1High-throughput screening of mouse gene knockouts identifies established and novel skeletal phenotypes显示文摘Screening gene function in vivo is a powerful approach to discover novel drug targets. We present high-throughput screening(HTS) data for 3 762 distinct global gene knockout(KO) mouse lines with viable adult homozygous mice generated using either gene-trap or homologous recombination technologies. Bone mass was determined from DEXA scans of male and female mice at 14 weeks of age and by microCT analyses of bones from male mice at 16 weeks of age. Wild-type(WT) cagemates/littermates were examined for each gene KO. Lethality was observed in an additional 850 KO lines. Since primary HTS are susceptible to false positive findings, additional cohorts of mice from KO lines with intriguing HTS bone data were examined. Aging,ovariectomy, histomorphometry and bone strength studies were performed and possible non-skeletal phenotypes were explored. 3Modelling the regenerative niche:a major challenge in biomaterials research显示文摘By definition,biomaterials are developed for clinical application.In the field of regenerative medicine their principal function is to play a significant,and,if possible,an instructive role in tissue healing.In the last analysis the latter involves targeting the‘regenerative niche’.The present paper will address the problem of simulating this niche in the laboratory and adopts a life science approach involving the harnessing of heterotypic cellular communication to achieve this,that is,the ability of cells of different types to mutually influence cellular functions.Thus,co-culture systems using human cells are the methodological focus and will concern four exemplary fields of regeneration,namely,bone,soft tissue,lower respiratory tract and airway regeneration.The working hypothesis underlying this approach is that in vitro models of higher complexity will be more clinically relevant than simple monolayer cultures of transformed cell lines in testing innovative strategies with biomaterials for regeneration.C.James Kirkpatrick 2015Regenerative Biomaterials2015,2,4:3
4Towards the Tipping Point for FAIR Implementation显示文摘This article explores the global implementation of the FAIR Guiding Principles for scientific management and data stewardship,which provide that data should be findable,accessible,interoperable and reusable.The implementation of these principles is designed to lead to the stewardship of data as FAIR digital objects and the establishment of the Internet of FAIR Data and Services(IFDS).If implementation reaches a tipping point,IFDS has the potential to revolutionize how data is managed by making machine and human readable data discoverable for reuse.Accordingly,this article examines the expansion of the implementation of FAIR Guiding Principles,especially how and in which geographies(locations)and areas(topic domains)implementation is taking place.A literature review of academic articles published between 2016 and 2019 on the use of FAIR Guiding Principles is presented.The investigation also includes an analysis of the domains in the IFDS Implementation Networks(INs).Its uptake has been mainly in the Western hemisphere.The investigation found that implementation of FAIR Guiding Principles has taken firm hold in the domain of bio and natural sciences.To achieve a tipping point for FAIR implementation,it is now time to ensure the inclusion of non-European ascendants and of other scientific domains.Apart from equal opportunity and genuine global partnership issues,a permanent European bias poses challenges with regard to the representativeness and validity of data and could limit the potential of IFDS to reach across continental boundaries.The article concludes that,despite efforts to be inclusive,acceptance of the FAIR Guiding Principles and IFDS in different scientific communities is limited and there is a need to act now to prevent dampening of the momentum in the development and implementation of the IFDS.It is further concluded that policy entrepreneurs and the GO FAIR INs may contribute to making the FAIR Guiding Principles more flexible in including different research epistemologies,especially through its GO CHANGE pillar.Mirjam van Reisen Mia Stokmans Mariam Basajja Antony Otieno Ong’ayo Christine Kirkpatrick Barend Mons 2020Data Intelligence2020,2,1:3
5Analysis of the in vitro degradation and the in vivo tissue response to bi-layered 3D-printed scaffolds combining PLA and biphasic PLA/bioglass components--Guidance of the inflammatory response as basis for osteochondral regeneration显示文摘The aim of the present study was the in vitro and in vivo analysis of a bi-layered 3D-printed scaffold combining a PLA layer and a biphasic PLA/bioglass G5 layer for regeneration of osteochondral defects in vivo Focus of the in vitro analysis was on the(molecular)weight loss and the morphological and mechanical variations after immersion in SBF.The in vivo study focused on analysis of the tissue reactions and differences in the implant bed vascularization using an established subcutaneous implantation model in CD-1 mice and established histological and histomorphometrical methods.Both scaffold parts kept their structural integrity,while changes in morphology were observed,especially for the PLA/G5 scaffold.Mechanical properties decreased with progressive degradation,while the PLA/G5 scaffolds presented higher compressive modulus than PLA scaffolds.The tissue reaction to PLA included low numbers of BMGCs and minimal vascularization of its implant beds,while the addition of G5 lead to higher numbers of BMGCs and a higher implant bed vascularization.Analysis revealed that the use of a bi-layered scaffold shows the ability to observe distinct in vivo response despite the physical proximity of PLA and PLA/G5 layers.Altogether,the results showed that the addition of G5 enables to reduce scaffold weight loss and to increase mechanical strength.Furthermore,the addition of G5 lead to a higher vascularization of the implant bed required as basis for bone tissue regeneration mediated by higher numbers of BMGCs,while within the PLA parts a significantly lower vascularization was found optimally for chondral regeneration.Thus,this data show that the analyzed bi-layered scaffold may serve as an ideal basis for the regeneration of osteochondral tissue defects.Additionally,the results show that it might be able to reduce the number of experimental animals required as it may be possible to analyze the tissue response to more than one implant in one experimental animal.Mike Barbeck Tiziano Serra Patrick Booms Sanja Stojanovic Stevo Najman Elisabeth Engel Robert Sader Charles James Kirkpatrick Melba Navarro Shahram Ghanaati 2017Bioactive Materials2017,2,4:3

10FAIR Practices in Africa显示文摘This article investigates expansion of the Internet of FAIR Data and Services(IFDS)to Africa,through the three GO FAIR pillars:GO CHANGE,GO BUILD and GO TRAIN.Introduction of the IFDS in Africa has a focus on digital health.Two examples of introducing FAIR are compared:a regional initiative for digital health by governments in the East Africa Community(EAC)and an initiative by a local health provider(Solidarmed)in collaboration with Great Zimbabwe University in Zimbabwe.The obstacles to introducing FAIR are identified as underrepresentation of data from Africa in IFDS at this moment,the lack of explicit recognition of situational context of research in FAIR at present and the lack of acceptability of FAIR as a foreign and European invention which affects acceptance.It is envisaged that FAIR has an important contribution to solve fragmentation in digital health in Africa,and that any obstacles concerning African participation,context relevance and acceptance of IFDS need to be removed.This will require involvement of African researchers and ICT-developers so that it is driven by local ownership.Assessment of ecological validity in FAIR principles would ensure that the context specificity of research is reflected in the FAIR principles.This will help enhance the acceptance of the FAIR Guidelines in Africa and will help strengthen digital health research and services.Mirjam van Reisen Mia Stokmans Munyaradzi Mawere Mariam Basajja Antony Otieno Ong’ayo Primrose Nakazibwe Christine Kirkpatrick Kudakwashe Chindoza 2020Data Intelligence2020,2,1:2
