1Primary duct closure versus T-tube drainage after laparoscopic common bile duct exploration:a meta-analysis显示文摘Background and aims:Laparoscopic common bile duct exploration(LCBDE)is considered a safe and effective method for the removal of bile duct stones.However,the choice of primary duct closure(PDC)or T-tube drainage(TTD)technique after LCBDE is still controversial.This study aimed to compare the safety and effectiveness of PDC and TTD after LCBDE.Methods:Studies published before May 1,2021 in Pub Med,Web of Science,and Cochrane Library databases were searched to screen out randomized controlled trials(RCTs)and cohort studies to compare PDC with TTD.Meta-analyses of fixed effect and random effect models were performed using Rev Man 5.3.Results:A total of 1865 patients were enrolled in six RCTs and ten cohort studies.Regarding RCTs,the PDC group was significantly better than the TTD group in terms of operation time,total postoperative complications,postoperative hospital stay,and hospitalization expenses(all P<0.05).Based on cohort studies of the subgroup,the PDC group had shorter operation time,shorter postoperative hospital stay,less intraoperative blood loss,and limited total postoperative complications.Statistically,there were no significant differences in bile leakage,retained stones,stone recurrence,bile duct stricture,postoperative pancreatitis,other complications,or postoperative exhaust time between the TTD and PDC groups.Conclusions:Based on the available evidence,compared with TTD,PDC is safe and effective,and can be used as the first choice after transductal LCBDE in patients with choledocholithiasis.Taifeng ZHU Haoming LIN Jian SUN Chao LIU Rui ZHANG 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,12:15
2Mechanisms involved in antineuralgic effects of Paeonia Lactiflora: prediction based on network pharmacology显示文摘目的:白芍的镇痛作用已被广泛应用于中医临床,但对其潜在机制知之甚少.本研究旨在从网络药理学的角度阐明其有效成分和镇痛机制.方法:通过中药系统药理学分析平台(TCMSP)筛选出其可能的活性成分及CAS.从Pubchem 中获得其SMILES,并通过在Swiss Target Prediction 数据库中反向对接获得潜在的靶点.从GeenCards 数据库中提取疼痛相关分子,采用韦恩图筛选出芍药治疗疼痛的预测靶点.机制分析采用String 构建蛋白-蛋白相互作用,运用DAVID 平台进行靶点GO 功能富集和KEGG 信号通路富集.结果:通过GO 和KEGG 分析我们发现疼痛相关的信号通路主要涉及血清素能突触、钙信号通路、调控TRP 通道的炎症介质.利用基于网络的系统生物学和分子对接分析,我们预测了白芍中的11 种活性成分和97 个潜在的治疗靶标.PRKCA、CASP3、ALOX15、SLC6A4、PRKCG、ALOX5、PRKCB、ALOX12、 EGFR、ADRB2、RYR3、RYR1、NOS2、PTAFR、PRKCQ、PRKCD 等基因参与了白芍的镇痛作用.结论:白芍可通过调节TRP 通道炎症介质、钙信号通路和5-羟色胺受体减轻疼痛.PRKCA、PRKCB、PRKCD、 PRKCQ 和PRKCG 可能是疼痛治疗的新靶标.Di Zhang Shengsuo Ma Jianxin Sun Bing Yang Haoming Lin Meijing Xie Meina Huang Guoping Zhao 2019TMR Clinical Research2019,2,2:8
3PCDH17 increases the sensitivity of colorectal cancer to 5-fluorouracil treatment by inducing apoptosis and autophagic cell death显示文摘5-Fluorouracil(5-FU)is known as a first-line chemotherapeutic agent against colorectal cancer(CRC),but drug resistance occurs frequently and significantly limits its clinical success.Our previous study showed that the protocadherin 17(PCDH17)gene was frequently methylated and functioned as a tumor suppressor in CRC.However,the relationship between PCDH17 and 5-FU resistance in CRC remains unclear.Here,we revealed that PCDH17 was more highly expressed in 5-FU-sensitive CRC tissues than in 5-FU-resistant CRC tissues,and high expression of PCDH17 was correlated with high BECN1 expression.Moreover,this expression profile contributed to superior prognosis and increased survival in CRC patients.Restoring PCDH17 expression augmented the 5-FU sensitivity of CRC in vitro and in vivo by promoting apoptosis and autophagic cell death.Furthermore,autophagy played a dominant role in PCDH17-induced cell death,as an autophagy inhibitor blocked cell death to a greater extent than the pancaspase inhibitor Z-VAD-FMK.PCDH17 inhibition by siRNA decreased