1Effects of Activin A on the Activities of the Mouse Peritoneal Macrophages显示文摘Activin A is a kind of pre-inflammatory factor that belongs to the transforming growth factor-β (TGF-β) superfamily. To investigate the effect and mechanism of activin A on the activities of mouse macrophages, the secretion of NO in the supernatant of cultured mouse peritoneal macrophages was examined by NO assay kit, and the expression of iNOS, ActRIIA and ARIP2 mRNA in mouse peritoneal macrophages was analyzed by RT-PCR.The results showed that activin A stimulated the secretion of NO and the expression of iNOS mRNA in non-activated mouse macrophages in a time- and dose-dependent manner. In contrast, activin A in the same concentration inhibited the secretion of NO in LPS-activated mouse macrophages in a dose-dependent manner.ActRIIA was highly expressed on macrophages, and activin A upregulated the ActRIIA mRNA expression in macrophages. 2Computational Identification of Novel Family Members of MicroRNA Genes in Arabidopsis thaliana and Oryza sativa显示文摘MicroRNAs (miRNAs) are a class of endogenous small RNAs that play important regulatory roles in both animals and plants, miRNA genes have been intensively studied in animals, but not in plants. In this study, we adopted a homology search approach to identify homologs of previously validated plant miRNAs in Arabidopsis thaliana and Oryza sativa. We identified 20 potential miRNA genes in Arabidopsis and 40 in O. sativa, providing a relatively complete enumeration of family members for these miRNAs in plants. In addition, a greater number of Arabidopsis miRNAs (MIR168, MIR159 and MIR172) were found to be conserved in rice. With the novel homologs, most of the miRNAs have closely related fellow miRNAs and the number of paralogs varies in the different miRNA families. Moreover, a probable functional segment highly conserved on the elongated stem of pre-miRNA fold-backs of MIR319 and MIR159 family was identified. These results support a model of variegated miRNA regulation in plants, in which miRNAs with different functional elements on their pre-miRNA fold-backs can differ in their function or regulation, and closely related miRNAs can be diverse in their specificity or competence to downregulate target genes. 4Fabrication of hexagonal gallium nitride films on silicon (111) substrates显示文摘Hexagonal gallium nitride films were successfully fabricated through ammoniating Ga2O3 films deposited on silicon (111 ) substrates by electrophoresis. The structure, composition, and surface morphology of the formed films were characterized by X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM),and transmission electron microscopy (TEM). The measurement results reveal that the polycrystalline GaN films with hexagonal wurtzite structure were successfully grown on the silicon (111) substrates. 5Influence of Tip Clearance on Performance of a Contra-Rotating Fan显示文摘Influences of tip clearance on the tip flow and associated loss mechanism in a contra-rotating axial flow fan hasbeen studied in the paper, based on three dimensional numerical results. The results with different tip clearanceare compared in terms of stage efficiency, relative total pressure loss coefficient, flow angle. It is found that theefficiency of the contra-rotating fan changes almost linearly with increment of the tip clearance, however, efficiencyof the rear rotor is observed to decrease more dramatically than that of the forward rotor given same tipclearance variation. The analysis on the flow structure indicates that the tip region flow field is qualitatively similarin both rotors. 7不同产地姜黄重金属含量测定显示文摘目的:测定姜黄药材中重金属的含量,为建立姜黄药材重金属限量标准提供参考依据。方法:采用电感耦合等离子体质谱法测定27批姜黄样品中Cu、As、Cd、Pb的含量。结果:不同产地姜黄药材中重金属的含量存在较大差异,姜黄样品中Cu、As、Cd、Pb元素的含量分别为:2.62~9.34μg/g,0.03~0.60μg/g,0.01~5.39μg/g,0.106~2.04μg/g。4种重金属中仅镉元素超标,超标率为40.7%。结论:姜黄中重金属镉超标情况严重,部分姜黄药材镉元素污染达到中度污染或重度污染。建议建立姜黄重金属限量标准,为姜黄的规范种植与安全评价提供依据。卢莹莹 徐露露 解清 乐志勇 白宗利 曹龙青 关锰 李得运 刘雅琼 叶敏 Ou Yangli 2019世界中医药2019,14,5:3
8Overcoming barriers to the clinical utilization of iPSCs:reprogramming efficiency,safety and quality显示文摘Differentiated cells can be reprogrammed into pluripotent stem cells,known as“induced pluripotent stem cells”(iPSCs),through the overexpression of defined transcription factors.The creation of iPSC lines has opened new avenues for patient-specific cell replacement therapies for regenerative medicine.However,the clinical utilization of iPSCs is largely impeded by two limitations.The first limitation is the low efficiency of iPSCs generation from differentiated cells.The second limitation is that many iPSC lines are not authentically pluripotent,as many cell lines inefficiently differentiate into differentiated cell types when they are tested for their ability to complement embryonic development.Thus,the“quality”of iPSCs must be increased if they are to be differentiated into specialized cell types for cell replacement therapies.Overcoming these two limitations is paramount to facilitate the widespread employment of iPSCs for therapeutic purposes.Here,we summarize recent progress made in strategies enabling the efficient production of high-quality iPSCs,including choice of reprogramming factors,choice of target cell type,and strategies to improve iPSC quality.Suying Cao Kyle Loh Yangli Pei Wei Zhang Jianyong Han 2012Protein & Cell2012,3,11:3
