12017版WHO甲状腺肿瘤分类解读显示文摘2017版WHO内分泌肿瘤分类根据肿瘤的病理、临床及基因学特点,对内分泌肿瘤的分类进行了全面的更新。新版WHO中甲状腺肿瘤分类总体可分为上皮性肿瘤、非上皮性肿瘤和继发肿瘤三大类(表1),其中最重要的进展在于甲状腺滤泡上皮细胞起源的高分化肿瘤。乳头和滤泡、良性和恶性分类的更新,并新增加了一组甲状腺交界性肿瘤。嗜酸细胞肿瘤从滤泡性肿瘤中剔除,成为一组独立的病变。低分化癌诊断标准进一步明确。刘志艳 周庚寅 Kennichi Kakudo Alfred K-Y Lam 2018中华病理学杂志2018,47,4:39
2Endothelin-1 stimulates contraction and migration of rat pancreatic stellate cells显示文摘AIM: To examine the ability of ET-1 to affect the cell functions of PSCs and the underlying molecular mechanisms.METHODS: PSCs were isolated from the pancreas of male Wistar rats after perfusion with collagenase, and cells between passages two and five were used. Expression of ET-1 and ET receptors was assessed by reverse transcription-PCR and immunostaining. Phosphorylation of myosin regulatory light chain (MLC), extracellular-signal regulated kinase (ERK), and Akt was examined by Western blotting. Contraction of PSCs was assessed on hydrated collagen lattices. Cell migration was examined using modified Boyden chambers. Cell proliferation was assessed by measuring the incorporation of 5-bromo-2'-deoxyuridine.RESULTS: Culture-activated PSCs expressed ETA and ETB receptors, and ET-1. ET-1 induced phosphorylation of MLC and ERK, but not Akt. ET-1 induced contraction and migration,but did not alter proliferation of PSCs. ET-1-induced contraction was inhibited by an ETA receptor antagonist BQ-123 and an ETB receptor antagonist BQ-788, whereas migration was inhibited by BQ-788 but not by BQ-123. A Rho kinase inhibitor Y-27632 abolished both contraction and migration.CONCLUSION: ET-1 induced contraction and migration of PSCs through ET receptors and activation of Rho-Rho kinase. 3Granular cell tumor of the pancreas:A case report and review of literature显示文摘Granular cell tumors,also called Abrikossof's tumors,were originally described by Abrikossof A in 1926.The f irst case of a pancreatic granular cell tumor was described in 1975 and only 6 cases have been reported.We describe a case of granular cell tumor in the pancreas showing pancreatic duct obstruction.Because imaging studies showed f indings compatible with those of pancreatic carcinoma,the patient underwent distal pancreatectomy.Histological examination showed that the tumor consisted of a nested growth of large tumor cells with ample granular cytoplasm and small round nuclei.The tumor cells expressed S-100 protein and were stained with neuron-specific enolase and periodic acid-Schiff,but were negative for desmin,vimentin,and cytokeratin.The resected tumor was diagnosed as a granular cell tumor.To our knowledge,this is the seventh case of Granular cell tumor of the pancreas to be reported.Atsushi Kanno Kennichi Satoh Morihisa Hirota Shin Hamada Jun Umino Hiromichi Itoh Atsushi Masamune Shinichi Egawa Fuyuhiko Motoi Michiaki Unno Kazuyuki Ishida Tooru Shimosegawa 2010World Journal of Gastrointestinal Oncology2010,2,2:5
4MSX2 overexpression inhibits gemcitabine-induced caspase-3 activity in pancreatic cancer cells显示文摘AIM: To evaluate the effect of MSX2 on gemcitabineinduced caspase-3 activation in pancreatic cancer cell line Panc-1.METHODS: Using V5-tagged MSX2 expression vector,stable transfectant of MSX2 was generated from Panc-1cells (Px14 cells). Cell viability under gemcitabine administration was determined by MTT assay relative to control cell line (empty-vector transfected Panc-1 cells;P-3EV cells). Hoechst staining was used for the detection of apoptotic cell. Activation of caspase-3 was assessed using Western blotting analysis and direct measurement of caspase-3 specific activities.RESULTS: MSX2 overexpression in Panc-1 cells resulted in decreased gemcitabine-induced caspase-3 activation and increased cell viability under gemcitabine treatment in Px14 cells.CONCLUSION: MSX2 exerts repressive effects on gemcitabine-induced apoptotic pathway. 8Effectiveness of therapeutic barium enema for diverticular hemorrhage显示文摘AIM:To evaluate the effectiveness of barium impaction therapy for patients with colonic diverticular bleeding.METHODS:We reviewed the clinical charts of patients in whom therapeutic barium enema was performed for the control of diverticular bleeding between August2010 and March 2012 at Yokohama Rosai Hospital.Twenty patients were included in the review,consisting of 14 men and 6 women.The median age of the patients was 73.5 years.The duration of the followup period ranged from 1 to 19 mo(median:9.8 mo).Among the 20 patients were 11 patients who required the procedure for re-bleeding during hospitalization,6patients who required it for re-bleeding that developed after the patient left the hospital,and 3 patients who required the procedure for the prevention of rebleeding.Barium(concentration:150 w%/v%)was administered per the rectum,and the leading edge of the contrast medium was followed up to the cecum by fluoroscopy.After confirmation that the ascending colon and cecum were filled with barium,the enema tube was withdrawn,and the patient’s position was changed every 20 min for 3 h.RESULTS:Twelve patients remained free of rebleeding during the follow-up period(range:1-19mo)after the therapeutic barium enema,including 9men and 3 women with a median age of 72.0 years.Re-bleeding occurred in 8 patients including 5 men and 3 women with a median age of 68.5 years:4developed early re-bleeding,defined as re-bleeding that occurs within one week after the procedure,and the remaining 4 developed late re-bleeding.The DFI(disease-free interval)decreased 0.4 for 12 mo.Only one patient developed a complication from therapeutic barium enema(colonic perforation).CONCLUSION:Therapeutic barium enema is effective for the control of diverticular hemorrhage in cases where the active bleeding site cannot be identified by colonoscopy.Mizue Matsuura Masahiko Inamori Atsushi Nakajima Yasuhiko Komiya Yumi Inoh Keigo Kawasima Mai Naitoh Yuji Fujita Akiko Eduka Noriyoshi Kanazawa Shiori Uchiyama Rie Tani Kennichi Kawana Setsuya Ohtani Hajime Nagase 2015World Journal of Gastroenterology2015,21,18:2
