1Preliminary study on modelling, fabrication by photo-chemical etching and in vivo testing of biodegradable magnesium AZ31 stents显示文摘Magnesium metal(Mg)is a promising material for stent applications due to its biocompatibility and ability to be resorbed by the body.Manufacturing of stents by laser cutting has become an industry standard.Our alternative approach uses photo-chemical etching to transfer a pattern of the stent onto a Mg sheet.In this study,we present three stages of creating and validating a stent prototype,which includes design and simulation using finite element analysis(FEA),followed by fabrication based on AZ31 alloy and,finally,in vivo testing in peripheral arteries of domestic pigs.Due to the preliminary character of this study,only six stents were implanted in two domestic farm pigs weighing 25-28 kg and they were evaluated after 28 days,with an interim follow-up on day 14.The left and right superficial femoral,the left iliac,and the right renal artery were selected for this study.The diameters of the stented artery segments were evaluated at the time of implantation,on day 14 and then,finally,on day 28,by quantitative vessel analysis(QVA)using fluoroscopic imaging.Optical Coherence Tomography(OCT)imaging displayed some malposition,breaks,stacking,and protrusion into the lumen at the proximal,distal,and mid-sections of the stented arteries.The stents degraded with time,but simultaneously became embedded in the intima.After 28 days,the animals were euthanized,and explanted vessels were fixed for micro-CT imaging and histology studies.Micro-CT imaging revealed stent morphological and volumetric changes due to the in-body degradation.An in vivo corrosion rate of 0.75 mm/year was obtained by the CT evaluation.The histology suggested no-life threatening effects,although moderate injury,inflammation,and endothelialization scores were observed.Bala Subramanya Pavan Kumar Kandala Guangqi Zhang Capucine LCorriveau Mark Paquin Madeleine Chagnon Dana Begun Vesselin Shanov 2021Bioactive Materials2021,6,6:2

12Chemical characterization of fine and ultrafine PM, direct and indirect genotoxicity of PM and their organic extracts on pulmonary cells显示文摘Particulate matter in ambient air constitutes a complex mixture of fine and ultrafine particles composed of various chemical compounds including metals, ions, and organics. A multidisciplinary approach was developed by studying physico-chemical characteristics and mechanisms involved in the toxicity of particulate atmospheric pollution. PM_(0.3–2.5) and PM_(2.5) including ultrafine particles were sampled in Dunkerque, a French industrialized seaside city. PM samples were characterized from a chemical and toxicological point of view. Physicochemical characterization evidenced that PM_(2.5) comes from several sources: natural ones, such as soil resuspension and marine sea-salt emissions, as well as anthropogenic ones, such as shipping traffic, road traffic, and industrial activities. Human BEAS-2 B lung cells were exposed to PM_(0.3–2.5), or to the Extractable Organic Matter(EOM) of PM_(0.3–2.5) and PM_(2.5). These exposures induced several mechanisms of action implied in the genotoxicity, such as oxidative DNA adducts and DNA Damage Response. The toxicity of PM-EOM was higher for the sample including the ultrafine fraction(PM_(2.5)) containing also higher concentrations of polycyclic aromatic hydrocarbons. 20Clinical significance of the loss of CD20 antigen on tumor cells in patients with relapsed or refractory follicular lymphoma显示文摘Aim:Anti-CD20 monoclonal antibody is a cornerstone therapy for follicular lymphoma.Following anti-CD20 therapy,a potential decrease in CD20 antigen,and therefore a loss of the tumor target might be expected.However,the incidence and clinical significance of CD20 loss on tumor cells in patients with relapsed or refractory follicular lymphoma are unknown.This study aims to investigate the incidence and outcome of patients with relapsed or refractory follicular lymphoma patients harboring the loss of the tumor target,CD20.Methods:All consecutive adult patients with relapsed or refractory follicular lymphoma referred to the Early Drug Department at Gustave Roussy were included.The main objectives were to assess the incidence and prognosis of the loss in expression of CD20 antigen on the surface of tumor cells on patient outcome.Results:Over the study period 2013-2018,131 patients were screened for clinical trials with B-cell malignancies in the early drug department of Gustave Roussy in France.Forty-four patients presented with relapsed or refractory follicular lymphoma and 32 had tumor biopsies at the time of relapse that were retained for analysis.The median(range)age was 67.5 years(55.3-75.3)and the median number of prior anti-cancer systemic therapies was 3(2-4).At the time of relapse,CD20 expression was positive in 84%of tumors(n=27)and negative in 16%of tumors(n=5).At a median follow-up of 18.3(0.6-83.3)months,CD20 negativity was associated with a poorer prognosis with a median overall survival of 8.9 months(95%CI:2.4-19.1)in comparison to CD20 positive patients(28.3 months,95%CI:25.1-75.3 months,P=0.019).Conclusion:The loss of the tumor target antigen,CD20,occurred in 16%of patients with relapse or refractory follicular lymphoma.Due to confounding factors in patients who received anti-CD20 immunotherapy,it was not possible to formally establish the prognostic significance of CD20 negativity.However,we suggest that a check for CD20 antigen positivity nevertheless be performed to adapt subsequent therapies for patients with relapsed or refractory follicular lymphoma.Jean-Marie Michot Alice Buet-Elfassy Maxime Annereau Julien Lazarovici Alina Danu Clémentine Sarkozy Claude Chahine Camille Bigenwald Jacques Bosq Julien Rossignol Patricia Romano-Martin Capucine Baldini David Ghez Peggy Dartigues Christophe Massard Vincent Ribrag 2021Cancer Drug Resistance2021,4,3:0
