| 1 | Glutamate and aspartate alleviate testicular/epididymal oxidative stress by supporting antioxidant enzymes and immune defense systems in boars显示文摘Several potential oxidative agents have damaging effects on mammalian reproductive systems.This study was conducted to investigate the effects of glutamate(Glu)and aspartate(Asp)supplementation on antioxidant enzymes and immune defense systems in the outer scrotum of boars injected with H2O2.A total of 24 healthy boars were randomly divided into 4 treatment groups:control(basal diet,saline-treated),H2O2(basal diet,H2O2-challenged outer scrotum(1 m L kg^–1 BW)),Glu(basal diet+2%Glu,H2O2-challenged),and Asp(basal diet+2%Asp,H2O2-challenged).Our results showed that both Glu and Asp supplementation improved testicular morphology and decreased the genital index in the H2O2-treated boars.Glu and Asp administration increased the antioxidant enzyme activities and affected the testicular inflammatory cytokine secretion but had no effect on sex hormone levels.Furthermore,the m RNA expression of CAT,Cu Zn SOD,and GPx4 was altered in the testes and epididymis of boars treated with Asp and Glu.Glu and Asp supplementation also modulated the expression of TGF-β1,IL-10,TNF-α,IL-6 and IL-1βin the testis and epididymis.These results indicate that dietary Glu and Asp supplementation might enhance antioxidant capacity and regulate the secretion and expression of inflammatory cytokines to protect the testes and epididymis of boars against oxidative stress. | Wenjie Tang Jian Wu Shunshun Jin Liuqin He Qinlu Lin Feijun Luo Xingguo He Yanzhong Feng Binsheng He Pingping Bing Tiejun Li Yulong Yin | 2020 | Science China(Life Sciences)2020,63,1: | 9 |
| 2 | Effects of soil stoichiometry of the CaCO_3/available phosphorus ratio on plant density in Artemisia ordosica communities显示文摘In soil, CaCO3 can reduce the availability of phosphorus in arid and semi-arid regions, thereby influencing plant growth. However, the effects of soil stoichiometry of the CaCO3/available phosphorus ratio (CaCO3/AP ratio) on plant density are unknown. In this study, we examined 4 Artemisia ordosica communities located in arid and semi-arid regions, and performed a pot experiment with an orthogonal design to examine the relationships between the soil CaCO3/AP ratio and plant density. The results showed that the soil stoichiometry of the CaCO3/AP ratio had a greater effect on growth of A. ordosica than either CaCO3 or AP as single factor. The influence of the soil CaCO3/AP ratio on density of A. ordosica was related to the climatic zone and the types and/or amplitudes of increases in the soil CaCO3/AP ratio. When the soil CaCO3/AP ratio was within the same amplitude and was less than 2.5, the A. ordosica density increased in the semi-arid region, whereas the density decreased with increasing soil CaCO3/AP ratios in the arid region. In the semi-arid region, when the soil CaCO3/AP ratio was within the same amplitude and the increase in the soil CaCO3 content was greater than the increase in soil AP, the A. ordosica density increased with increasing soil CaCO3/AP ratios. This means that the relationship between the soil CaCO3/AP ratio and the A. ordosica density remained the same. In the arid region, an increase in the soil CaCO3/AP ratio as a result of increased soil CaCO3 content and decreased soil AP resulted in increased density of A. ordosica. Therefore, the relationship between soil CaCO3/AP ratios and density of A. ordosica was altered by these changes in soil chemistry. To address these two questions, we performed a two-factor (soil nitrate: ammonium ratio and plant density) randomized block design and a uniform-precision rotatable central composite design pot experiments to examine the relationships between soil nitrate: ammonium ratios, the carbon: nitrogen ratios and growth rate of Artemisia sphaerocephala seedlings. Under adequate nutrient status, both soil nitrate: ammonium ratios and plant density influenced the carbon: nitrogen ratios and growth rate of A. sphaerocephala seedlings. Under the lower soil nitrate: ammonium ratios, with the increase of soil nitrate: ammonium ratios, the growth rates of plant height and shoot biomass of A. sphaerocephala seedlings decreased significantly; with the increase of plant carbon: nitrogen ratios, the growth rates of shoot biomass of A. sphaerocephala seedlings decreased significantly. Soil nitrate: ammonium ratios affected the carbon: nitrogen ratios of A. sphaerocephala seedlings by plant nitrogen but not by plant carbon. Thus, soil nitrate: ammonium ratios influenced the carbon: nitrogen ratios of A. sphaerocephala seedlings, and hence influenced its growth rates. Under moderate water stress and adequate nutrient status, both soil N:P and plant density influenced the N:P ratios and growth rates of A. ordosica. With the increase of soil N:P ratios, the growth rates of A. ordosica seedlings decreased significantly. With the increase of soil N:P ratios, N:P ratios in A. ordosica seedlings increased significantly. While the nitrogen concentrations in the plant increased slightly, the phosphorus concentrations significantly decreased. With the increase of plant density, the shoot N:P ratios and growth rates significantly decreased, which resulted from soil N:P ratios. Thus, soil N:P ratios influenced the N:P ratios in A. ordosica seedlings, and hence, influenced its growth. | 11 | Phenotypic plasticity of Artemisia ordosica seedlings in response to