9Spatially defined single-cell transcriptional profiling characterizes diverse chondrocyte subtypes and nucleus pulposus progenitors in human intervertebral discs显示文摘A comprehensive understanding of the cellular heterogeneity and molecular mechanisms underlying the development,homeostasis,and disease of human intervertebral disks(IVDs)remains challenging.Here,the transcriptomic landscape of 108108 IVD cells was mapped using single-cell RNA sequencing of three main compartments from young and adult healthy IVDs,including the nucleus pulposus(NP),annulus fibrosus,and cartilage endplate(CEP).The chondrocyte subclusters were classified based on their potential regulatory,homeostatic,and effector functions in extracellular matrix(ECM)homeostasis.Notably,in the NP,a PROCR+resident progenitor population showed enriched colony-forming unit-fibroblast(CFU-F)activity and trilineage differentiation capacity.Finally,intercellular crosstalk based on signaling network analysis uncovered that the PDGF and TGF-βcascades are important cues in the NP microenvironment.In conclusion,a single-cell transcriptomic atlas that resolves spatially regulated cellular heterogeneity together with the critical signaling that underlies homeostasis will help to establish new therapeutic strategies for IVD degeneration in the clinic.Yibo Gan Jian He Jun Zhu Zhengyang Xu Zhong Wang Jing Yan Ou Hu Zhijie Bai Lin Chen Yangli Xie Min Jin Shuo Huang Bing Liu Peng Liu 2021Bone Research2021,9,3:3
10Enhanced strength and ductility in Ti46Al4Nb1Mo alloys via boron addition显示文摘To improve the strength and ductility of Ti Al alloys by second phase, Ti46 Al4 Nb1 Mo alloys doped with different B content(0.4%, 0.8%, 1.2%, 1.6% and 2.0%, atomic percent, hereafter in at.%, referred to as TNMx B) were prepared. Macro/microstructure evolution, mechanical properties and deformation mechanisms of the alloys were studied systematically. Results showed that the microstructure of TNM-0.4 B and TNM-0.8 B alloy remained columnar dendrites, and the secondary dendritic arms of columnar grains were more obvious. When the content of B is 1.2%, the columnar dendrites transformed to equiaxed grains, and theα2/γ lamellae colony size was further refined in TNM-1.6 B and TNM-2.0 B alloy. The morphologies and kinds of the borides were changed with increasing B content, XRD results showed that Ti B phase appeared with 1.6%B content, and both Ti B and Ti B2 phase formed in TNM-2.0 B alloy. There were straight and curved Ti B phases located around grain boundaries in TNM-0.4 B and TNM-0.8 B alloy, and when the content of B increased to 1.2%, the curved Ti B phases were reduced, while the tiny and straight Ti B phases increased. With further increasing B content to 1.6% and 2.0%, the tiny and straight Ti B phases were coarser. Compressive testing results showed that the mechanical properties of the TNM alloy were enhanced with increasing B content. The maximum strength and strain of TNM alloy were 2339 MPa and33.7% with 1.6% B addition. 11DNA damage of germ cell of rat induced by nitrotoluene chemicals显示文摘dinitrotoluene(2,4 DNT), 2,6 dinitrotoluene(2,6 DNT) and 4 nitrotoluene(4 NT) are typical pollutants in the Songhua River of Northeast China. Sertoli/germ cell cocultures and single cell gel electrophoresis(SCGE) are applied to investigate whether they have genotoxicity on DNA damage of germ cell of Kunming male rat. The results showed that all three nitrotoluene compounds tested could induce DNA single strand breaks of the germ cell. 13Inhibition of aberrant Hif1αactivation delays intervertebral disc degeneration in adult mice显示文摘The intervertebral disc(IVD) is the largest avascular tissue. Hypoxia-inducible factors(HIFs) play essential roles in regulating cellular adaptation in the IVD under physiological conditions. Disc degeneration disease(DDD) is one of the leading causes of disability, and current therapies are ineffective. This study sought to explore the role of HIFs in DDD pathogenesis in mice. The findings of this study showed that among HIF family members, Hif1α was significantly upregulated in cartilaginous endplate(EP) and annulus fibrosus(AF) tissues from human DDD patients and two mouse models of DDD compared with controls. Conditional deletion of the E3 ubiquitin ligase Vhl in EP and AF tissues of adult mice resulted in upregulated Hif1α expression and age-dependent IVD degeneration. Aberrant Hif1α activation enhanced glycolytic metabolism and suppressed mitochondrial function. On the other hand, genetic ablation of the Hif1α gene delayed DDD pathogenesis in Vhl-deficient mice. Administration of 2-methoxyestradiol(2ME2), a selective Hif1α inhibitor, attenuated experimental IVD degeneration in mice. 17Physical and Thermal Properties of P_2O_5-Al_2O_3-BaO-La_2O_3 Glasses显示文摘The physical and thermal properties of P2O5-Al2O3-BaO-La2O3 glasses were investigated. The effects of glass compositions on the transition temperature, thermal expansion coefficient, density, hardness and refractive index of glasses were studied. The highest hardness of the glasses is 4143.891 MPa and the lowest thermal expansion coefficient of the glasses is 71.770×10-7/℃.
