1Complete chloroplast genome sequence of Magnolia grandiflora and comparative analysis with related species显示文摘Magnolia grandiflora is an important medicinal,ornamental and horticultural plant species.The chloroplast(cp) genome of M.grandiflora was sequenced using a 454 sequencing platform and the genome structure was compared with other related species.The complete cp genome of M.grandiflora was 159623 bp in length and contained a pair of inverted repeats(IR) of 26563 bp separated by large and small single copy(LSC,SSC) regions of 87757 and 18740 bp,respectively.A total of 129 genes were successfully annotated,18 of which included introns.The identity,number and GC content of M.grandiflora cp genes were similar to those of other Magnoliaceae species genomes.Analysis revealed 218 simple sequence repeat(SSR) loci,most composed of A or T,contributing to a bias in base composition.The types and abundances of repeat units in Magnoliaceae species were relatively conserved and these loci will be useful for developing M.grandiflora cp genome vectors.In addition,results indicated that the cp genome size in Magnoliaceae species and the position of the IR border were closely related to the length of the ycf1 gene.Phylogenetic analyses based on 66 shared genes from 30 species using maximum parsimony(MP) and maximum likelihood(ML) methods provided strong support for the phylogenetic position of Magnolia.The availability of the complete cp genome sequence of M.grandiflora provides valuable information for breeding of desirable varieties,cp genetic engineering,developing useful molecular markers and phylogenetic analyses in Magnoliaceae.LI XiWen GAO HuanHuan WANG YiTao SONG JingYuan HENRY Robert WU HeZhen HU ZhiGang YAO Hui LUO HongMei LUO Kun PAN HongLin CHEN ShiLin 2013Science China(Life Sciences)2013,56,2:17
2High precision in-situ Pb isotopic analysis of sulfide minerals by femtosecond laser ablation multi-collector inductively coupled plasma mass spectrometry显示文摘An in-situ microanalysis of Pb isotopic compositions in sulfide minerals is carried out by using femtosecond laser-ablation multi-collector inductively coupled plasma mass spectrometry(fs LA-MC-ICP-MS).High-temperature-activated carbon was used to filter Hg contained in the carrier gas,which reduced the Hg background signal by 48%and also lowered the detection limit of the analysis.Fractionation and mass discrimination effects existing in the ICP-MS analytical processes were corrected using an internal reference Tl in conjunction with an external reference NIST SRM 610.The proposed method was used to analyze the Pb isotopic compositions of chalcopyrite,pyrite,and sphalerite from the Dulong Sn-Zn-In polymetallic ore district.The results showed that in this ore district,the sulfide minerals and different grains of the same sulfide mineral show a large variation in Pb content up to 1000-fold.The studied pyrites show relatively higher Pb contents and homogeneous Pb isotopic compositions,whereas the sphalerites have low Pb contents but most variable Pb isotopic compositions.It is suggested that the large variation of Pb isotopic composition may reflect a late hydrothermal superimposition on the primary sulfide formation.In addition,radiogenic Pb accumulated by radioactive decay of trace amounts of U over time in the host minerals may also be one of the causes for the large variation range of Pb content and Pb isotopic composition of those low-Pb sphalerites.Chalcopyrite and sphalerite grains with Pb content greater than 10 ppm presented a consistent Pb isotopic distribution,whereas all the sulfide grains with Pb content greater than 100 ppm had consistent Pb isotopic composition within 2s measurement uncertainties.The in-situ analysis of Pb isotopic composition agreed well with the results obtained by conventional chemical methods within2s measurement uncertainties,indicating that the data obtained by fs LA-MC-ICP-MS are reliable.Additionally,this study indicates that the Pb isotopic composition could truthfully record the source of ore-forming minerals only for sulfide minerals with high Pb content.On the contrary,the Pb isotopic composition of low-Pb sulfide minerals may be affected by trace amounts of U in the host minerals that may lead to a highly radiogenic Pb isotope ratio.Alternatively,it is also possible that late fluid metasomatic overprinting may alter the Pb isotopic compositions.YUAN HongLin YIN Cong LIU Xu CHEN KaiYun BAO ZhiAn ZONG ChunLei DAI MenNing LAI ShaoCong WANG Rong JIANG ShaoYong 2015Science China Earth Sciences2015,58,10:14

