1Biological character of human adipose-derived adult stem cells and influence of donor age on cell replication in culture显示文摘To investigate the biological character of human adipose-derived adult stem cells (hADAS cells) when cultured in vitro and the relationship between hADAS cell’s replication activity and the donor’s age factor, and to assess the stem cells as a new source for tissue engineering. hADAS cells are isolated from human adipose tissue of different age groups (from adolescents to olds: <20 years old, 21―40 years old, 41―60 years old and >61 years old groups). The protein markers (CD29, CD34, CD44, CD45, CD49d, HLA-DR, CD106) of hADAS cells were detected by flow cytometry (FCM) to identify the stem cell, and the cell cycle was examined for P20 hADAS cells to evaluate the safety of the subculture in vitro. The generative activity of hADAS cells in different age groups was also examined by MTT method. The formula “ log2T D = t logN t ? logN 0” was used to get the time doubling (TD) of the cells. The results showed that the cells kept heredity stabilization by chromosome analysis for at least 20 passages. The TD of these cells increased progressively by ageing, and the TD of the <20 years old group was lower than that of the >61 years old group (statistical analysis of variance (ANOVA), P=0.002, P<0.05). 2Effects of vegetation on flow conveyance and sediment transport capacity显示文摘In-stream and riparian vegetation may significantly affect flow and sediment transport in vegetated channels. A hydraulic model has been developed in this paper to compute the flow discharge in channels with rigid and flexible vegetation under emergent and submerged conditions. An empirical formula has also been presented to determine the bed-load discharge in vegetated channels. The model has been tested against experimental and field data available in the literature. 3Development of the straw biochar returning concept in China显示文摘Biochar produced from straw has been shown to improve soil physicochemical properties.This review introduces the fundamental concepts,the broad applications,and underlying theory of straw biochar returning.Current developments in biochar industry and the production practices prevalent among enterprises in China are critiques.This review analyzes current knowledge gaps,challenges,and opportunities in the industrial application of straw biochar returning.Biochar standards,the quantitative and qualitative analysis methods for biochar,and high-value-added products that are based on biochar are critically examined with goal of providing recommendations for future studies.We propose production and modification of biochar that is application oriented to enhance its fitness for purpose as well as long-term and large-space-scale field study to better understand its impact on soil properties and ecotoxicology.Finally,we make prospects for the future development of SBR,including constructing a standard system about straw biochar returning and promoting self-discipline of biochar industry and the establishment of a biochar-based agricultural production model.Jun Meng Tianyi He Edmond Sanganyado Yu Lan Weiming Zhang Xiaori Han Wenfu Chen 2019Biochar2019,1,2:9
4Monoclonal Antibody-Based Enzyme Linked Immunosorbent Assay for the Analysis of Jasmonates in Plants显示文摘Methyl jasmonate (MeJA) and its free-acid form, jasmonic acid (JA) are naturally occurring plant growth regulators widely distributed in higher plants. In order to improve the sensitivity for the analysis of MeJA at low levels in small amounts of plant samples, a monoclonal antibody (MAb) (designated as MAb 3E5D7C4B6) against MeJA was derived from a JA-bovine serum albumin (BSA) conjugate as an immunogen. The antibody belongs to the IgG1 subclass with a κ type light chain and has a dissociation constant of approximately 6.07 × 10?9 M. MAb3E5D7C4B6 is very specific to MeJA. It was used to develop a direct competitive enzyme-linked immunosorbent assay (dcELISA), conventional and simplified indirect competitive ELISAs (icELISA). JA was derivatized into MeJA for the ELISA analysis. The IC50 value and detection range for MeJA were, respectively, 34 and 4-257 ng/mL by the conventional icELISA, 21 and 3-226 ng/mL by the simplified icELISA and 5.0 and 0.7-97.0 ng/mL by the dcELISA. The dcELISA was more sensitive than either the conventional or simplified icELISA. The assays were used to measure the content of jasmonates as MeJA in tobacco leaves under drought stress or inoculated with tobacco mosaic virus and tomato leaves inoculated with tomato mosaic virus or Lirioinyza sativae Blanchard as compared with the corresponding healthy leaves. 