1Extracellular histones stimulate collagen expression in vitro and promote liver fibrogenesis in a mouse model via the TLR4-MyD88 signaling pathway显示文摘BACKGROUND Liver fibrosis progressing to liver cirrhosis and hepatic carcinoma is very common and causes more than one million deaths annually.Fibrosis develops from recurrent liver injury but the molecular mechanisms are not fully understood.Recently,the TLR4-MyD88 signaling pathway has been reported to contribute to fibrosis.Extracellular histones are ligands of TLR4 but their roles in liver fibrosis have not been investigated.AIM To investigate the roles and potential mechanisms of extracellular histones in liver fibrosis.METHODS In vitro,LX2 human hepatic stellate cells(HSCs)were treated with histones in the presence or absence of non-anticoagulant heparin(NAHP)for neutralizing histones or TLR4-blocking antibody.The resultant cellular expression of collagen I was detected using western blotting and immunofluorescent staining.In vivo,the CCl4-induced liver fibrosis model was generated in male 6-week-old ICR mice and in TLR4 or MyD88 knockout and parental mice.Circulating histones were detected and the effect of NAHP was evaluated.RESULTS Extracellular histones strongly stimulated LX2 cells to produce collagen I.Histone-enhanced collagen expression was significantly reduced by NAHP and TLR4-blocking antibody.In CCl4-treated wild type mice,circulating histones were dramatically increased and maintained high levels during the duration of fibrosisinduction.Injection of NAHP not only reduced alanine aminotransferase and liver injury scores,but also significantly reduced fibrogenesis.Since the TLR4-blocking antibody reduced histone-enhanced collagen I production in HSC,the CCl4 model with TLR4 and MyD88 knockout mice was used to demonstrate the roles of the TLR4-MyD88 signaling pathway in CCl4-induced liver fibrosis.The levels of liver fibrosis were indeed significantly reduced in knockout mice compared to wild type parental mice.CONCLUSION Extracellular histones potentially enhance fibrogenesis via the TLR4–MyD88 signaling pathway and NAHP has therapeutic potential by detoxifying extracellular histones.Zhi Wang Zhen-Xing Cheng Simon T Abrams Zi-Qi Lin ED Yates Qian Yu Wei-Ping Yu Ping-Sheng Chen Cheng-Hock Toh Guo-Zheng Wang 2020World Journal of Gastroenterology2020,26,47:3

3HIF-1α signaling: Essential roles in tumorigenesis and implications in targeted therapies显示文摘The hypoxic microenvironment is an essential characteristic of most malignant tu-mors.Notably,hypoxia-inducible factor-1 alpha(HIF-1a)is a key regulatory factor of cellular adaptation to hypoxia,and many critical pathways are correlated with the biological activity of organisms via HIF-1a.In the intra-tumoral hypoxic environment,HIF-1αis highly expressed and contributes to the malignant progression of tumors,which in turn results in a poor prog-nosis in patients.Recently,it has been indicated that HiF-1αinvolves in various critical pro-cesses of life events and tumor development via regulating the expression of HiF-1a target genes,such as cell proliferation and apoptosis,angiogenesis,glucose metabolism,immune response,therapeutic resistance,etc.Apart from solid tumors,accumulating evidence has re-vealed that HiF-1αis also closely associated with the development and progression of hemato-logical malignancies,such as leukemia,lymphoma,and multiple myeloma.Targeted inhibition of HiF-1a can facilitate an increased sensitivity of patients with malignancies to relevant ther-apeutic agents.In the review,we elaborated on the basic structure and biological functions of HIF-1a and summarized their current role in various malignancies.It is expected that they will have future potential fortargeted therapy.Yan Zhao Cheng Xing Yating Deng Can Ye Hongling Peng 2024Genes & Diseases2024,11,1:1

8Magnesium ion-implantation-based gallium nitride p-i-n photodiode for visible-blind ultraviolet detection显示文摘In this work, a GaN p-i-n diode based on Mg ion implantation for visible-blind UV detection is demonstrated.With an optimized implantation and annealing process, a p-GaN layer and corresponding GaN p-i-n photodiode are achieved via Mg implantation. As revealed in the UV detection characterizations, these diodes exhibit a sharp wavelength cutoff at 365 nm, high UV/visible rejection ratio of 1.2 × 10~4, and high photoresponsivity of 0.35 A/W, and are proved to be comparable with commercially available GaN p-n photodiodes. Additionally, a localized states-related gain mechanism is systematically investigated, and a relevant physics model of electricfield-assisted photocarrier hopping is proposed. 