2SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. 3Celastrol targets adenylyl cyclase-associated protein 1 to reduce macrophages-mediated inflammation and ameliorates high fat diet-induced metabolic syndrome in mice显示文摘Metabolic syndrome is a clustering of metabolic disorder with unclear molecular mechanism.Increasing studies have found that the pathogenesis and progression of metabolic syndrome are closely related to inflammation.Here,we report celastrol,a traditional Chinese medicine,can improve high fat diet-induced metabolic syndrome through suppressing resistin-induced inflammation.Mechanistically,celastrol binds to adenylyl cyclase associated protein 1(CAP1)and inhibits the interaction between CAP1 and resistin,which restrains the cyclic adenylate monophosphate(cAMP)-protein kinase A(PKA)-nuclear factor kappa-B(NF-kB)signaling pathway and ameliorates high fat diet-induced murine metabolic syndrome.Knockdown of CAP1 in macrophages abrogated the resistin-mediated inflammatory activity.In contrast,overexpression of CAP1 in macrophages aggravated inflammation.Taken together,our study identifies celastrol,which directly targets CAP1 in macrophages,might be a promising drug candidate for the treatment of inflammatory metabolic diseases,such as metabolic syndrome.Yuyu Zhu Ning Wan Xinni Shan Guoliang Deng Qiang Xu Hui Ye Yang Sun 2021Acta Pharmaceutica Sinica B2021,11,5:9
4Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis.However,little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins,including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types,and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels.The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies.Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry,providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations.Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu 2022Cell Research2022,32,1:8
5Study on variational aerosol fields over Beijing and its adjoining areas derived from Terra-MODIS and ground sunphotometer observation显示文摘This paper presents a comprehensive observa-tion technique on derived aerosols data from mobile sun-photometer graph and Terra-Moderate Resolution Imaging Spectroradiometer (MODIS) observation. Research results suggest that after being treated by the variational technique with respect to sunphotometer observations, the Terra- MODIS remote sensing aerosol data are remarkably im-proved, thus for the first time revealing features of the influ-ence of aerosols and pollution emissions of Beijing and its adjoining areas (Hebei, Shandong, etc. provinces). The re-gional impact features of aerosols are related with the pe-ripheral U-shape topography of Beijing. 6Secondary instability control of compressible flow by suction for a swept wing显示文摘The crossflow instability of a three-dimensional boundary layer is a main factor affecting the transition around the swept-wing.The three-dimensional boundary layer flow affected by the saturated crossflow vortex is very sensitive to the high frequency disturbances,which foreshadows that the swept wing flow transition will happen.The primary instability of the compressible flow over a swept wing is investigated with nonlinear parabolized stability equations (NPSE).The Floquet theory is then applied to the analysis of the influence of localized steady suction on the secondary instability of crossflow vortex.The results show that suction can significantly suppress the growth of the crossflow mode as well as the secondary instability modes.XU GuoLiang XIAO ZhiXiang FU Song 2011Science China(Physics,Mechanics & Astronomy)2011,54,11:6

10Development of an Intelligent Vehicle Experiment System显示文摘Existing vehicle experiment systems tend to focus on the research of vehicle dynamics by conducting performance tests on every system or some parts of the vehicle so as to improve the entire performance of the vehicle.Virtual technology is widely utilized in various vehicle test-beds.These test-beds are mainly used to simulate the driving training,conduct the research on drivers' behaviors,or give virtual demonstrations of the transportation environment.However,the study on the active safety of the running vehicle in the virtual environment is still insufficient.A virtual scene including roads and vehicles is developed by using the software Creator and Vega,and radars and cameras are also simulated in the scene.Based on dSPACE's rapid prototyping simulation and its single board DS1103,a simulation model including vehicle control signals is set up in MATLAB/Simulink,the model is then built into C code,and the system defined file(SDF) is downloaded to the DS1103 board through the experiment debug software ControlDesk and is kept running.Programming is made by mixing Visual C++ 6.0,MATLAB API and Vega API.Control signals are read out by invoking library function MLIB/MTRACE of dSPACE.All the input,output,and system state values are acquired by arithmetic and are dynamically associated with the running status of the virtual vehicle.An intelligent vehicle experiment system is thus developed by virtue of program and integration.The system has not only the demonstration function,such as general driving,cruise control,active avoiding collision,but also the function of virtual experiment.Parameters of the system can be set according to needs,and the virtual test results can be analyzed and studied and used for the comparison with the existing models.The system reflects the running of the intelligent vehicle in the virtual traffic environment,at the same time,the system is a new attempt performed on the intelligent vehicle travel research and provides also a new research method for the development of intelligent vehicles.YANG Xinhong GAO Feng LIU Guoliang WANG Guofu XU Guoyan 2010Chinese Journal of Mechanical Engineering2010,23,6:5