the autophagy response and 5-FU sensitivity.Mechanistically,we showed that c-Jun NH2-terminal kinase(JNK)activation was a key determinant in PCDH17-induced autophagy.The compound SP600125,an inhibitor of JNK,suppressed autophagy and 5-FU-induced cell death in PCDH17-reexpressing CRC cells.Taken together,our findings suggest for the first time that PCDH17 increases the sensitivity of CRC to 5-FU treatment by inducing apoptosis and JNK-dependent autophagic cell death.PCDH17 may be a potential prognostic marker for predicting 5-FU sensitivity in CRC patients.Shuiping Liu Haoming Lin Da Wang Qiang Li Hong Luo Guoxiong Li Xiaohui Chen Yongqiang Li Peng Chen Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Mingming Zhang Xuemeng Han Qiujie Li Liuxi Chen Ying Liu Xiaying Chen Guohua Li Yu Xiang Ting Duan Jiao Feng Jianshu Lou Xingxing Huang Qin Zhang Ting Pan Lili Yan Ting Jin Wenzheng Zhang Lvjia Zhuo Yitian Sun Tian Xie Xinbing Sui 2019Signal Transduction and Targeted Therapy2019,4,1:4

15Ambient Temperature and Years of Life Lost:A National Study in China显示文摘Although numerous studies have investigated premature deaths attributable to temperature,effects of temperature on years of life lost(YLL)remain unclear.We estimated the relationship between temperatures and YLL,and quantified the YLL per death caused by temperature in China.We collected daily meteorological and mortality data,and calculated the daily YLL values for 364 locations(2013–2017 in Yunnan,Guangdong,Hunan,Zhejiang,and Jilin provinces,and 2006–2011 in other locations)in China.A time-series design with a distributed lag nonlinear model was first employed to estimate the location-specific associations between temperature and YLL rates(YLL/100,000 population),and a multivariate meta-analysis model was used to pool location-specific associations.Then,YLL per death caused by temperatures was calculated.The temperature and YLL rates consistently showed U-shaped associations.A mean of 1.02(95%confidence interval:0.67,1.37)YLL per death was attributable to temperature.Cold temperature caused 0.98 YLL per death with most from moderate cold(0.84).The mean YLL per death was higher in those with cardiovascular diseases(1.14),males(1.15),younger age categories(1.31 in people aged 65–74 years),and in central China(1.34)than in those with respiratory diseases(0.47),females(0.87),older people(0.85 in people R75 years old),and northern China(0.64)or southern China(1.19).The mortality burden was modified by annual temperature and temperature variability,relative humidity,latitude,longitude,altitude,education attainment,and central heating use.Temperatures caused substantial YLL per death in China,which was modified by demographic and regional characteristics.Tao Liu Chunliang Zhou Haoming Zhang Biao Huang Yanjun Xu Lifeng Lin Lijun Wang Ruying Hu Zhulin Hou Yize Xiao Junhua Li Xiaojun Xu Donghui Jin Mingfang Qin Qinglong Zhao Weiwei Gong Peng Yin Yiqing Xu Jianxiong Hu Jianpeng Xiao Weilin Zeng Xing Li Siqi Chen Lingchuan Guo Zuhua Rong Yonghui Zhang Cunrui Huang Yaodong Du Yuming Guo Shannon Rutherford Min Yu Maigeng Zhou Wenjun Ma 2021The Innovation2021,2,1:1
16Assessment of gully erosion susceptibility using different DEM-derived topographic factors in the black soil region of Northeast China显示文摘As a primary sediment source,gully erosion leads to severe land degradation and poses a threat to food and ecological security.Therefore,identification of susceptible areas is critical to the prevention and control of gully erosion.This study aimed to identify areas prone to gully erosion using four machine learning methods with derived topographic attributes.Eight topographic attributes(elevation,slope aspect,slope degree,catchment area,plan