10The deubiquitylase UCHL3 maintains cancer stem-like properties by stabilizing the aryl hydrocarbon receptor显示文摘Cancer stem cells(CSCs)exhibit highly aggressive and metastatic features and resistance to chemotherapy and radiotherapy.Aryl hydrocarbon receptor(AhR)expression varies among non-small cell lung cancers(NSCLCs),and the mechanisms that support abnormal AhR expression in CSCs remain elusive.Here,we identified ubiquitin carboxyl terminal hydrolase L3(UCHL3),a DUB enzyme in the UCH protease family,as a bona fide deubiquitylase of the AhR in NSCLC.UCHL3 was shown to interact with,deubiquitylate,and stabilize AhR in a manner dependent on its deubiquitylation activity.Moreover,we showed that UCHL3 promotes the stem-like characteristics and potent tumorigenic capacity of NSCLC cells.UCHL3 increased AhR stability and the binding of AhR to the promoter regions of the“stemness”genes ATP-binding cassette subfamily G member 2(ABCG2),KLF4,and c-Myc.Depletion of UCHL3 markedly downregulated the“stemness”genes ABCG2,KLF4,and c-Myc,leading to the loss of selfrenewal and tumorigenesis in NSCLCs.Furthermore,the UCHL3 inhibitor TCID induced AhR degradation and exhibited significantly attenuated efficacy in NSCLC cells with stem cell-like properties.Additionally,UCHL3 was shown to indicate poor prognosis in patients with lung adenocarcinoma.In general,our results reveal that the UCHL3 deubiquitylase is pivotal for AhR protein stability and a potential target for NSCLC-targeted therapy.Lianlian Ouyang Bin Yan Yating Liu Chao Mao Min Wang Na Liu Zuli Wang Shouping Liu Ying Shi Ling Chen Xiang Wang Yan Cheng Ya Cao Desheng Xiao Lingqiang Zhang Shuang Liu Yongguang Tao 2020Signal Transduction and Targeted Therapy2020,5,1:1
11Landsat-Based Monitoring of Landscape Dynamics in Arctic Permafrost Region显示文摘Ice-rich permafrost thaws as a result of Arctic warming,and the land surface collapses to form characteristic thermokarst landscapes.Thermokarst landscapes can bring instability to the permafrost layer,affecting regional geomorphology,hydrology,and ecology and may further lead to permafrost degradation and greenhouse gas emissions.Field observations in permafrost regions are often limited,while satellite imagery provides a valuable record of land surface dynamics.Currently,continuous monitoring of regional-scale thermokarst landscape dynamics and disturbances remains a challenging task.In this study,we combined the Theil–Sen estimator with the LandTrendr algorithm to create a process flow for monitoring thermokarst landscape dynamics in Arctic permafrost region on the Google Earth Engine platform.A robust linear trend analysis of the Landsat Tasseled Cap index time series based on the Theil–Sen estimator and Mann–Kendall test showed the overall trends in greenness,wetness,and brightness in northern Alaska over the past 20 years.Six types of disturbances that occur in thermokarst landscape were demonstrated and highlighted,including long-term processes(thermokarst lake expansion,shoreline retreat,and river erosion)and short-term events(thermokarst lake drainage,wildfires,and abrupt vegetation change).These disturbances are widespread throughout the Arctic permafrost region and represent hotspots of abrupt permafrost thaw in a warming context,which would destabilize fragile thermokarst landscapes rich in soil organic carbon and affect the ecological carbon balance.The cases we present provide a basis for understanding and quantifying specific disturbance analyses that will facilitate the integration of thermokarst processes into climate models.Yating Chen Aobo Liu Xiao Cheng 2022Journal of Remote Sensing2022,,1:0
12Target prediction and activity verification for the antidepressant action of Huangqin(Radix Scutellariae Baicalensis)显示文摘OBJECTIVE:To decipher the antidepressant targets and mechanisms of Huangqin(Radix Scutellariae Baicalensis)(RSB)by a novel computational system based on prediction and experimental verification.METHODS:The putative targets of RSB against depression were identified from Traditional Chinese Medicine Systems Pharmacology(TCMSP)and Drug Bank.Next,protein-protein interaction network of the anti-depression targets of RSB were identified,and differentially expressed genes(DEGs)of depression were mined from the NCBI database.Then,Kyoto Encyclopedia of Genes and Genomes and Gene Ontology were used to analysis the common targets.Finally,the selected pathways and functions were verified by experimentation.RESULTS:Thirty active compounds in RSB were predicted with high confidence by TCMSP and DrugBank,and seventy-one DEGs were identified in the GEO database.Besides,eight core target proteins were screened out by descending order of degree value,including ACHE,IL6,SLC6A4,FOS,SLC6A3,MAOB,DPP4,and JUN.These target genes were further found to be associated with pathways involved in neuronal apoptosis,such as pathways in cancer,Toll-like receptor signaling pathway,and TNF