4Metal Sealing Performance of Subsea X-tree Wellhead Connector Sealer显示文摘The metal sealing performance of subsea X-tree wellhead connectors is crucial for the safety and reliability of subsea X-trees. In order to establish the theoretical relation between metal sealing ring's contact stress and its structural parameters and working pressure, a mechanical analysis method for double-cone sealing of high pressure vessels is applied in analyzing the metal sealing ring under the condition of preload and operation. As a result, the formula of the unit sealing load for the metal sealing ring under operation with residual preload is shown in this paper, which ensures that the metal sealing ring has an excellent sealing effect and can prevent the metal sealing ring from yielding. Besides, while analyzing the sealing process of the metal sealing ring, the change rule of contact stress and working pressure is concluded here, putting forward that the structural parameters of the metal sealing ring are the major factors affecting the change rule. Finally, the analytical solution through theoretical analysis is compared with the simulation result through finite element analysis in a force feedback experiment, and both are consistent with each other, which fully verifies for the design and calculation theory on metal sealing ring's contact stress and its structural parameters and working pressure deduced in this paper. 6Construction of an integrated map and location of a bruchid resistance gene in mung bean显示文摘Bruchid beetle(Callosobruchus chinensis) poses a serious threat to the production and storage of mung bean(Vigna radiata). Mapping bruchid resistance(Br) will provide an important basis for cloning the responsible gene(s) and elucidating its functional mechanism, and will also facilitate marker-assisted selection in mung bean breeding. Here, we report the construction of the genetic linkage groups of mung bean and mapping of the Br1 locus using an RIL population derived from a cross between Berken, a bruchid-susceptible line, and ACC41, a bruchid-resistant line. A total of 560 markers were mapped onto 11 linkage groups,with 38.0% of the markers showing distorted segregation. The lengths of the linkage groups ranged from 45.2 to 117.0 c M with a total coverage of 732.9 c M and an average interval of1.3 c M between loci. Br1 was located on LG9 between BM202(0.7 c M) and Vr2-627(1.7 c M).Based on 270 shared SSR markers, most of the linkage groups were assigned to specific chromosomes. 7Physical picture of the optical memory effect显示文摘The optical memory effect is an interesting phenomenon that has attracted considerable attention in recent decades. Here, we present a new physical picture of the optical memory effect, in which the memory effect and the conventional spatial shift invariance are united. Based on this picture we depict the role of thickness, scattering times, and anisotropy factor and derive equations to calculate the ranges of the angular memory effect(AME) of different scattering components(ballistic light, singly scattered, doubly scattered, etc.), and hence a more accurate equation for the real AME ranges of volumetric turbid media. A conventional random phase mask model is modified according to the new picture. 