1内皮型一氧化氮合酶脱偶联的研究进展显示文摘血管内皮功能障碍(endothelial dysfunction)是多种心脑血管疾病的共同病理机制,其突出表现为内皮依赖性血管舒张功能障碍,主要由NO减少及氧自由基增加所致。最新研究发现,内皮型一氧化氮合酶脱偶联(eNOS uncoup ling)是导致NO水平下降和氧自由基水平升高的重要机制,是高血压、糖尿病、动脉粥样硬化等疾病中内皮功能障碍的重要原因。通过纠正eNOS脱偶联可有效改善内皮功能,有望为保护血管内皮功能提供有效途径。郑建普 卞卡 可燕 Murad Ferid 2006中国药理学通报2006,22,6:29

4Intrahepatic endometriosis as differential diagnosis: Case report and literature review显示文摘Intrahepatic endometriosis is one of the rarest forms of atypical endometriosis; only eighteen cases have been reported in the English literature. We describe the case of a 32-year-old woman, who presented with persistent, non-cyclical upper right quadrant abdominal pain, a central liver cyst, and no history of endometriosis. Three years previous, she was diagnosed with an intrahepatic cyst. The lesion progressed and two laparoscopic deroofing-operations were performed, yet the diagnosis of intrahepatic endometriosis was never reached. She presented in our clinic with further progress of the cyst as well as obstruction of the intrahepatic biliary system. The magnetic resonance imaging showed a 9.5 cm × 12 cm, lobulated intrahepatic cyst. We performed an ultrasonic pericystectomy. Immunostaining confirmed intrahepatic endometriosis. Only one of the previously described eighteen patients with intrahepatic endometriosis presented with cyclical pain in the upper right abdominal quadrant accompanying menstruation. This lack of a 'typical' clinic makes it challenging to diagnose extragonadal endometriosis without histopathology. A previous history of endometriosis was described in only twelve cases, thus the diagnosis of this condition should not be limited to patients with a known history of endometriosis. Six of 18 patients were postmenopausal, demonstrating this condition is not limited to women of reproductive age. A preoperative diagnosis was only reached in seven of the previously described cases, highlighting the importance of preoperative biopsies. Yet due to the potential adverse effects, a transhepatic biopsy must be discussed individually. Although rare, intrahepatic endometriosis should always be considered as a differential diagnosis in women with recurrent hepatic cysts, regardless of age or previous medical history. 5Gallbladder function and dynamics of bile flow in asymptomatic gallstone disease显示文摘AIM:To investigate the effects of gallbladder stones on motor functions of the gallbladder and the dynamics of bile flow in asymptomatic gallstone disease.METHODS:Quantitative hepatobiliary scintigraphy was performed to detect the parameters of gallbladder motor function [gallbladder ejection fraction(GBEF),gallbladder visualization time(GBVT),gallbladder time to peak activity(GBTmax),gallbladder half emptying time(GBT1/2),and transit time of bile to duodenum(TTBD)] in 24 patients with asymptomatic cholelithiasis who were diagnosed incidentally during routine abdominal ultrasonographic examination and 20 healthy subjects with normal gallbladder.RESULTS:Even though there was no significant difference in the clinical and laboratory parameters between the patient and control groups,all parameters of gallbladder function except TTBD were found to differ significantly between the two groups.GBEF in the patient group was decreased(P=0.000) and GBVT,GBTmax,GBT1/2 in the patient group were longer(P=0.000,P=0.015,P=0.001,respectively).CONCLUSION:Our results showed that even if there were not any clinical and laboratory findings,gallbladder filling and emptying could be impaired in patients with gallstone disease.Sevim Süreyya eri Feride Meltem zbek Celal eri Bahattin Baykal Hasan Erol Eroglu Zeynep Baykal Mustafa Yildiz Semahat Saglam Ahmet Yesildag 2009World Journal of Gastroenterology2009,15,22:8