11Analysis of the secondary instability of the incompressible flows over a swept wing显示文摘The crossflow instability of a three-dimensional (3-D) boundary layer is an important factor which affects the transition over a swept-wing.In this report,the primary instability of the incompressible flow over a swept wing is investigated by solving nonlinear parabolized stability equations (NPSE).The Floquet theory is applied to study the dependence of the secondary and high-frequency instabilities on curvature,Reynolds number and angle of swept (AOS).The computational results show that the curvature in the present case has no significant effect on the secondary instabilities.It is generally believed that the secondary instability growth rate increases with the magnitude of the nonlinear mode of crossflow vortex.But,at a certain state,when the Reynolds number is 3.2 million,we find that the secondary instability growth rate becomes smaller even when the magnitude of the nonlinear mode of the crossflow vortex is larger.The effect of the angle of swept at 35,45 and 55 degrees,respectively,is also studied in the framework of the secondary linear stability theory.Larger angles of swept tend to decrease the spanwise spacing of the crossflow vortices,which correspondingly helps the stimulation of 'z' mode.XU GuoLiang XIAO ZhiXiang FU Song 2011Science China(Physics,Mechanics & Astronomy)2011,54,4:5
12Vertical characteristics and source identification of PM_(10) in Tianjin显示文摘Ambient PM 10 (particulate matter with a diameter less than 10μm) concentrations were measured on a 255 meter tower in Tianjin,China.The samples were collected at four vertical levels (10,40,120 and 220 m).Vertical characteristics for PM 10 samples were studied.The results showed that the concentrations of PM 10 and constituent species had a negative correlation with the sampling height.The highest concentrations of PM 10 and species were obtained at the 10 m level,and the lowest concentrations were measured at the 220 m level.For the fractions of species to total mass,SO 4 2- and NO 3- had higher values (fraction) at greater height;while Ca had a higher fraction at lower height.Possible source categories for the PM 10 ambient dataset were identified by the principal component analysis method.The possible source categories included crustal dust,vehicles,cement dust,and incineration as well as secondary sulfate and nitrate sources.Analysis of meteorological factors on PM 10 concentrations indicated that wind speed and inversion may be the main factors contributing to different concentrations of PM 10 at different heights.Zhimei Xiao Jianhui Wu Suqin Han Yufen Zhang Hong Xu Xiaoyong Zhang Guoliang Shi Yinchang Feng 2012Journal of Environmental Sciences2012,24,1:5
13Genome-wide prediction in a hybrid maize population adapted to Northwest China显示文摘Genome-wide prediction is a promising approach to boost selection gain in hybrid breeding.Our main objective was to evaluate the potential and limits of genome-wide prediction to identify superior hybrid combinations adapted to Northwest China.A total of 490 hybrids derived from crosses among 119 inbred lines from the Shaan A and Shaan B heterotic pattern were used for genome-wide prediction of ten agronomic traits.We tested eight different statistical prediction models considering additive(A)effects and in addition evaluated the impact of dominance(D)and epistasis(E)on the prediction ability.Employing five-fold cross validation,we show that the average prediction ability ranged from 0.386 to 0.794 across traits and models.Six parametric methods,i.e.ridge regression,LASSO,Elastic Net,Bayes B,Bayes C and reproducing kernel Hilbert space(RKHS)approach,displayed a very similar prediction ability for each trait and two non-parametric methods(random forest and support vector machine)had a higher prediction performance for the trait rind penetrometer resistance of the third internode above ground(RPR_TIAG).The models of A+D RKHS and A+D+E RKHS were slightly better for predicting traits with a relatively high non-additive variance.Integrating trait-specific markers into the A+D RKHS model improved the prediction ability of grain yield by 3%,from 0.528 to 0.558.Of all 6328 potential hybrids,selection of the top 44 hybrids would lead to a 6%increase in grain yield compared with Zhengdan 958,a commercially successful hybrid variety.In conclusion,our results substantiate the value of genome-wide prediction for hybrid breeding and suggest dozens of promising single crosses for developing high-yielding hybrids for Northwest China.Guoliang Li Yuan Dong Yusheng Zhao Xiaokang Tian Tobias Würschum Jiquan Xue Shaojiang Chen Jochen C.Reif Shutu Xu Wenxin Liu 2020The Crop Journal2020,8,5:4