signaling.The cell proliferation assay and wound-healing assay results showed that RSB does not affect PC12 cell proliferation and chemotaxis.Unexpectedly,RSB protected PC12 cells from oxidative stress induced by H2O2 via inhibiting autophagy and apoptosis.We revealed significant changes in mice treated with 400 mg/kg RSB compared with the lipopolysaccharide mice.The possible mechanism for the antidepressive action of RSB is by reducing the expression of LC3-B in CA1 neurons.CONCLUSIONS:Our research partially expounds the mechanism of the antidepressant effect of RSB by the combination of network pharmacology prediction and experimental verification.Furthermore,it is also conducive to the application of Traditional Chinese Medicine within modern medicine.LI Ganggang LU Ye HE Pei ZHANG Shiyue CHENG Yating ZHANG Shaodan PEI Lin 2021Journal of Traditional Chinese Medicine2021,41,6:0
13An Approach for User’s Trajectory Prediction Based on the Trajectory Cluster显示文摘The existing user's trajectory prediction methods considered little about the interrelation among users and would fail if the user historical trajectory data were lack. This paper presents a user's trajectory prediction model and corresponding algorithms by the historical trajectories of users based on the trajectory cluster. 14Engineering the thermostability of D-lyxose isomerase from Caldanaerobius polysaccharolyticus via multiple computer-aided rational design for efficient synthesis of D-mannose显示文摘D-Mannose is an attractive functional sugar that exhibits many physiological benefits on human health.The demand for low-calorie sugars and sweeteners in foods are increasingly available on the market.Some sugar isomerases,such as D-lyxose isomerase(D-LIase),can achieve an isomerization reaction between D-mannose and D-fructose.However,the weak thermostability of D-LIase limits its efficient conversion from D-fructose to D-mannose.Nonetheless,few studies are available that have investigated the molecular modification of D-LIase to improve its thermal stability.In this study,computer-aided tools including FireProt,PROSS,and Consensus Finder were employed to jointly design D-LIase mutants with improved thermostability for the first time.Finally,the obtained five-point mutant M5(N21G/E78P/V58Y/C119Y/K170P)showed high thermal stability and cat-alytic activity.The half-life of M5 at 65◦C was 10.22 fold,and the catalytic efficiency towards 600 g/L of D-fructose was 2.6 times to that of the wild type enzyme,respectively.Molecular dynamics simulation and intramolecular forces analysis revealed a thermostability mechanism of highly rigidity conformation,newly formed hydrogen bonds andπ-cation interaction between and within protein domains,and redistributed surface electrostatic charges for the mutant M5.This research provided a promising D-LIase mutant for the industrial production of D-mannose from D-fructose.Hao Wu Ming Yi Xiaoyi Wu Yating Ding Minghui Pu Li Wen Yunhui Cheng Wenli Zhang Wanmeng Mu 2023Synthetic and Systems Biotechnology2023,8,2:0
15Establishment of new convenient two-line system for hybrid production by targeting mutation of OPR3 in allopolyploid Brassica napus显示文摘The two-line pollination control system,which usually depends on the utilization of thermosensitive or photoperiod genic male-sterile lines,has been widely used in various crops.However,this system is susceptible to instability issues caused by uncontrollable weather fluctuations.A stable and handy two-line pollination control system is highly desirable in many crop species for heterosis exploitation.Oxophytodienoic acid reductase 3(OPR3)was proven to be involved in jasmonate biosynthesis.In the present study,CRISPR/Cas9(Clustered Regularly Interspaced Short Palindromic Repeat)was utilized to mutate two OPR3 homologs in Brassica napus.After two OPR3 homologs were simultaneously mutated,mutants exhibited complete male sterility,and fertility could be easily restored by exogenous MeJA treatment.Hybrids produced from crosses between the opr3 sterile lines and normal varieties exhibited heterosis.This new two-line system based on OPR3 mutation provides higher stability and convenience than traditional systems.By using exogenous MeJA treatment to restore fertility,the system enables more precise control of male fertility transition,which has great potential to significantly contribute to the maneuverable production of hybrid seeds in rapeseed as well as other Brassica species crops.Hongtao Cheng Mengyu Hao Shifei Sang Yunfei Wen Yating Cai Hui Wang Wenxiang Wang Desheng Mei Qiong Hu 2023Horticulture Research2023,10,12:0
