1Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. 2Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy.Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma 2018Cancer Communications2018,38,1:6
3Effects of electroacupuncture at Zusanli (ST 36) on neurons in the colonic myenteric plexus in rats with irritable bowel syndrome with constipation显示文摘We studied the effects of electroacupuncture at Zusanli (ST 36) on neurons in the colonic myenteric plexus and on defecation in rats with irritable bowel syndrome with constipation (IBS-C). We also used intragastric administration of pinaverium bromide as a positive control treatment to reveal the pathway mediating the onset of IBS-C. Both electroacupuncture and pinaverium bromide greatly improved defecation in rats with IBS-C. Immunohistochemical staining of the enteric nervous system neuronal marker protein gene product 9.5 in the colonic myenteric plexus showed that electroacupuncture by itself, or in combination with pinaverium bromide, increased the number of neurons and the staining intensity of protein gene product 9.5 in the colonic myenteric plexus. 4Single-cell transcriptomes of peripheral blood cells indicate and elucidate severity of COVID-19显示文摘The blood and immune system of coronavirus disease 2019(COVID-19)infected patients are dysfunctional,and numerous studies have been conducted to resolve their characteristics and pathogenic mechanisms.Nevertheless,the variations of immune responses along with disease severity have not been comprehensively documented.Here,we profiled the single-cell transcriptomes of 96,313 peripheral blood mononuclear cells(PBMCs)derived from 12 COVID-19 patients(including four moderate,four severe and four critical cases)and three healthy donors.We showed that proliferative CD8 effector T cells with declined immune functions and cytotoxicity accumulated in the critical stage.By contrast,the quantity of natural killer(NK)cells was significantly reduced,while they exhibited enhanced immune activities.Notably,a gradually attenuated responseto COVID-19 along with disease severity was observed in monocytes,in terms of cellular composition,transcriptional discrepancy and transcription factor regulatory network.Furthermore,we identified immune cell-type dependent cytokine signatures distinguishing the severity of COVID-19 patients.In addition,cell interactions between CD8 effector T/NK cells and monocytes mediated by inflammatory cytokines were enhanced in moderate and severe stages,but weakened in critical cases.Collectively,our work uncovers the cellular and molecular players underlying the disordered and heterogeneous immune responses associated with COVID-19 severity,which could provide valuable insights for the treatment of critical COVID-19 patients.Xiaowei Xie Xuelian Cheng Gaoxiang Wang Biao Zhang Mengyao Liu Liting Chen Hui Cheng Sha Hao Jianfeng Zhou Ping Zhu Tao Cheng 2021Science China(Life Sciences)2021,64,10:3

6Global analysis of lysine succinylation in patchouli plant leaves显示文摘Lysine succinylation is a novel,naturally occurring posttranslational modification(PTM)in living organisms.Global lysine succinylation identification has been performed at the proteomic level in various species;however,the study of lysine succinylation in plant species is relatively limited.Patchouli plant(P.cablin(Blanco)Benth.,Lamiaceae)is a globally important industrial plant and medicinal herb.In the present study,lysine succinylome analysis was carried out in patchouli plants to determine the potential regulatory role of lysine succinylation in patchouli growth,development,and physiology.The global succinylation sites and proteins in patchouli plants were screened with an immunoprecipitation affinity enrichment technique and advanced mass spectrometry-based proteomics.Several bioinformatic analyses,such as function classification and enrichment,subcellular location predication,metabolic pathway enrichment and protein−protein interaction networking,were conducted to characterize the functions of the identified sites and proteins.In total,1097 succinylation sites in 493 proteins were detected in patchouli plants,among which 466 succinylation sites in 241 proteins were repeatedly identified within three independent experiments.The functional characterization of these proteins indicated that the tricarboxylic acid(TCA)cycle,oxidative phosphorylation,photosynthesis processes,and amino acid biosynthesis may be regulated by lysine succinylation.In addition,these succinylated proteins showed a wide subcellular location distribution,although the chloroplast and cytoplasm were the top two preferred cellular components.Our study suggested the important role of lysine succinylation in patchouli plant physiology and biology and could serve as a useful reference for succinylation studies in other medicinal plants.Xiaobing Wang Xiuzhen Chen Junren Li Xuanxuan Zhou Yanting Liu Liting Zhong Yun Tang Hai Zheng Jiyun Liu Ruoting Zhan Likai Chen 2019Horticulture Research2019,6,1:2