curvature,profile curvature,stream power index,and topo-graphic wetness index)were derived as feature variables controlling gully occurrence from digital elevation models with four different pixel sizes(5.0 m,12.5 m,20.0 m,and 30.0 m).A gully inventory map of a small agricultural catchment in Heilongjiang,China,was prepared through a combination of field surveys and satellite imagery.Each topographic attribute dataset was randomly divided into two portions of 70%and 30%for calibrating and validating four machine learning methods,namely random forest(RF),support vector machines(SVM),artificial neural network(ANN),and generalized linear models(GLM).Accuracy(ACC),area under the receiver operating characteristic curve(AUC),root mean square error(RMSE),and mean absolute error(MAE)were calculated to assess the performance of the four machine learning methods in predicting spatial distribution of gully erosion susceptibility(GES).The results suggested that the selected topographic attributes were capable of predicting GES in the study catchment area.A pixel size of 20.0 m was optimal for all four machine learning methods.The RF method described the spatial relationship between the feature variables and gully occurrence with the greatest accuracy,as it returned the highest values of ACC(0.917)and AUC(0.905)at a 20.0 m resolution.The RF was also the least sensitive to resolutions,followed by SVM(ACC=0.781-0.891,AUC=0.724-0.861)and ANN(ACC=0.744-0.808,AUC=0.649-0.847).GLM performed poorly in this study(ACC=0.693-0.757,AUC=0.608-0.703).Based on the spatial distribution of GES determined using the optimal method(RF+pixel size of 20.0 m),16%of the study area has very high level susceptibility classes,whereas areas with high,moderate,and low levels of susceptibility make up approximately 24%,30%,and 31%of the study area,respectively.Our results demonstrate that GES assessment with machine learning methods can successfuly identify areas prone to gully erosion,providing reference information for future soil conservation plans and land management.In addition,pixel size(resolution)is the key consideration when preparing suitable datasets of feature variables for GES assessment.Donghao Huang Lin Su Lili Zhou Yulu Tian Haoming Fan 2023International Soil and Water Conservation Research2023,11,1:0
17Fucosyltransferase 9 promotes neuronal differentiation and functional recovery after spinal cord injury by suppressing the activation of Notch signaling显示文摘Individuals with spinal cord injury(SCI)suffer from permanent disabilities such as severe motor,sensory and autonomic dysfunction.Neural stem cell transplantation has proven to be a potential strategy to promote regeneration of the spinal cord,since NSCs can produce neurotrophic growth factors and differentiate into mature neurons to reconstruct the injured site.However,it is necessary to optimize the differentiation of NSCs before transplantation to achieve a better regenerative outcome.Inhibition of Notch signaling leads to a transition from NSCs to neurons,while the underlying mechanism remains inadequately understood.Our results demonstrate that overexpression of fucosyltransferase 9(Fut9),which is upregulated by Wnt4,promotes neuronal differentiation by suppressing the activation of Notch signaling through disruption of furin-like enzyme activity during S1 cleavage.In an in vivo study,Fut9-modified NSCs efficiently differentiates into neurons to promote functional and histological recovery after SCI.Our research provides insight into the mechanisms of Notch signaling and a potential treatment strategy for SCI.Jiewen Chen Xiaolin Zeng Wenwu Zhang Gang Li Haoming Zhong Chengzhong Xu Xiang Li Tao Lin 2023Acta Biochimica et Biophysica Sinica2023,55,10:0