8Mutation profile and its correlation with clinicopathology in Chinese hepatocellular carcinoma patients显示文摘Background:Hepatocellular carcinoma(HCC)is one of the most common causes of cancer worldwide.Although many studies have focused on oncogene characteristics,the genomic landscape of Chinese HCC patients has not been fully clarified.Methods:A total of 165 HCC patients,including 146 males and 19 females,were enrolled.The median age was 55 years(range,27-78 years).Corresponding clinical and pathological information was collected for further analysis.A total of 168 tumor tissues from these patients were selected for next-generation sequencing(NGS)-based 450 panel gene sequencing.Genomic alterations including single nucleotide variations(SNV),short and long insertions and deletions(InDels),copy number variations,and gene rearrangements were analyzed.Tumor mutational burden(TMB)was measured by an algorithm developed in-house.The top quartile of HCC was classified as TMB high.Results:A total of 1,004 genomic alterations were detected from 258 genes in 168 HCC tissues.TMB values were identified in 160 HCC specimens,with a median TMB of 5.4 Muts/Mb(range,0-28.4 Muts/Mb)and a 75%TMB of 7.7 Muts/Mb.The most commonly mutated genes were TP53,TERT,CTNNB1,AXIN1,RB1,TSC2,CCND1,ARID1A,and FGF19.SNV was the most common mutation type and C:G>T:A and guanine transformation were the most common SNVs.Compared to wild-type patients,the proportion of Edmondson grade III-IV and microvascular invasion was significantly higher in TP53 mutated patients(P<0.05).The proportion of tumors invading the hepatic capsule was significantly higher in TERT mutated patients(P<0.05).The proportion of Edmondson grade I-II,alpha fetoprotein(AFP)<25μmg/L,and those without a history of hepatitis B was significantly higher in CTNNB1 mutated patients(P<0.05).CTNNB1 mutations were associated with TMB high in HCC patients(P<0.05).Based on correlation analysis,the mutation of TP53 was independently correlated with microvascular invasion(P=0.002,OR=3.096)and Edmondson grade III-IV(P=0.008,OR=2.613).The mutation of TERT was independently correlated with tumor invasion of the liver capsule(P=0.001,OR=3.030),and the mutation of CTNNB1 was independently correlated with AFP(<25μmg/L)(P=0.009,OR=3.414).Conclusions:The most frequently mutated genes of HCC patients in China were TP53,TERT,and CTNNB1,which mainly lead to the occurrence and development of HCC by regulating the P53 pathway,Wnt pathway,and telomere repair pathway.There were more patients with microvascular invasion and Edmondson III-IV grade in TP53 mutated patients and more patients with hepatic capsule invasion in TERT mutated patients,while in CTNNB1 mutated patients,there were more patients with Edmondson I-II grade,AFP<25μmg/L,and a non-hepatitis B background.Also,the TMB values were significantly higher in CTNNB1 mutated patients than in wild type patients.Shuo Wang Huasheng Shi Tao Liu Manjiang Li Sanshun Zhou Xuan Qiu Zusen Wang Weiyu Hu Weidong Guo Xiaoqian Chen Honglin Guo Xiaoliang Shi Junping Shi Yunjin Zang Jingyu Cao Liqun Wu 2021Hepatobiliary Surgery and Nutrition2021,10,2:8

11A genome-wide landscape of mRNAs,lncRNAs,and circRNAs during subcutaneous adipogenesis in pigs显示文摘Background: Preadipocyte differentiation plays a critical role in subcutaneous fat deposition in pigs.However,the roles of different RNAs,such as messenger RNAs(mRNAs),long non-coding RNAs(lncRNAs),and circular RNAs(circRNAs) in the differentiation process of subcutaneous preadipocytes,are still largely unclear.In the present study,a transcriptome analysis,including the analysis of mRNAs,lncRNAs,and circRNAs,during different differentiation stages,namely,day 0(D0),day 2(D2),day 4(D4),and day 8(D8),of subcutaneous preadipocytes from Chinese Erhualian pigs was performed.Results: A total of 1554,470,1344,1777,and 676 differentially expressed(DE) mRNAs,112,58,95,136,and 93 DE lncRNAs,and 902,787,710,932,and 850 DE circRNAs were identified between D2 and D0,between D4 and D2,between D8 and D4,between D4 and D0,and between D8 and D0,respectively.Furthermore,functional enrichment analysis showed that the common DE mRNAs during the entire differentiation process were mainly involved in lipid metabolic and cell differentiation processes.Additionally,co-expression network analysis identified the potential lncRNAs related to adipogenesis,e.g.,MSTRG.131380 and MSTRG.62128.Conclusions: Our study provides new insights of the expression changes of RNAs during adipogenic differentiation,which might contribute to the phenotype of subcutaneous adipogenesis.These results greatly improve our understanding of the molecular mechanisms regulating subcutaneous fat deposition in pigs.Xin Liu Kaiqing Liu Baosen Shan Shengjuan Wei Dongfeng Li Haiyin Han Wei Wei Jie Chen Honglin Liu Lifan Zhang 2019Journal of Animal Science and Biotechnology2019,10,1:5