9Phase-field simulation tending to depict practical electrodeposition process in lithium-based batteries显示文摘Lithium dendrite growth due to uneven electrodeposition usually leads to the potential hazard of internal short circuit and shorter lifetime of lithium-based batteries. Extensive efforts have been devoted to explore the effects of single or two factors on dendrite growth, involving the diffusion coefficient, exchange current density, electrolyte concentration, temperature, and applied voltage. However, these factors interrelate during battery operation, signifying that a understanding of how they jointly influence the electrodeposition is of paramount importance for the effective suppression of dendrites. Here, we incorporate the dependent relationships among key factors into the phase-field model to capture their synergistic effects on electrodeposition. All the simulations are implemented in our self-written MATLAB code under a unified modeling framework. Following this, five groups of experimentally common dendrite patterns are reproduced and the corresponding electrodeposition driving forces are identified. Unexpectedly, we find that with the decrease of the ratio of exchange current density(or applied voltage) to diffusion coefficient, the electrodeposition morphology changes from needle-like dendrites to columnar dendrites and to uniform deposition. 14Effect of Roucongrong(Herba Cistanches Deserticolae) decoction on the substantia nigra through Wnt/β-catenin signaling pathway in rats with Parkinson's disease induced by 6-hydroxydopamine hydrochloride显示文摘OBJECTIVE:To investigate the effect of Roucongrong(Herba Cistanches Deserticolae)decoction on the substantia nigra in rats with Parkinson’s disease(PD)induced by 6-hydroxydopamine hydrochloride(6-OHDA).To further determine whether the Wnt/β-catenin signaling pathway is involved in the action.METHODS:A rat model of PD was established by intracranial injection of 6-OHDA.Subsequently,three concentrations of Roucongrong(Herba Cistanches Deserticolae)decoction were prepared and administered to rats by gavage therapy for 14 d.Behavioral changes were measured in PD rats.In vivo tyrosine hydroxylase(TH)levels in the substantia nigra were examined by immunohistochemistry.Additionally,gene and protein expression levels of members of the Wnt/β-catenin signaling pathway were examined by Western blotting and polymerase chain reaction.Lastly,a Wnt/β-catenin inhibitor was used to investigate the mechanism of action in 1-methyl-4-phenylpyridinium(MPP+)-treated MES23.5 cells in vitro.RESULTS:Roucongrong(Herba Cistanches Deserticolae)decoction improved performance in the stride and gait adjustment tests in PD rats.It also increased TH in the substantia nigra and altered the expression of genes and proteins in the Wnt/β-catenin signaling pathway.Wnt/β-catenin inhibitor reduced the effect of Roucongrong(Herba Cistanches Deserticolae)decoction in MPP+-treated MES23.5 cells.CONCLUSION:Roucongrong(Herba Cistanches Deserticolae)decoction may promote neuronal survival in PD in vivo and in vitro by increasing TH content in the substantia nigra and by activating the Wnt/β-catenin signaling pathway.XU Qian QIN Wei WU Fangzhen LIN Yao HONG Liting CHEN Dan HU Xuefeng CAI Jing 2021Journal of Traditional Chinese Medicine2021,41,5:1
15Shen-Qi-Wan Protects the Renal Peritubular Capillary Injury from Adenine-mediated Damage by Upregulating Aquaporin 1显示文摘Background:Shen-Qi-Wan(SQW),a commonly used prescription against chronic kidney disease(CKD)in Tradi-tional Chinese Medicine(TCM),has a nephroprotective action in adenine-induced kidney injury.However,the mechanism of SQW in renal injury remains unclear.Objective:This study was undertaken to measure the effect of SQW on peritubular capillary injury both in vivo and in vitro assays.Methods:The effect of SQW on the peritubular capillary injury was evaluated according to measuring hypothalamic-pituitary-adrenal(HPA)function,inflammatory cytokines,VEGF(vascular endothelial growth fac-tor),CD34(Cluster of differentiation 34),and AQP1(Aquaporin 1)levels in the kidney,cell migration,and lumen forming capacity.Results:SQW supplementation could ameliorate dysfunction of the HPA and renal function loss induced by adenine.SQW also significantly inhibited the inflammatory cytokines including MCP-1(monocyte chemotactic protein-1)and VCAM-1(vascular cell adhesion molecule-1)level.Alternatively,SQW administration showed an ameliorating effect from the toxicity and alleviated the injury of capillaries around renal tubules instigated by adenine through increasing AQP1 mRNA and protein level.SQW medicated the serum enhanced the migration and lumen formation ability of HMEC-1 cells,and significantly increased AQP1 protein level.Moreover,AQP1 knockdown efficiently inhibited the migration and lumen formation ability in HMEC-1 cells,and weakened the effect of SQW medicated serum.Conclusion:These results suggested that SQW attenuated peritubular capillary injury in adenine-induced CKD model rats through boosting angiogenesis in endothelial cell,and AQP1 may be a potential target of SQW for treating the renal injury.Yuting Bao Yehui Zhang Yuanxiao Yang Xueming Chen Luning Lin Yunbo Fu Liting Ji Changyu Li 2022Clinical Complementary Medicine and Pharmacology2022,2,1:1

17HSCS:a hybrid shared cache scheduling scheme for multiprogrammed workloads显示文摘The traditional dynamic random-access memory (DRAM)storage medium can be integrated on chips via modem emerging 3D-stacking technology to architect a DRAM shared cache in multicore systems.Compared with static random-access memory (SRAM),DRAM is larger but slower.In the existing research,a lot of work has been devoted to improving the workload performance using SRAM and stacked DRAM together in shared cache systems,ranging from SRAM structure improvement to optimizing cache tags and data access.However,little attention has been paid to designing a shared cache scheduling scheme for multiprogrammed workloads with different memory footprints in multicore systems.Motivated by this,we propose a hybrid shared cache scheduling scheme that allows a multicore system to utilize SRAM and 3D-stacked DRAM efficiently, thus achieving better workload performance.This scheduling scheme employs (1)a cache monitor,which is used to collect cache statistics;(2)a cache evaluator,which is used to evaluate the cache information during the process of programs being executed;and (3)a cache switcher,which is used to self-adaptively choose SRAM or DRAM shared cache modules.A cache data migration policy is naturally developed to guarantee that the scheduling scheme works correctly.Extensive experiments are conducted to evaluate the workload performance of our proposed scheme.The experimental resuits showed that our method can improve the multiprogrammed workload performance by up to 25%compared with state-of-the-art methods (including conventional and DRAM cache systems).Jingyu ZHANG Chentao WU Dingyu YANG Yuanyi CHEN Xiaodong MENG Liting XU Minyi GUO 2018Frontiers of Computer Science2018,12,6:1
