foreland Basin as examples,formation condition and mechanism of high-efficiency tight sandstone gas reservoir in Kuqa area are studied through a comparative analysis of typical tight sandstone gas reservoir in Sichuan Basin and Ordos Basin.The results show that the formation condition of deep gas reservoir of tight sandstone in Kuqa foreland basin includes four factors:i.e.,overpressure gas charging,fracture development,“early-oil and late-gas”accumulation process and favorable preservation condition.The overpressure gas charging and fracture development are the most important factors for formation of high-efficiency tight gas reservoirs in Kuqa Foreland Basin.High-quality source rocks,high sourcereservoir pressure difference,and overpressure filling induced thereby are preconditions for formation of tight sandstone with high gas saturation.The fracture development controls gas migration,accumulation,and high yield of tight sandstone gas reservoir.The reservoir wettability changed by the early oil charging is beneficial to late natural gas charging,and the preservation condition of high-quality gypsum cap rocks is the key factor for gas reservoirs to maintain overpressure and high gas saturation.Matching of above four favorable factors leads to the tight sandstone gas reservoir with high abundance,high gas saturation and high gas production in Kuqa Foreland Basin,which is very different from other basins.Under the condition of little difference in physical property of tight sandstone reservoir,excessive source-reservoir pressure difference,facture development,preservation condition and current formation overpressure are the most significant factors to be considered in exploration and evaluation of deep tight sandstone gas. | Xuesong Lu Mengjun Zhao Keyu Liu Qingong Zhuo Junjia Fan Zhichao Yu Yanjie Gong | 2018 | Petroleum Research2018,3,4: | 4 |
| 3 | Detection of Porcine Epidemic Diarrhea Virus in Guangxi Province from 2011 to 2014 and Sequence Analysis of Its M Gene显示文摘Detection of pigs epidemic diarrhea virus( PEDV) was conducted on 331 piglets diarrhea fecal samples collected in Nanning,Yulin and other 12 areas of Guangxi Province from January of 2011 to April of 2014 by the method of reverse transcription-polymerase chain reaction( RT-PCR). The results showed that the positive samples of PEDV were 210 and the positive rate was 63. 44%. The clone and sequencing of M gene was carried out on 25 positive samples. PEDV reference strains were selected from Gene Bank to conduct the sequence homology alignment analysis and the phylogenetic tree of M gene. The M gene homology and amino acid sequence identity between 25 isolated strains and 51 reference strains were 96. 0%- 99. 6% and 94. 3%- 99. 6%,respectively. The genetic variation analysis of M gene showed that the genetic relationship of PEDV prevalent strains in Guangxi Province from 2013 to 2014 was close to that of the prevalent strains in Beijing,Anhui,Wuhan,Hebei and Guangdong from 2010 to 2013,and which were far from that of the Chinese early isolates CH / S( Gen Bank number: JN547228),vaccine strain CV777( Gen Bank number: AF353511) and Attenuated DR13( Gen Bank number: JQ023162). | 4 | Human health risk assessment of groundwater nitrate at a two geomorphic units transition zone in northern China显示文摘To assess groundwater nitrate contamination and its human health risks,489 unconfined groundwater samples were collected and analyzed from Zhangjiakou,northern China.The spatial distribution of principle hydrogeochemical results showed that the average concentrations of ions in descend order was HCO3-,SO42-,Na+,Ca2+,Cl-,NO3-,Mg2+and K+,among which the NO3-concentrations were between 0.25 and 536.73 mg/L with an average of 29.72 mg/L.In total,167 out of 489 samples(34%)exceeded the recommended concentration of 20 mg/L in Quality Standard for Groundwater of China.The high NO3-concentration groundwater mainly located in the northern part and near the boundary of the two geomorphic units.As revealed by statistical analysis,the groundwater chemistry was more significantly affected by anthropogenic sources than by the geogenic sources.Moreover,human health risks of groundwater nitrate through oral and dermal exposure pathways were assessed by model,the results showed that about 60%,50%,32%and 26%of the area exceeded the acceptable level(total health index>1)for infants,children,adult males and females,respectively.The health risks for different groups of people varied significantly,ranked:infants>children>adult males>adult females,suggesting that younger people are more susceptible to nitrate contamination,while females are more resistant to nitrate contamination than males.To ensure the drinking water safety in Zhangjiakou and its downstream areas,proper management and treatment of groundwater will be necessary to avoid the health risks associated with nitrate contamination. | Huiliang Wang Keyu Lu Chenyang Shen Xiaoguang Song Bin Hu Gang Liu | 2021 | Journal of Environmental Sciences2021,33,12: | 2 |