5Whole-genome sequencing provides insights into the genetic diversity and domestication of bitter gourd(Momordica spp.)显示文摘Bitter gourd(Momordica charantia)is a popular cultivated vegetable in Asian and African countries.To reveal the characteristics of the genomic structure,evolutionary trajectory,and genetic basis underlying the domestication of bitter gourd,we performed whole-genome sequencing of the cultivar Dali-11 and the wild small-fruited line TR and resequencing of 187 bitter gourd germplasms from 16 countries.The major gene clusters(Bi clusters)for the biosynthesis of cucurbitane triterpenoids,which confer a bitter taste,are highly conserved in cucumber,melon,and watermelon.Comparative analysis among cucurbit genomes revealed that the Bi cluster involved in cucurbitane triterpenoid biosynthesis is absent in bitter gourd.Phylogenetic analysis revealed that the TR group,including 21 bitter gourd germplasms,may belong to a new species or subspecies independent from M.charantia.Furthermore,we found that the remaining 166 M.charantia germplasms are geographically differentiated,and we identified 710,412,and 290 candidate domestication genes in the South Asia,Southeast Asia,and China populations,respectively.This study provides new insights into bitter gourd genetic diversity and domestication and will facilitate the future genomics-enabled improvement of bitter gourd.Junjie Cui Yan Yang Shaobo Luo Le Wang Rukui Huang Qingfang Wen Xiaoxia Han Nansheng Miao Jiaowen Cheng Ziji Liu Changyuan Zhang Chengcheng Feng Haisheng Zhu Jianwen Su Xinjian Wan Fang Hu Yu Niu Xiaoming Zheng Yulan Yang Dai Shan Zhensheng Dong Weiming He Narinder P.S.Dhillon Kailin Hu 2020Horticulture Research2020,7,1:7

7Qiguiyishen decoction reduced the accumulation of extracellular matrix in the kidneys of rats with adriamycin-induced nephropathy显示文摘OBJECTIVE: To investigate the effect of Qiguiyishen decoction(QGYS) on the severity of nephropathy.METHODS: Twenty-four rats were randomly divided into four groups(Ⅰ, Ⅱ, Ⅲ, Ⅳ) according to the random number table. Group Ⅰ as control group did not establish nephropathy model. Groups Ⅱ,Ⅲ, and Ⅳ were intravenously administered Adriamycin(7.5 mg/kg) through the tail vein to establish nephropathy model. QGYS was prepared with the extracts of Huangqi(Radix Astragali Mongolici),Danggui(Radix Angelicae Sinensis),Niuxi(RadixAchyran-this Bidentatae), and Chuanxiong(Rhizoma Chuanxiong).Group Ⅳ was administered QGYS (2mL·kg-1·d-1), group Ⅲ was administered benazepril(10 mL·kg-1·d-1), and group Ⅰ, Ⅱ was administered water(2mL·kg-1·d-1)once daily for eight weeks.RESULTS: QGYS reduced the excretion of urinary protein and N-acetyl-β-D-glucosaminidase and alleviated the accumulation of extracellular matrix(ECM) in renal tissue. 8Research on mode localization of reticulated shell structures显示文摘Reticulated shell structures (RSSs) are characterized as cyclically periodic structures. Mistuning of RSSs will induce structural mode localization. Mode localization has the following two features: some modal vectors of the structure change remarkably when the values of its physical parameters (mass or stiffness) have a slight change; and the vibration of some modes is mainly restricted in some local areas of the structure. In this paper, two quantitative assessment indexes are introduced that correspond to these two features. The first feature is studied through a numerical example of a RSS, and its induced causes are analyzed by using the perturbation theory. The analysis showed that internally, mode localization is closely related to structural frequencies and externally, slight changes of the physical parameters of the structure cause instability to the RSS. A scaled model experiment to examine mode localization was carried out on a Kiewit single-layer spherical RSS, and both features of mode localization are studied. Eight tests that measured the changes of the physical