different levels of calcium carbonate in soil显示文摘Plant phenotypic plasticity is a common feature that is crucial for explaining interspecific competition, dynamics and biological evolution of plant communities. In this study, we tested the effects of soil CaCO_3(calcium carbonate) on the phenotypic plasticity of a psammophyte, Artemisia ordosica, an important plant species on sandy lands in arid and semi-arid areas of China, by performing pot experiments under different CaCO_3 contents with a two-factor randomized block design and two orthogonal designs. We analyzed the growth responses(including plant height, root length, shoot-leaf biomass and root biomass) of A. ordosica seedlings to different soil CaCO_3 contents. The results revealed that, with a greater soil CaCO_3 content, A. ordosica seedlings gradually grew more slowly, with their relative growth rates of plant height, root length, shoot-leaf biomass and root biomass all decreasing significantly. Root N/P ratios showed significant negative correlations with the relative growth rates of plant height, shoot-leaf biomass and root length of A. ordosica seedlings; however, the relative growth rate of root length increased significantly with the root P concentration increased, showing a positive correlation. These results demonstrate that soil CaCO_3 reduces the local P availability in soil, which produces a non-adaptive phenotypic plasticity to A. ordosica seedlings. In addition, arterial blood gas analysis showed arterial blood acidosis, reduced pH values, increased alkali reserve negative values, decreased peripheral blood 5-hydroxytryptamine content, and increased 5-hydroxytryptamine expression in cerebellar Purkinje cells. Following lidocaine injection to block the habenular nucleus, abnormalities in breath, genioglossal electromyogram, and blood gas values disappeared, and peripheral blood 5-hydroxytryptamine content returned to levels prior to electric stimulation. | 15 | Crosstalk between IL-15Rα^(+)tumor-associated macrophages and breast cancer cells reduces CD8+T cell recruitment显示文摘Background:Interleukin-15(IL-15)is a promising immunotherapeutic agent owing to its powerful immune-activating effects.However,the clinical benefits of these treatments are limited.Crosstalk between tumor cells and immune cells plays an important role in immune escape and immunotherapy drug resistance.Herein,this study aimed to obtain in-depth understanding of crosstalk in the tumor microenvironment for providing potential therapeutic strategies to prevent tumor progression.Methods:T-cell killing assays and co-culture models were developed to determine the role of crosstalk between macrophages and tumor cells in breast cancer resistant to IL-15.Western blotting,histological analysis,CRISPR-Cas9 knockout,multi-parameter flow cytometry,and tumor cell-macrophage co-injection mouse models were developed to examine the mechanism by which IL-15Rα^(+)tumor-associated macrophages(TAMs)regulate breast cancer cell resistance to IL-15.Results:We found thatmacrophages contributed to the resistance of tumor cells to IL-15,and tumor cells induced macrophages to express high levels of theαsubunit of the IL-15 receptor(IL-15Rα).Further investigation showed that IL-15Rα^(+)TAMs reduced the protein levels of chemokine CX3C chemokine ligand 1(CX3CL1)in tumor cells to inhibit the recruitment of CD8^(+)T cells by releasing the IL-15/IL-15Rαcomplex(IL-15Rc).Administration of an IL-15Rc blocking peptide markedly suppressed breast tumor growth and overcame the resistance of cancer cells to anti-programmed cell death protein 1(PD-1)antibody immunotherapy.Interestingly,Granulocyte-macrophage colony-stimulating factor(GMCSF)inducedγchain(γc)expression to promote tumor cell-macrophage crosstalk,which facilitated tumor resistance to IL-15.Additionally,we observed that the non-transcriptional regulatory function of hypoxia inducible factor-1alpha(HIF-1α)was essential for IL-15Rc to regulateCX3CL1 expression in tumor cells.Conclusions:The IL-15Rc-HIF-1α-CX3CL1 signaling pathway serves as a crosstalk between macrophages and tumor cells in the tumormicroenvironment of breast cancer.Targeting this pathway may provide a potential therapeutic strategy for enhancing the efficacy of cancer immunotherapy. | Wenlong Zhang Qing Zhang Nanfei Yang Qian Shi Huifang Su Tingsheng Lin Zhonglei He Wenxin Wang Hongqian Guo Pingping Shen | 2022 | Cancer Communications2022,42,6: | 0 |

| 17 | Effect of Deuterium Ion Implantation on Microstructures of Fe-M (M=V, W, Ta) Alloys显示文摘To study the effect of tungsten, vanadium and tantalum on the microstructures in CLAM (China Low Activation Martensitic) steel after irradiation respectively, the microstructures of Fe-M (M= V ,W, Ta) model alloys were investigated after implanted deuterium ions using an ion accelerator at 773 K. After implanted deuterium ion, TEM (Transmission Electron Microscope) observation and EDX (Energy Dispersive X-ray Spectrom) analysis have been carried out. The result showed that tiny voids were observed in all model alloys after implanted the same dose of deuterium ions. The swelling rate in FeTa alloy was the smallest among the three alloys. Unlike FeW and FeV alloys, there was the segregation in FeTa alloy under a fluence of 5×1017 D + /cm2 at 773 K. A theoretical analysis showed that the void growth in FeTa alloy slowed down due to tantalum segregation near voids. | 20 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment.