| 11 | Unveiling mitophagy-mediated molecular heterogeneity and development of a risk signature model for colorectal cancer by integrated scRNA-seq and bulk RNA-seq analysis显示文摘Background:Accumulating researchers have recognized mitophagy as a key player in tumors,but few studies have investigated its role in the tumor microenvironment(TME).Advances in the technology of single-cell RNA sequencing(scRNA-seq)have allowed unveiling the concealed features of the TME at cellular resolution.This study aimed to elucidate the role of mitophagy within the TME of colorectal cancer(CRC)and to establish a mitophagy-mediated risk model.Methods:We assessed mitophagy-related pathway activities at both single-cell and tissue levels.Subsequently,an unsupervised clustering algorithm was employed to identify mitophagy-mediated subtypes.Furthermore,we developed a mitophagy-mediated risk signature(MMRS)using least absolute shrinkage and selection operator(LASSO)Cox analysis and constructed a MMRS model incorporating the risk score and clinical variables.Subsequently,we used quantitative reverse transcription polymerase chain reaction analysis to verify the expression of the screened genes.Results:We retrieved and annotated a total of 14,719 cells from eight samples in the scRNA-seq GSE132465 data set.The activities of mitophagy-related pathways were uniformly upregulated in cancer cells.Integrating with bulk RNA-seq data,we identified two mitophagy-mediated clusters(C1 and C2)with distinct characteristics and prognoses.C2 was identified as a mitophagy-high cluster.Then,we developed a five-gene MMRS via LASSO Cox analysis in The Cancer Genome Atlas(TCGA)cohort.We utilized the GSE39582 cohort to validate the efficacy of our model.The expression of CX3CL1 and INHBB was upregulated in CRC tissues.Conclusions:The present study identified two mitophagy-mediated CRC subtypes with distinct features.Our MMRS may provide potential therapeutic strategies for CRC.The findings of our work offer novel insights into the involvement of mitophagy in CRC. | Han Gao Qi Zou Linyun Ma Keyu Cai Yi Sun Li Lu Donglin Ren Bang Hu | 2023 | Gastroenterology Report2023,11,1: | 0 |

| 14 | External rectal prolapse:abdominal or perineal repair for men?A retrospective cohort study显示文摘Background:External rectal prolapse is a relatively rare disease,in which male patients account for a minority.The selection of abdominal repair or perineal repair for male patients has rarely been investigated.Methods:Fifty-one male patients receiving abdominal repair(laparoscopic ventral rectopexy)or perineal repair(Delorme or Altemeier procedures)at the Sixth Affiliated Hospital of Sun Yat-sen University(Guangzhou,China)between March 2013 and September 2019 were retrospectively analysed.We compared the recurrence,complication rate,post-operative defecation disorder,length of stay,and quality of life between the abdominal and perineal groups.Results:Of the 51 patients,45 had a complete follow-up,with a median of 48.5 months(range,22.8–101.8 months).A total of 35 patients were under age 40 years.The complication rate associated with abdominal repair was less than that associated with perineal repair(0%vs 20.7%,P=0.031)and the recurrence rate was also lower(9.5%vs 41.7%,P=0.018).Multivariate analysis showed that perineal repair(odds ratio,9.827;95%confidence interval,1.296–74.50;P=0.027)might be a risk factor for recurrence.Moreover,only perineal repair significantly improved post-operative constipation status(preoperative vs post-operative,72.4%vs 25.0%,P=0.001).There was no reported mortality in either of the groups.No patient’s sexual function was affected by the surgery.Conclusions:Both surgical approaches were safe in men.Compared with perineal repair,the complication rate and recurrence rate for abdominal repair were lower.However,perineal repair was better able to correct constipation. | Bang Hu Qi Zou Zhenyu Xian Dan Su Chao Liu Li Lu Minyi Luo Zixu Chen Keyu Cai Han Gao Hui Peng Wuteng Cao Donglin Ren | 2022 | Gastroenterology Report2022,10,1: | 0 |

| 18 | Feature extraction of gear-localized defect using adaptive lifting scheme and local gradient maps显示文摘In this paper,the adaptive lifting scheme (ALS) and local gradient maps (LGM) are proposed to isolate the transient feature components from the gearbox vibration signals. Based on entropy minimization rule,the ALS is employed to change properties of an initial wavelet and design adaptive wavelet. Then LGM is applied to characterize the transient feature components in detail signal of decomposition results using ALS. In the present studies, the orthogonal Daubechies 4 (Db 4) wavelet is used as the initial wavelet. The proposed method is applied to both simulated signals and vibration signals acquired from a gearbox for periodic impulses detection. The two conventional methods (cepstrum analysis and Hilbert envelope analysis) and the orthogonal Db4 wavelet are also used to analyze the same signals for comparison.
