| 1 | High-affinity mutant Interleukin-13 targeted CAR T cells enhance delivery of clickable biodegradable fluorescent nanoparticles to glioblastoma显示文摘Glioblastoma(GBM),the deadliest form of brain cancer,presents long-standing problems due to its localization.Chimeric antigen receptor(CAR)T cell immunotherapy has emerged as a powerful strategy to treat cancer.IL-13-receptor-α2(IL13Rα2),present in over 75%of GBMs,has been recognized as an attractive candidate for antiglioblastoma therapy.Here,we propose a novel multidisciplinary approach to target brain tumors using a combination of fluorescent,therapeutic nanoparticles and CAR T cells modified with a targeted-quadruplemutant of IL13(TQM-13)shown to have high binding affinity to IL13Rα2-expressing glioblastoma cells with low off-target toxicity.Azide-alkyne cycloaddition conjugation of nanoparticles to the surface of T cells allowed a facile,selective,and high-yielding clicking of the nanoparticles.Nanoparticles clicked onto T cells were retained for at least 8 days showing that the linkage is stable and promising a suitable time window for in vivo delivery.T cells clicked with doxorubicin-loaded nanoparticles showed a higher cytotoxic effect in vitro compared to bare T cells.In vitro and in vivo T cells expressing TQM-13 served as delivery shuttles for nanoparticles and significantly increased the number of nanoparticles reaching brain tumors compared to nanoparticles alone.This work represents a new platform to allow the delivery of therapeutic nanoparticles and T cells to solid tumors. | Gloria B.Kim Virginia Aragon-Sanabria Lauren Randolph Hali Jiang Joshua A.Reynolds Becky S.Webb Achuthamangalam Madhankumar Xiaojun Lian James R.Connor Jian Yang Cheng Dong | 2020 | Bioactive Materials2020,5,3: | 6 |
| 2 | Effects of plant growth regulators, carbon sources and pH values on callus induction in Aquilaria malaccensis leaf explants and characteristics of the resultant calli显示文摘The endangered tropical tree, Aquilaria malaccensis, produces agarwood for use in fragrance and medicines. Efforts are currently underway to produce valuable agarwood compoundsn tissue culture. The purpose of this study was to develop an optimal growth medium, specifically, the best hormone combination for callus suspension culture. Using nursery-grown A. malaccensis, sterilized leaf explants were first incubated on basic Murashige and Skoog(MS) gel medium containing 15g/L sucrose and at pH 5.7. Different auxin types including 1-naphthaleneacetic acid(NAA), 2,4-dichlorophenoxyacetic acid(2,4-D), and indole-3-butyric acid(IBA), were tested at various concentrations(0.55, 1.1 and 1.65 μM) using the basic medium. Leaf explants were incubated for 30 days in the dark. Callus induced by 1.1 μM NAA had the highest biomass dry weight(DW) of 17.3 mg; however the callus was of a compact type. This auxin concentration was then combined with either 6-benzylaminopurine(BAP) or kinetin at 0.55, 1.1, 2.2 or 3.3 μM to induce growth of friable callus. The 1.1μM NAA + 2.2μM BAP combination produced friable callus with the highest biomass(93.3mg DW). When testing the different carbon sources and pHs, sucrose at 15g/L and pH at 5.7 yielded highest biomasses at 87.7mg and 83 mg DW, respectively. Microscopic observations revealed the arrangement of the friable cells as loosely packed with relatively large cells, while for the compact callus, the cells were small and densely packed. | 3 | Protective effects of Rutin against methanol induced acute toxic optic neuropathy:an experimental study显示文摘AIM:To determine the effects of Rutin on methanol induced optic neuropathy and compare the results with the effects of ethanol.METHODS:Totally 30 rats were divided into 5 groups,with 6 rats in each group as follows:healthy controls(C),methotrexate(MTX),methotrexate+methanol(MTM),methotrexate+methanol+ethanol(MTME) and methotrexate+ methanol+Rutin(MTMR).In all rabbits except those of the control group,MTX,diluted in sterile serum physiologic,0.3 mg/kg per oral was applied for 7 d by the aid of a tube.After this procedure to the rats of MTM,MTME and MTMR groups,20% methanol with a dose of 3 g/kg per oral was given by the aid of a tube.In MTME group,4 h after the application of methanol,20% ethanol was applied by the same way with a dose of 0.5 g/kg.On the other hand,in MTMR group 4 h after the application of methanol,Rutin,which was dissolved in distilled water,was applied by the same way with a dose of 50 mg/kg.RESULTS:There were statistically significant differences in tissue 8-hydroxy-2 deoxyguanine(8-OHdG),interleukin-1β(IL-1β),tumor necrosis factor-alpha(TNF-α),malondialdehyde(MDA),myeloperoxidase(MPO).glutathione peroxidase(t GSH) and superoxide dismutase(SOD) levels between groups(P<0.001).In MTMR group tissue 8-OHdG,IL-1β,MDA,and MPO levels were similar with the healthy controls but significantly different than the other groups.In histopathological evaluations,in MTX group there was moderate focal destruction,hemorrhage and decrease innumber of astrocytes and oligodendrocytes;in MTM group there was severe destruction and edema with decrease in number of astrocytes and oligodendrocytes;in MTME group there was mild hemorrhage,mild edema,mildly dilated blood vessels with congestion while in MTMR group,optic nerve tissue was resembling the healthy controls.CONCLUSION:Rutin may prevent methanol-induced optic neuropathy via anti-inflammatory effects and decreasing the oxidative stress.New treatment options are warranted in this disease to avoid loss of vision in patients. | Nurdan Gamze Tasli Ferda Keskin Cimen Yücel Karakurt Turgay Ucak Renad Mammadov Bahadir Suleyman Nezahat Kurt Halis Suleyman | 2018 | International Journal of Ophthalmology(English edition)2018,11,5: | 2 |