6Curcumin induces differentiation of embryonic stem cells through possible modulation of nitric oxide-cyclic GMP pathway显示文摘Curcumin,an active ingredient of dietary spice used in curry,has been shown to exhibit anti-oxidant,anti-inflammatory and anti-proliferative properties.Using EB directed differentiation protocol of H-9 human em-bryonic stem(ES)cells;we evaluated the effect of cur-cumin(0-20μmol/L)in enhancing such differentiation.Our results using real time PCR,western blotting and immunostaining demonstrated that curcumin signifi-cantly increased the gene expression and protein levels of cardiac specific transcription factor NKx2.5,cardiac troponin I,myosin heavy chain,and endothelial nitric oxide synthase during ES cell differentiation.Further-more,an NO donor enhanced the curcumin-mediated induction of NKx2.5 and other cardiac specific proteins.Incubation of cells with curcumin led to a dose depend-ent increase in intracellular nitrite to the same extent as giving an authentic NO donor.Functional assay for second messenger(s)cyclic AMP(cAMP)and cyclic GMP(cGMP)revealed that continuous presence of cur-cumin in differentiated cells induced a decrease in the baseline levels of cAMP but it significantly elevated baseline contents of cGMP.Curcumin addition to a cell free assay significantly suppressed cAMP and cGMP degradation in the extracts while long term treatment of intact cells with curcumin increased the rates of cAMP and cGMP degradation suggesting that this might be due to direct suppression of some cyclic nucleo-tide-degrading enzyme(phosphodiesterase)by curcu-min.These studies demonstrate that polyphenol curcu-min may be involved in differentiation of ES cells partly due to manipulation of nitric oxide signaling.Kalpana Mujoo Lubov ENikonoff Vladislav G Sharin Nathan SBryan Alexander YKots Ferid Murad 2012Protein & Cell2012,3,7:5
7Keratinocyte 生长 factor-2 和自体同源的浆液加强处于在老鼠的角膜损坏的间充质的干细胞的再生效果显示文摘AIM:To investigate the healing process after severe corneal epithelial damage in rats treated with mesenchymal stem cells(MSCs)cultured with or without keratinocyte growth factor(KGF-2)and autologous serum(AS)on amniotic membrane(AM).Many patients are blind and devastated by severe ocular surface diseases due to limbal stem cell deficiency.Bone marrow-derived MSCs are potential sources for cellbased tissue engineering to repair or replace the corneal tissue,having the potential to differentiate to epithelial cells.METHODS:The study included 5 groups each including 10 female'Sprague Dawley'rats in addition to20 male rats used as bone marrow donors.Group I rats received AM+MSCs,Group II rats AM+MSCs cultured with KGF-2,Group III rats AM+MSCs cultured with KGF-2+AS,Group IV rats only AM and Group V rats,none.AS was derived from blood drawn from male rats and bone marrow was obtained from the femur and tibia bones of the same animals.Therapeutic effect was evaluated with clinical,histopathological and immunohistochemical assessment.MSC engraftment was demonstrated via detection of donor genotype(Y+)in the recipient tissue(X)with polymerase chain reaction.RESULTS:Corneal healing was significantly better in Groups I-III rats treated with MSC transplantation compared to Group IV and Group V rats with supportive treatment only.The best results were obtained in Group III rats with 90%transparency,70%lack of neovascularization,and 100%epithelium damage limited to less than 1/4 of cornea.CONCLUSION:We suggest that culture of MSCs with KGF-2 and AS on AM is effective in corneal repair in case of irreversible damage to limbal stem cells.Ferda Alpaslan Pinarli Gülsen Okten Umit Beden Tunc Fisgin Mehmet Kefeli Nurten Kara Feride Duru Leman Tomak 2014International Journal of Ophthalmology(English edition)2014,7,2:4
8Co-doping of hydroxyapatite with zinc and fluoride improves mechanical and biological properties of hydroxyapatite显示文摘Hydroxyapatite(HA) co-doped with Zn2+ tand F- ions was synthesized by precipitation method for the first time in this study.FTIR spectroscopy revealed Zn2+ tand F ions incorporation into HA structure.Co-doping of Zn2+ tand F- ions decreased unit cell volume of HA and decreased grain sizes.Zn2+ tor 5 mol% F addition into HA significantly improved its density.Microhardness was increased with Zn2+ taddition and further increase was detected with F co-doping.Zn2+ tand F co-doped samples had higher fracture toughness than pure HA.Zn2+ tincorporation to the structure resulted in an increase in cell proliferation and ALP activity of cells,and further increase was observed with 1 mol%F addition.With superior mechanical properties and biological response 2Zn1 F is a good candidate for biomedical applications.Idil Uysal Feride Severcan Aysen Tezcaner Zafer Evis 2014Progress in Natural Science:Materials International2014,24,4:4