12A Fast Separation Method for Isotope Analysis Based on Compressed Nitrogen Gas and Ion-Exchange Chromatography Technique--A Case Study of Sr-Nd Isotope Measurement显示文摘High-purity N_2 was used to increase the mobile phase flow rate during ion purification of ion-exchange resin. This was performed to improve the efficiency of isotope separation and purification, and to meet the efficiency requirements of rapid multiple-collector-inductively coupled plasma mass spectrometry(MC-ICPMS) analysis. For Cu isotope separation, our results indicated that at a gas flow rate >60 m L/min, the separation chromatographic peaks broadened and the recovery rate decreased to <99.2%. On the other hand, no significant change in the Cu peaks was observed at a gas flow rate of 20 m L/min and the recovery rate was determined to be >99.9%. The Cu isotope ratio, measured by the standard-sample bracketing method, agreed with reference data within a ±2 SD error range. The separation time was reduced from the traditional 10 h(without N_2) to 4 h(with N_2), indicating that the efficiency was more than doubled. Moreover, Sr and Nd isotope separation in AGV-2(US Geological Survey andesite standard sample) accelerated with a 20 m L/min gas flow, demonstrating that with the passage of N_2, the purified liquid comprised Rb/Sr and Sm/Nd ratios of <0.000 049 and <0.000 001 5, respectively. This indicated an effective separation of Rb from Sr and Sm from Nd. MC-ICPMS could therefore be applied to accurately determine Sr and Nd isotope ratios. 13Simultaneous Measurement of Major, Trace Elements and Pb Isotopes in Silicate Glasses by Laser Ablation Quadrupole and Multi-Collector Inductively Coupled Plasma Mass Spectrometry显示文摘A method was developed for the rapid in situ analysis of major and trace elements and Pb isotopes in silicate glass samples that combines laser ablation quadrupole and multi-collector inductively coupled plasma mass spectrometry(LA-Q-ICP-MS/MC-ICP-MS). Major, trace elements, and Pb isotope ratio compositions were clearly affected by laser conditions. Using a laser spot size of 160 μm, a laser ablation frequency of 15 Hz, an energy density 18 J/cm^2, and a 1 : 9 ratio of laser ablation aerosol to the corresponding makeup gas, we obtained accurate major and trace element contents and Pb isotope ratios. Using Ca as the internal standard element, and GSE-1G and NIST 610 as the external standards for calibration, element contents generally matched the preferred values within 15%. Higher relative errors for some elements(e.g., Cr, Ga, Ge) may have been caused by lower than recommended values in some standards. The exponential law correction method for Tl normalization, with optimum adjusted Tl ratio, was utilized to obtain Pb isotopic data with good precision and accuracy. Pb isotopic ratios of the glass reference materials were in good agreement with the reference or published values to within 2 s measurement uncertainties, and the analytical precision was better than 0.17%(e.g., ^(208)Pb/^(206)Pb). 14Effects of freeze–thaw cycles on soil N_2O concentration and flux in the permafrost regions of the Qinghai–Tibetan Plateau显示文摘Nitrous oxide(N_2 O) is one of the most important greenhouse gases in the atmosphere; freeze–thaw cycles(FTCs) might strongly influence the emission of soil N_2 O on the Qinghai–Tibetan Plateau(QTP). However, there is a lack of in situ research on the characteristics of soil N_2 O concentration and flux in response to variations in soil properties caused by FTCs.Here, we report the effect of FTC-induced changes in soil properties on the soil N_2 O concentration and flux in the permafrost region of the higher reaches of the Shule River Basin on the northeastern margin of the QTP. We measured chemical properties of the topsoil, activities of soil microorganisms, and air temperature(AT), as well as soil N_2 O concentration and flux, over an annual cycle from July 31, 2011, to July 30, 2012. The results showed that soil N_2 O concentration was significantly affected by soil temperature(ST), soil moisture(SM), soil salinity(SS), soil polyphenol