16The cross-talk between methylation and phosphorylation in lymphoid-specific helicase drives cancer stem-like properties显示文摘Posttranslational modifications(PTMs)of proteins,including chromatin modifiers,play crucial roles in the dynamic alteration of various protein properties and functions including stem-cell properties.However,the roles of Lymphoid-specific helicase(LSH),a DNA methylation modifier,in modulating stem-like properties in cancer are still not clearly clarified.Therefore,exploring PTMs modulation of LSH activity will be of great significance to further understand the function and activity of LSH.Here,we demonstrate that LSH is capable to undergo PTMs,including methylation and phosphorylation.The arginine methyltransferase PRMT5 can methylate LSH at R309 residue,meanwhile,LSH could as well be phosphorylated by MAPK1 kinase at S503 residue.We further show that the accumulation of phosphorylation of LSH at S503 site exhibits downregulation of LSH methylation at R309 residue,which eventually promoting stem-like properties in lung cancer.Whereas,phosphorylation-deficient LSH S503A mutant promotes the accumulation of LSH methylation at R309 residue and attenuates stem-like properties,indicating the critical roles of LSH PTMs in modulating stem-like properties.Thus,our study highlights the importance of the crosstalk between LSH PTMs in determining its activity and function in lung cancer stem-cell maintenance.Na Liu Rui Yang Ying Shi Ling Chen Yating Liu Zuli Wang Shouping Liu Lianlian Ouyang Haiyan Wang Weiwei Lai Chao Mao Min Wang Yan Cheng Shuang Liu Xiang Wang Hu Zhou Ya Cao Desheng Xiao Yongguang Tao 2020Signal Transduction and Targeted Therapy2020,5,1:0

18Sequentially releasing self-healing hydrogel fabricated with TGFβ3-microspheres and bFGF to facilitate rat alveolar bone defect repair显示文摘Resorption and loss of alveolar bone leads to oral dysfunction and loss of natural or implant teeth. Biomimetic delivery of growth factors based on stem cell recruitment and osteogenic differentiation, as the key steps in natural alveolar bone regenerative process, has been an area of intense research in recent years. A mesoporous self-healing hydrogel(DFH) with basic fibroblast growth factor(bFGF) entrapment and transforming growth factor β3(TGFβ3)-loaded chitosan microspheres(CMs) was developed. The formulation was optimized by multiple tests of self-healing, in-bottle inversion, SEM, rheological, swelling rate and in vitro degradation. In vitro tubule formation assays, cell migration assays, and osteogenic differentiation assays confirmed the ability of DFH to promote blood vessels, recruit stem cells, and promote osteogenic differentiation. The optimum DFH formula is 0.05 ml 4ArmPEG-DF(20%) added to 1 ml CsGlu(2%) containing bFGF(80 ng) and TGFβ3-microspheres(5 mg). The results of in vitro release studied by Elisa kit, indicated an 95% release of b FGF in7 d and long-term sustained release of TGFβ3. For alveolar defects rat models, the expression levels of CD29 and CD45, the bone volume fraction, trabecular number, and trabecular thickness of new bone monitored by Micro-CT in DFH treatment groups were significantly higher than others(*P < 0.05, vs Model). HE and Masson staining show the same results.In conclusion, DFH is a design of bionic alveolar remodelling microenvironment, that is in early time microvessels formed by b FGF provide nutritious to recruited endogenous stem cells, then TGFβ3 slowly released speed up the process of new bones formation to common facilitate rat alveolar defect repair. 20Nuclear localization of human MEIOB requires its NLS in the OB domain and interaction with SPATA22显示文摘MEIOB is a vital protein in meiotic homologous recombination and plays an indispensable role in human gametogenesis.In mammals,MEIOB and its partner SPATA22 form a heterodimer,ensuring their effective localization on single-strand DNA(ssDNA)and proper synapsis processes.Mutations in human MEIOB(hMEIOB)cause human infertility attributed to the failure of its interaction with human SPATA22(hSPATA22)and ssDNA binding.However,the detailed mechanism is still unclear.In our study,truncated or full-length hMEIOB and hSPATA22 are traced by fused expression with fluorescent proteins(i.e.,copGFP or mCherry),and the live cell imaging system is used to observe the expression and localization of the proteins.When transfected alone,hMEIOB accumulates in the cytoplasm.Interestingly,a covered NLS in the OB domain of hMEIOB is identified,which can be exposed by hSPATA22 and is necessary for the nuclear localization of hMEIOB.When hSPATA22 loses its hMEIOB interacting domain or NLS,the nuclear localization of hMEIOB is aborted.Collectively,our results prove that the NLS in the OB domain of hMEIOB and interaction with hSPATA22 are required for hMEIOB nuclear localization.Yating Xu Zhen Chen Panfeng Wu Wei Qu Hanqing Shi Muyang Cheng Yumin Xu Tingyu Jin Cong Liu Cong Liu Yi Li Mengcheng Luo 2023Acta Biochimica et Biophysica Sinica2023,55,1:0