14Multicenter phaseⅡstudy of apatinib single or combination therapy in HER2-negative breast cancer involving chest wall metastasis显示文摘Objective:Breast cancer(BC)with chest wall metastasis(CWM)usually shows rich neovascularization.This trial explored the clinical effect of apatinib on human epidermal growth factor receptor 2(HER2)-negative advanced BC involving CWM.Methods:This trial involved four centers in China and was conducted from September 2016 to March 2020.Patients received apatinib 500 mg/d[either alone or with endocrine therapy if hormone receptor-positive(HR+)]until disease progression or unacceptable toxicity.Progression-free survival(PFS)was the primary endpoint.Results:We evaluated 26 patients for efficacy.The median PFS(mPFS)and median overall survival(mOS)were4.9[range:2.0-28.5;95%confidence interval(95%CI):2.1-8.3]months and 18(range:3-55;95%CI:12.9-23.1)months,respectively.The objective response rate(ORR)was 42.3%(11/26),and the disease-control rate was76.9%(20/26).In the subgroup analysis,HR+patients compared with HR-negative patients had significantly improved mPFS of 7.0(95%CI:2.2-11.8)months vs.2.3(95%CI:1.2-3.4)months,respectively(P=0.001);and mPFS in patients without or with chest wall radiotherapy was 6.4(95%CI:1.6-19.5)months vs.3.0(95%CI:1.3-4.6)months,respectively(P=0.041).In the multivariate analysis,HR+status was the only independent predictive factor for favorable PFS(P=0.014).Conclusions:Apatinib was highly effective for BC patients with CWM,especially when combined with endocrine therapy.PFS improved significantly in patients with HR+status who did not receive chest wall radiotherapy.However,adverse events were serious and should be carefully monitored from the beginning of apatinib treatment.Huiping Li Cuizhi Geng Hongmei Zhao Hanfang Jiang Guohong Song Jiayang Zhang Yaxin Liu Xinyu Gui Jing Wang Kun Li Zhongsheng Tong Fangyuan Zhao Junlan Yang Guoliang Chen Qianyu Liu Xu Liang 2021Chinese Journal of Cancer Research2021,33,2:4
15Comparison of the oncological,perioperative and functional outcomes of partial nephrectomy versus radical nephrectomy for clinical T1b renal cell carcinoma:A systematic review and metaanalysis of retrospective studies显示文摘Objective:To conduct a meta-analysis assessing the perioperative,functional and oncological outcomes of partial nephrectomy(PN)and radical nephrectomy(RN)for T1b tumours.The primary endpoints were the oncological outcomes.The secondary endpoints were the perioperative and functional outcomes.Methods:A systematic literature review was performed by searching multiple databases through February 2019 to identify eligible comparative studies according to the Preferred Reporting Items for Systematic Review and Meta-analysis statement.Identified reports were assessed according to the Newcastle-Ottawa Scale for nonrandomized controlled trials.Results:Overall,13 retrospective cohort studies were included in the analysis.Patients undergoing PN were younger(weighted mean difference[WMD]3.49 years,95%confidence interval[CI]5.16 to1.82;p<0.0001)and had smaller masses(WMD0.45 cm,95%CI0.59 to0.31;p<0.0001).There were no differences in the oncological outcome,which was demonstrated by progression-free survival(hazard ratio[HR]0.70;pZ0.22),cancerspecific mortality(HR 0.91;pZ0.57)and all-cause mortality(HR 1.01;pZ0.96).The two procedures were similar in estimated blood loss(WMD16.47 mL;pZ0.53)and postoperative complications(risk ratio[RR]1.32;pZ0.10),and PN provided better renal function preservation and was related to a lower likelihood of chronic kidney disease onset(RR 0.38;pZ0.006).Conclusion:PN is an effective treatment for T1b tumours because it offers similar surgical morbidity,equivalent cancer control,and better renal preservation compared to RN.Yucong Zhang Gongwei Long Haojie Shang Beichen Ding Guoliang Sun Wei Ouyang Man Liu Yuan Chen Heng Li Hua Xu Zhangqun Ye 2021Asian Journal of Urology2021,8,1:4

19Numerical study on the thermal performance of packed-bed latent heat thermal energy storage system with biomimetic alveoli structure capsule显示文摘Bionics provides a positive and beneficial impact on the development of various materials and systems, which has been widely used in energy storage, heat transfer enhancement, and solar thermochemical reactions. In this paper, the idea of heat storage unit with biomimetic alveoli structure is proposed and introduced to increase the heat transfer area and improve the thermal performance of the packed-bed latent heat thermal energy storage(LHTES) system. The effects of heat storage unit structure on temperature distribution, liquid fraction, thermal storage capacity, and other performance indexes of the conventional model and biomimetic model are analyzed. The numerical results indicate that the biomimetic alveoli structure can improve the thermal response of heat storage unit, increase the specific surface area, and achieve a faster heat storage process of the packed-bed compared with the conventional structure. Among various existing models, the double-cell model has a significant improvement.Compared with the conventional model, the maximum increase of liquid fraction and charging efficiency are 9.8% and 6.4%,respectively.