oxidase(SPO), soil alkaline phosphatase(SAP), and soil culturable actinomycetes(SCA), ranked as SM>SS>ST>SPO>SAP>SCA, whereas ST significantly increased soil N_2 O flux, compared with SS. 16Internal structures and high-velocity frictional properties of Longmenshan fault zone at Shenxigou activated during the 2008 Wenchuan earthquake显示文摘This paper reports internal structures of a wide fault zone at Shenxigou,Dujiangyan,Sichuan province,China,and high-velocity frictional properties of the fault gouge collected near the coseismic slip zone during the 2008 Wenchuan earthquake.Vertical offset and horizontal displacement at the trench site were 2.8 m(NW side up)and 4.8 m(right-lateral),respectively.The fault zone formed in Triassic sandstone,siltstone,and shale about 500 m away from the Yingxiu-Beichuan fault,a major fault in the Longmenshan fault system.A trench survey across the coseismic fault,and observations of outcrops and drill cores down to a depth of 57 m revealed that the fault zone consists of fault gouge and fault breccia of about0.5 and 250–300 m in widths,respectively,and that the fault strikes N62°E and dips 68° to NW.Quaternary conglomerates were recovered beneath the fault in the drilling,so that the fault moved at least 55 m along the coseismic slip zone,experiencing about 18 events of similar sizes.The fault core is composed of grayish gouge(GG) and blackish gouge(BG) with very complex slip-zone structures.BG contains low-crystalline graphite of about 30 %.High-velocity friction experiments were conducted at normal stresses of 0.6–2.1 MPa and slip rates of 0.1–2.1 m/s.Both GG and BG exhibit dramatic slip weakening at constant high slip rates that can be described as an exponential decay from peak friction coefficient lpto steadystate friction coefficient lssover a slip-weakening distance Dc.Deformation of GG and BG is characterized by overlapped slip-zone structures and development of sharp slickenside surfaces,respectively.Comparison of our data with those reported for other outcrops indicates that the high-velocity frictional properties of the Longmenshan fault zones are quite uniform and the high-velocity weakening must have promoted dynamic rupture propagation during the Wenchuan earthquake.Yu Wang Shengli Ma Toshihiko Shimamoto Lu Yao Jianye Chen Xiaosong Yang Honglin He Jiaxiang Dang Linfeng Hou Tetsuhiro Togo 2014Earthquake Science2014,27,5:3

18Genetic diversity assessment of a set of introduced mung bean accessions(Vigna radiata L.)显示文摘Genetic resources from other countries or regions play an important role in broadening the genetic background of local breeding varieties. Here we describe observations on the adaptability of mung bean germplasm obtained from the United States Department of Agriculture and their genetic diversity assessment using SSR markers. Several accessions were shown to be mixtures,based on their phenotypes for some characters. Most accessions were able to complete their lifecycles when grown in Beijing, China, making them ideal for crossbreeding without day length control. High diversity was revealed by the SSR markers, with an average of 4.2 alleles per locus and a PIC value of 0.650 per locus. STRUCTURE analysis divided the accessions into six groups.There was no obvious trend of accessions forming groups according to their geographical origin,owing mainly to germplasm exchange and an uneven distribution of accessions. 19Determination of Hf–Sr–Nd isotopic ratios by MC-ICP-MS using rapid acid digestion after flux-free fusion in geological materials显示文摘In this study, we established a rapid acid digestion for determining Hf–Sr–Nd isotopic ratios of geological samples by using MC-ICP-MS. Conditions of 1600 °C for 1 min and 1400 °C for 1 min were adopted for fusing intrusive rocks and extrusive rocks, respectively. The rapid acid digestion technique is superior in digestion time compared with high-pressure PTFE bomb method. The procedural blanks of the method were also lower than that flux fusion. Replicate analyses of international certified reference materials(CRMs) indicate that isotopic ratios of ^(176)Hf/^(177)Hf,^(87)Sr/^(86)Sr and^(143)Nd/^(144)Nd agree well with previously published data.