parameters were carried out in the experiment. Since many modes make their contribution in structural dynamic response, six strong vibration modes were tested at random in the experimental analysis. The change and localization of the six modes are analyzed for each test. 9Uniform single atomic Cu_(1)-C_(4)sites anchored in graphdiyne for hydroxylation of benzene to phenol显示文摘For single-atom catalysts(SACs),the catalyst supports are not only anchors for single atoms,but also modulators for geometric and electronic structures,which determine their catalytic performance.Selecting an appropriate support to prepare SACs with uniform coordination environments is critical for achieving optimal performance and clarifying the relationship between the structure and the property of SACs.Approaching such a goal is still a significant challenge.Taking advantage of the strong d-πinteraction between Cu atoms and diacetylenic in a graphdiyne(GDY)support,we present an efficient and simple strategy for fabricating Cu single atoms anchored on GDY(Cu_(1)/GDY)with uniform Cu_(1)-C_(4)single sites under mild conditions.The Cu atomic structure was confirmed by combining synchrotron radiation X-ray absorption spectroscopy,X-ray photoele ctron spectroscopy and density functional theory(DFT)calculations.The as-prepared Cu1/GDY exhibits much higher activity than state-of-the-art SACs in direct benzene oxidation to phenol with H_(2)O_(2)reaction,with turnover frequency values of 251 h^(-1)at room temperature and 1889 h^(-1)at 60℃,respectively.Furthermore,even with a high benzene conversion of86%,high phenol sele ctivity(96%)is maintaine d,which can be ascribed to the hydrophobic and oleophyllic surface nature of Cu1/GDY for benzene adsorption and phenol desorption.Both experiments and DFT calculations indicate that Cu_(1)-C_(4)single sites are more effective at activating H_(2)O_(2)to form Cu=O bonds,which are important active intermediates for benzene oxidation to phenol.Jia Yu Changyan Cao Hongqiang Jin Weiming Chen Qikai Shen Peipei Li Lirong Zheng Feng He Weiguo Song Yuliang Li 2022National Science Review2022,9,9:3
10Hundred-Joule-level,nanosecond-pulse Nd:glass laser system with high spatiotemporal beam quality显示文摘A 100-J-level Nd:glass laser system in nanosecond-scale pulse width has been constructed to perform as a standard source of high-fluence-laser science experiments. The laser system, operating with typical pulse durations of 3–5 ns and beam diameter 60 mm, employs a sequence of successive rod amplifiers to achieve 100-J-level energy at 1053 nm at3 ns. The frequency conversion can provide energy of 50-J level at 351 nm. 12HPLC and GC methods development for the analysis of key intermediate for synthesis of dicamba显示文摘The microreactor based hydroxylation process of 1,2,4-trichlorobezene for producing 2,5-dichlorphenol,the key intermediate of dicamba,is energy efficient and cost effective.But the 2,5-dicholorphneol is present in a mixed state after production.The reaction mixture contained the main by-product 2,4-dichlorophneol,low-content by-product 3,4-dichlorophneol,and other impurities.The difficulty in separation and analysis limits the application of this process widely.The current work aimed at establishing effective analysis methods by gas chromatography(GC)and high performance liquid chromatography(HPLC).The GC method was not able to separate 2,5-dichlorophenol and 2,4-dichlorophenol completely,but the developed HPLC method worked efficiently.The linear correlation coefficients of 2,5-dichlorophenol and 2,4-dichlorophenol were both higher than 0.999,and the average recovery was 100.33% for 2,5-dichlorophenol and 100.13% for 2,4-dichlorophenol,respectively.The relative standard deviations from precision tests were both less than 1%.The contents of 2,5-dichlorophenol and 2,4-dichlorophenol were determined with external standard method.The HPLC method has the advantages of simple operation,good separation efficiency,high accuracy and precision,and was successfully applied for both qualitative and quantitative analysis of 2,5-dichlorophenol and 2,4-dichlorophenol of the sample solution.Zhenming Zhang He Zhao Xiaohui Wang Weiming Ni Fengsheng Gao Jianrong Wang Minjin Liu Yongli Li 2021Chinese Journal of Chemical Engineering2021,34,5:2
